Isolation and molecular characterization of bacteria to heavy metals isolated from soil samples in Bokaro Coal Mines, India

In recent years, environmental pollution by coal mining is a long-established human activity affecting all levels of life with various environmental impacts by generating heavy metals. The presence of heavy metals even in trace amount is toxic and detrimental to all living organisms. The coal mine area in Bokaro is one of the “Toxic Hotspot” in India. Bacteria have evolved uptake and efflux mechanisms to adapt in heavy metals contaminated environments and thus represent a potential source for bioremediation processes. In the present study, we isolated and characterized eight heavy metal resistant bacteria (NK-1 to 8) from soil sample in Bokaro coal mines, India. Isolates were selected based on high level of heavy metal resistance and its biochemical characterization. The following bacteria were identified based on 16S rRNA gene sequencing Enterobacter ludwigii (KM029957; NK-1), Klebsiella pneumonia (KM029958; NK-2), Enterobacter ludwigii (KM029959; NK-3), Enterobacter ludwigii (KM029960; NK-4), Klebsiella oxytoca (KM029961; NK-5), Enterobacter cloacae (KM029962; NK-6), Acinetobacter gyllenbergii (KM029963; NK-7), Enterobacter cloacae (KM029964; NK-8). A high degree of metal resistance associated with multiple antibiotic resistances was also detected in the selected isolate which was confirmed by the presence of plasmid. These isolates can further be used for bioremediation of heavy metals from contaminated site.

International audience | Chronic exposure to pollutants may represent a threat for wildlife. We tested whether adult survival rate, breeding probability and breeding success the year of sampling and the following year were affected by blood levels of mercury or persistent organic pollutants in Svalbard black-legged kittiwake Rissa tridactyla, by using capture–mark–recapture models over a five-year period. Survival rate was negatively linked to HCB levels in females, to chlordane mixture and oxychlordane, tended to decrease with increasing PCBs or DDE levels, but was unrelated to mercury. Breeding probability decreased with increasing mercury levels during the sampling year and with increasing CHL or HCB levels during the following year, especially in males observed as breeders. Surprisingly, the probability of raising two chicks increased with increasing HCB levels. Although levels of these legacy pollutants are expected to decline, they represent a potential threat for adult survival rate and breeding probability, possibly affecting kittiwake population dynamics.

Phylogenetic study based on16S rRNA gene sequence showed that the isolate has 100 % of similarity with several Bradyrhizobium and was accordingly designated as Bradyrhizobium sp. SR1. Plasmid profiling revealed the presence of a large plasmid (>50 kb) in SR1 not cured under nonselective conditions. Its transfer to Escherichia coli by electroporation failed to induce β-triketone degrading capacity,suggesting that degrading genes possibly located on this plasmid cannot be expressed in E. coli or that they are not plasmid borne. The evaluation of the SR1 ability to degrade various synthetic (mesotrione and tembotrione) and natural (leptospermone) triketones showed that this strain was also able to degrademesotrione. Although SR1 was able to entirely dissipate both herbicides, degradation rate of sulcotrione was ten times higher than that of mesotrione, showing a greater affinity of degrading-enzyme system to sulcotrione. Degradation pathway of sulcotrione involved the formation of 2-chloro-4-mesylbenzoic acid (CMBA), previously identified in sulcotrione degradation, and of a new metabolite identified as hydroxy-sulcotrione.Mesotrione degradation pathway leads to the accumulation of-methylsulfonyl-2-nitrobenzoic acid(MNBA) and 2-amino-4 methylsulfonylbenzoic acid(AMBA), two well-known metabolites of this herbicide. Along with the dissipation of β-triketones, one could observe the decrease in 4-hydroxyphenylpyruvate dioxygenase(HPPD) inhibition, indicating that toxicity was due to parent molecules, and not to the formed metabolites. This is the first report of the isolation of bacterial strain able to transform two β-triketones.

The increasing presence of plastic pollution in marine ecosystems has become a major concern. In the environment, plastics break down into smaller and smaller pieces of microplastics. Methods of microplastic recovery are needed to reduce the dangers they can pose to a variety of organisms. An elutriation device was manufactured and optimized to achieve maximum microplastic recovery. The parameters flow rate and diameter of elutriation column were varied and their domain of variation was determined. A composite factorial experimental design was generated using MODDE 10.1 and was undergone. The optimal values of flow rate and column diameter were determined to be 385Lh−1 and 5.06cm respectively, under constraints, to achieve a maximum feasible microplastics recovery percentage of 50.2%. The elutriation process can be improved through further testing, and can be tested in the field to compare its efficiency to that of manual microplastics filtration.

The mining sector is growing in parallel with societal demands for minerals. One of the most important environmental issues and economic burdens of industrial mining on land is the safe storage of the vast amounts of waste produced. Traditionally, tailings have been stored in land dams, but the lack of land availability, potential risk of dam failure and topography in coastal areas in certain countries results in increasing disposal of tailings into marine systems. This review describes the different submarine tailing disposal methods used in the world in general and in Norway in particular, their impact on the environment (e.g. hyper-sedimentation, toxicity, processes related to changes in grain shape and size, turbidity), current legislation and need for future research. Understanding these impacts on the habitat and biota is essential to assess potential ecosystem changes and to develop best available techniques and robust management plans.

Marine litter is a significant environmental problem inherently linked to individuals’ purchasing, use and disposal behaviour. This research examined 176 British schoolchildren’s (aged 8–13years) baseline marine litter understanding and self-reported actions, and tested the impact of an educational intervention. All children participated in the educational intervention and completed a pre- and post-intervention questionnaire. At baseline, children were quite concerned about marine litter and recognised some of the causes and impacts of the problem. Children also reported taking a number of actions to help solve the problem. After the intervention, children were significantly more concerned, had a better understanding of the causes and negative impacts, and reported engaging in more actions to reduce the potential causes of marine litter. Understanding the perceptions and behaviours of children is crucial as they represent current and future actors and a potentially important source of social influence among their peers, parents and community.

Cracks are the main cause for structural failure. One way to circumvent costly manual maintenance and repair is to incorporate an autonomous self-healing mechanism in concrete. This study exploited the potential to apply calcite-precipitating bacteria as a crack-healing agent in concrete. These bacteria were prepared in different cell concentrations and incorporated in the concrete mix. Compressive strength tests were performed at the stage of 28th day of curing. The effects of different cell concentrations of Bacillus sphaericus on concrete, reducing the crack, were studied. We used mortar cubes with 30mL of bacteria/mortar cube and sequentially increased up to 50mL (10, 20, 30, 40 and 50mL) in the ratio of mortar cubes in 1:6. The concrete grade used for the study was M25. At last, we had made concrete blocks of size 150×150×150 mm with concrete of grade M25. For those blocks, the compressive strength and non-destructive tests such as, rebound hammer and ultrasonic pulse velocity tests were performed. The results obtained in the work are that the compressive strength of blocks of size 150×150×150 mm is good when compared to control concrete. When load is applied to control concrete, the crack gets developed earlier and when bacterial concrete is used, the crack does not develop at an early stage.

Mountain flood that causes landslide and other geological disasters can damage the fragile ecological environment in mountain areas. In this paper, threshold and distribution model of accumulated temperature based on snowmelt flood magnitude are designed in mountainous watershed. Meanwhile, input data for this model make use of ample reliable data that include remote sensing and so on. In detail, this model simulates the average watershed temperature by using the meteorologic re-analysis data of the National Center for Atmospheric Research and calculates the average snow depth by using hyperspectral remote sensing data. In addition, the model related data comprise long-term observation experiments of the watershed, including the characteristics of accumulated snow and result of correlation between runoff and infiltration in runoff simulation experiment through distributed hydrological models (i.e., Soil and Water Assessment Tool and Distributed Hydrology Soil Vegetation Model). Finally, the average accumulated temperature of the watershed that causes snowmelt flood can be obtained through the aforementioned method, and the characteristics of the accumulated temperature distribution of the watershed area are determined based on the temperature lapse rate. The characteristics of accumulated temperature distribution can provide decision-making reference for monitoring the ecological environment in mountain areas and preventing and reducing disasters.
