Localization of hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) and 2,4,6-trinitrotoluene (TNT) in poplar and switchgrass plants using phosphor imager autoradiography

Phosphor imager autoradiography is a technique for rapid, sensitive analysis of the localization of xenobiotics in plant tissues. Use of this technique is relatively new to research in the field of plant science, and the potential for enhancing visualization and understanding of plant uptake and transport of xenobiotics remains largely untapped. Phosphor imager autoradiography is used to investigate the uptake and translocation of the explosives 1,3,5-trinitro-1,3,5-triazine (RDX) and 2,4,6-trinitrotoluene within Populus deltoides × nigra DN34 (poplar) and Panicum vigratum Alamo (switchgrass). In both plant types, TNT and/or TNT-metabolites remain predominantly in root tissues while RDX and/or RDX-metabolites are readily translocated to leaf tissues. Phosphor imager autoradiography is further investigated for use in semi-quantitative analysis of uptake of TNT by switchgrass. Phosphor imager autoradiography allows for rapid localization and quantification of RDX, TNT, and/or metabolites in plant tissues.

Pools of Zn, Cu, Cd and Co in leaf, stem and root tissues of Sarcocornia fruticosa, Sarcocornia perennis, Halimione portulacoides and Spartina maritima were analyzed on a bimonthly basis, in a Tagus estuary salt marsh. All the major concentrations were found in the root tissues, being the concentrations in the aboveground organs neglectable for sediment budget proposes, as seen by the low root-aboveground translocation. Metal annual accumulation, root turnovers and cycling coefficients were also assessed. S. maritima showed the higher root turnovers and cycling coefficients for most of the analyzed metals, making this a phytostabilizer specie. By contrast the low root turnover, cycling coefficient and low root necromass generation makes S. perennis the most suitable specie for phytoremediation processes. Although the high amounts of metal return to the sediments, due to root senescence, salt marshes can still be considered sinks of heavy metals, cycling heavy metals mostly between sediment and root.

Two common sorrel (Rumex acetosa) accessions, one from a Zn–Pb contaminated site (CS accession) and the other from an uncontaminated site (UCS accession), were hydroponically exposed to a mixture of heavy metals (Pb2+ + Zn2+ + Cd2+) with and without EDTA at an equimolar rate. The metallicolous CS accession showed a higher tolerance to metal treatment in the absence of the chelating agent, whereas the UCS accession was especially tolerant to EDTA treatment alone. Combination of metal and EDTA treatment resulted in a higher Pb accumulation in shoots of both accessions although plants hardly showed phytotoxic symptoms. Cd and Zn uptake was not augmented by EDTA addition to the polymetallic medium. Chelant-assisted Pb accumulation was 70% higher in the CS accession than in the UCS accession, despite the fact that the former accession evapotranspired less water than the UCS accession. These results support the existence of a non-selective apoplastic transport of metal chelates by R. acetosa roots, not related to transpiration stream.

In this study, the ability of the organic fraction of municipal solid wastes (OFMSW) to enhance heavy metal uptake of maize shoots compared with ethylenediamine disuccinic acid (EDDS) was tested on soil contaminated with heavy metals. Soils treated with OFMSW and EDDS significantly increased the concentration of heavy metals in maize shoots (increments of 302%, 66%, 184%, 169%, and 23% for Cr, Cu, Ni, Zn, and Pb with respect to the control and increments of 933%, 482%, 928%, 428%, and 5551% for soils treated with OFMSW and EDDS, respectively). In soil treated with OFMSW, metal uptake was favored because of the high presence of dissolved organic matter (DOM) (41.6× than soil control) that exhibited ligand properties because of the high presence of carboxylic acids. Because of the toxic effect of EDDS on maize plants, soil treated with OFMSW achieved the highest extraction of total heavy metals.

The aim of this work was to investigate if engineered endophytes can improve phytoremediation of co-contaminations by organic pollutants and toxic metals. As a model system, yellow lupine was inoculated with the endophyte Burkholderia cepacia VM1468 possessing (a) the pTOM-Bu61 plasmid, coding for constitutive trichloroethylene (TCE) degradation, and (b) the ncc-nre Ni resistance/sequestration system. Plants were exposed to Ni and TCE and (a) Ni and TCE phytotoxicity, (b) TCE degradation and evapotranspiration, and (c) Ni concentrations in the roots and shoots were determined. Inoculation with B. cepacia VM1468 resulted in decreased Ni and TCE phytotoxicity, as measured by 30% increased root biomass and up to 50% decreased activities of enzymes involved in anti-oxidative defence in the roots. In addition, TCE evapotranspiration showed a decreasing trend and a 5 times higher Ni uptake was observed after inoculation.

In the present work, the relationships between plant consortia, consisting of 1-4 metallicolous pseudometallophytes with different metal-tolerance strategies (Thlaspi caerulescens: hyperaccumulator; Jasione montana: accumulator; Rumex acetosa: indicator; Festuca rubra: excluder), and their rhizosphere microbial communities were studied in a mine soil polluted with high levels of Cd, Pb and Zn. Physiological response and phytoremediation potential of the studied pseudometallophytes were also investigated. The studied metallicolous populations are tolerant to metal pollution and offer potential for the development of phytoextraction and phytostabilization technologies. T. caerulescens appears very tolerant to metal stress and most suitable for metal phytoextraction; the other three species enhance soil functionality. Soil microbial properties had a stronger effect on plant biomass rather than the other way around (35.2% versus 14.9%). An ecological understanding of how contaminants, ecosystem functions and biological communities interact in the long-term is needed for proper management of these fragile metalliferous ecosystems.

Incubation and pot experiments using poplar (Populus nigra L. cv. Wolterson) were performed in order to evaluate the questionable efficiency of EDDS-enhanced phytoextraction of Cu from contaminated soils. Despite the promising conditions of the experiment (low contamination of soils with a single metal with a high affinity for EDDS, metal tolerant poplar species capable of producing high biomass yields, root colonization by mycorrhizal fungi), the phytoextraction efficiency was not sufficient. The EDDS concentrations used in this study (3 and 6 mmol kg-1) enhanced the mobility (up to a 100-fold increase) and plant uptake of Cu (up to a 65-fold increase). However, despite EDDS degradation and the competition of Fe and Al for the chelant, Cu leaching cannot be omitted during the process. Due to the low efficiency, further research should be focused on other environment-friendly methods of soil remediation.

The effects of soil amendments on the phytoextraction of ∑DDT (DDT + DDD + DDE) from soil ([∑DDT] not, vert, similar 1500 ng/g) by a pumpkin variety of Cucurbita pepo ssp pepo were tested and the patterns of ∑DDT storage throughout the plant shoot were examined. The soil amendments did not increase the total amount of ∑DDT extracted into plant shoots, but new information about ∑DDT distribution in the plants was obtained. As observed previously, the ∑DDT concentration in plant leaves (mean 290 ng/g) was significantly lower than in plant stems (mean 2600 ng/g). Further analysis revealed that ∑DDT composition was consistent throughout the plant shoot and that ∑DDT concentration in leaves and stems decreased exponentially as distance from the root increased, which was previously unknown. This new information about the patterns of ∑DDT uptake and translocation within pumpkin plants highlights the need for appropriate plant sampling strategies in future POPs phytoextraction research. Patterns of ∑DDT storage in a pumpkin plant are elucidated and specific surfactant and mycorrhizal soil amendments did not increase the total amount of ∑DDT phytoextracted into plant shoots.

Arsenic (As) contamination of the environment has emerged as a concerning issue recently for which phytoremediation has been suggested as a viable solution. Hydrilla verticillata (L.f.) Royle is a widely distributed rapidly growing aquatic weed possessing significant potential to accumulate As and is thus a potential candidate for the purpose of As phytoremediation. In the present study, an investigation of thiol metabolism was conducted in H. verticillata, which revealed differential effects upon exposure to arsenite [As(III)] and arsenate [As(V)]. The accumulation of arsenic was found to be higher upon exposure to As(III) than to As(V). Besides, As(III) was found to induce the activities of enzymes, such as cysteine synthase and γ-glutamylcysteine synthetase and the amounts of cysteine and glutathione (GSH) to higher levels than that observed with As(V). The activity of glutathione-S-transferase was, however, stimulated to a higher level upon exposure to As(V) than As(III). The activity of arsenate reductase was found to increase upon As(V) exposure at all concentrations and durations. In addition, a significant stimulation in the activity of phytochelatin synthase was noticed in vitro with an increase in As/GSH concentration and time of incubation. Arsenic detoxification in H. verticillata thus appeared to involve an induction of thiol synthesis and consumption in a coordinated manner, though differentially upon exposure to As(III) and As(V). The information gained through this study would help in better designing of the pilot experiment at the field level depending on the chemical composition of the contaminated water.

Two species of sunflower, i.e., Tithonia diversifolia and Helianthus annuus, were investigated for their potential to remove heavy metals from contaminated soils. Dried and mature T. diversifolia (Mexican flower) seeds were collected along roadsides, while H. annuus (sunflower) seeds were sourced from the Department of PBST, University of Agriculture Abeokuta, Nigeria. The contaminants were added as lead nitrate (Pb (NO3)2) and zinc nitrate (Zn (NO3)2) at 400 mg/kg which represents upper critical soil concentration for both Pb and Zn. The results indicated that T. diversifolia mopped up substantial concentrations of Pb in the above-ground biomass compared to concentrations in the roots. The concentrations in the leaf compartment were 87.3, 71.3, and 71.5 mg/kg at 4, 6, and 8 weeks after planting (AP), respectively. In roots, it was 99.4 mg/kg, 97.4 mg/g, and 77.7 mg/kg while 79.3, 77.8, and 60.7 mg/kg were observed in the stems at 4, 6, and 8 weeks AP, respectively. Observations with H. annuus followed the pattern found with T. diversifolia, showing significant (p < 0.05) accumulation of Pb in the above-ground biomass. Results obtained from Zn contaminated soils showed significant (p < 0.05) accumulation in the above-ground compartments of T. diversifolia and H. annuus compared with root. However, the highest accumulation of Zn was observed in the leaf. The translocation factor and enrichment coefficient of Pb and Zn with these plant species are greater than 1, indicating that these metals moved more easily in these plants. However, this result also showed that the translocation of Zn from root to the shoot of the two plants was higher than Pb. In conclusion, this experiment showed that these plants accumulated substantial Pb and Zn in their shoots (leaf and stem) at 4 weeks AP which diminished with time. This implies that the efficiency of these plants in cleaning the contaminated soils was at the early stage of their growth.
