Perfluorooctane sulfonate alternatives and metabolic syndrome in adults: New evidence from the Isomers of C8 Health Project in China Полный текст

Chlorinated polyfluoroalkyl ether sulfonates (Cl-PFESAs), are ubiquitous alternatives to perfluorooctane sulfonate (PFOS), a widely used poly- and perfluoroalkyl substance (PFAS). Despite in vivo and in vitro evidence of metabolic toxicity, no study has explored associations of Cl-PFESAs concentrations with metabolic syndrome (MetS) in a human population. To help address this data gap, we quantified 32 PFAS, including 2 PFOS alternative Cl-PFESAs (6:2 and 8:2 Cl-PFESAs) in serum from 1228 adults participating in the cross-sectional Isomers of C8 Health Project in China study. The odds ratios (ORs) and 95% confidence intervals (CIs) of MetS and its various components were estimated using individual PFAS as a continuous or categorical predictor in multivariate regression models. The association between the overall mixture of PFAS and MetS was examined using probit Bayesian Kernel Machine Regression (BKMR-P). Greater serum PFAS concentrations were associated with higher odds of MetS and demonstrated a statistically significant dose-response trend (P for trend < 0.001). For example, each ln-unit (ng/mL) increase in serum 6:2 Cl-PFESA was associated with a higher prevalence of MetS (OR = 1.52, 95% CI: 1.25, 1.85). MetS was also 2.26 (95% CI: 1.59, 3.23) times more common in the highest quartile of serum 6:2 Cl-PFESA concentration than the lowest, and particularly high among women (OR = 6.41, 95% CI: 3.65, 11.24). The BKMR-P analysis showed a positive association between the overall mixture of measured PFAS and the odds of MetS, but was only limited to women. While our results suggest that exposure to Cl-PFESAs was associated with MetS, additional longitudinal studies are needed to more definitively address the potential health concerns of these PFOS alternatives.

As one of the most dominant ecosystems of urban green space, turfgrasses provide a wide range of ecosystem services. However, little is known about the interactions of microbial communities in turfgrass soils and how these interactions respond to expanding development of impervious surfaces during watershed urbanization. In this study, we analyzed bacterial communities and their co-occurrence patterns in turfgrass soils along an urbanization gradient as measured by the proportion of impervious surfaces in Jiulong River watershed in Fujian, China. Results show that the diversity and network size of bacterial communities negatively associated with impervious surfaces. The bacterial communities showed non-random co-occurrence patterns, with more intra-module connections observed for urbanized networks. The co-occurrence network with distinct modules of soil samples with contrasting land cover imperviousness suggested different functional organizations with altered microbial nitrogen processes. Structural equation modelling revealed that watershed impervious surfaces had indirect impacts on microbial connectivity by altering soil properties, including pH, temperature, moisture, C/N and nitrate (NO₃⁻). Moreover, impervious surfaces affected microbial connectivity far more than human population density. Our study highlights the significance of human disturbances in affecting microbial interactions and assemblies in turfgrass ecosystems through impervious surfaces and provides benefits for sustainable urban planning and management at a watershed scale.

Anthropogenic noise is an emergent ecological pollutant in both terrestrial and aquatic habitats. Human population growth, urbanisation, resource extraction, transport and motorised recreation lead to elevated noise that affects animal behaviour and physiology, impacting individual fitness. Currently, we have a poor mechanistic understanding of the effects of anthropogenic noise, but a likely candidate is the neuroendocrine system that integrates information about environmental stressors to produce regulatory hormones; glucocorticoids (GCs) and androgens enable rapid individual phenotypic adjustments that can increase survival. Here, we carried out two field-based experiments to investigate the effects of short-term (30 min) and longer-term (48 h) motorboat-noise playback on the behaviour, GCs (cortisol) and androgens of site-attached free-living orange-fin anemonefish (Amphiprion chrysopterus). In the short-term, anemonefish exposed to motorboat-noise playback showed both behavioural and hormonal responses: hiding and aggression increased, and distance moved out of the anemone decreased in both sexes; there were no effects on cortisol levels, but male androgen levels (11-ketotestosterone and testosterone) increased. Some behaviours showed carry-over effects from motorboat noise after it had ceased, and there was no evidence for a short-term change in response to subsequent motorboat-noise playback. Similarly, there was no evidence that longer-term exposure led to changes in response: motorboat noise had an equivalent effect on anemonefish behaviour and hormones after 48 h as on first exposure. Longer-term noise exposure led to higher levels of cortisol in both sexes and higher testosterone levels in males, and stress-responses to an additional environmental challenge in both sexes were impaired. Circulating androgen levels correlated with aggression, while cortisol levels correlated with hiding, demonstrating in a wild population that androgen/glucocorticoid pathways are plausible proximate mechanisms driving behavioural responses to anthropogenic noise. Combining functional and mechanistic studies are crucial for a full understanding of this global pollutant.

Adult coho salmon (Oncorhynchus kisutch) prematurely die when they return from the ocean to spawn in urban watersheds throughout northwestern North America. The available evidence suggests the annual mortality events are caused by toxic stormwater runoff. The underlying pathophysiology of the urban spawner mortality syndrome is not known, and it is unclear whether closely related species of Pacific salmon are similarly at risk. The present study co-exposed adult coho and chum (O. keta) salmon to runoff from a high traffic volume urban arterial roadway. The spawners were monitored for the familiar symptoms of the mortality syndrome, including surface swimming, loss of orientation, and loss of equilibrium. Moreover, the hematology of both species was profiled by measuring arterial pH, blood gases, lactate, plasma electrolytes, hematocrit, and glucose. Adult coho developed behavioral symptoms within a few hours of exposure to stormwater. Various measured hematological parameters were significantly altered compared to coho controls, indicating a blood acidosis and ionoregulatory disturbance. By contrast, runoff-exposed chum spawners showed essentially no indications of the mortality syndrome, and measured blood hematological parameters were similar to unexposed chum controls. We conclude that contaminant(s) in urban runoff are the likely cause of the disruption of ion balance and pH in coho but not chum salmon. Among the thousands of chemicals in stormwater, future forensic analyses should focus on the gill or cardiovascular system of coho salmon. Because of their distinctive sensitivity to urban runoff, adult coho remain an important vertebrate indicator species for degraded water quality in freshwater habitats under pressure from human population growth and urbanization.

Coastal urban infrastructures are proliferating across the world, but knowledge about their emergent impacts is still limited. Here, we provide evidence that urban artificial reefs have a high potential to accumulate the diverse forms of litter originating from anthropogenic activities around cities. We test the hypothesis that the structural complexity of urban breakwaters, when compared with adjacent natural rocky intertidal habitats, is a driver of anthropogenic litter accumulation. We determined litter abundances at seven sites (cities) and estimated the structural complexity in both urban breakwaters and adjacent natural habitats from northern to central Chile, spanning a latitudinal gradient of ∼15° (18°S to 33°S). Anthropogenic litter density was significantly higher in coastal breakwaters when compared to natural habitats (∼15.1 items m−2 on artificial reefs versus 7.4 items m−2 in natural habitats) at all study sites, a pattern that was temporally persistent. Different litter categories were more abundant on the artificial reefs than in natural habitats, with local human population density and breakwater extension contributing to increase the probabilities of litter occurrence by ∼10%. In addition, structural complexity was about two-fold higher on artificial reefs, with anthropogenic litter density being highest at intermediate levels of structural complexity. Therefore, the spatial structure characteristic of artificial reefs seems to enhance anthropogenic litter accumulation, also leading to higher residence time and degradation potential. Our study highlights the interaction between coastal urban habitat modification by establishment of artificial reefs, and pollution. This emergent phenomenon is an important issue to be considered in future management plans and the engineering of coastal ecosystems.

Trees remove air pollutants through dry deposition processes depending upon forest structure, meteorology, and air quality that vary across space and time. Employing nationally available forest, weather, air pollution and human population data for 2010, computer simulations were performed for deciduous and evergreen trees with varying leaf area index for rural and urban areas in every county in the conterminous United States. The results populated a national database of annual air pollutant removal, concentration changes, and reductions in adverse health incidences and costs for NO2, O3, PM2.5 and SO2. The developed database enabled a first order approximation of air quality and associated human health benefits provided by trees with any forest configurations anywhere in the conterminous United States over time.Comprehensive national database of tree effects on air quality and human health in the United States was developed.

Novel approaches for estimating the emissions of perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) to surface waters are explored. The Danube River catchment is used to investigate emissions contributing to riverine loads of PFOS and PFOA and to verify the accuracy of estimates using a catchment-scale dynamic fugacity-based chemical transport and fate model (STREAM-EU; Spatially and Temporally Resolved Exposure Assessment Model for European basins). Model accuracy evaluation performed by comparing STREAM-EU predicted concentrations and monitoring data for the Danube and its tributaries shows that the best estimates for PFOS and PFOA emissions in the Danube region are obtained by considering the combined contributions of human population, wealth (based on local gross domestic product (GDP)) and wastewater treatment. Human population alone cannot explain the levels of PFOS and PFOA found in the Danube catchment waters. Introducing wealth distribution information in the form of local GDPs improves emission estimates markedly, likely by better representing emissions resulting from consumer trends, industrial and commercial sources. For compounds such as PFOS and PFOA, whose main sink and transport media is the aquatic compartment, a major source to freshwater are wastewater treatment plants. Introducing wastewater treatment information in the emission estimations also further improves emission estimates.

In England and Wales, steroid estrogens: estrone, estradiol and ethinylestradiol have previously been identified as the main chemicals causing endocrine disruption in male fish. A national risk assessment is already available for intersex in fish arising from estrogens under current flow conditions. This study presents, to our knowledge, the first set of national catchment-based risk assessments for steroid estrogen under future scenarios. The river flows and temperatures were perturbed using three climate change scenarios (ranging from relatively dry to wet). The effects of demographic changes on estrogen consumption and human population served by sewage treatment works were also included. Compared to the current situation, the results indicated increased future risk:the percentage of high risk category sites, where endocrine disruption is more likely to occur, increased. These increases were mainly caused by changes in human population. This study provides regulators with valuable information to prepare for this potential increased risk.

Since early 1980s, chemical pollution has become a serious environmental problem in rapidly developing China. This study reviewed the policy and legal framework for monitoring and management of chemical pollutants in China, with reference to the relevant experience in other jurisdictions. Although efforts in environmental monitoring of chemical contamination have been substantially increased over the last decade, China is lagging behind in terms of nationwide monitoring of chemical contamination in human population, and standardization of sampling and analytical protocols. While actively participating in various international treaties and conventions related to pollution control, China also has a very comprehensive set of environmental laws and policies. These include the newly enacted legislation on the control of new chemicals (i.e., China REACH) and the development of a set of National Environmental Standards. In addition to environmental education, these new measures will further enhance the control of chemical pollutants and facilitate effective law enforcement.

Human exposure to organic contaminants is widespread. Many of these contaminants show adverse health effects on human population. Human biomonitoring (HBM) follows the levels and the distribution of biomarkers of exposure (BoE), but it is usually done in a targeted manner. Suspect and non-targeted screening (SS/NTS) tend to find BoE in an agnostic way, without preselection of compounds, and include finding evidence of exposure to predicted, unpredicted known and unknown chemicals. This study describes the application of high-resolution mass spectrometry (HRMS)-based SS/NTS workflow for revealing organic contaminants in urine of a cohort of 200 children from Slovenia, aged 6–9 years. The children originated from two regions, urban and rural, and the latter were sampled in two time periods, summer and winter. We tentatively identified 74 BoE at the confidence levels of 2 and 3. These BoE belong to several classes of pharmaceuticals, personal care products, plasticizers and plastic related products, volatile organic compounds, nicotine, caffeine and pesticides. The risk of three pesticides, atrazine, amitraz and diazinon is of particular concern since their use was limited in the EU. Among BoE we tentatively identified compounds that have not yet been monitored in HBM schemes and demonstrate limited exposure data, such as bisphenol G, polyethylene glycols and their ethers. Furthermore, 7 compounds with unknown use and sources of exposure were tentatively identified, either indicating the entry of new chemicals into the market, or their metabolites and transformation products. Interestingly, several BoE showed location and time dependency. Globally, this study presents high-throughput approach to SS/NTS for HBM. The results shed a light on the exposure of Slovenian children and raise questions on potential adverse health effects of such mixtures on this vulnerable population.
