The detection of dioxin- and estrogen-like pollutants in marine and freshwater fishes cultivated in Pearl River Delta, China

In this study we aimed to assess the dioxin- and estrogen-like activities of contaminants extracted from twenty species of freshwater and seawater fishes, using luciferase reporter assays. Transfected MCF7 cells were treated with sample extracts and luciferase activities were then measured at 24-h of post-treatment. The mean values of the detected dioxin- and estrogen-like activities in the freshwater fishes were 25.3 pg TEQ/g ww and 102.3 pM EEQ/g ww whereas in the seawater fishes, the values were 46.2 pg TEQ/g ww and 118.8 pM EEQ/g ww. Using sample-relevant dosage of estrogen, inductions of cell proliferation markers (i.e. retinoblastoma, cyclin D) and stimulations of cell growth were revealed by Western blotting, colony formation and BrdU uptake assays. A cotreatment with TCDD significantly reduced these effects. Using the sample extracts with different dioxin- and estrogen-like activities, similar observation was revealed. The data highlighted the mixture effect of food contaminants on human health.

This paper reports on the partitioning behaviour of 15 perfluorinated compounds (PFCs), including C₄–C₁₀ sulfonates and C₅–C₁₄ carboxylic acids, between water, sediment and fish (European chub, Leuciscus cephalus) in the Orge River (nearby Paris). Total PFC levels were 73.0±3.0ngL⁻¹ in water and 8.4±0.5ngg⁻¹ in sediment. They were in the range 43.1–4997.2ngg⁻¹ in fish, in which PFC tissue distribution followed the order plasma>liver>gills>gonads>muscle. Sediment–water distribution coefficients (logKd) and bioaccumulation factors (logBAF) were in the range 0.8–4.3 and 0.9–6.7, respectively. Both distribution coefficients positively correlated with perfluoroalkyl chain length. Field-based biota–sediment accumulation factors (BSAFs) are also reported, for the first time for PFCs other than perfluorooctane sulfonate. logBSAF ranged between −1.3 and 1.5 and was negatively correlated with the perfluoroalkyl chain length in the case of carboxylic acids.

Muscle tissue was collected from ewes and lambs derived from farms throughout Scotland and sample concentrations of five endocrine disrupting compound groups were determined. Farms of origin were categorised according to geographic region. There were few statistically-significant differences with region or distance from cities. However, the magnitude of the difference between the highest and lowest mean values in ewe muscle from different regions exceeded 30% for 13 of the 15 compounds that were consistently detected in muscle, with animals derived from the industrialised region having the highest mean values for 11 of the 13 compounds. A less marked trend was apparent in the lamb muscle (8 of 13 highest were in the industrialised region). The physiological effects of such small differences in exposure to mixtures of pollutants remain to be determined.

The Subantarctic Kerguelen Islands (49°S, 70°E) contain freshwater ecosystems among the most isolated in the world. Concentrations of polychlorinated biphenyls (PCBs) were assessed in the muscle of 48 brook trout and 38 brown trout caught during summer and spring 2006 in the rivers, lakes and ponds of Kerguelen. The sum of 29 PCBs averaged 404 and 358ngg⁻¹ lipid, and dioxin-like PCB was 19 and 69ngg⁻¹ lipid, in brook and brown trout, respectively. The values showed a high variability and some fish accumulated PCBs at levels similar to those of fish from impacted areas. While inter-sex differences were limited, the season and the morphotype appeared to have the most influence. Fish captured in summer had muscle PCB concentrations about three times higher than those caught in spring and the ‘river’ morphotype of brook trout showed the highest PCB levels.

Cape monkfish (Lophius vomerinus) is one of the long-lived species and top predators in the northern Benguela region. Studies on bioaccumulation of mercury (Hg) in cape monkfish are limited. This study compared the total Hg concentration between monkfish muscle and liver tissue; and related the monkfish total Hg concentrations to fish body size and capture locations (depth and latitude). Monkfish specimens (n = 529) were collected from 2016 to 2018. The mean total Hg concentration was 0.126 ± 0.005 mg/kg in muscle tissues and 0.106 ± 0.005 mg/kg in liver tissues. No significant differences were observed between total Hg concentrations of muscles and liver tissues. Differences in Hg concentrations between monkfish length classes were observed. No significant correlation was found between total Hg concentrations and latitude. A significant increase of total Hg concentration with the depth was observed. The concentrations of Hg were below the World Health Organization (WHO limits for fish (0.5 mg/kg).

Muscle and liver tissues of nine fish species were analyzed to assess butyltin and phenyltin contamination. The samples were collected from three basins located in the Southern Baltic Sea coastal zone that each represent different potential for organotin pollution. Maximum total concentrations of butyltin compounds (BTs) in the fish muscles and livers were 715 and 1132ng Sn g−1 d.w., respectively, whereas triphenyltin (TPhT) was not detected. In the muscle samples, the predominant compound in the sum of butyltins was tributyltin (TBT), while in the liver samples, tributyltin degradation products were found in the majority. The results demonstrate that 6–7years after the implementation of the total ban on harmful organotin use in antifouling paints, butyltins remain present in fishes from the Polish coast of the Baltic Sea. According to the HELCOM recommendation, eight samples exceeded the good environmental status boundary for tributyltin in seafood.

This study is one of several baseline studies that will provide basic and reliable information about the content of undesirable substances in important species of fish caught in Norwegian waters. Concentrations of metals in the muscle and liver of more than 800 Northeast Arctic cod caught at 32 sites in the Barents Sea are reported. The highest concentration of both mercury in the muscle and cadmium in the liver was found in cod caught in the western part of the Barents Sea, while the highest concentration of total arsenic was found in cod from the eastern part. The arsenic concentrations varied greatly among individual fish, ranging from 0.3 to 170mgkg−1 wet weight in the muscle. Such high levels of total arsenic have never previously been reported in any fish, and the primary factor for these high concentrations is likely to be the shrimp in the cod diet.

Wild fish (Cyprinus carpio, Mugil cephalus, Platichthys flesus) from an estuary of the NE Atlantic coast were investigated for plastic contamination (N = 128). From the 1289 particles recovered from fish samples, 883 were plastics. Among these, 84% were fibres and 97% were microplastics. Thirty-six polymers were identified. The number of microplastics (mean ± SD) per individual fish (MP/fish) was 8 ± 6 in C. carpio, 10 ± 9 in M. cephalus and 2 ± 2 in P. flesus. The means of MP/fish per body site were 6 ± 7 in gastrointestinal tract, 0.5 ± 1.1 in gills, 0.3 ± 0.7 in liver and 0.6 ± 1.2 in muscle samples. A few large fibres in liver (≤ 4841 μm) and muscle (≤ 5810 μm) samples were found. The results evidence the existence of high fish contamination by microplastics and reinforce the need of further research on plastic pollution in estuaries.

Total mercury concentrations ([THg]) were determined in muscle and liver of the bat ray (Myliobatis californica), shovelnose guitarfish (Pseudobatos productus) and banded guitarfish (Zapteryx exasperata). Generalized linear models (GLM) were used to determine the effects of size and sex in [THg] and showed that both are determinants of [THg] in these species. The [THg] in both tissues significantly increased with length especially in sexually mature organisms with a steeper slope for mature male than mature female. This may relate to elasmobranchs sexual dimorphism driven variation in growth rates. Median muscle [THg] was significantly greater than liver in each ray species but there were some individuals with higher liver [THg] than muscle. There were individuals with muscle [THg] higher than the advisory thresholds of 0.2 and 0.5mgkg−1ww (2.4 and 11% of the bat ray; 2.1 and 10% of the shovelnose guitarfish; 12.6 and 45% of the banded guitarfish, respectively).

The occurrence of endocrine disrupting chemicals (EDCs) has been determined in two widely consumed fish species from Persian Gulf i.e., Epinephelus coioides and Platycephalus indicus by applying a validated analytical for the simultaneous detection of fourteen EDCs. The concentrations of all detected EDCs were greater in the liver than in the muscle (except for bisphenol A in P. indicus), suggesting a prolonged exposure of the fishes to these pollutants in the Persian Gulf. Specifically, the results showed that di (2-ethylhexyl) phthalate (DEHP) was the compound detected most frequently and at the highest concentration in both species. DEHP levels in ranged from 6.68 to 297.48 μg g-dw⁻¹ and from 13.32 to 350.52 μg g-dw⁻¹, in muscle and in liver, respectively. A risk assessment study was conducted, and demonstrated that consuming two fish based- meals per week may result in a moderate risk especially for vulnerable population groups.
