Characterization of phthalate exposure in relation to serum thyroid and growth hormones, and estimated daily intake levels in children exposed to phthalate-tainted products: A longitudinal cohort study

No information is available on the long-term effects on thyroid and growth hormones of children exposed to phthalate-tainted products, despite the infamous 2011 Taiwan phthalate episode. We investigated estimated daily intake levels and their long-term effects on serum thyroid and growth hormone levels in children.We recruited 166 children (2–18 years old) in three visits who provided specimens and filled out a questionnaire from the Risk Assessment of Phthalate Incident in Taiwan (RAPIT) project study from 2012 to 2016. Morning spot urine samples were analyzed for nine phthalate metabolites. Serum thyroid (triiodothyronine [T₃], thyroxine [T₄], and free T₄) and growth hormone (insulin-like growth factor-1 [IGF-1] and its binding protein 3 [IGF-BP3]) levels were measured. A generalized estimating equation model was used to evaluate associations between phthalate metabolite levels and children’s thyroid and growth hormone levels.The median metabolite levels of monomethyl phthalate (MMP), Σdibutyl phthalate (DBP), and Σdi-(2-ethylhexyl) phthalate (DEHP) at visits 1, 2, and 3 were 6.59, 10.5, and 21.0 ng/mL, 0.15, 0.24, and 0.20 nmol/mL, and 0.15, 0.17, and 0.12 nmol/mL, respectively. After adjusting for potential confounding factors, we found that levels of urinary MMP were negatively associated with T₃ (β = −0.013, p = 0.047), T₄ (β = −0.016, p = 0.006), free T₄ (β = −0.012, p = 0.002), and IGF-BP3 (β = −0.025, p = 0.003). Urinary mono-ethyl phthalate (MEP) was negatively associated with IGF-1 (β = −0.027, p = 0.029) and IGF-BP3 (β = −0.016, p = 0.018). In addition, serum free T₄ was positively associated with urinary mono-2-ethyl-5-hydroxy hexyl phthalate (MEHHP) (β = 0.016, p = 0.043), mono-2-ethyl-5-oxohexyl phthalate (MEOHP) (β = 0.015, p = 0.024), and ΣDEHPm (β = 0.019, p = 0.020).Our findings support the hypothesis that specific phthalates disturb the hemostasis of thyroid and growth hormone levels in children exposed to phthalate-tainted products.

Bisphenol A (BPA) is widely used by manufacturers and in consumer products. Its release in the environment may affect male reproductive function. In this study, we examined the effect of low dose (0.1 mg/kg BW), short term exposure during puberty (PD21-35) on adult rat male reproduction. The results indicated that such exposure reset growth hormone (GH) and follicular stimulating hormone (FSH) homeostasis and resulted in a significantly higher level of serum testosterone without affecting serum luteinizing hormone level. QPCR and Western blot results showed that BPA significantly up-regulated selective genes/proteins in the Leydig cell steroidogenic pathway, including steroidogenic acute regulatory protein, cytochrome P450 11A1, cytochrome P450 17A, and low-density lipoprotein receptor. RNA-Seq analysis of testicular RNAs showed that BPA significantly affected the gene profiles of multiple testicular interstitial populations without affecting germ cells. Also, GO- and KEGG-analysis suggested that IGF1-related PI3K/AKT signaling was activated, which was confirmed by the increased phosphorylation of IRS1, AKT1 and CREB. The results indicated that a low-dose, short-term BPA exposure during puberty affected the adult male rat pituitary (GH and FSH) and testis (testosterone) homeostasis.

Brominated disinfection byproducts (Br-DBPs) are formed during the disinfection process of water containing bromine ions, such as marine aquaculture water. Little attention has been paid to Br-DBPs with anthropogenic chemicals as precursors. This study summarized the sodium hypochlorite (NaClO) oxidation of three frequently used pharmaceuticals, including two antibiotics, norfloxacin (NOR) and sulfamethoxazole (SMX), and the growth hormone estrogen 17β-estradiol (E2). Transformations of the pharmaceuticals were found to be faster in marine aquaculture water than in distilled water. Several Br-DBPs and Cl-DBPs were identified for NOR, SMX, and E2. It was shown that the carboxyl group, piperazine ring, C3, and C8 atoms were the primary reaction sites on NOR. The aniline moiety and S-N bond were identified to be the reaction sites on SMX. The C2, C4, C9, and C16 atoms were the potential reaction centers on E2. Preliminary calculation by QSAR model indicated that the value of logKow significantly increased with an increase in the number of bromine atoms in the Br-DBPs. The results of the bioconcentration factors (BCF) analysis suggested that the bioaccumulation of Br-DBPs were greater than that chlorinated DBPs (Cl-DBPs) in distilled water.

2,4,6-Trichlorophenol (2,4,6-TCP) is a common waste among the resulting chlorophenols generated in the production of common products classified as an extremely toxic, mutagenic, carcinogenic and highly persistent xenobiotic in the environment. To evaluate the impact of 2,4,6-TCP in aquatic systems, the catfish species Clarias batrachus has been selected to test its toxicity due to its high market value and consumption in India. Here is presented the impact of this compound on different physiological parameters of fish: haematological parameters (haemoglobin, total erythrocyte count, total leucocyte count and mean corpuscular haemoglobin), biochemical parameters (total serum protein and total serum glucose), growth and reproductive parameters (condition factor, hepatosomatic index, maturity index, specific growth rate, growth hormone, 17β-estradiol and testosterone), exposed to two concentrations of 2,4,6-TCP (0.5 mg/L and 1 mg/L — 1/10th and 1/20th of the LC₅₀) for a period of 15, 30 and 45 days. The results showed that C. batrachus even when exposed to the lower concentration (0.5 mg/L) for the shortest time (15 days) negatively impacted the organism in all the assessed parameters. This was highlighted by the Integrated Biomarker Response index (IBR), showing worse scores for the treatments (up to 20 × worse than the control). This work highlights the importance of continued research on the impact of 2,4,6-TCP, on an important commercial, supported by the high environmental persistence of this compound that can reach the same range of tested concentrations.
