Detection of sea polluters with change in conditioned reflexive responses of fish

This work is based on biotests being experimented at sea organisms - fish Serranus scriba. We were studying the activity of the mentioned fish in sublethal concentrations of organochloric pesticide - lindane. Besides the classic biotests, we used some new methods for neurotoxic testing that include the influence of this pesticide on circadian activity of fish as well as the variations in obtaining the positive conditioned reflexive responses. The results of these experiments show that this kind of fish reacts at the subtile changes of toxic concentration with the change of quality and rhythm in its conditioned reflexive reactions, that appeared earlier than other global signs of intoxication.

In this work influence of different concentrations of linear active substance (LAS-DBS) on activity of Serranus scriba CUV have been studied. Desynchronization of circadian rhythms is in direct correlation with LAS growing concentrations. The high disproportion was found between maximum permitted concentrations (3 mg/l) and our results which show the changes in fish behaviour at 0.2 mg/l LAS.

The experiment was performed on marine Teleost Serranus scriba with lower concentrations of lindane then MPC (due to our valid law). The aim of this study was to find the effects of lindane to fish activities connected with nutrition. This motive is among the strongest. It includes parameters as: number of fish movements in the presence of prey or without it and the number of onslaughts to the prey during one hour. The results have shown that the increasing of lindane concentration caused considerable decreasing and even complete cessation of activity. Interest for prey and decreasing activity of predators in the presence of prey (typical for Serranus - as a fish of prey and the hunter) endures till the moment of convulsion.

A total of 1345 specimens belonging to 58 different species of wild fish and seafood from the western Mediterranean Sea were analyzed to assess total mercury levels and to estimate which species meet the EU recommendations for human consumption (0.5 μg g⁻¹ ww) in all cases. All fish species were caught off the Mediterranean coasts and intended for human consumption. All specimens were collected from local markets located in Spain, Italy and France that sell fish caught by local fishermen (Eivissa, Menorca, Mallorca, Alacant, L'Ampolla, Ametlla de Mar, Marseille, Genoa, Civitavecchia, Alghero) at different time periods. Mercury concentrations were measured by thermal decomposition-gold amalgamator-atomic absorption spectrometry. Only thirteen species were found that did not exceed 0.5 μg g⁻¹ ww in any specimen analyzed. These safe species were sardines (Sardina pilchardus), anchovies (Engraulis encrasicolus), blue whiting (Micromesistius poutassou), picarel (Spicara smaris), blackspot seabream (Pagellus bogaraveo), gilthead seabream (Sparus aurata), pearly razorfish (Xyrichtys novacula), surmullet (Mullus surmuletus), painted comber (Serranus scriba), brown meagre (Sciaena umbra), salema (Sarpa salpa), common dolphinfish (Coryphaena hippurus) and squid (Loligo vulgaris). These species occupy different trophic levels, have different lengths and average weights, but show a low mercury concentration than others living in the same environments. Potential human consumption of these species as sole source of fish would imply estimated weekly intakes representing between 49% and 70% of the recommended provisional tolerable weekly intake of methylmercury in the worst case. Health authorities should pay specific attention to species that do not meet EU thresholds and make appropriate precautionary health recommendations, especially for pregnant women and children.

There is limited research on the ingestion of microplastic particles (MPs) by fish from the southern part of the Mediterranean Sea. This study provides the occurrence of small MPs (≤3 μm) in the gastrointestinal tract and muscle of adult benthopelagic fish Serranus scriba (L.1758), caught along Tunisian coasts. MPs were extracted from selected tissues using a potassium hydroxide digestion method (KOH 10%) and then quantified, and their chemical structure was characterized through Raman microspectroscopy. The results highlighted that MPs were present in all samples. The average abundance of MPs per gram of fish tissue identified through successive filters of 3 μm, 1.2 μm, and 0.45 μm differed significantly among the sites. The properties of the MPs extracted indicated that polyethylene-vinyl-acetate (PEVA: 33.45%), high density polyethylene (HD-PE: 17.33%), and fragments were the most abundant plastic types and shape found, respectively. Among those, most MPs were found at a size class of 3–1.2 μm (∼60%), especially in the muscle, suggesting a high transfer of MPs into the human diet. Our field work also aimed to explore the effects observed in the gastrointestinal tract with a battery of biomarkers assessing oxidative stress and neurotoxicity. The preliminary results of this study showed the existence of a link between small MPs, sites, and their associated urban activities and induced oxidative stress. However, more detailed studies are required to evaluate the transfer of MPs into tissues and the potential impacts of this transfer on human health.
