Comparative study of 17 β-estradiol on endocrine disruption and biotransformation in fingerlings and juveniles of Japanese sea bass Lateolabrax japonicus

Estrogenic contaminants in the aquatic environment are associated with endocrine disruption and feminization in fish. The effects of endocrine disrupting chemicals (EDCs) on fish have been well documented. However, very few studies have focused on 17 β-estradiol (E2) and its effects on endocrine system and biotransformation in a single prolonged exposure. This study investigated changes in the levels of serotonin (5-hydroxytryptamine) and acetyl choline esterase (AchE) in brain, cortisol in plasma and Ethoxyresorufin-O-deethylase (EROD) activity in gill of two different size groups (fingerlings and juveniles) of Japanese sea bass (Lateolabrax japonicus) upon exposure to two sub-lethal concentrations (200 and 2000ngL−1) of E2 for 30d. The results indicate that cortisol level and EROD activity significantly increased in both groups, whereas serotonin level increased in juveniles and decreased in fingerlings due to E2 exposure. The correlation analysis revealed that E2 significantly affected the endocrine and biotransformation systems in both age groups.

Southern parts of the Caspian Sea have been faced with a diverse range of oil pollutants. Therefore, this study was designed to evaluate the effects of relevant environmental concentrations of benzo[α]pyrene (BαP) on liver, gill, and blood of Caspian White fish. To this end, 150 fingerling fish (6.5 ± 0.8 g) were exposed to under, near and over environmental concentrations of BαP (i.e. 50, 100, and 200 ppb, respectively) and two control groups for 21 days. Following exposure to BαP, generally, DNA damage increased in the liver and gill cells as well as the frequency of micro- and bi-nucleated erythrocytes in a time and concentration-dependent pattern. In addition, the liver and gill tissues displayed several histopathological lesions. Together, the findings are warning the health status of the Caspian Sea due to an ever-increasing concentration of BαP through using Caspian White fish as an ecological model.

Immobilized microalgae are a promising approach to incorporate microalgae in recirculating aquaculture system (RAS) for water purification. In the present study, two types of biosorbents including sodium alginate–immobilized Scenedesmus spp. and Chlorella spp. beads (algal beads) and sodium alginate beads without microalgae (alginate beads) were prepared. In the first experiment (static test), the potential of two biosorbents to remove different concentrations of total ammonia nitrogen (TAN) and total phosphorus (TP) from water was investigated. In the second experiment, two prepared biosorbents were used as biofilter in a RAS for rearing African cichlid (Labidochromis lividus) for 30 days. The survival rate and growth indices of fingerling fish and removal efficiency of two biosorbents for TAN, NO₃—N, and TP were determined. The results of static test showed that the removal efficiency and uptake capacity of the two biosorbents for TAN and TP increased during 30 days of the experiment, and these values for the algal beads were higher than the alginate beads. The TAN removal efficiency of the two biosorbents increased with increasing TAN concentration from 0.5 to 5 mg L⁻¹. The application of algal beads in the RAS improved the survival rate, final weight, final length, weight gain, and daily growth index (DGI%) indices of fish compared to those cultured in the RAS containing the alginate beads and the control (P<0.05). The algal and alginate beads decreased the TAN concentration by 42.85% and 28.57% compared to the control after 30 days of cultivation period, respectively. The uptake of nitrate was not observed by the two biosorbents during cultivation period. The TP removal efficiency of algal beads reached 44.90% after 30 days. The findings of this study indicated that the sodium alginate–immobilized microalgae could be considered as a suitable biofilter to be incorporated into a RAS to improve water quality and consequently enhance the growth and health of fish.

The present study assessed the impact of fingerling stocking of Indian major carps (Gibelion catla, Labeo rohita, and Cirrhinus mrigala) on fish yield in 24 small reservoirs of Central India. The time series data on fish stocking and yield were collected from the State Fisheries Department, Govt. of Chhattisgarh, for the year 2008 to 2016. The fish yield increased significantly (p < 0.05) from 87.11 to 379.13 kg/ha/year due to stocking indicating significant impact on fish production. The relationships between area, stocking density, and fish yield of reservoirs were also assessed. Stocking had a significant positive impact on fish yield (t test, p < 0.05). The important water quality parameters were also studied to have a conspicuous picture of eco-hydrological characteristics of these selected small reservoirs. The study forms a baseline for the small tropical reservoir fisheries of Central India (Chhattisgarh state) and suggests regional strategies for sustainable fisheries enhancement and management, employment generation, and nutritional security of a large number of fish-eating population.

Acute toxicity (96 h LC₅₀) of phenol was analyzed in the cat fish Mystus vittatus in static bio-assay over a 96-h exposure period using probit method. The 24, 48, 72, and 96 h LC₅₀ values (with 95% confidence limits) of phenol for fingerling catfish were found out as 13.98, 13.17, 12.62, and 12.21 mg/l respectively. Investigations pertaining to the histopathological sections have shown high degree of pathological lesions observed in various parts like gill, liver intestine, and kidney of the fish species. Analysis of gill section revealed observable changes in the experimental species such as fusion, malformation at the tip of secondary lamellae, vacuolation, hyperplasia, and epithelial damage. Exposure of phenol showed cytoplasmic vacuolation, tissue damage, and loss of hepatic cell wall in the liver of experimental organism. Lesions of tissue damage at the epithelial site, inflammation, and clumping of adjacent villi made of columnar epithelium have been observed in the intestine of fish, and also the excretory part of the fish kidney revealed various changes like glomerular atrophy, damage of Bowman’s capsule, vacuolization, and degeneration of renal epithelium. The current study on histological changes observed in the experimental organisms has thrown light on the current scenario which poses threat and danger to the whole aquatic ecosystem, and this study plays a vital role in assessing the aquatic pollution.

Deltamethrin is one of the most commonly used pyrethroids in the world, and it has a high toxic potential, mainly on aquatic organism. Thus, the purpose of this study was to evaluate LC₅₀ values of deltamethrin on tambaqui (Colossoma macropomum) fingerlings and to investigate genotoxic effects and histopathological responses. Fish were exposed to different concentrations of deltamethrin (0, 6.16 × 10⁻³; 6.44 × 10⁻²; 1.34 × 10⁻¹, and 1.93 × 10⁻¹ mg L⁻¹) for 96 h. In addition, a genotoxicity analysis was carried out on peripheral blood erythrocytes and histopathological changes were classified by the severity degree of damage and organ functioning. The 96 h LC₅₀ value for tambaqui was estimated at 5.56 × 10⁻² mg L⁻¹ using a static test system. Nuclear abnormalities in exposed fish included micronuclei, blebbed, notched, 8-shaped, and binucleated nuclei forms. Deltamethrin significantly induced a notched nucleus compared to other abnormalities. A histopathological examination showed hepatic lesions and gill damage. Deltamethrin was found to be highly toxic; it induced genotoxicity and caused liver and gill inflammation in tambaqui.

Organophosphates (e.g. chlorpyrifos ethyl) and carbamates (e.g. fenobucarb) are commonly used to control a wide range of pests in rice fields of the Mekong Delta in Vietnam. This study assesses the combined effect of chlorpyrifos ethyl (CPF) and fenobucarb (F), applied at concentrations used by rice farmers, on the brain acetylcholinesterase (AChE) activity in climbing perch fingerlings from rice fields in the Mekong Delta. It also investigates if Pyridine-2-aldoxime methiodide (2-PAM) can be used to reactivate cholinesterase that has been blocked by CPF. Three days after spraying, the water concentrations of both insecticides decreased quickly below the detection levels. However, the brain AChE activity in fish was inhibited for more than 7 days. The results indicate a quicker but less prolonged inhibition of the brain AChE activity by the mixture than by only CPF. The inhibition levels were above 70 % only during the first 48 h, which could explain why all fish survived. 2-PAM resulted in a significant reactivation of the cholinesterase blocked by a combination of CPF and F, and it is proposed that the reactivation by 2-PAM could provide a way to assess the AChE inhibition levels in organisms, when no unexposed individuals are available as controls. The results indicate that the current use of CPF and F in rice farming in the Mekong Delta is likely to cause negative effects on non-target organisms. Many of these effects may be sub-lethal, and there is a need to develop biomarkers that are relevant, inexpensive and easy to apply. The results show that brain AChE in climbing perch fingerling is a relevant biomarker for monitoring of exposure to, and sub-lethal impacts from organophosphates and carbamates under tropical conditions in developing countries.

The present study was carried out to assess the contamination levels of heavy metals (Pb, Cd, Cr, Cu, Hg, Al, Ni, Co, Zn, Mn) in surface water and sediment of the Halda River. The observed order of heavy metal concentration in water for Al > Ni > Zn > Mn > Cu > Cd > Pb > Cr > Co > Hg (mg/l) and for sediments Al > Mn > Zn > Ni > Cr > Pb > Cu > Co > Cd > Hg (mg/kg), respectively. The concentrations of Pb, Cd, Cr, Cu, Hg, Al, Ni, Co, Zn, and Mn in water, whereas in sediment Pb, Cu, Al, Ni, Co, Zn, and Mn were found above the permissible limit (WHO 2004; USEPA 2006; EPA 1986, 2002 and ECR 1997). Significant variations in the concentrations of Al and Ni were found in water (p < 0.05) while Cr, Cu, Pb, Co, Mn, and Ni showed substantial changes in sediment (p < 0.05). Principal component analysis (PCA) and correlation matrix revealed anthropogenic intrusions of Pb, Cd, Cr, Cu, Hg, Al, Ni, Co, Zn, and Mn in water and sediment. In case of water, very strong linear relationship was found in Hg vs Pb (0.941), Mn vs Zn (0.939), and Ni vs Cu (0.922) at the significance level 0.01. In sediment, very strong linear relationships were found in Mn vs Cr (0.999), Co vs Ni (0.999), Ni vs Cu (0.994), Zn vs Pb (0.993), Co vs Cu (0.992), Cu vs Cr (0.990), Mn vs Cu (0.989), Mn vs Ni (0.975), Mn vs Co (0.975), Ni vs Cr (0.974), Co vs Cr (0.972), Mn vs Pb (0.951), Cr vs Pb (0.948), Zn vs Cr (0.944), and Mn vs Zn (0.941) at the significance level 0.01 which direct that their common origin entirely from industrial effluents, municipal wastes, and agricultural activities. The study shows that seasonal water flows/water discharge (pre-monsoon, monsoon, and post-monsoon) have an impact on the mobility of metals. Elevated levels of metals were detected during monsoon in sediments (Pb, Cr, Cu, Al, Ni, Co, Zn, Mn) and post-monsoon in water (Cd, Hg, Ni, Co, Mn). The detection of high-risk metals in the Halda River may demonstrate that metals can cause significant effects on fry and fingerlings of the Gangetic carp fishery and prawn fishery (via sub-lethal and lethal effects and bioaccumulation or secondary poisoning of metals to fish and prawn).

The combined effect of Vitashield 40EC (chlorpyrifos ethyl-CPF) and Bassa 50EC (fenobucarb-F) was compared with the effects from exposure to the two pesticides separately, by measuring the brain acetylcholinesterase (AChE) activity in climbing perch fingerlings (Anabas testudineus). The experiment was conducted under controlled laboratory conditions and included three treatments containing 0.173 mg/L of CPF, 1.137 mg/L of F, 0.173 mg/L of CPF + 1.137 mg/L of F (M), and a control. The inhibition of the brain AChE activity in fish exposed to F was weaker and shorter than in fish exposed to CPF. The inhibition by the mixture of CPF and F was significantly lower and less prolonged than the inhibition by only CPF but significantly higher than the inhibition by only F.

The interfacial properties depicting the micellization behaviour of the cationic amphiphiles (surfactants) belonging to the class of quaternary ammonium salts varying in degree of hydrophobicity were evaluated using tensiometry, conductivity and fluorescence spectrophotometric methods at 303.15 K. The impact of the amphiphilic nature of these amphiphiles as a function of their concentration is accounted against the selective microbial strains using the well-diffusion approach. Also, its influence on the histological (shrinkage/curling of lamellae, necrosis, haemorrhage, hyperplasia of villi in gills and intestine) alterations and haematological (blood parameters) changes in fingerling of Cirrhinus mrigala (C. mrigala) offers an insight into the stern damages reported as aquatic toxicity. The lesions exhibited moderate to severe alterations that are further correlated with the semi-quantitative mean alteration value (MAV). The in vitro and in vivo findings are explained significantly in terms of amphiphilic hydrophobicity which followed the order: C₁₆TAB > C₁₂TAB. All the observed outcomes are rationalized by the structural assessment of the selected amphiphiles as specified by the computational simulation approach using density functional theory (DFT) with B3LYP method and 3-21G basis source set. This work also portrays the biodegradability of these cationic amphiphiles and their fate on the environment.
