Decrease in catalase activity of Folsomia candida fed a Bt rice diet

Here we report the effects of three Bt-rice varieties and their non-Bt conventional isolines on biological traits including survival, reproduction, and the activities of three antioxidant enzymes superoxide dismutase, catalase and peroxidase, in the Collembolan, Folsomia candida. The reproduction was significantly lower when fed Kemingdao and Huahui1 than those feeding on their non-GM near-isogenic varieties Xiushui and Minghui63 respectively, this can be explained by the differences of plant compositions depended on variety of rice. The catalase activity of F. candida was significantly lower when fed the Bt-rice variety Kemingdao compared to the near-isogenic non-Bt-rice variety Xiushui. This suggests that some Bt-rice varieties may impose environmental stress to collembolans. We emphasize that changes in activity of antioxidant enzymes of non-target organisms are important in understanding the ecological consequences for organisms inhabiting transgenic Bt-rice plantations.

As part of the tier 2 of a site-specific risk assessment, this study was the first reporting an intensive in situ application of the bait-lamina assay; two exposure periods (7 and 14 days) were tested during four seasons in ten different sites, within a uranium mine area and at two different depths. The most contaminated sites (by deposition of sludge from the effluent treatment pond) were discriminated after 14 days of exposure because extremely low percentages of feeding activity were recorded. Previous sub-lethal ecotoxicological assays, already had demonstrated that the habitat function of these soils is compromised. Nevertheless, seasonality has proved to have a significant influence on responses. Thus to strength conclusions about the impact of contaminants, the in situ bait-lamina assay should be performed on different annual seasons, at least for temperate regions. It was also found that some environmental parameters (e.g. soil moisture and litter) can act as confounding factors in the bait-lamina assay. Structural and functional responses of a benthic macroinvertebrate assemblage to pulses of the insecticide imidacloprid were assessed in outdoor stream mesocosms. Imidacloprid pulses reduced invertebrate abundance and community diversity in imidacloprid-dosed streams compared to control streams. These results correlated well with effects of imidacloprid on leaf litter decomposition and feeding rates of Pteronarcys comstocki, a stonefly, in artificial streams. Reductions in oxygen consumption of stoneflies exposed to imidacloprid were also observed in laboratory experiments. Our findings suggest that leaf litter degradation and single species responses can be sensitive ecotoxicological endpoints that can be used as early warning indicators and biomonitoring tools for pesticide contamination. The data generated illustrates the value of mesocosm experiments in environmental assessment and how the consideration of functional and structural endpoints of natural communities together with in situ single species bioassays can improve the evaluation and prediction of pesticide effects on stream ecosystems. Combining organism-level responses with community-level processes for the evaluation and prediction of pesticide effects on stream ecosystems.

The objective of this study was to evaluate (qualitatively and quantitatively) the occurrence of antibiotic residue in pasteurized milk in Brazil. Pasteurized milk samples (n = 252) were collected monthly from Nov. 2010–Oct. 2011 from 21 commercial establishments (brands). A screening test (Delvotest® SP-NT) was applied to those samples. In positive (n = 19) and/or suspect samples (n = 24), we quantified oxytetracycline (OTC) and tetracycline (TC) by high-performance liquid chromatography with diode array detector (HPLC-DAD). OTCs were detected in all positive samples and TCs in six. In the 24suspected samples, OTCs were detected in 23 and TCs were not found in 8. Of the milk brands evaluated (n = 21), the presence of antibiotic residue was not detected in 4; in the other brands, both positive and suspect samples were verified. Results indicate the presence of antibiotic residue above legal limits. According to actual milk consumption in Brazil (441 mL/kg BW/day), in only 9 of the 17 brands of milk with antibiotic residue, the estimated daily intake was at or less than the maximum recommended by the European Union. The screening test used was effective to identify the presence of antibiotic residue (OTC and TC), confirmed by HPLC-DAD. The OTC is the predominant antimicrobial used by dairy farmers. Ingestion of contaminated milk by OTC and TC can increase the resistance of microorganisms to antibiotics.

For sheep grazing pastures in areas of mineralisation and former metalliferous mining activity, an excessive intake of potentially harmful elements (PHEs) such as fluorine (F) and lead (Pb) can result in clinical and subclinical toxicity. The prime aim of our work was to calculate the intakes of both of these PHEs by sheep grazing pastures in the mineralised/mined Peak District area of Derbyshire. The bi-monthly sampling of topsoils (0-15 cm depth) and the faeces of sheep from fields at seven farms was undertaken for a 1-year period. These samples were analysed for titanium that allowed the rates of soil ingestion (and hence also herbage ingestion since we assume that the sheep have an overall diet of 1 kg dry matter (DM)/day) to be determined. Our findings were then combined with previously published soil and soil-free pasture herbage F and Pb concentrations determined from the seven farms to calculate the intakes of both PHEs. The results show seasonal variations of soil ingestion at the seven farms ranging from <0.1 to 20.1 % of the DM intake (median = 3 %), with the highest rates of ingestion being associated with the winter-spring (i.e. December-April) period. Our calculations show that at some farms, sheep can be exposed to dietary concentrations in excess of recommended guidelines potentially throughout the whole year, though livestock movement to less-contaminated pastures would lessen any toxic impact. Because the soil concentrations are greater than those associated with soil-free pasture herbage, ingested soils are the main dietary source of Pb and (especially) F to sheep. However, subjecting freshly sampled topsoils to sequential extraction procedures undertaken in the laboratory indicates that the majority of Pb and (especially) F may not be readily soluble in the ovine digestion system, so reducing the quantities of both PHEs available for absorption.

PURPOSE: Chlor-alkali plants are one of the most important point sources of mercury to aquatic environment. The problem of Hg contamination has been studied in a region, Rm Valcea (Romania), impacted by the wastewater discharge of a chlor-alkali plant. The purpose of the present study is to evaluate the current status of mercury pollution in the Babeni reservoir (Olt River) and the exposure of local population via fish consumption to mercury originating from the chlor-alkali plant. METHODS: Sediments were collected from Valcea, Govora and Babeni reservoirs. Grain size distribution, organic content and total mercury (THg) concentrations were analysed in sediments. Fish were purchased from local anglers, and the scalp hair was collected from volunteers. THg in sediment, fish and hair samples was determined using an atomic absorption spectrophotometer for Hg determination. Monomethylmercury (MMHg) was analysed in the muscle and liver tissues by species-specific isotope dilution and capillary gas chromatography hyphenated to inductively coupled plasma mass spectrometer. RESULTS: High mercury concentrations were found in the sediments and in fish from Babeni reservoir, with a median of 2.1 mg/kg (IQR = 3.2) in sediments and a mean value of 1.8 ± 0.8 mg/kg_ww in fish muscle. MMHg concentrations in fish were well above the WHO guidelines for fish consumption. Local population consuming fish from the Babeni reservoir had THg concentrations in hair significantly higher than those consuming fish from upstream reservoirs and/or from the shops and reached a median value of 2.5 mg/kg (IQR = 3.6). CONCLUSIONS: The remnant pollution in the fish of this reservoir, and probably many other lakes and reservoirs receiving Hg polluted wastewater, represents a considerable health risk for the local fish consumers.
