Polycyclic aromatic hydrocarbon (PAH) extracts of fine particles (PM₂.₅) collected from combustion of seven wood species and briquettes were tested for mutagenic activities using Ames test with Salmonella typhimurium TA98 and TA100. The woods were Pinus pinaster (maritime pine), Eucalyptus globulus (eucalypt), Quercus suber (cork oak), Acacia longifolia (golden wattle), Quercus faginea (Portuguese oak), Olea europea (olive), and Quercus ilex rotundifolia (Holm oak). Burning experiments were done using woodstove and fireplace, hot start and cold start conditions. A mutagenic response was recorded for all species except golden wattle, maritime pine, and briquettes. The mutagenic extracts were not correlated with high emission factors of carcinogenic PAHs. These extracts were obtained both from two burning appliances and start-up conditions. However, fireplace seemed to favour the occurrence of mutagenic emissions. The negative result recorded for golden wattle was interesting, in an ecological point of view, since after confirmation, this invasive species, can be recommended for domestic use.

We studied the effectiveness of remediation on microbial endpoints, namely microbial biomass and activity, microbial and plant species richness, of an As-contaminated mine spoil, amended with compost (C) alone and in combination with beringite (B) or zerovalent iron grit (Z), to increase organic matter content and reduce trace elements mobility, and to allow Holcus lanatus and Pinus pinaster growth. Untreated spoil showed the lowest microbial biomass and activity and hydrolase activities, and H. lanatus as sole plant species, whereas the presented aided phytostabilisation option, especially CBZ treatment, significantly increased microbial biomass and activity and allowed colonisation by several plant species, comparable to those of an uncontaminated sandy soil. Microbial species richness was only increased in spoils amended with C alone. No clear correlation occurred between trace element mobility and microbial parameters and plant species richness. Our results indicate that the choice of indicators of soil remediation practices is a bottleneck. Organo-mineral amendment and revegetation of a gold mine spoil increased microbial activity but did not increase microbial species richness.

Due to relatively high chelant dosages and potential environmental risks it is necessary to explore different approaches in the remediation of metal-contaminated soils. The present study focussed on the removal of metals (As, Cd, Cu, Pb and Zn) from a multiple metal-contaminated soil by growing Brassica carinata plants in succession to spontaneous metallicolous populations of Pinus pinaster, Plantago lanceolata and Silene paradoxa. The results showed that the growth of the metallicolous populations increased the extractable metal levels in the soil, which resulted in a higher accumulation of metals in the above-ground parts of B. carinata. Root exudates of the three metallicolous species were analysed to elucidate their possible role in the enhanced metal availability. The presence of metals stimulated the exudation of organic and phenolic acids as well as flavonoids. It was suggested that root exudates played an important role in solubilising metals in soil and in favouring their uptake by roots.

The effects of municipal sewage sludge applied on topsoil and understory vegetation (Molinia caerulea (L.) Moench) were studied in a maritime pine forest located in the South West of France (Landes of Gascogne). Understory response to sludge application is important as sludge addition to forest could increase competition with pine and affect herbivorous wildlife through incorporation of heavy metals in the food chain. The experiment was conducted in a young stand of maritime pines. The experimental design consisted of three 0.1-ha plots. One plot received composted sludge, one plot received liquid sludge, and one control plot received no sludge. Liquid sludge and composted sludge were applied on the basis of 3Â tons dry matter sludge per hectare and per year. After 2Â years of sludge application, we observed the following: (1) a significant increase in total concentrations of the following major and trace elements in the topsoil (layer 0–20Â cm) [organic carbon (+140%), nitrogen (+140%), and lead (+80%)] and (2) no significant accumulation of trace elements in M. caerulea except nickel, which increased moderately (+40%) following application of composted sludge. These initial results need to be completed (1) by the assessment of long-term effects and dynamics of trace elements with additional applications of sludge and (2) by analyzing secondary understory species to determine if understory response to sludge application is more dependant on species than on soil parameters and sludge type.

The purpose of the present study was to evaluate the effect of planting trees (Pinus pinaster or Eucalyptus globulus) and amending with wastes (sewage sludges and paper mill residues) on the nutrient content of mine soils and under field conditions. The studied soils were located in a settling pond and a mine tailing within a former copper mine. The soil samples were analyzed for several physico-chemical characteristics and the concentration of nutrients. The untreated settling pond soil had levels of N and K adequate only for the growth of eucalyptuses and pines, and moreover, the concentration of Ca and P were undetectable. The untreated mine tailing soil presented the same condition, also with adequate levels of Ca and Mg for eucalyptuses and pines. Planting these trees increased the concentration of Mg in the settling pond up to adequate levels only for such trees. Amending with wastes increased the phytoavailable concentration of all nutrients up to adequate levels for most plant species. In conclusion, it is recommended to amend mine soils with wastes rich in nutrients and re-amend after some time because they raise them up to adequate levels for most plants but are depleted over time. It is possible to increase the concentration of all nutrients in mine soils by adding organic wastes, even to values adequate for most plant species.

São Domingos belongs among the most important historic Iberian Pyrite Belt Cu mines. The anthrosoil is contaminated by a very high content of heavy metals and metalloids. The study was focused on evaluating the interaction of some chemical elements (Ca, Mg, Fe, Mn, Cu, Pb, Zn, Ag, Cd, Ni, Co, As, Sb) in the system soil vs. five autochthonous dominant plant species: Pinus pinaster Aiton, Quercus rotundifolia Lam., Agrostis sp., Juncus conglomeratus L. and Juncus effusus L. The plants are heavily contaminated by Cu, Pb, As and Zn. The bioconcentration factor proved that they exhibit features of metal tolerant excluders. The trees are accumulators of Ag, whereas the graminoids are hyper-accumulators of Ag and Juncus effusus of Co. The translocation factor confirmed that the selected elements are immobilised in the roots except for Mn and Zn in Pinus pinaster and Mn in Quercus rotundifolia and Juncus conglomeratus. The bioaccumulation of Mn, Zn and Cu at low pH increases. The increased content of Ca and Mg in the soil inhibits, in the case of some metals and metalloids, their intake to plants. Although the studied plants, despite their fitness and vitality at the contaminated sites, are not suitable for phytoextraction (except Co and Ag), they can be used for phytostabilisation at the mining habitats.

Domestic biofuel combustion is one of the major sources of regional and local air pollution, mainly regarding particulate matter and organic compounds, during winter periods. Mutagenic and carcinogenic activity potentials of the ambient particulate matter have been associated with the fraction of polycyclic aromatic hydrocarbons (PAH) and their oxygenated (OPAH) and nitrogenated (NPAH) derivatives. This study aimed at assessing the mutagenicity potential of the fraction of this polycyclic aromatic compound in particles (PM₁₀) from domestic combustion by using the Ames assays with Salmonella typhimurium TA98 and TA100. Seven biofuels, including four types of pellets and three agro-fuels (olive pit, almond shell and shell of pine nuts), were tested in an automatic pellet stove, and two types of wood (Pinus pinaster, maritime pine, and Eucalyptus globulus, eucalypt) were burned in a traditional wood stove. For this latter appliance, two combustion phases—devolatilisation and flaming/smouldering—were characterised separately. A direct-acting mutagenic effect for the devolatilisation phase of pine combustion and for both phases of eucalypt combustion was found. Almond shell revealed a weak direct-acting mutagenic effect, while one type of pellets, made of recycled wastes, and pine (devolatilisation) presented a cytotoxic effect towards strain TA100. Compared to the manually fired appliance, the automatic pellet stove promoted lower polyaromatic mutagenic emissions. For this device, only two of the studied biofuels presented a weak mutagenic or cytotoxic potential.

Diflubenzuron (DFB) is an insecticide commonly used to control forest pests. The objectives of this study were to assess the effect of diflubenzuron on the development of Pinus pinaster seedlings and Pisolithus tinctorius under laboratory conditions and to study the possible protective role of this ectomycorrhizal fungus against the effects of diflubenzuron. In vitro experiments revealed that diflubenzuron inhibited fungal growth at all tested concentrations (0.01, 0.1, 1, 10 and 100 mg L⁻¹). Root growth was inhibited at the two highest diflubenzuron concentrations. The activity of the antioxidant defence system of non-inoculated P. pinaster increased at 1 and 10 mg DFB kg⁻¹ substrate, and inoculation increased the threshold to the highest concentration. The protective role of the ectomycorrhizal fungus was seen in the increase of CAT activity. This study revealed that despite causing no mortality, diflubenzuron has the ability to cause sub-lethal damage to P. pinaster. The disproportionate use of this insecticide may lead to higher amounts of its residues in soil and the biosphere, endangering trees, fungi and their symbiosis.

Exposure to chemical pollution can cause significant damage to plants by imposing conditions of oxidative stress. Plants combat oxidative stress by inducing antioxidant metabolites, enzymatic scavengers of activated oxygen and heat shock proteins. The accumulation of these proteins, in particular heat shock protein 70 and heme oxygenase, is correlated with the acquisition of thermal and chemical adaptations and protection against oxidative stress. In this study, we used Pinus pinaster Ait. collected in the areas of Priolo and Aci Castello representing sites with elevated pollution and reference conditions, respectively. The presence of heavy metals and the levels of markers of oxidative stress (lipid hydroperoxide levels, thiol groups, superoxide dismutase activity and expression of heat shock protein 70, heme oxygenase and superoxide dismutase) were evaluated, and we measured in field-collected needles the response to environmental pollution. P. pinaster Ait. collected from a site characterized by industrial pollution including heavy metals had elevated stress response as indicated by significantly elevated lipid hydroperoxide levels and decreased thiol groups. In particular, we observed that following a chronic chemical exposure, P. pinaster Ait. showed significantly increased expression of heat shock protein 70, heme oxygenase and superoxide dismutase. This increased expression may have protective effects against oxidative stress and represents an adaptative cellular defence mechanism. These results suggest that evaluation of heme oxygenase, heat shock protein 70 and superoxide dismutase expression in P. pinaster Ait. could represent a useful tool for monitoring environmental contamination of a region and to better understand mechanisms involved in plant defence and stress tolerance.
