In this study, the effectiveness of bioremediating 1,2-dichloroethane (DCA)-contaminated groundwater under different oxidation–reduction processes was evaluated. Microcosms were constructed using indigenous bacteria and activated sludge as the inocula and cane molasses and a slow polycolloid-releasing substrate (SPRS) as the primary substrates. Complete DCA removal was obtained within 30 days under aerobic and reductive dechlorinating conditions. In anaerobic microcosms with sludge and substrate addition, chloroethane, vinyl chloride, and ethene were produced. The microbial communities and DCA-degrading bacteria in microcosms were characterized by 16S rRNA-based denatured-gradient-gel electrophoresis profiling and nucleotide sequence analyses. Real-time polymerase chain reaction was applied to evaluate the variations in Dehalococcoides spp. and Desulfitobacterium spp. Increase in Desulfitobacterium spp. indicates that the growth of Desulfitobacterium might be induced by DCA. Results indicate that DCA could be used as the primary substrate under aerobic conditions. The increased ethene concentrations imply that dihaloelimination was the dominate mechanism for DCA biodegradation.

Although elevated ground-level O3 has a species–specific impact on plant growth, the differences in soil biota responses to O3 pollution among rice cultivars are rarely reported. Using O3 Free-Air Concentration Enrichment, the responses of the rhizospheric bacterial communities in the O3-tolerant (YD6) and the O3-sensitive (IIY084) rice cultivars to O3 pollution and their differences were assessed by pyrosequencing at rice tillering and anthesis stages. Elevated ground-level O3 negatively influenced the bacterial community in cultivar YD6 at both rice growth stages by decreasing the bacterial phylogenetic diversities and response ratios. In contrast, in cultivar IIY084, the bacterial community responded positively at the rice tillering stage under O3 pollution. However, several keystone bacterial guilds were consistently negatively affected by O3 pollution in two rice cultivars. These findings indicate that continuously O3 pollution would negatively influence rice agroecosystem and the crop cultivar is important in determining the soil biota responses to elevated O3.

This study was performed to evaluate the abundance and diversity of Enterococcus sp. and the distribution of biomarker genes in Enterococcus faecalis in Port Blair Bay, Andaman and Nicobar Islands. The Enterococcus sp. densities at the seven sampling stations were highly influenced by tidal fluctuations and season. The distributions and diversities of species varied in the inner and outer regions of Port Blair Bay. Among the 1816 total isolates, the occurrence of fecal Enterococcus was high (1.78×104CFU/100mL) in Phoenix Bay. Moreover, 67.76% of the isolates were identified as Enterococcus, and the most frequently identified species were E. hirae, E. avium and E. faecalis. Assessments of antibiotic resistance and biomarker genes revealed the maximum occurrence in the Aberdeen Bay isolates. The most prevalent biomarker genes observed in the E. faecalis isolates were gelE and asa1, whereas cyl was not found among the isolates. In silico sequence analysis of biomarker genes of E. faecalis also revealed that they are evolutionarily well conserved with those of earlier reports. Further, multivariate analysis distinguished the JB, PB and OS stations from the other stations according to distinctive microbial densities and compositions. In addition, the Shannon-Wiener diversity indices and box-whisker plots further facilitated and supported the multivariate results.

The main aim of the study was to evaluate the role of scallop hatcheries as source of the floR and cmlA genes. A number of 133 and 121 florfenicol-resistant strains were isolated from scallop larval cultures prior to their transfer to seawater and from effluent samples from 2 commercial hatcheries and identified by 16S rRNA gene sequence analysis, observing a predominance of the Pseudomonas, Pseudoalteromonas and Halomonas genera and exhibiting an important incidence of co-resistance to streptomycin, oxytetracycline and co-trimoxazole. A high percentage of strains from both hatcheries carried the floR gene (68.4% and 89.3% of strains), whereas a lower carriage of the cmlA gene was detected (27.1% and 54.5% of strains). The high prevalence of floR-carrying bacteria in reared scallop larvae and hatchery effluents contributes to enrich the marine resistome in marine environments, prompting the need of a continuous surveillance of these genes in the mariculture environments.

The Alang–Sosiya shipbreaking yard (ASSBY) is considered the largest of its kind in the world, and a major source of anthropogenic pollutants. The aim of this study was to investigate the impact of shipbreaking activities on the bacterial community structure with a combination of culture-dependent and culture-independent approaches. In the culture-dependent approach, 200 bacterial cultures were isolated and analyzed by molecular fingerprinting and 16S ribosomal RNA (r-RNA) gene sequencing, as well as being studied for degradation of polycyclic aromatic hydrocarbons (PAHs). In the culture-independent approach, operational taxonomic units (OTUs) were related to eight major phyla, of which Betaproteobacteria (especially Acidovorax) was predominantly found in the polluted sediments of ASSBY and Gammaproteobacteria in the pristine sediment sample. The statistical approaches showed a significant difference in the bacterial community structure between the pristine and polluted sediments. To the best of our knowledge, this is the first study investigating the effect of shipbreaking activity on the bacterial community structure of the coastal sediment at ASSBY.

The present study aims to optimize the slow release biostimulant ball (BSB) for bioremediation of contaminated coastal sediment using response surface methodology (RSM). Different bacterial communities were evaluated using a pyrosequencing-based approach in contaminated coastal sediments. The effects of BSB size (1–5cm), distance (1–10cm) and time (1–4months) on changes in chemical oxygen demand (COD) and volatile solid (VS) reduction were determined. Maximum reductions of COD and VS, 89.7% and 78.8%, respectively, were observed at a 3cm ball size, 5.5cm distance and 4months; these values are the optimum conditions for effective treatment of contaminated coastal sediment. Most of the variance in COD and VS (0.9291 and 0.9369, respectively) was explained in our chosen models. BSB is a promising method for COD and VS reduction and enhancement of SRB diversity.

Using enrichment culture technique, three isolates marked as ODB-1, ODB-2 and ODB-3, were selected from oil contaminated seawater. 16S rDNA gene sequencing indicated that ODB-1 affiliated with Pseudomonas sp. while ODB-2 and ODB-3 affiliated with Brevundimonas sp. Subsequently, the bacterial cells were immobilized on the surface of expanded graphite (EG), expanded perlite (EP) and bamboo charcoal (BC). Among the three isolates, ODB-1 showed a strong binding to the bio-carriers through extracellular polysaccharides, while ODB-2 and ODB-3 made the adhesion to bio-carrier through direct physical adsorption. The immobilized bacteria exhibited good salinity tolerance compared with the planktonic bacteria. Their total diesel oil removal rates were more than 85% after 6 days’ incubation. Adsorption–biodegradation process played an important role in the oil-pollution remediation. EG-bacteria system was treated as a promising remediation method, which achieved nearly 100% removal of diesel oil. Thereinto, over 83% removal of diesel oil owed to biodegradation.

Intertidal benthic ecosystems in estuaries are productive sites where microbial processes play critical roles in nutrients mineralization, primary production and trophic web. In this groundwork study we analyzed the bacterial community of intertidal biofilms from Río de la Plata beaches with different anthropogenic impacts. Several environmental parameters were measured and bacterial assemblages were analyzed by 16S-rDNA pyrosequencing. The average OTU found per sample was 527.3±122.5, showing similar richness and diversity among them. However, sites having the highest and lowest salinity displayed higher bacterial diversity. Assemblages from a site nearby an oil refinery, showing the lowest salinity and oxygen concentration, were clearly distinct from the rest. The weight of this splitting relied on OTUs belonging to Thauera, known by its ability to metabolize aromatic compounds. Our results suggest that intertidal bacterial assemblages would be structured by major estuarine variables such as salinity, and that anthropogenic-induced environmental parameters might also be relevant.

High density polyethylene (HDPE) is the most commonly found non-degradable solid waste among the polyethylene. In this present study, HDPE degrading various fungal strains were isolated from the polyethylene waste dumped marine coastal area and screened under in vitro condition. Based on weight loss and FT-IR Spectrophotometric analysis, two fungal strains designated as VRKPT1 and VRKPT2 were found to be efficient in HDPE degradation. Through the sequence analysis of ITS region homology, the isolated fungi were identified as Aspergillus tubingensis VRKPT1 and Aspergillus flavus VRKPT2. The biofilm formation observed under epifluorescent microscope had shown the viability of fungal strains even after one month of incubation. The biodegradation of HDPE film nature was further investigated through SEM analysis. HDPE poses severe environmental threats and hence the ability of fungal isolates was proved to utilize virgin polyethylene as the carbon source without any pre-treatment and pro-oxidant additives.

Biofiltration has been widely used to reduce organic matter and control the formation of disinfection by-products in drinking water. Backwashing might affect the biofilters’ performance and the attached microbiota on filter medium. In this study, the impacts of backwashing on the removal of dissolved organic carbon (DOC), dissolved organic nitrogen (DON) and N-nitrosamine precursors by a pilot-scale biological activated carbon (BAC) filtration system were investigated. The impacts of backwashing on biomass and microbial community structure of BAC biofilm were also investigated. Phospholipid fatty acid (PLFA) analysis showed that backwashing reduced nearly half of the attached biomass on granular activated carbon (GAC) particles, followed by a recovery to the pre-backwashing biomass concentration in 2 days after backwashing. Backwashing was found to transitionally improve the removal of DOC, DON and N-nitrosamine precursors. MiSeq sequencing analysis revealed that backwashing had a strong impact on the bacterial diversity and community structure of BAC biofilm, but they could gradually recover with the operating time after backwashing. Phylum Proteobacteria was the largest bacterial group in BAC biofilm. Microorganisms from genera Bradyrhizobium, Hyphomicrobium, Microcystis and Sphingobium might contribute to the effective removal of nitrogenous organic compounds by drinking water biofilter. This work could add some new insights towards the operation of drinking water biofilters and the biological removal of organic matter.
