A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the simultaneous analysis of 6 ectoparasiticides – 2 synthetic pyrethroids (deltamethrin, cypermethrin) and 4 macrocyclic lactones (abamectin, doramectin, ivermectin and eprinomectin) in biosolids. The method was used to investigate the occurrence of these ectoparasiticides in beef cattle feedlot wastes in Australia from 5 commercial feedlot operations which employ varying waste management practices. Deltamethrin and cypermethrin were not detected in any of the samples while abamectin, ivermectin, doramectin and eprinomectin were detected in some of the samples with concentrations ranging from 1 to 36 μg/kg dry weight (d.w.) freeze dried feedlot waste. Levels of macrocyclic lactones detected in the feedlot wastes varied and were dependent on sample type. The effect of seasonal variations and waste management practices were also investigated in this study.

Antibiotics including three β-lactams, two fluoroquinolones and two macrolides, which were the top seven most prescribed antibiotics in Dalian, China, were selected to investigate their occurrence in six municipal wastewater treatment plants (WWTPs) and their distribution in the effluent-receiving waters of the Yellow Sea. Four WWTPs employing different treatment technologies were selected to explore the mechanism of antibiotics elimination during wastewater treatment. Results showed that fluoroquinolones and macrolides were dominant species in both WWTPs effluents and the surveyed coastal waters. Biodegradation was the main pathway for β-lactams removal, however, primary treatment performed better than biological treatment for fluoroquinolones removal. Concentrations of macrolides increased dramatically after the biological treatment, which was probably due to the release of macrolides enclosed in feces particles. In the surveyed coastal waters, reduction of antibiotic concentration with distance was observed. Potential environmental risk caused by the occurrence of these antibiotics should be evaluated in future work.

We assessed the capacity of real-time PCR markers to identify the origin of contamination in shellfish. Oyster, cockles or clams were either contaminated with fecal materials and host-associated markers designed from Bacteroidales or Catellicoccus marimammalium 16S RNA genes were extracted from their intravalvular liquid, digestive tissues or shellfish flesh. Extraction of bacterial DNA from the oyster intravalvular liquid with FastDNA spin kit for soil enabled the selected markers to be quantified in 100% of artificially contaminated samples, and the source of contamination to be identified in 13 out of 38 naturally contaminated batches from European Class B and Class C areas. However, this protocol did not enable the origin of the contamination to be identified in cockle or clam samples. Although results are promising for extracts from intravalvular liquid in oyster, it is unlikely that a single protocol could be the best across all bacterial markers and types of shellfish.

The prevalence of antibiotic resistance and the implicated mechanisms of resistance were evaluated in Enterococcus spp. and Escherichia coli, isolated from a total of 250 faecal samples of echinoderms collected from Azorean waters (Portugal). A total of 144 enterococci (120 Enterococcus faecium, 14 E. hirae, 8 E. faecalis, 2 E. gallinarum) and 10 E. coli were recovered. High percentages of resistance in enterococci were found for erythromycin, ampicillin, tetracyclin and ciprofloxacin. The erm(A) or erm(B), tet(M) and/or tet(L), vat(D), aac(6′)-aph(2″) and aph(3′)-IIIa genes were found in isolates resistant to erythromycin, tetracycline, quinupristin/dalfopristin, high-level gentamicin and high-level kanamycin, respectively. Resistance in E. coli isolates was detected for streptomycin, amikacin, tetracycline and tobramycin. The aadA gene was found in streptomycin-resistant isolates and tet(A)+tet(B) genes in tetracycline-resistant isolates. The data recovered are essential to improve knowledge about the dissemination of resistant strains through marine ecosystems and the possible implications involved in transferring these resistances either to other animals or to humans.

Marine fish farms could cause environmental disturbances on the sediment due to uneaten food and fish faeces that impact the marine benthos. Polychaete assemblages are considered good indicators of environmental perturbations. The present study aimed to establish groups of polychaetes as potential indicators of fish farm pollution. This study was carried out in ten fish farms along the Spanish coast. Changes in polychaete assemblage were analyzed with meta-analysis and multivariate techniques. Abundance, richness and diversity showed significant decreases under fish farm conditions. Distribution patterns of polychaetes responded to combinations of physicochemical variables. The main ones are sulfide concentration, silt and clays percentage, and stable nitrogen isotope ratio. The results showed that some families are tolerant, Capitellidae, Dorvilleidae, Glyceridae, Nereididae, Oweniidae and Spionidae; while others are sensitive to fish farm pollution, Magelonidae, Maldanidae, Nephtyidae, Onuphidae, Paralacydoniidae, Paraonide, Sabellidae and also Cirratulidae in spite of being reported as a tolerant family.

Wild birds have been shown to be significant sources of numerous types of pathogens that are relevant to humans and agriculture. The presence of large numbers of migratory birds in such a sensitive and important ecosystem as the Platte River in central Nebraska, USA, could potentially serve a significant source of bird-derived pathogens in the water/sediment and riverine environment. In 2009 and 2010, a study was completed to investigate the potential water-quality impacts of Sandhill Cranes and Snow Geese on the microbial water quality of the central Platte River during their spring migration period. Fecal material, river-bottom sediment, and water samples were collected from January through May of each year during the spring migration season of Sandhill Cranes in the Central Flyway of North America. Results indicate that several types of fecal indicator bacteria and from a range of viral, protozoan, and bacterial pathogens, Campylobacter jejuni were present in Sandhill Crane excreta, and at significantly higher frequency and densities in water and sediments when the Sandhill Cranes were present, particularly during evening roosts within the Platte River environment. Therefore, further investigation of the health significance of avian pathogens is warranted for the Platte River in Central Nebraska during migration of Sandhill Cranes and other waterfowl.

Colistin is a peptide antibiotic widely used as a food additive in animal farming, specially swine and poultry, and also has recently been applied in human medicine to treat infections caused by multiresistant gram-negative bacteria strains. When orally administered, colistin is eliminated in feces virtually unaltered; thus, it may reach water bodies and wastewater treatment facilities in its active form. Apart from the risks associated with development of antimicrobial resistance and environmental toxicity issues, the presence of antimicrobials in wastewater can, additionally, interfere in biological processes commonly used to treat them. Nitrifying bacteria are among the most sensitive microorganisms to inhibitory compounds, including pharmaceuticals, and are useful as biosensors to access contaminant toxicity information in wastewater treatment plants. Therefore, in order to assess the colistin acute toxicity to the microorganisms involved in the nitrification processes, the nitritation and nitratation kinetics were monitored under different colistin concentrations. The results showed that only ammonia-oxidizing bacteria are sensitive to the antibiotic, presenting an IC50 of 10.8 mg L⁻¹ of colistin when used as a commercial formulation and 67.0 mg L⁻¹ when used as raw colistin sulfate. For nitrite-oxidizing bacteria, even the highest colistin concentration used in the assays (316 mg L⁻¹) was not sufficient to inhibit the process. According to these results, the colistin concentrations expected in animal farming wastewater, when its dosage is used as a growth promoter, would not be enough to keep nitrification from taking place. Nevertheless, when used in higher concentrations, such as for therapeutic purposes, it could endanger the maintenance of the process.

The effective disposal of redundant elephant dung (ED) is important for environmental protection and utilization of resource. The aim of this study was to remove a toxic-azo dye, Reactive Red (RR) 120, using this relatively cheap material as a new adsorbent. The FTIR-ATR spectra of ED powders before and after the sorption of RR 120 and zero point charge (pHzpc) of ED were determined. The sorption capacity of ED for removing of RR 120 were carried out as functions of particle size, adsorbent dose, pH, temperature, ionic strength, initial dye concentration, and contact time. Sorption isotherm, kinetic, activation energy, thermodynamic, and desorption parameters of RR 120 on ED were studied. The sorption process was found to be dependent on particle size, adsorbent dose, pH, temperature, ionic strength, initial dye concentration, and contact time. FTIR-ATR spectroscopy indicated that amine and amide groups have significant role on the sorption of RR 120 on ED. The pHzpc of ED was found to be 7.3. Sorption kinetic of RR 120 on ED was well described by sigmoidal Logistic model. The Langmuir isotherm was well fitted to the equilibrium data. The maximum sorption capacity was 95.71 mg g(-1). The sorption of RR 120 on ED was mainly physical and exothermic according to results of D-R isotherm, Arrhenius equation, thermodynamic, and desorption studies. The thermodynamic parameters showed that this process was feasible and spontaneous. This study showed that ED as a low-cost adsorbent had a great potential for the removal of RR 120 as an alternative eco-friendly process.

For sheep grazing pastures in areas of mineralisation and former metalliferous mining activity, an excessive intake of potentially harmful elements (PHEs) such as fluorine (F) and lead (Pb) can result in clinical and subclinical toxicity. The prime aim of our work was to calculate the intakes of both of these PHEs by sheep grazing pastures in the mineralised/mined Peak District area of Derbyshire. The bi-monthly sampling of topsoils (0-15 cm depth) and the faeces of sheep from fields at seven farms was undertaken for a 1-year period. These samples were analysed for titanium that allowed the rates of soil ingestion (and hence also herbage ingestion since we assume that the sheep have an overall diet of 1 kg dry matter (DM)/day) to be determined. Our findings were then combined with previously published soil and soil-free pasture herbage F and Pb concentrations determined from the seven farms to calculate the intakes of both PHEs. The results show seasonal variations of soil ingestion at the seven farms ranging from <0.1 to 20.1 % of the DM intake (median = 3 %), with the highest rates of ingestion being associated with the winter-spring (i.e. December-April) period. Our calculations show that at some farms, sheep can be exposed to dietary concentrations in excess of recommended guidelines potentially throughout the whole year, though livestock movement to less-contaminated pastures would lessen any toxic impact. Because the soil concentrations are greater than those associated with soil-free pasture herbage, ingested soils are the main dietary source of Pb and (especially) F to sheep. However, subjecting freshly sampled topsoils to sequential extraction procedures undertaken in the laboratory indicates that the majority of Pb and (especially) F may not be readily soluble in the ovine digestion system, so reducing the quantities of both PHEs available for absorption.

The impact of the insecticide chlorpyrifos (CPF) on the mammalian digestive system has been poorly described. The present study aimed at evaluating the effect of chronic, low-dose exposure to CPF on the composition of the gut microbiota in a Simulator of the Human Intestinal Microbial Ecosystem: the SHIME® and in rats. The SHIME® comprises six reactor vessels (stomach to colon). The colonic segments were inoculated with feces from healthy humans. Then, the simulator was exposed to a daily dose of 1 mg of CPF for 30 days. The changes over time in the populations of bacteria were examined at different time points: prior to pesticide exposure (as a control) and after exposure. In parallel, pregnant rats were gavaged daily with 1 mg/kg of CPF (or vehicle) until the pups were weaned. Next, the rats were gavaged with same dose of CPF until 60 days of age (adulthood). Then, samples of different parts of the digestive tract were collected under sterile conditions for microbiological assessment. Chronic, low-dose exposure to CPF in the SHIME® and in the rat was found to induce dysbiosis in the microbial community with, in particular, proliferation of subpopulations of some strains and a decrease in the numbers of others bacteria. In compliance with European guidelines, the use of the SHIME® in vitro tool would help to (1) elucidate the final health effect of toxic agents and (2) minimize (though not fully replace) animal testing. Indeed, certain parameters would still have to be studied further in vivo.
