Influence of different types of nanomaterials on their bioaccumulation in a paddy microcosm: A comparison of TiO2 nanoparticles and nanotubes

We investigated the environmental fate and bioaccumulation of TiO2 nanomaterials in a simplified paddy microcosm over a period of 17 days. Two types of TiO2 nanomaterials, nanoparticles (TiO2-NP) and nanotubes (TiO2-NT), were synthesized to have a negative surface charge. Ti concentrations in the environmental media (water, soil), crops (quillworts, water dropworts), and some lower and higher trophic organisms (biofilms, algae, plant-parasitic nematodes, white butterfly larva, mud snail, ricefish) were quantified after exposure periods of 0, 7, and 17 days. The titanium levels of the two nanomaterials were the highest in biofilms during the exposure periods. Bioaccumulation factors indicated that TiO2-NP and TiO2-NT were largely transferred from a prey (e.g., biofilm, water dropwort) to its consumer (e.g., nematodes, mud snail). Considering the potential entries of such TiO2 nanomaterials in organisms, their bioaccumulation throughout the food chain should be regarded with great concern in terms of the overall health of the ecosystem.

Pine wood nematode, Bursaphelenchus xylophilus, is a plant parasitic nematode which causes severe damage to several Pinus species. Two natural compounds, dipropyl trisulfide (DPTS) and methyl propyl trisulfide (MPTS), showed strong nematicidal activity against the pine wood nematode, presenting 4.24 and 17.81 μg/mL LC₅₀ values, respectively. However, hydrophobicity and low stability have limited their practical use in the field as nematicides. To overcome these problems, chitosan-coated nanoemulsions of DPTS and MPTS were developed. The optimum chitosan concentration for the delivery system of the two sulfides was 0.5%. Optimized chitosan-coated nanoemulsions of sulfides have a uniform size distribution (mean diameter = 203.7 and 207.7 nm, mean polydispersity index = 0.176 and 0.178) with sufficient colloidal stability (mean zeta potential = +40 and +45 mV). The LC₅₀ values of DPTS and MPTS nanoemulsions coated with 0.5% chitosan against the pine wood nematode were 5.01 and 16.60 μg/mL, respectively. In addition, chitosan coating improved the long-term storage stability and persistence of nematicidal activity of the nanoemulsions. This study indicates that the chitosan-coated nanoemulsion is a suitable formulation for sulfides as novel nematicides against the pine wood nematode for field application.

Nanoparticulate ZnO is one of the most commonly applied nanomaterials. As ZnO is more soluble than many other oxide nanoparticles, its toxicity beyond the nanoparticle-specific effects can be attributed to the dissolved ionic zinc. The investigation of uptake and toxicity of nano-ZnO in the plant-feeding nematode, Xiphinema vuittenezi, which was used in previous studies as a biological model organism, was aimed. The establishment of the role of dissolved zinc and nanoparticle-specific effects in the toxicity was also the objective of our study. Zn uptake was found to be significantly higher for bulk and nano-ZnO than for ZnSO₄ solution; however, treatments caused loss of potassium in the worms in a dissolved-zinc-dependent manner. The toxicity was the lowest for bulk ZnO, and it was very similar for nano-ZnO and ZnSO₄ solution. Accordingly, the toxicity of ZnO nanoparticles is a combination of dissolved-zinc-caused toxicity and nanoparticle-specific effects.
