Possible overestimation of bisphenol analogues in municipal wastewater analyzed with GC-MS

This work for the first time showed that sulfated BPA could be directly analyzed as BPA with GC-MS after the derivatization with N,O-bis-(trimethylsilyl) trifluoroacetamide (BSTFA)+1% trimethylchlorosilane (TMCS), i.e. the deconjugation step was not necessary. This was because sulfated BPA indeed could be simultaneously deconjugated and derivatized to BPA derivative during derivatization, suggesting that any co-elution of BPA and sulfated BPA during sample extraction led to BPA overestimation in the GC-MS method with BSTFA +1% TMCS as the derivative reagent. Using BPA 4,4′-disulfates (BPA diS) as the pure standard, the co-elution phenomena of sulfated BPA was confirmed with two widely used elution solvents (i.e. methanol and ethyl acetate) or their mixed solutions with different ratios, which further suggested if only sulfated BPA existed in any wastewater sample, BPA was likely over-determined. To further confirm this finding, both influent and effluent samples collected from a local municipal wastewater treatment plant were analyzed, which clearly showed the overestimation of BPA in the two wastewaters due to co-existence of sulfated BPA in the wastewater samples. In addition to BPA, the results also showed the overestimation of other nine bisphenol analogues. As sulfated micropollutants including estrogens, androgens, phytoestrogens, etc., have been widely found in municipal wastewater, the overestimating phenomenon observed in this study may also be extended to determination of other micropollutants, which should be addressed in future.

Genistein is a phytoestrogen, which is structurally similar to 17β-estradiol. It is present in plants, food, and as a contaminant in effluents. In this article, we demonstrate the effects of embryonic exposure to three different concentrations of genistein (10 μg/L, 40 μg/L, and 80 μg/L) which is similar to those found in effluents. Zebrafish eggs were exposed during the first 72 h post-fertilization (hpf). Heart rate was evaluated at 48 hpf and mortality rate was assessed during the first 72 hpf. The light/dark (LDT) and open field (OFT) behavioral tests were applied to the larvae (6 dpf), and the novel tank (NTT), social preference (SPT), light-dark (LDT), and sexing tests were performed on adult fish (90 dpf). Embryonic exposure to genistein caused anxiolytic-like behavior in both larvae and adult animals. In adult stage, we observed an increase in locomotor activity and antisocial behavior in the concentration of 40 μg/L. There was an increase in the mortality rate in all concentrations when compared to the control and an increase in heart rate at the concentration of 80 μg/L. Exposure to 10 μg/L generated a higher frequency of females when compared to the control group. Our results show that exposure to genistein during the embryonic phase brings damage in the short and long term as it increases the mortality rate and leads to behavioral disorders both in the larval stage, with perpetuation until adult stage. The anxiolytic-like effect and less social interaction are effects that harm fish survival.

Phytoestrogens are naturally plant-derived compounds that could bind to estrogen receptors and mimic estrogenic effects. Previous studies showed a positive association between phytoestrogens and hypothyroidism; however, little is known on phytoestrogens and thyroid hormones. This study was designed to investigate the associations between urinary phytoestrogens and thyroid hormone levels. Based on the US National Health and Nutrition Examination Survey (NHANES) 2007–2010, 4103 participants were recruited in this cross-sectional study. Linear regression models and multiple linear regressions models were applied to examine the relationships between urinary phytoestrogens and thyroid hormone levels. Urinary O-desmethylangolensin (O-DMA) was found to be correlated with serum FT₄ levels in the female 20–60-year-of-age group (β=0.018, 95% CI: 0.006, 0.031). Higher enterolactone (ENT) levels were significantly positively associated with TSH levels in the 12–19-year-of-age female group (β=0.196, 95% CI: 0.081, 0.311). In the male group, enterodiol (END) was significantly positively correlated with TSH and TT₃ in the 12–19-year-of-age group, respectively (TT₃: β=3.444, 95% CI: 0.150, 6.737; TSH: β=0.104, 95% CI: 0.005, 0.203). However, equol (EQU) levels were negatively associated with TT₄ (12–19-year-of-age: β=− 0.166, 95% CI: − 0.279, − 0.034; 20–60-year-of-age: β=− 0.132, 95% CI: − 0.230, − 0.034). Our study provided epidemiological evidence that urinary phytoestrogens were powerfully associated with thyroid hormone levels. The results also supported that phytoestrogens acted as endocrine disruptors. It is imperative and important to pay attention to the intake of phytoestrogens.

Phytoestrogens have been widely praised for their health-promoting effects, whereas synthetic environmental estrogens are considered a toxicological risk to human health. The aim of this study was therefore to compare in vitro the estrogenic, cytotoxic, and genotoxic profiles of three common estrogen-like endocrine-disrupting chemicals: the phytoestrogens 8-prenylnaringenine (8-PN) and genistein and the synthetic xenoestrogen tartrazine. As assessed by a yeast bioreporter assay and estrogen-dependent proliferative response in human mammary gland adenocarcinoma cell line (MCF-7), 8-PN showed the highest estrogen-like activity of the three compounds, followed by tartrazine and genistein. After 24-h incubation on MCF-7 cells, all three compounds exhibited low cytotoxicity in the lactate dehydrogenase assay and no genotoxicity in the micronucleus assay. These results demonstrate that 8-PN, genistein and tartrazine possess variable estrogenic activity but display little cellular toxicity in short-term tests in vitro. No difference between phytoestrogens and a synthetic xenoestrogen could be established.

A pan-European monitoring campaign of the wastewater treatment plant (WWTP) effluents was conducted to obtain a concise picture on a broad range of pollutants including estrogenic compounds. Snapshot samples from 75 WWTP effluents were collected and analysed for concentrations of 150 polar organic and 20 inorganic compounds as well as estrogenicity using the MVLN reporter gene assay. The effect-based assessment determined estrogenicity in 27 of 75 samples tested with the concentrations ranging from 0.53 to 17.9 ng/L of 17-beta-estradiol equivalents (EEQ). Approximately one third of municipal WWTP effluents contained EEQ greater than 0.5 ng/L EEQ, which confirmed the importance of cities as the major contamination source. Beside municipal WWTPs, some treated industrial wastewaters also exhibited detectable EEQ, indicating the importance to investigate phytoestrogens released from plant processing factories. No steroid estrogens were detected in any of the samples by instrumental methods above their limits of quantification of 10 ng/L, and none of the other analysed classes of chemicals showed correlation with detected EEQs. The study demonstrates the need of effect-based monitoring to assess certain classes of contaminants such as estrogens, which are known to occur at low concentrations being of serious toxicological concern for aquatic biota.

Everyone needs emotional support at some point in life, but the needs might not always be met. The present study was aimed to investigate the prevalence of unmet needs of emotional support in adults and to identify social, environmental and health attributes in a national and population-based setting in recent years. Data was retrieved from the US National Health and Nutrition Examination Surveys, 2005–2008, including demographics, blood pressure readings, self-reported emotional support needs in the last 12 months, self-reported ever health conditions and urinary environmental chemical concentrations. Statistical analyses included chi-square test, t test, survey-weighted logistic regression modeling and population attributable risk (PAR) estimation. Of 6733 American adults aged 40–80, 1273 (21.0 %) needed more emotional support in the past year. They tended to be aged 40–60, female, Mexican American, other Hispanic, education less than high school, or poverty income ratio 5+. People with higher levels of butyl paraben, ethyl paraben, methyl paraben, 1-hydroxynaphthalene, 2-hydroxynaphthalene, or 9-hydroxyfluorene (but not heavy metals, arsenic, phenols, phthalates, pesticides, or phytoestrogens) or historical diabetes, asthma, arthritis, stroke, thyroid disorder, chronic bronchitis, sleep complaint/disorder, or trouble seeing needed more emotional support. Significant risk associations from environmental chemicals mentioned above have remained after adjusting for historical health conditions as potential mediators. This is the first time examining prevalence of the unmet emotional support in adults and identifying the social, environmental and health attributes. Removal of parabens and polyaromatic hydrocarbons and increasing healthcare for people with health conditions to accommodate emotional support should be considered.
