Metal toxicity to lettuce Lactuca sativa was determined following mixture exposure based on the concepts of concentration addition (CA) and response addition (RA). On the basis of conventional models assuming no interaction between mixture components, Ag+ was the most toxic, followed by Cu2+ and Zn2+. Furthermore, ion–ion interactions were included in quantitatively estimating toxicity of interactive mixtures of Cu2+–Zn2+ and Cu2+–Ag+ by linearly expanding the CA and RA models. About 80–92% of the variability in the root growth could be explained by this approach. Estimates by the extended models indicate significant alleviative effects of Zn2+ on Cu2+ toxicity whereas Cu2+ did not significantly affect Zn2+ toxicity. According to the extended CA model, Cu2+ significantly reduced Ag+ toxicity while Ag+ enhanced Cu2+ toxicity. Similar effects were not found by the extended RA model. These interactions might result from their individual uptake mechanisms and toxic actions as published in literature.

In order to evaluate Cd tolerance in wide-ranging sources of alfalfa (Medicago sativa) and to identify Cd tolerant genotypes which may potentially be useful for restoring Cd-contaminated environments, thirty-six accessions of alfalfa were screened under hydroponic culture. Our results showed that the relative root growth rate varied from 0.48 to 1.0, which indicated that different alfalfa accessions had various responses to Cd stress. The candidate fragments derived from differentially expressed metallothionein (MT) genes were cloned from leaves of two Cd tolerant genotypes, YE and LZ. DNA sequence and the deduced protein sequence showed that MsMT2a and MsMT2b had high similarity to those in leguminous plants. DDRT-PCR analysis showed that MsMT2a expressed in both YE and LZ plants under control and Cd stress treatment, but MsMT2b only expressed under Cd stress treatment. This suggested that MsMT2a was universally expressed in leaves of alfalfa but expression of MsMT2b was Cadmium (Cd) inducible.

Gene expression responses of paper birch (Betula papyrifera) leaves to elevated concentrations of CO2 and O3 were studied with microarray analyses from three time points during the summer of 2004 at Aspen FACE. Microarray data were analyzed with clustering techniques, self-organizing maps, K-means clustering and Sammon's mappings, to detect similar gene expression patterns within sampling times and treatments. Most of the alterations in gene expression were caused by O3, alone or in combination with CO2. O3 induced defensive reactions to oxidative stress and earlier leaf senescence, seen as decreased expression of photosynthesis- and carbon fixation-related genes, and increased expression of senescence-associated genes. The effects of elevated CO2 reflected surplus of carbon that was directed to synthesis of secondary compounds. The combined CO2 + O3 treatment resulted in differential gene expression than with individual gas treatments or in changes similar to O3 treatment, indicating that CO2 cannot totally alleviate the harmful effects of O3. Clustering analysis of birch leaf gene expression data reveals differential responses to O3 and CO2.

Tropospheric ozone causes severe oxidative stress in plants. To investigate the transcriptional responsiveness of adult trees to ozone, fully-expanded sun and shade leaves of mature beech trees were harvested at four time points over the entire vegetation period in 2005 and 2006. Microarray analyses were conducted on leaves from trees grown in the field under ambient and twice-ambient ozone concentrations at Kranzberger Forst (Bavaria). Beech trees changed their transcript levels in response to ozone. In the years 2005 and 2006 different transcription patterns were observed; this may have been a result of different weather conditions and ozone uptake. Furthermore, we obtained differences in mRNA expression patterns between shade and sun leaves. In the ozone-treated sun leaves of 2005, slightly up- and down-regulated transcript levels were detected, particularly in the spring and autumn, whereas shade leaves clearly exhibited reduced mRNA levels, particularly at the end of the vegetation period. In 2006, this pattern could not be confirmed, and in the autumn, four other transcripts were slightly up-regulated in ozone-treated shade leaves. In addition, two other transcripts were found to be influenced in sun leaves in the spring/summer. While we detected changes in the levels of only a few transcripts, the observed effects were not identical in both years. In conclusion, elevated ozone exhibited very small influence on the transcription levels of genes of mature beech trees. At the transcriptional level, leaves of mature beech trees barely react to double ambient ozone concentrations; differences are detected primarily between sun/shade leaves and between different growing seasons.

Plants react to O3 threat by setting up a variety of defensive strategies involving the co-ordinated modulation of stress perception, signalling and metabolic responses. Although stomata largely controls O3 uptake, differences in O3 tolerance cannot always be ascribed to changes in stomatal conductance but cell protective and repair processes should be taken into account. O3-driven ROS production in the apoplast induces a secondary, active, self-propagating generation of ROS, whose levels must be finely tuned, by many enzymatic and non-enzymatic antioxidant systems, to induce gene activation without determining uncontrolled cell death. Additional signalling molecules, as ethylene, jasmonic and salicylic acid are also crucial to determine the spreading and the containment of leaf lesions. The main recent results obtained on O3 sensing, signal transduction, ROS formation and detoxification mechanisms are here discussed. A dissection of the complex network of interacting mechanisms which determine the cell fate under ozone stress.

In urbanized regions with expansive impervious surfaces and often low vegetation cover, air pollution due to motor vehicles and other combustion sources, is a problem. The poor air quality days in Montreal, Quebec are mainly due to fine particulate matter and ozone. Businesses using wood ovens are a source of particulates. Careful vegetation selection and increased green roof usage can improve air quality. This paper reviews different green roofs and the capability of plants in particulate matter (PM), ozone (O3) as well as nitrogen dioxide (NO2) level reductions. Both the recommended green roof category and plants to reduce these pollutants in Montreal's zone 5 hardiness region are provided. Green roofs with larger vegetation including shrubs and trees, or intensive green roofs, remove air pollutants to a greater extent and are advisable to implement on existing, retrofitted or new buildings. PM is most effectively captured by pines. The small Pinus strobus ‘Nana’, Pinus mugho var. pumilio, Pinus mugho ‘Slowmound’ and Pinus pumila ‘Dwarf Blue’ are good candidates for intensive green roofs. Drought tolerant, deciduous broadleaved trees with low biogenic volatile organic compound emissions including Japanese Maple or Acer palmatum ‘Shaina’ and ‘Mikawa-Yatsubusa’ are options to reduce O3 levels. Magnolias are tolerant to NO2 and it is important in their metabolic pathways. The small cold-tolerant Magnolia ‘Genie’ is a good option to remove NO2 in urban settings and to indirectly reduce O3 formation. Given the emissions by Montreal businesses' wood ovens, calculations performed based on their respective complex roof areas obtained via Google Earth Pro indicates 88% Pinus mugho var. pumilio roof coverage can annually remove 92.37 kg of PM10 of which 35.10 kg is PM2.5. The removal rates are 4.00 g/m2 and 1.52 g/m2 for PM10 and PM2.5, respectively. This paper provides insight to addressing air pollution through urban rooftop greening.

Current risk assessment models for metals such as the biotic ligand model (BLM) are usually applied to individual metals, yet toxic metals are rarely found singly in the environment. In the present research, the toxicity of Cu and Zn alone and together were studied in wheat (Triticum aestivum L.) using different Ca2+ and Mg2+ concentrations, pH levels and Zn:Cu concentration ratios. The aim of the study was to better understand the toxicity effects of these two metals using BLMs and toxic units (TUs) from single and combined metal toxicity data. The results of single-metal toxicity tests showed that toxicity of Cu and Zn tended to decrease with increasing Ca2+ or Mg2+ concentrations, and that the effects of pH on Cu and Zn toxicity were related not only to free Cu2+ and Zn2+ activity, respectively, but also to other inorganic metal complex species. For the metal mixture, Cu–Zn interactions based on free ion activities were primarily additive for the different Ca2+ and Mg2+ concentrations and levels of pH. The toxicity data of individual metals derived by the BLM, which incorporated Ca2+ and Mg2+ competition and toxicity of inorganic metal complexes in a single-metal toxicity assessment, could predict the combined toxicity as a function of TU. There was good performance between the predicted and observed effects (root mean square error [RMSE] = 7.15, R2 = 0.97) compared to that using a TU method with a model based on free ion activity (RMSE = 14.29, R2 = 0.86). The overall findings indicated that bioavailability models that include those biochemistry processes may accurately predict the toxicity of metal mixtures.

The effects of elevated concentrations of atmospheric tropospheric ozone (O3) on DNA damage in five trembling aspen (Populus tremuloides Michx.) clones growing in a free-air enrichment experiment in the presence and absence of elevated concentrations of carbon dioxide (CO2) were examined. Growing season mean hourly O3 concentrations were 36.3 and 47.3 ppb for ambient and elevated O3 plots, respectively. The 4th highest daily maximum 8-h ambient and elevated O3 concentrations were 79 and 89 ppb, respectively. Elevated CO2 averaged 524 ppm (+150 ppm) over the growing season. Exposure to O3 and CO2 in combination with O3 increased DNA damage levels above background as measured by the comet assay. Ozone-tolerant clones 271 and 8L showed the highest levels of DNA damage under elevated O3 compared with ambient air; whereas less tolerant clone 216 and sensitive clones 42E and 259 had comparably lower levels of DNA damage with no significant differences between elevated O3 and ambient air. Clone 8L was demonstrated to have the highest level of excision DNA repair. In addition, clone 271 had the highest level of oxidative damage as measured by lipid peroxidation. The results suggest that variation in cellular responses to DNA damage between aspen clones may contribute to O3 tolerance or sensitivity. Ozone tolerant clones and sensitive Populus tremuloides clones show differences in DNA damage and repair.

Background, aim, and scope In papermaking, there is a continuous interest both to minimize fresh water consumption and to reduce discharges into the environment. These general trends mean an increase in the amounts of detrimental substances, such as resin and fatty acids, in papermaking process waters. Resin acids, in particular, are responsible for much of the toxicity typically present in paper mill effluents and, for this reason, the routine and rapid monitoring of these compounds in various process streams is necessary. This also means that there is a continuous need to develop straightforward offline and online techniques to clarify problems occurring, for example, as a result of the introduction of more intensively closed systems of water circulation. In the present study, we describe the use of a novel, online, sample enrichment technique followed by atmospheric pressure chemical ionization-mass spectrometry (APCI-MS) suitable for monitoring the concentration levels of common resin and fatty acids in papermaking process waters. Materials and methods The representative process water samples were taken from the grinding zone of a thermomechanical pulping mill. The samples were first preconcentrated in a precolumn C18, and the analytes were transferred online to MS. The high intensive [M-H]⁻ ion was used for the identification of each analyte since, according to the present ionization method, no other fragmentation was observed. Laboratory-scale, online measurements with an online sample feed were carried out by connecting a centrifugal pump and a ceramic filter to the APCI-MS. Results Quality parameters, such as repeatability, linearity, and limit of detection (LOD), were determined by using dehydroabietic acid (DHAA) in order to evaluate the suitability of the method for the rapid screening of concentration levels. This method provided satisfactory linearity and a good correlation between analyte concentration and peak area. The suitability of the system for the continuous analysis of the same acids was evaluated in laboratory-scale, online experiments. In all cases, the response to changes in the analyte concentration was linear, and the repeatability of the system was also satisfactory. Discussion Only a few studies have been published on the analysis of resin and fatty acids with MS techniques. The present method was applied to the monitoring of dehydroabietic, oleic, and stearic acids. The quality parameters were highly comparable with those reported earlier, and the LOD values of the DHAA were below the levels usually encountered in process waters. The quality parameters were only slightly higher than those obtained by the traditional methods of analysis, probably due to the absence of an effective sample clean-up before analysis. Conclusions The results of the laboratory-scale, online experiments indicated that the online enrichment APCI-MS system is a suitable alternative for monitoring the concentration levels of selected resin and fatty acids in papermaking process waters. The method can be used, for example, to provide useful information about the concentration levels of these acids in different stages of the process, thus signaling possibly impending problems. In general, faster and simpler measurements are needed to meet the requirements for a reduction in fresh water usage in papermaking. Recommendations and perspectives Compared to the conventional methods used for this purpose, the main benefits of the method are rapidity of measurement, simplicity of use, and absence of the need for multistage sample treatments (short analysis time). For this reason, this online method is more suitable for the control of papermaking by analyzing the concentration levels of interfering substances (i.e., selected resin and fatty acids) than an offline analysis detailing all the individual extractives-based compounds in process streams. It is also obvious that the technique can easily be modified for other environmental pollutants as well.

The aim of this study was to address the lack of information concerning the air quality in the city of Uberlândia, Minas Gerais, Brazil. In this study, we conducted an unprecedented experiment involving the in situ biomonitoring of air genotoxicity in the city center during and after a public transportation strike using the Tradescantia micronucleus test. The frequency of micronuclei was significantly higher in the city center compared with the reference site (Mann–Whitney test, p < 0.05), with the highest MN levels being observed during public transport stoppage (Kruskal–Wallis, Dunn p < 0.01). In addition, the multiple linear regression analyses revealed that the low circulation of buses during public transport stoppage and the increase in the concentration of particulate matter from the increased flow of vehicles in the city center during the strike positively influenced the MN frequency. The climatic factors did not change during the biomonitoring period, reflecting the fact that climatic factors did not influence the MN frequency.
