How can environmental conditions influence dicofol genotoxicity on the edible Asiatic clam, Meretrix meretrix?

Genotoxic effects of dicofol on the edible clam Meretrix meretrix were investigated through a mesocosm experiment. Individuals of M. meretrix, were exposed to environmental concentration (D1 = 50 ng/L) and supra-environmental concentration (D2 = 500 ng/L) of dicofol for 15 days, followed by the same depuration period. DNA damage (i.e., strand breaks and alkali-labile sites) was evaluated at day 1, 7 and 15, during uptake and depuration, using Comet assay (alkaline version) and nuclear abnormalities (NAs) as genotoxicity biomarkers. The protective effects of dicofol against DNA damage induced by ex vivo hydrogen peroxide (H₂O₂) exposure were also assessed. Comet assay results revealed no significant DNA damages under dicofol exposure, indicating 1) apparent lack of genotoxicity of dicofol to the tested conditions and/or 2) resistance of the animals due to optimal adaptation to stress conditions. Moreover, ex vivo H₂O₂ exposure showed an increase in the DNA damage in all the treatments without significant differences between them. However, considering only the DNA damage induced by H₂O₂ during uptake phase, D1 animals had significantly lower DNA damage than those from other treatments, revealing higher protection against a second stressor. NAs data showed a decrease in the % of cells with polymorphic, kidney shape, notched or lobbed nucleus, along the experiment. The combination of these results supports the idea that the clams used in the experiment were probably collected from a stressful environment (in this case Pearl River Delta region) which could have triggered some degree of adaptation to those environmental conditions, explaining the lack of DNA damages and highlighting the importance of organisms’ origin and the conditions that they were exposed during their lives.

Microplastics ingested by two bivalve species Perna viridis and Meretrix meretrix collected from three estuaries viz. Ariyankuppam, Panithittu, and Chunnambar in Pondicherry, India was analysed for the first time in this research. Nile Red dye was used for microplastic detection. A survey of 50 local families was conducted to determine the frequency and quantity in which they consume mussels/clams. On an average, the number of microplastics per gram of soft tissue (wet weight) is 0.18 ± 0.04, 1.84 ± 0.61, and 1.76 ± 0.48; and the number of microplastics per bivalve is 0.50 ± 0.11, 1.75 ± 0.35, and 4.80 ± 1.39 respectively for Ariyankuppam, Panithittu, and Chunnambar. 61.02% and 77.42% of the particles belonged to the size group of <100 μm in M. meretrix and P. viridis respectively. A moderate positive correlation of r (18) = 0.6985, p < 0.05 was calculated between bivalve weight and microplastic particles. An average person belonging to the local community is likely to ingest 3917.79 ± 144.71 microplastic particles per year through mussel consumption.

Plastic debris in the oceans is a major and growing problem in global environmental pollution. There are increasing concerns that plastic debris is a source of contaminant, either added during manufacturing or adsorbed from the environment. However, there is little information about the acute toxicity of leachates from plastic debris on marine organisms. In this study, we conducted experiments to evaluate the toxicity of leachates from two single-use polyethylene plastic bags (PB1 and PB2) with the embryo and larvae of the commercial clam Meretrix meretrix. Results showed that fertilization of the embryos was not affected by plastic leachates, but the developments of D-veliger larvae, including survival, deformity, and shell height, were significantly affected by plastic leachates from both PB1 and PB2 compared to the controls of filtered seawater. We speculate that compounds leaching from plastic bags are responsible for the observed toxicity. Therefore, leaching toxicity from plastic debris should be considered when assessing the risks of plastic pollution in the oceans.

Little knowledge is available on impact of microplastics (MPs) on the bivalve larvae at different developmental stages throughout their life history, especially for metamorphic stage. Therefore, this study aims to evaluate the toxic responses of carboxylated (PS-COOH) and amino (PS-NH2) polystyrene MPs on the developing clam larvae at three key life stages, i.e., fertilized eggs, D-veliger larvae, and umbo larvae. PS-COOH and PS-NH2 significantly decreased the hatching rates by 5.79–39.5% and developmental rates by 4.78–7.86% of the clam larvae relative to the unexposed clam larvae. The toxicity of MPs followed the order: hatching stage > metamorphosis > D-veliger larvae stage, showing stage-dependent toxic effects. Moreover, PS-NH2 with a smaller hydrodynamic diameter showed a greater toxicity to the developing larvae compared to PS-COOH. Our study highlighted the stage-dependent toxic effects of MPs on the developing clam larvae, thus posing ecological risks to population succession of marine bivalves and aquatic ecosystems.

Surface sediments and clam Meretrix meretrix were collected from a northern estuarine region in Dongying City, China. Sediments were analysed for heavy metals (Hg, As, Cd, Cr, Cu, Pb, and Zn) and the clams were tested for metallothioneins (MTs) and lysosomal membrane stability (LMS). The heavy metal total concentrations decreased in the order of Cr>Zn>Cu>Pb>As>Cd>Hg. The results of Bureau Communautaire de Référence (BCR) sequential extraction of heavy metals showed that the geochemical speciation of all heavy metals was dominated by residual fraction. According to the responses of biomarkers in M. meretrix, the modified potential ecological risk index (PERI-B) can more accurately reflect heavy metals pollution. PERI-B showed all sediment samples have low or moderate risk, except at site S10 (considerable risk), and the main contribution of ecological risk heavy metals were Cd and Hg.

The effects of artificial shell reef (ASR) on the benthic macroinvertebrates were studied in Shuangdao Bay, Yellow Sea, China. Results showed that the biomass of macroinvertebrates in the ASR increased with the age of the ASR. Based on self-organizing map (SOM), the macroinvertebrate community of short-term artificial reef (SAR), medium-term artificial reef (MAR) and long-term artificial reef (LAR) emerged as a cluster, which may indicate that the benthic community in the ASR formed after three years. The age of the ASR was the main factor affecting the benthic community. The macroinvertebrates belonged to six phyla, Platyhelminthes, Nemertea, Annelida, Mollusca, Arthropoda and Echinodermata, among which the latter four were the ones that contributed the most for abundance. The biomass of Mollusca increased dramatically with age. The dissimilarity of the species composition of Mollusca was mainly caused by Meretrix meretrix and Protothaca jedoensis. The two species accounted for 15.61%, 28.05% and 75.11% of the macroinvertebrate biomass found in SAR, MAR and LAR, respectively. The ASR might be served as a bivalve stock enhancement tool. We conclude that ASR could assemble macrobenthos effectively and increase the environmental quality of the adjacent area, being a valid option for marine habitat restoration purposes.

A quinquennial seasonal study (2015–2019) has been conducted to evaluate the bioaccumulation pattern of trace metals in Meretrix meretrix. The concentration of trace metals in the clam was observed as Cr > Cu > Ni > Zn > Pb > Cd > Hg, (Body> Mantle > Gills), similar to sediments. Contamination Factor of Cu and Cr in sediments showed strong association with the corresponding metal concentration in the body (r = 0.687, r = 0.962), mantle (r = 0.880, r = 0.956) and gills (r = 0.937, r = 0.863). Bioconcentration Factor was high for Cr followed by Ni. Mean Metal Concentration Rate (MMCR) of Cr was high and Hg was low (Body>Mantle>Gills). Our study establishes that the trace metal intake by Meretrix meretrix is associated with seasonal variation, physicochemical factors, sediment texture, chemical speciation and the metabolic stress created within the species induced from increased demand for protein synthesis. The latter resulted in the augmented rate of accumulation of Cu and Cr.

This study investigated accumulation of Escherichia coli and aerobic colony count in three types of shellfish species. The results indicated that the capability of accumulating E. coli and aerobic colony count for Sinonovacula constricta was stronger than that for Meretrix meretrix and Tegillarca granosa, and capability of accumulating E. coli for M. meretrix was slightly stronger than that for T. granosa. However, no significant difference was observed in the capability of accumulating aerobic colony count between M. meretrix and T. granosa. Moreover, accumulation of E. coli in S. constricta is affected by contaminated seawater and E. coli were accumulated much faster and more in S. constricta when the seawater contaminated more serious. Meanwhile, the results suggested that the populations of E. coli in S. constricta changed in accordance with the weather. This is the first study to investigate the differences of accumulating E. coli in three types of shellfish species.

The present study is an attempt to assess the heavy metal contamination in the marine environment of the Arabian Gulf of Saudi Arabia. The concentrations of heavy metals in water and the soft tissues of the bivalve species Meretrix meretrix Linnaeus, 1758, from different stations along the Arabian Gulf coastline, were determined during the summer season of 2008. Bioaccumulation of some heavy metals (Cd, Pb, Cu, and Zn) in fresh parts of the clam (M. meretrix) was measured by an atomic absorption spectrophotometer. The average concentrations of heavy metals in the clam tissues were 0.224-0.908, 0.294-2.496, 3.528-8.196, and 12.864-24.56 mg/kg wet weight for Cd, Pb, Cu, and Zn, respectively. In water, the mean concentration values of these metals were arranged in the following descending order: Pb > Cu > Zn > Cd. The heavy metal concentrations in tissues of M. meretrix were within the acceptable standards set by the US Environmental Protection Agency, the Commission Européenne, and the Food and Drug Administration of the USA. From the human public health point of view, these results seem to show no possibility of acute toxicities of Cd, Cu, Pb, and Zn if the edible clam is consumed. It is recommended that relevant authorities should carry out a continual assessment on the levels of these pollutants in the studied area.

The distribution of the toxic elements As, Cd, and Pb in nine different types of seafood from Shenzhen, China, was investigated by using inductively coupled plasma mass spectrometry (ICP-MS). The results revealed that the concentrations of arsenic (As) in fish (Lutjanus erythropterus, Paralichthys olivaceus) and in bivalve (Meretrix meretrix) and cadmium (Cd) in scallop (Argopecten irradians) exceed the limits established by food safety regulations in China and EU (European Union). Furthermore, the bioavailability of As, Cd, and lead (Pb) in mice after 20-day oral ingestion of Crassostrea rivularis was investigated, and the total rate of absorption of toxic elements in samples from the liver and kidney tissues and blood was determined. The results of this in vivo trial indicated that the oral bioavailability of As, Cd, and Pb was approximately 0.33, 0.45, and 0.74%, respectively.
