peer reviewed | The northwestern shelf of the Black Sea has been affected by eutrophication and bottom hypoxia since the sixties. Consequently, the macrozoobenthos has suffered a well-established decline in biodiversity. However, the nature of the current benthic communities remains questionable. From 1995 to 2017, we compiled species and abiotic data for 138 sites over the shelf. Through an appropriate multivariate analytical approach, we identified benthic community changes solely due to organic pollution variations. Our results show signs of recovery with an increase in biodiversity and proportion of species vulnerable to organic enrichment. These changes were related to a decrease in riverine loads and subsequent eutrophication. However, some long-lived species typical of the area still did not exhibit noticeable recovery, which suggests that either the recovery process has not yet been achieved or some environmental conditions are still not met to warrant a sea floor ecosystem state substantially healthy. | 14. As a widely used pure elemental carbon in colloidal particles, carbon black was listed as a group 2B carcinogen by IARC in 2010. The most available mechanism information about carbon black and carcinogenesis are from in vivo or in vitro studies. However, few studies concerned the nanoparticle's real-ambient exposure causing systemic change and further affecting the target organ. Herein, we used an ex vivo biosensor assay to investigate the transcriptome change of primary bronchial epithelial cells after treatment with the plasma from workers with long-term occupational carbon black exposure history. Based on ex vivo biosensor assay and transcriptome sequencing, we found the effect of internal systemic environment on epithelial cells after carbon black exposure was an inflammatory response, which mainly activates cell cycle-related pathways. After exposure to carbon black, the internal systemic environment could activate cancer-related pathways like epithelial-mesenchymal transition, hypoxia, TNF-α signaling via NF-κB. The hub genes in the carbon black group (CDC20 and PLK1) and their correlation with the systemic environment were uncovered by constructing the protein-protein interaction network. Inflammatory cytokines, especially CRP, were strongly correlated with the expression of CDC20 and PLK1. Besides, we also find a strong correlation between CDC20 and cytokinesis-block micronucleus endpoints in peripheral blood (rho = 0.591, P < 0.001). Our results show that long-term carbon black exposure might activate cell cycle-related pathways through circulating inflammation and increase the risk of cancer, while the oxidative stress caused by diesel exhaust particles are mainly related to PAHs exposure. After exposure to carbon black, the systemic environment could activate cancer-related pathways like diesel exhaust particles, increasing the risk of lung cancer. These attempts might provide a further understanding of the indirect effect of chronic occupational inhaled carbon black exposure on pulmonary carcinogenesis.

Hypoxia (low oxygen) often occurs in aquatic ecosystems that receive effluent from municipal wastewater treatment plants (WWTP). The combination of hypoxia and WWTP effluent could impair fish health, because WWTP effluent contains multiple contaminants that could disrupt the physiological pathways fish use to cope with hypoxia, but the interactive effects of these stressors on fish physiology are poorly understood. We have examined this issue by exposing mummichog killifish (Fundulus heteroclitus) to hypoxia (5 and 2 kPa O₂) and/or 100% WWTP effluent for 21 days in a full factorial design. We then measured hypoxia tolerance, whole-animal metabolism, gill morphology, haematology, and tissue metabolites. In clean water, killifish responded to chronic hypoxia with improvements in hypoxia tolerance, as reflected by increases in time to loss of equilibrium at 0.5 kPa (tLOE). These improvements occurred in association with increases in the exposed surface of gill lamellae that resulted from a regression of interlamellar cell mass (ILCM). Concurrent exposure to wastewater attenuated the increases in tLOE and gill remodeling in chronic hypoxia, and nearly depleted brain glycogen stores. Therefore, exposure to WWTP effluent can disrupt the physiological mechanisms fish use to cope with chronic hypoxia and impair hypoxia tolerance. Our research suggests that the combination of stressors near WWTPs can have interactive effects on the physiology and health of fish.

Nitrogen (N) pollution and the resulting eutrophication can have deleterious consequences on estuaries, such as hypoxia, fish kills, and loss of biotic diversity. An understanding of N sources and cycling in estuaries is fundamental to determining how to effectively manage these ecologically and commercially important areas. We applied a multiple-isotopic approach to examine the transformations and sources of the N pools in the Pearl River Estuary (PRE) during winter. The surface water in the West PRE was characterized by low salinity and high NO₃⁻, while that in the east had high salinity and low NO₃⁻. The NO₃⁻ in the West PRE was largely regulated by a conservative mixing process. In contrast, assimilation and nitrification dominated in the East PRE, which was attributed to the long water-residence time. For the first time, the source contributions of NO₃⁻ and NH₄⁺ were estimated by isotope mixing models. Our results suggest that river discharge and nitrification contributed 81% and 12% to the NO₃⁻ pool, respectively. A major portion (68%) of the NH₄⁺ was from river discharge, with the remainder likely from sewage and the aquitard-aquifer system. Our study demonstrates that internal nitrification can potentially be of pivotal importance in determining the NO₃⁻ level in an estuary and its export to coastal waters.

2021
Jaiswal, Deepa | Pandey, Jitendra
Riverine ecosystems can have tipping points at which the system shifts abruptly to alternate states, although quantitative characterization is extremely difficult. Here we show, through critical analysis of two different reach scale (25 m and 50 m) studies conducted downstream of two point sources, two tributaries (main stem and confluences) and a 630 km segment of the Ganga River, that human-driven benthic hypoxia/anoxia generates positive feedbacks that propels the system towards a contrasting state. Considering three positive feedbacks-denitrification, sediment-P- and metal-release as level determinants and extracellular enzymes (β-D-glucosidase, protease, alkaline phosphatase and FDAase) as response determinants, we constructed a ‘river ecosystem resilience risk index (RERRI)’ to quantitatively characterize tipping points in large rivers. The dynamic fit intersect models indicated that the RERRI<4 represents a normal state, 4–18 a transition where recovery is possible, and >18 an overstepped condition where recovery is not possible. The resilience risk index, developed for the first time for a lotic ecosystem, can be a useful tool for understanding the tipping points and for adaptive and transformative management of large rivers.

Hypoxia is a major stressor in aquatic environments and it is frequently linked with excess nutrients resulting from sewage effluent discharges and agricultural runoff, which often also contain complex mixtures of chemicals. Despite this, interactions between hypoxia and chemical toxicity are poorly understood. We exposed male three-spined stickleback during the onset of sexual maturation to a model anti-androgen (flutamide; 250 μg/L) and a pesticide with anti-androgenic activity (linuron; 250 μg/L), under either 97% or 56% air saturation (AS). We assessed the effects of each chemical, alone and in combination with reduced oxygen concentration, by measuring the transcription of spiggin in the kidney, as a marker of androgen signalling, and 11 genes in the liver involved in some of the molecular pathways hypothesised to be affected by the exposures. Spiggin transcription was strongly inhibited by flutamide under both AS conditions. In contrast, for linuron, a strong inhibition of spiggin was observed under 97% AS, but this effect was supressed under reduced air saturation, likely due to interactions between the hypoxia inducible factor and the aryl hydrocarbon receptor (AhR) pathways. In the liver, hypoxia inducible factor 1α was induced following exposure to both flutamide and linuron, however this was independent of the level of air saturation. This work illustrates the potential for interactions between hypoxia and pollutants with endocrine or AhR agonist activity to occur, with implications for risk assessment and management.

Hypoxia off the Changjiang Estuary (CE) and its adjacent waters is purported to be the most severe in China, attracting considerable concern from both the scientific community and the general public. Currently, continuous observations of dissolved oxygen (DO) levels covering hypoxia from its appearance to disappearance are lacking. In this study, twelve consecutive monthly cruises (from February 2015 to January 2016) were conducted. The consecutive spatiotemporal variations in hypoxia throughout the annual cycle were elucidated in detail, and the responses of annual variations in hypoxia to the different influential factors were explored. Overall, hypoxia experienced a consecutive process of expanding from south to north, then disappearing from north to south. The annual variations in hypoxia were mainly contingent on stratification variations. Among different stages, there was significant heterogeneity in the dominant factors. Specifically, low-DO waters initially appeared from the intrusion of nearshore Kuroshio branch current (NKBC), as NKBC intrusion provided a low-DO background and triggered stratification. Thereafter, stratification was enhanced and gradually expanded northward, which promoted the extension of low-DO areas. The formation of hypoxia was regionally selective, and more intense organic matter decomposition at local regions facilitated the occurrence and discontinuous distribution of hypoxia. Hypoxic zones were observed at the Changjiang bank and Zhejiang coastal region from August (most extensively at 14,800 km²) to October. Thereafter, increased vertical mixing facilitated the dissipation of hypoxia from north to south.

Hydrocarbons contamination and hypoxia are two stressors that can coexist in coastal ecosystems. At present, few studies evaluated the combined impact of these stressors on fish physiology and behavior. Here, we tested the effect of the combination of hypoxia and petrogenic hydrocarbons on the anti-predator locomotor performance of fish. Specifically, two groups of European sea bass (Dicentrarchus labrax) were exposed to clean water (Ctrl) or oil-contaminated water (Oil). Subsequently, fish of both groups were placed in normoxic (norx) or hypoxic (hyp) experimental tanks (i.e. four groups of fish were formed: Ctrl norx, Ctrl hyp, Oil norx, Oil hyp). In these tanks, escape response was elicited by a mechano-acoustic stimulus and recorded with a high speed camera. Several variables were analyzed: escape response duration, responsiveness (percentage of fish responding to the stimulation), latency (time taken by the fish to initiate a response), directionality (defined as away or toward the stimulus), distance-time variables (such as speed and acceleration), maneuverability variables (such as turning rate), escape trajectory (angle of flight) and distancing of the fish from the stimulus. Results revealed (i) effects of stressors (Ctrl hyp, Oil norx and Oil hyp) on the directionality; (ii) effects of Oil norx and Oil hyp on maneuverability and (iii) effects of Oil hyp on distancing. These results suggest that individual stressors could alter the escape response of fish and that their combination could strengthen these effects. Such an impact could decrease the probability of prey escape success. By investigating the effects of hydrocarbons (and the interaction with hypoxia) on the anti-predator behavior of fish, this work increases our understanding of the biological impact of oil spill. Additionally, the results of this study are of interest for oil spill impact evaluation and also for developing new ecotoxicological tools of ecological significance.

Wind induced flow velocity patterns and associated thermal destratification can drive to hypoxia reduction in large shallow lakes. The effects of wind induced hydrodynamic changes on destratification and hypoxia reduction were investigated at the Meiling bay (N 31° 22′ 56.4″, E 120° 9′ 38.3″) of Lake Taihu, China. Vertical flow velocity profile analysis showed surface flow velocities consistency with the wind field and lower flow velocity profiles were also consistent (but with delay response time) when the wind speed was higher than 6.2 m/s. Wind field and temperature found the control parameters for hypoxia reduction and for water quality conditions at the surface and bottom profiles of lake. The critical temperature for hypoxia reduction at the surface and the bottom profile was ≤24.1C° (below which hypoxic conditions were found reduced). Strong prevailing wind field (onshore wind directions ESE, SE, SSE and E, wind speed ranges of 2.4–9.1 m/s) reduced the temperature (22C° to 24.1C°) caused reduction of hypoxia at the near surface with a rise in water levels whereas, low to medium prevailing wind field did not supported destratification which increased temperature resulting in increased hypoxia. Non-prevailing wind directions (offshore) were not found supportive for the reduction of hypoxia in study area due to less variable wind field. Daytime wind field found more variable (as compared to night time) which increased the thermal destratification during daytime and found supportive for destratification and hypoxia reduction. The second order exponential correlation found between surface temperature and Chlorophyll-a (R²: 0.2858, Adjusted R-square: 0.2144 RMSE: 4.395), Dissolved Oxygen (R²: 0.596, Adjusted R-square: 0.5942, RMSE: 0.3042) concentrations. The findings of the present study reveal the driving mechanism of wind induced thermal destratification and hypoxic conditions, which may further help to evaluate the wind role in eutrophication process and algal blooms formation in shallow water environments.Wind field is the key control factor for thermal destratification and hypoxia reduction. 24.1C° is the critical/threshold temperature for hypoxia, Chlorophyll-a and NH3-N concentrations of the shallow freshwater lake.
