Adverse effects induced by ecgonine methyl ester to the zebra mussel: A comparison with the benzoylecgonine

Cocaine and its metabolites are the prevalent psychotropic substances in aquatic environment. However, to date the knowledge on their adverse effects to non-target organisms is inadequate. The aims of this study were to investigate sub-lethal effects induced by the ecgonine methyl ester (EME) to the freshwater bivalve Dreissena polymorpha and to compare its toxicity to that by benzoylecgonine (BE), the other main cocaine metabolite. EME sub-lethal effects were investigated by 14 days in-vivo exposures and a multi-biomarker approach. Slight variations in biomarker responses were found at 0.15 μg/L treatment. 0.5 μg/L EME treatment induced destabilization of lysosome membranes, an overall inactivation of defense enzymes, increases in lipid peroxidation, protein carbonylation and DNA fragmentation, but no variations in fixed genetic damage. The use of a biomarker response index (BRI) showed that at 0.5 μg/L both cocaine metabolites had the same toxicity to zebra mussels specimens.

We tested antioxidant responses of the green microalga Pseudokirchneriella subcapitata exposed to different concentrations of the three antibiotics erythromycin (ETM), ciprofloxacin (CPF) and sulfamethoxazole (SMZ). Measurements included the level of lipid peroxidation, the total antioxidative capacity and three major antioxidant mechanisms: the ascorbate–glutathione cycle, the xanthophyll cycle and the enzyme activities of catalase (CAT), superoxide dismutase (SOD), guaiacol glutathione peroxidase (GPX) and glutathione-S-transferase (GST). Three antibiotics significantly affect the antioxidant system of P. subcapitata, but in different ways the alga was more tolerant to CPF and SMZ exposures than to ETM exposure. ETM caused reductions in AsA and GSH biosynthesis, ascorbate–glutathione cycle, xanthophylls cycle and antioxidant enzyme activities. The toxicity of CPF seems to be mainly overcome via induction of the ascorbate–glutathione cycle and CAT, SOD and GPX activities, while the toxicity of SMZ on the photosynthetic apparatus is predominantly reduced by the xanthophyll cycle and GST activity.

The present study was conducted to assess morphological, biochemical and yield responses of palak (Beta vulgaris L. cv Allgreen) to ambient and elevated levels of CO2 and O3, alone and in combination. As compared to the plants grown in charcoal filtered air (ACO2), growth and yield of the plants increased under elevated CO2 (ECO2) and decreased under combination of ECO2 with elevated O3 (ECO2 + EO3), ambient O3 (ACO2 + AO3) and elevated O3 (EO3). Lipid peroxidation, ascorbic acid, catalase and glutathione reductase activities enhanced under all treatments and were highest in EO3. Foliar starch and organic carbon contents increased under ECO2 and ECO2 + EO3 and reduced under EO3 and ACO2 + AO3. Foliar N content declined in all treatments compared to ACO2 resulting in alteration of C/N ratio. This study concludes that ambient level of CO2 is not enough to counteract O3 impact, but elevated CO2 has potential to counteract the negative effects of future O3 level.

Fluoxetine (FLX) the active pharmaceutical ingredient (API) in Prozac® is a widely prescribed psychoactive drug which ubiquitous occurrence in the aquatic environment is associated to a poor removal rate in waste-water treatment plant (WWTP) systems. This API acts as a selective serotonin reuptake inhibitor (SSRI) frequently reported to cause disrupting effects in non-target species. The objective of this study includes a multibiomarker response evaluation on mussel Mytilus galloprovincialis during two weeks exposure to 75 ng L−1 FLX assessing antioxidant enzymes activities – superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST); lipid peroxidation (LPO), acetylcholinesterase (AChE) neurotoxic response and endocrine disruption through alkali-labile phosphates (ALP) indirect measurement of vitellogenin-like proteins. Results show transient tissue-specific enzymatic responses and damage affecting mostly mussel gills. However, the clear ALP levels inhibition throughout time in both sex-differentiated gonads gives evidence to FLX reinforced action as an endocrine disruptor rather than an oxidative or neurotoxic inducer.

This study investigated the relationship between contaminant body burden and the oxidative stress status of the gills and livers of two wild fish species in the Furnas Hydroelectric Power Station (HPS) reservoir (Minas Gerais, Brazil). Gills and livers presented similar pathways of metals and organochlorine bioaccumulation. During June, organochlorines were associated with lipid peroxidation (LPO), indicating oxidative stress due to the inhibition of the antioxidant enzymes superoxide dismutase and glutathione peroxidase. In the most polluted areas, metal concentrations in the liver were associated with metallothionein. During December, contaminants in the gills and liver were associated with catalase activity and LPO. Aldrin/dieldrin was the contaminant most associated with oxidative damage in the livers of both species. This integrated approach shed light on the relationship between adverse biological effects and bioaccumulation of contaminants inputted by intensive agricultural practices and proved to be a suitable tool for assessing the environmental quality of man-made reservoirs.

Organic pollutants effects on lichens have not been addressed. Rehydration is critical for lichens, a burst of free radicals involving NO occurs. Repeated dehydrations with organic pollutants could increase oxidative damage. Our aim is to learn the effects of cumene hydroperoxide (CP) during lichen rehydration using Ramalina farinacea (L.) Ach., its photobiont Trebouxia spp. and Asterochloris erici. Confocal imaging shows intracellular ROS and NO production within myco and phycobionts, being the chloroplast the main source of free radicals. CP increases ROS, NO and lipid peroxidation and reduces chlorophyll autofluorescence, although photosynthesis remains unaffected. Concomitant NO inhibition provokes a generalized increase of ROS and a decrease in photosynthesis. Our results suggest that CP induces a compensatory hormetic response in Ramalina farinacea that could reduce the lichen's antioxidant resources after repeated desiccation-rehydration cycles. NO is important in the protection from CP.

The combined effects of decreased pH and increased temperature values on antioxidant enzyme activities and lipid peroxidation were evaluated for the first time in the clam Chamelea gallina and the mussel Mytilus galloprovincialis, two bivalve species that are widespread along the northwestern coast of the Adriatic Sea. For 7days, bivalves were exposed to three pH values (8.1, 7.7 and 7.4) at two temperatures (22 and 28°C). Three independent experiments were carried out at salinity values of 28, 34 and 40psu. Superoxide dismutase, catalase and glutathione S-transferase activities as well as lipid peroxidation were measured in the gills and digestive gland of the bivalves. The results demonstrated that the experimental conditions significantly influenced the biochemical parameters of the bivalves, although the variation pattern varied depending on the species and tissues analysed.

Biomarkers, which can detect changes at the biochemical level, have been used for many years as early warning tools in environmental monitoring. In order to distinguish between natural variability and the potential effects of anthropogenic pollution, it is essential to identify background levels and establish the potential influence of abiotic (season, temperature and salinity) and biotic (gametogenesis) factors. In this study, we examined various biomarkers of stress (glutathione S-transferase (GST)), reproduction (vitellin-like proteins) and damage (lipid peroxidation (LPO) and DNA damage (DNA)) in marine mussels (Mytilus spp.) from a known pristine hybrid zone. Levels of all biomarker expression varied between seasons and appeared to be linked to the reproductive cycle. Oxidative stress in winter, with low GST expression and a higher expression of LPO and DNA damage displayed could be explained by low food availability. These data provide vital baseline information for future ecotoxicological and environmental monitoring studies.

Although aluminum (Al) is considered innocuous to living beings, exposure to high concentrations can elicit damage. Al has been found to cause liver pathologies in various animal models. Its mechanisms of toxicity are unclear; presumably, it interacts with protein sulfhydryl groups and promotes reactive oxygen species formation causing oxidative stress. Lipid peroxidation, protein carbonyl content, and activity of superoxide dismutase, catalase, and caspase-3 were determined in liver of Cyprinus carpio exposed to 0.05, 120, and 239.42 mg Al L⁻¹ for 12, 24, 48, 72, and 96 h. Al induced increased lipid peroxidation and protein carbonyl content as well as changes in enzymatic activity, indicating it elicits oxidative stress and apoptosis in common carp liver.

The development of human society entails increased industrial activity and consequently the release of a large number of chemical substances including solvents, detergents and pharmaceutical products which reach water bodies through the discharge of industrial effluents, damaging the organisms living in these ecosystems. This study aimed to determine oxidative stress induced on the common carp Cyprinus carpio by effluent from a pharmaceutical plant that manufactures nonsteroidal anti-inflammatory drugs. The median lethal concentration and subsequently the lowest observed adverse effect level were determined. Carp were exposed to the latter value (0.1173 %) for different exposure periods (12, 24, 48, 72 and 96 h), and the following biomarkers were evaluated in gill, brain, liver and blood: hydroperoxide content (HPC), lipid peroxidation (LPX), protein carbonyl content (PCC), and the activity of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). Statistically significant increases with respect to the control group (P < 0.05) were observed in HPC, LPX and PCC particularly in gill of effluent-exposed specimens. SOD, CAT and GPx activity in gill also increased with respect to the control group. This particular industrial effluent is therefore concluded to induce oxidative stress on C. carpio, this damage being most evident in gill. © 2013 Springer Science+Business Media Dordrecht.
