Organophosphorus pesticides exert estrogen receptor agonistic effect determined using Organization for Economic Cooperation and Development PBTG455, and induce estrogen receptor-dependent adipogenesis of 3T3-L1 adipocytes

Various chemicals containing pesticides can induce adipogenesis and cause obesity. Organophosphorus pesticides have been used for pest control. Here, we investigated the estrogen receptor (ER)-dependent adipogenesis-inducing effect of representative organophosphorus pesticides (OPs), diazinon, phoxim, terbufos and tolclofos-methyl in 3T3-L1 adipocytes. Four OPs exhibited ER agonistic effect, determined using the OECD Performance Based Test Guideline No. 455; in vitro ER stably transfected transactivation assay using ERα-HeLa-9903 cell line, through binding affinity to ERα. Additionally, they increased lipid droplet accumulation in a dose-dependent manner, which was suppressed by ICI182,780, a well-known ER antagonist. Four OPs treatment induced peroxisome proliferator-activated receptor gamma (PPARγ), CCAAT/enhancer-binding protein alpha (C/EBPα), and perilipin expression. Furthermore, PPARγ, C/EBPα and perilipin expression was inhibited by co-treatment with ICI182,780. The increased mRNA expression of lipoprotein lipase and fatty acid synthase by four OPs was suppressed by co-treatment with ICI182,780. These results indicated that diazinon, phoxim, terbufos, and tolclofos-methyl might have adipogenesis-inducing effect mediated by interacting with ER.

Dioxins are highly injurious environmental pollutants with proven toxicological effects on both animals and humans, but to date their effects on plants still need to be studied in detail. We identified a dioxin-inducible caleosin/peroxygenase isoform, PdPXG4, that is mostly expressed in leaves of date palm seedlings and exhibits a specific reductase activity towards the 13-hydroperoxide of C18:2 and C18:3 (HpODE and HpOTrE, respectively). After exposure to TCDD, lipid droplets (LDs) isolated from TCDD-exposed leaves were about 6.5–15.7-fold more active in metabolizing 13-HpOTrE compared with those isolated from non-exposed leaves. A characteristic spectrum of leaf dioxin-responsive oxylipins (LDROXYL) was detected in dioxin-exposed seedlings. Of particular importance, a group of these oxylipins, referred to as Class I, comprising six congeners of hydroxides fatty acids derived from C18:2 and C18:3, was exclusively found in leaves after exposure to TCDD. The TCDD-induced oxylipin pattern was confirmed in vitro using terbufos, a typical inhibitor towards the PdPXG4 peroxygenase activity. Of particular interest, the response of terbufos-pretreated protoplasts to TCDD was drastically reduced. Together, these findings suggest that PdPXG4 is implicated in the establishment of a dioxin-specific oxylipin signature in date palm leaves soon after their exposure to these pollutants.

In this study, we reported the occurrence of eight organophosphorus pesticides (OPPs) in the East China Sea. Forty samples were collected and analysed with a high volume solid phase extraction method (Hi-throat/Hi-volume SPE) in the early summer of 2020. All the target OPPs were detected in the surface water at one or more stations in the East China Sea, and the concentrations of ΣOPPs were in the range 0.0775–3.09 ng/L (mean: 0.862 ± 0.624 ng/L). Terbufos sulfone and fenthion were the main pollutants in this area, probably resulting from pesticide use in China and other countries. The off-shore input from coastal regions was suggested to be a major source of OPP pollution in the East China Sea, and the movement of ocean currents played an important role in their transportation because around 0.86 t OPPs passed through the Tsushima Strait from the East China Sea each month. An ecological risk assessment showed that these OPPs presented a high risk to species in the East China Sea, whereas they posed no health risk to humans under both the median and high exposure scenario.

Several epidemiological studies show the association between organophosphate pesticides (OPs) and the risk of non-Hodgkin lymphoma (NHL), yet various research results remain controversial. To explore the hazard of OPs exposure to human health, three kinds of OPs (Terbufos, Malathion, and Diazinon) that are non-halogenated aliphatic compounds were included in the meta-analysis. We searched PubMed and Web of Science Databases for articles published from 1985 to February 2017. The databases were also searched for eligible studies through a manual references search. The random-effect model was utilized to compute the odds ratios (ORs) and 95% confident intervals (CIs). A total of ten observational studies (five cohort, four case-control, and one nested case-control) were included in our meta-analysis, with a pooled OR of 1.22 (95% CI 1.04 to 1.43) of Malathion, Terbufos and Diazinion. The general heterogeneity for OR was moderate (Ph = 0.032, I² = 41.2%). The OR estimates in the subset analyses were utilized to compare the association between the three kinds of OPs and NHL; Terbufos (OR = 1.07, 95% CI 0.85 to 1.36) and Malathion (OR = 1.17, 95% CI 0.82 to 1.67) had a statistically non-significant relationship, whereas Diazinon (OR = 1.39, 95% CI 1.11 to 1.73) was significantly associated with an increased NHL risk. Because immune dysfunction was thought to increase NHL risk, the toxicity levels in the immune system of the three types of OPs were compared. Malathion attacked immune cells via a direct effect and Diazinon disrupted the neuro-immune system, which involves the cholinergic system of lymphocytes via indirect immune damage, whereas an immunotoxic effect involving Terbufos was not reported. Overall, the present meta-analysis indicated a statistically significant association between Diazinon exposure and NHL risk.

To examine the link between corn agriculture and the observed decline of the endangered southern bell frog (SBF), the effects of two corn crop pesticides on larval growth and development were investigated. Tadpoles were exposed to terbufos sulfone (10 μg/L), a major breakdown product of the insecticide terbufos, and the herbicide atrazine (25 μg/L) individually and as a mixture until the completion of metamorphosis. Atrazine did not interact synergistically with terbufos sulfone or result in significant effects on growth and development alone, although there was some indication of accelerated metamorphosis in the pilot study. Terbufos sulfone alone and as a mixture (terbufos/atrazine) significantly slowed larval development and ultimately delayed metamorphosis. The observed developmental effects from an environmentally relevant concentration of terbufos sulfone indicates a risk posed by this persistent degradation product to the endangered SBF, which breeds and develops in the rice bays adjacent to corn fields treated with pesticides.

Organophosphate pesticides (OPPs) are one type of the most massively used pesticides and ubiquitously detected in aquatic environments, which may pose potential risks to the aquatic organisms and human health. In the present study, the spatiotemporal distribution and potential risks of OPPs were investigated in overlying water and surficial sediments from urban waterways of Guangzhou. For all studied sites, in general, four target OPPs (i.e., malathion, chlorpyrifos, terbufos, and diazinon) were present in the overlying water, with malathion and chlorpyrifos as major components. Higher concentrations of the four OPPs were found for the water and sediments collected in the dry season compared to the wet season, possibly because of the dilution effect of heavy rains. The results of Pearson’s analyses and principal coordinate analyses (PCoA) suggested similar sources for target OPPs in the water and sediments across the Guangzhou urban waterways. Potential ecological risks of the OPPs to three representative taxons (algae, aquatic invertebrates, and fish) were evaluated via toxic units (TUs) and risk quotients (RQs), while risk assessment on human health was performed using hazard index (HI). Although TU results showed no acute risks to the aquatic organisms in the overlying water and surface sediments, RQ results of the mixture showed high risks to the aquatic invertebrate and fish in all water samples. Individual HI values and cumulative HI values were on the order of 10⁻⁶–10⁻³ for children and adults, suggesting no potential risks to either children or adults through drinking and bathing.

Along the southeastern coast of Costa Rica, a variety of pesticides are intensively applied to produce export-quality plantains and bananas. In this region, and in other agricultural areas, fish kills are often documented by local residents and/or in the national news. This study examines principal exposure pathways, measured environmental concentrations, and selected toxicity thresholds of the three most prevalent pesticides (chlorpyrifos, terbufos, and difenoconazole) to construct a deterministic risk assessment for fish mortality. Comparisons of observed pesticide concentrations, along with estimated biological effects and observations during actual fish kills, highlight gaps in knowledge in correlating pesticide environmental concentration and toxicity in tropical environments. Observations of fish kill events and measured pesticide concentrations in the field, along with other water quality indicators, suggest that a number of environmental conditions can interact to cause fish mortality and that current species toxicity datasets may not be applicable for estimating toxicological or other synergistic effects, especially in tropical environments.

This study assesses the ecological risks (ERA) of pesticides to aquatic organisms in the River Madre de Dios (RMD), which receives surface runoff water from banana, pineapple, and rice plantations on the Caribbean coast of Costa Rica. Water samples collected over 2 years at five sites in the RMD revealed a total of 26 pesticides. Their toxicity risk to aquatic organisms was assessed using three recent ERA models. (1) The PERPEST model showed a high probability (>50 %) of clear toxic effects of pesticide mixtures on algae, macrophytes, zooplankton, macroinvertebrates, and community metabolism and a low probability (<50 %) of clear effects on fish. (2) Species sensitivity distributions (SSD) showed a moderate to high risk of three herbicides: ametryn, bromacil, diuron and four insecticides: carbaryl, diazinon, ethoprophos, terbufos. (3) The multi-substance potentially affected fraction (msPAF) model showed results consistent with PERPEST: high risk to algae (maximum msPAF: 73 %), aquatic plants (61 %), and arthropods (25 %) and low risk to fish (0.2 %) from pesticide mixtures. The pesticides posing the highest risks according to msPAF and that should be substituted with less toxic substances were the herbicides ametryn, diuron, the insecticides carbaryl, chlorpyrifos, diazinon, ethoprophos, and the fungicide difenoconazole. Ecological risks were highest near the plantations and decreased progressively further downstream. The risk to fish was found to be relatively low in these models, but water samples were not collected during fish kill events and some highly toxic pesticides known to be used were not analyzed for in this study. Further sampling and analysis of water samples is needed to determine toxicity risks to fish during peaks of pesticide mixture concentrations. The msPAF model, which estimates the ecological risks of mixtures based on their toxic modes of action, was found to be the most suitable model to assess toxicity risks to aquatic organisms in the RMD. The PERPEST model was found to be a strong tool for screening risk assessments. The SSD approach is useful in deriving water quality criteria for specific pesticides. This study, through the application of three ERA models, clearly shows that pesticides used in plantations within the RMD watershed are expected to have severe adverse effects on most groups of aquatic organisms and that actions are urgently needed to reduce pesticide pollution in this high biodiversity ecosystem.

Degradation and mineralization behavior of selected nematicides was studied in soil samples from fields cultivated with banana, potato, and coffee. Degradation assays in most of the studied soils revealed shorter half-lives for carbofuran (CBF) and ethoprophos (ETP) in samples with a history of treatment with these compounds, which may have been caused by enhanced biodegradation. A short half-life value for CBF degradation was also observed in a banana field with no previous exposure to this pesticide, but with a recent application of the carbamate insecticide oxamyl, which supports the hypothesis that preexposure to oxamyl may cause microbial adaptation towards degradation of CBF, an observation of a phenomenon not yet tested according to the literature reviewed. Mineralization assays for CBF and terbufos (TBF) revealed that history of treatment with these nematicides did not cause higher mineralization rates in preexposed soils when compared to unexposed ones, except in the case of soils from coffee fields. Mineralization half-lives for soils unexposed to these pesticides were significantly shorter than most reports in the literature in the same conditions. Mineralization rates for soils with a previous exposure to these pesticides were also obtained, adding to the very few reports found. This paper contributes valuable data to the low number of reports dealing with pesticide fate in soils from tropical origin.
