Combined influences of transgenerational effects, temperature and insecticide on the moth Spodoptera littoralis

Climate warming is expected to impact the response of species to insecticides. Recent studies show that this interaction between insecticides and temperature can depend on other factors. Here, we tested for the influence of transgenerational effects on the Insecticide × Temperature interaction in the crop pest moth Spodoptera littoralis. Specifically, we analysed reaction norms among experimental clutches based on a split-plot design crossing the factors temperature, insecticide and clutch. The study was performed on 2280 larvae reared at four temperatures (23, 25, 27 and 29 °C), and their response to the insecticide deltamethrin (three concentrations and a control group) was tested. Temperature had a global influence with effects on larval survival, duration of development, pupal body mass, and significant reaction norms of the clutches for temperature variations of only 2 °C. In addition to the expected effect of deltamethrin on mortality, the insecticide slightly delayed the development of S. littoralis, and the effects on mortality and development differed among the clutches. Projection models integrating all the observed responses illustrated the additive effects of deltamethrin and temperature on the population multiplication rate. Variation in the response of the clutches showed that transgenerational effects influenced the impact of insecticide and temperature. Although no evidence indicated that the Insecticide × Temperature interaction depended on transgenerational effects, the studies on the dependence of the Insecticide × Temperature interaction on other factors continue to be crucial to confidently predict the combined effects of insecticides and climate warming.

The effects of insecticides at low residual doses on both the targeted crop pest species and beneficial insects have become a major concern. In particular, these low doses can induce unexpected positive (hormetic) effects on pest insects, such as surges in population growth exceeding what would have been observed without pesticide application. Methomyl and chlorpyrifos are two insecticides commonly used to control the population levels of the cotton leafworm Spodoptera littoralis, a major pest moth. The aim of the present study was to examine the effects of sublethal doses of these two pesticides, known to present a residual activity and persistence in the environment, on the moth physiology. Using a metabolomic approach, we showed that sublethal doses of methomyl and chlorpyrifos have a systemic effect on the treated insects. We also demonstrated a behavioral disruption of S. littoralis larvae exposed to sublethal doses of methomyl, whereas no effects were observed for the same doses of chlorpyrifos. Interestingly, we highlighted that sublethal doses of both pesticides did not induce a change in acetylcholinesterase activity in head of exposed larvae

Medicinal and aromatic plants represent an outstanding source of green active ingredients for a broad range of real-world applications. In the present study, we investigated the insecticidal potential of the essential oils obtained from three medicinal and aromatic plants of economic importance in Algeria, Artemisia campestris, Pulicaria arabica, and Saccocalyx satureioides. Gas chromatography coupled with mass spectrometry (GC-MS) was used to study the essential oil chemical compositions. The three essential oils were tested against a mosquito vectoring filariasis and arboviruses, i.e., Culex quinquefasciatus, a fly pest acting also as pathogens vector, Musca domestica, and an agricultural moth pest, i.e., Spodoptera littoralis, using WHO and topical application methods, respectively. The essential oil from A. campestris, containing β-pinene (15.2%), α-pinene (11.2%), myrcene (10.3%), germacrene D (9.0%) (Z)-β-ocimene (8.1%) and γ-curcumene (6.4%), showed remarkable toxicity against C. quinquefasciatus (LC₅₀ of 45.8 mg L⁻¹) and moderate effects (LD₅₀ of 99.8 μg adult⁻¹) against M. domestica. Those from P. arabica and S. satureioides, containing epi-α-cadinol (23.9%), δ-cadinene (21.1%), α-cadinol (19.8%) and germacrene D-4-ol (8.4%), and thymol (25.6%), α-terpineol (24.6%), borneol (17.4%) and p-cymene (11.4%), respectively, were more active on S. littoralis showing LD₅₀ values of 68.9 and 61.2 μg larva⁻¹, respectively. Based on our results, the essential oil from A. campestris may be further considered a candidate ingredient for developing botanical larvicides.

Echinophora spinosa (Apiaceae) is a psammophilous species growing along Mediterranean coastal sand dunes. This species secretes essential oils made up of monoterpenoids, phenylpropanoids, and polyacetylenes, which may represent effective, eco-friendly and safe active ingredients for the preparation of green insecticides. Based on this, in the present study, we selected a French accession of E. spinosa growing on the coastal dunes of Corsica, extracting the essential oils from aerial parts and roots by hydrodistillation. The essential oils were analyzed by GC-MS for chemical composition and screened for their insecticidal activity on three target insects, Culex quinquefasciatus, Spodoptera littoralis, and Musca domestica. The essential oil from roots was dominated by the phenylpropanoid myristicin and the monoterpene hydrocarbon terpinolene, with a minor contribution of (Z)-falcarinol. The oil from the aerial parts was characterized by monoterpenes, mainly p-cymene, a-phellandrene, and α-pinene. The root essential oil exhibited promising toxicity on the target insects, with special reference to C. quinquefasciatus larvae (LC₅₀ = 15.7 mg L⁻¹), while on M. domestica and S. littoralis, it showed LD₅₀ of 38.3 μg adult⁻¹ and 55.6 μg larva⁻¹, respectively. Based on our findings, E. spinosa can be viewed as a new potential crop to produce insecticidal essential oils in arid salty areas. However, synergistic and antagonistic effects of the major constituents of both oils tested here deserve future research. Further studies on the efficacy of stable formulations of the E. spinosa root essential oil, with special reference to micro- and nanoformulations, are ongoing.

Effects of low concentrations of pesticides, with no or moderate mortality of targeted species, are poorly studied even though these low concentrations are common under natural conditions. Studying their effects is critical because they can induce positive hormetic responses, possibly leading to greater pest multiplication and promoting the evolution of pest resistance. Here, we investigated the responses of the pest moth Spodoptera littoralis to low concentrations of deltamethrin, and tested for variation in effects of the pesticide between developmental stages and sexes. Indeed, we show that a given concentration of deltamethrin has different effects between stages, and even between sexes. Two experimental concentrations led to very high mortality early in S. littoralis development (4th larval instar), but only to low mortality rates in adults. Moreover, our highest experimental concentration had only detrimental effects in adult females, but improved the reproductive success of adult males. Model projections showed that the lethality from treatments at the 4th larval instar was the predominant effect. Because of the high multiplication rate of S. littoralis, it was also found that treatments with very similar effects on larval mortality can lead to either population extinction or rapid pest resurgence.

In the attempt to exploit the potential of the monoecious fiber hemp cv. Futura 75 in new fields besides textile, cosmetics and food industry, its crop-residue given by leaves and inflorescences was subjected to hydrodistillation to obtain the essential oils. These are niche products representing an ideal candidate for the development of natural insecticides for the control and management of mosquito vectors, houseflies and moth pests. After GC-MS analysis highlighting a safe and legal chemical profile (THC in the range 0.004–0.012% dw), the leaf and inflorescence essential oils were investigated for the insecticidal potential against three insect targets: the larvae of Culex quinquefasciatus and Spodoptera littoralis and the adults of Musca domestica. The essential oil from inflorescences, showing (E)-caryophyllene (21.4%), myrcene (11.3%), cannabidiol (CBD, 11.1%), α-pinene (7.8%), terpinolene (7.6%), and α-humulene (7.1%) as the main components, was more effective than leaf oil against these insects, with LD₅₀ values of 65.8 μg/larva on S. littoralis, 122.1 μg/adult on M. domestica, and LC₅₀ of 124.5 μl/l on C. quinquefasciatus larvae. The hemp essential oil moderately inhibited the acetylcholinesterase (AChE), which is a target enzyme in pesticide science. Overall, these results shed light on the future application of fiber hemp crop-residue for the development of effective, eco-friendly and sustainable insecticides.

Pesticides have long been used as the main solution to limit agricultural pests, but their widespread use resulted in chronic or diffuse environmental pollutions, development of insect resistances, and biodiversity reduction. The effects of low residual doses of these chemical products on organisms that affect both targeted species (crop pests) but also beneficial insects became a major concern, particularly because low doses of pesticides can induce unexpected positive—also called hormetic—effects on insects, leading to surges in pest population growth at greater rate than what would have been observed without pesticide application. The present study aimed to examine the effects of sublethal doses of deltamethrin, one of the most used synthetic pyrethroids, known to present a residual activity and persistence in the environment, on the peripheral olfactory system and sexual behavior of a major pest insect, the cotton leafworm Spodoptera littoralis. We highlighted here a hormetic effect of sublethal dose of deltamethrin on the male responses to sex pheromone, without any modification of their response to host-plant odorants. We also identified several antennal actors potentially involved in this hormetic effect and in the antennal detoxification or antennal stress response of/to deltamethrin exposure.
