Neuromuscular, retinal, and reproductive impact of low-dose polystyrene microplastics on Drosophila

Facing the challenge of global microplastics (MPs) pollution, full characterization of MPs biohazards is urgent. Recent intensive studies revealed that the toxicity depends on the material, size, and exposure concentration of MP. To better elucidate MPs biohazards, we investigated the impact of polystyrene-MPs of size 0.1 μm at a low dose of 50 μg/L on the neuromuscular, retinal, and reproductive phenotypes of fruit fly model, by voltage-clamped electrophysiology, electroretinogram, and reproductive assay, respectively. We found that MPs decreased the frequency of spontaneous junction currents of synapse and altered the receptor potential amplitude of the retina. Furthermore, MPs lowered the rate of embryo-laying of fruit flies. The differential gene expression of ligand-receptor interaction, endocytosis, phototransduction, and Toll/Imd signaling pathways might underlie these MPs-induced phenotypes. These findings call for further investigation on the potential biohazards of low-dose MPs.

Bis(2-ethylhexyl) phthalate (DEHP) is the most common plasticizer. Previous studies have shown DEHP treatment accelerates neurological degeneration, suggesting that DEHP may impact retinal sensitivity to light, neurotransmission, and copulation behaviors. Although its neurotoxicity and antifertility properties have been studied, whether DEHP exposure disrupts vision and how DEHP influences neuromuscular junction (NMJ) have not been reported yet. Moreover, the impact of DEHP on insect courtship behavior is still elusive. Fruit flies (Drosophila melanogaster) were treated with series concentrations of DEHP and observed for lifespan, motor function, electroretinogram (ERG), electrophysiology of neuromuscular junction (NMJ), courtship behaviors, and relevant gene expression. Our results confirmed the DEHP toxicity on lifespan and capacity of motor function and updated its effect on copulation behaviors. Additionally, we report for the first time that DEHP exposure may harm vision by affecting the synaptic signaling between the photoreceptor and the laminar neurons. Further, DEHP treatment altered both spontaneous and evoked neurotransmission properties. Noteworthy, the effect of DEHP exposure on the copulation behavior is sex-dependent, and we proposed potential mechanisms for future investigation.

Toxicity of bisphenol A on morphological and life-history traits of model insect Drosophila melanogaster was reported in our previous work. In the present study, we have analyzed the adversity of bisphenol A on the reproductive behavior of adult and on the expression of selected genes in the larva and adult stage of fruit fly exposed to bisphenol A (0.007 g/2 ml. or 3.5 mg/ml), in addition to determination of LC50 value of bisphenol A in larva and pupal stage. We employed both the quantitative reverse transcriptase PCR and droplet digital PCR for analyzing the expression profile of seven genes namely, decapentaplegic, vestigial, wingless, foraging, insulin-like receptor, doublesex, and fruitless. We found bisphenol A has more adverse effects on male sexual behavior than females. Moreover, we observed significant downregulation of all the selected genes in treated larvae except, fruitless in male where it showed significant upregulation. On contrary among the treated adult flies, significant downregulation of all target genes in both sexes is evident, except, doublesex and fruitless in males which showed significant upregulation. We did not observe any deviation of male: female sex ratio from 1:1 under bisphenol A exposure. All these results suggest bisphenol A adversely affects the optimum functioning of genes which are involved in the regulation of metabolic pathways, behavioral pattern, stress response, endocrine homeostasis, neural functioning, and the development of the specific organ in Drosophila melanogaster. Our result not only provides a foundation to study further the bisphenol A toxicity on different pivotal genes in Drosophila but also suggests the use of the droplet digital PCR technology in toxicity measurement at the molecular level in eukaryotic model systems.

The peach fruit fly (PFF), Bactrocera zonata, is a serious insect pest infesting fruits and vegetables. The insecticidal activity of three monoterpenes, namely, (R)-camphor, (R)-carvone, and (1R,2S,5R)-menthol, was evaluated on the second-instar larvae of B. zonata. In addition, the latent effects of monoterpenes on pupation, adult emergence, deformation, oviposition, adult longevity, and ovarian development were also examined. The three tested monoterpenes showed pronounced insecticidal activity against B. zonata larvae with (R)-carvone being the most potent toxicant. When the second-instar larvae of B. zonata were treated with monoterpenes at concentrations of 20, 50, and 70 mg/kg for 72 h, significant reduction in pupation and adult emergence was observed. The three monoterpenes caused complete suppression of adult emergence at 100 mg/kg. Moreover, monoterpenes induced complete inhibition of egg deposition at all tested concentrations. Some adult deformations were also noticed at 20, 50, and 70 mg/kg. However, (R)-carvone was more effective than (1R,2S,5R)-menthol and (R)-camphor on the examined biological parameters. On the other hand, histological examination of the ovaries of emerged females from larvae that fed on diet treated with (R)-carvone, (1R,2S,5R)-menthol, and (R)-camphor at 20 and 50 mg/L indicated that both concentrations caused retardation in the development of ovarioles. It is clear that all the egg chambers are empty; the germarium region is constricted at base due to the failure of oocyte formation. Many vacant spaces were present between ovarioles.

Cadmium (Cd) can be transferred and accumulated in spiders, posing a survival risk to them. To analyze potential biological damage caused by Cd accumulation and relevant detoxification strategies employed by spiders in response to Cd exposure, we conducted transcriptome analysis of the 5th instar spider P. pseudoannulata, a common spider species playing a vital role in natural pest control in agricultural fields of southern China. We obtained 92,778 unigenes with an average length of 1104 bp and identified 302, 655, and 424 differentially expressed genes (DEGs) in the spiders fed with Cd-containing fruit flies for 2, 5, and 8 days, respectively. Results showed that the body mass of Cd-containing P. pseudoannulata were reduced when compared with controls, presumably due to delayed maturation of tissues and organs. Meanwhile, functional analysis of DEGs indicated that Cd may have a negative effect on neural signal transduction and molt cycle of the spider. For defense strategies, detoxification enzymes like glutathione S-transferase (GST), catalase (CAT), superoxide dismutase (SOD), and P450, and typical proteins like heat shock protein and metallothionein were all differentially expressed in response to Cd stress. Besides, innate immune responses like toll-like receptor signaling pathways were also upregulated. Multiple critical Cd-responsive genes involved in biological damage, detoxification, and immune response were identified, providing referable foundation for further research on Cd toxicity to P. pseudoannulata.

Methylmercury (MeHg) is a well-known environmental pollutant associated with neurological and developmental deficits in animals and humans. However, epidemiological data showed that people living in the Amazon region although exposed to MeHg do not present these effects probably due to the protective effect of certain foods. We hypothesized here if guarana, a highly caffeinated fruit and consumed on a daily basis by Amazon people, could have some protective effect against MeHg toxicity using two complementary approaches. To assess locomotor impairment and sleep disruption, we used fruit fly (Drosophila melanogaster) model, and to evaluate neuroinflammation, we used human SH-SY5Y neural cells by measuring inflammatory cytokines levels. Results showed that guarana had a protective effect on the locomotor activity of male fruit flies reducing the excessive sleepiness caused by MeHg and increasing daily activity. Also, guarana increased the viability of flies and attenuated neural cells mortality. In addition, guarana reduced all pro-inflammatory cytokines levels increased by MeHg, along with caspase-1, caspase -3, caspase-8, and 8-dOHG levels, whereas increased the anti-inflammatory (IL-10) cytokine levels, which was decreased by MeHg. Our study provides new insights on the protective effects of guarana on the viability, locomotor activity, sleep, and activity patterns in vivo and the in vitro neuronal anti-inflammatory effect against MeHg toxicity.
