Clearfield® wheat (Triticum aestivum) have helped eliminate the toughest grasses and broadleaf weeds in Spain since 2005. This crop production system includes other tolerant cultivars to the application of imidazolinone (IMI) herbicides. However, the continuous use and off-label rates of IMI herbicides can contribute to the development of resistance in Lolium rigidum and other weed species. In this research, the main objectives were to study the resistance mechanisms to acetolactate synthase (ALS) and acetyl coenzyme A carboxylase (ACCase) inhibitors in a L. rigidum accession (LrR) from a Clearfield® wheat field, with a long history rotating these IMI-tolerant crops and compare them with those present in the IMI-tolerant wheat. The resistance to ACCase inhibitors in LrR was due to point mutations (Ile1781Leu plus Asp2078Gly) of the target site gene plus an enhanced herbicide metabolism (EHM), on the other hand, in wheat accessions was due only by EHM. Mechanisms involved in the resistance to ALS inhibitors were both point mutations of the target gene and EHM in the IMI-tolerant wheat, while only evidence of mutation (Trp574Leu) was found in the multiple herbicide resistant L. rigidum accession. This research demonstrates that if crop rotation is not accompanied by the use of alternative sites of action in herbicide-tolerant crops, resistant weeds to herbicide to which crops are tolerant, can easily be selected. Moreover, repeated and inappropriate use of Clearfield® crops and herbicide rotations can lead to the evolution of multiple resistant weeds, as shown in this study, and have also inestimable environmental impacts.

Arbuscular mycorrhizal fungi display desired potential to boost crop productivity and drought acclimation. Yet, whether nanoparticles can be incorporated into arbuscular mycorrhizal fungi for better improvement and its relevant morphologic and anatomical evidences are little documented. Pot culture experiment on wheat (Triticum aestivum L.) was conducted under drought stress (30% FWC) as well as well watered conditions (80% FWC) that involved priming of wheat seeds with iron nanoparticles at different concentrations (5mg L⁻¹, 10 mg L⁻¹ and 15 mg L⁻¹) with and without the inoculation of Glomus intraradices. The effects of treatments were observed on morphological and physiological parameters across jointing, anthesis and maturity stage. Root colonization and nanoparticle uptake trend by seeds and roots was also recorded. We observed strikingly high enhancement in biomass up to 109% under drought and 71% under well-watered conditions, and grain yield increased to 163% under drought and 60% under well-watered conditions. Iron nanoparticles at 10 mg L⁻¹ when combined with Glomus intraradices resulted in maximum wheat growth and yield, which mechanically resulted from higher rhizosphere colonization level, water use efficiency and photosynthetic rate under drought stress (P < 0.01). Across growth stages, optical micrograph observations affirmed higher root infection rate when combined with nanoparticles. Transmission electron microscopy indicated the penetration of nanoparticles into the seeds and translocation across roots whereas energy dispersive X-ray analyses further confirmed the presence of Fe in these organs. Iron nanoparticles significantly enhanced the growth-promoting and drought-tolerant effects of Glomus intraradices on wheat.

Per- and polyfluoroalkyl substances (PFASs) have been found to be widely present in soil. Dissolved organic matter (DOM) in soil are supposed to greatly affect the bioavailability of PFASs in soil. Herein, hydroponic experiments were conducted to understand the impacts of two kinds of typical DOM, bovine serum albumin (BSA) and humic acid (HA), on the uptake and translocation of legacy PFASs and their emerging alternatives, perfluorooctane sulfonic acid (PFOS), perfluorooctane acid (PFOA), perfluorohexane sulfonic (PFHxS) and 6:2 chlorinated polyfluoroalkyl ether sulfonate (6:2 Cl-PFESA) in wheat (Triticum aestivum L.). The results indicated that both HA and BSA significantly inhibited the bioaccumulation and translocation of PFASs in the roots and shoots of wheat, and the impacts of BSA were greater than HA. This difference was explained by the greater binding affinities of the four PFASs with BSA than with HA, as evidenced by the equilibrium dialysis and isothermal titration calorimetry (ITC) analyses. It was noting that inhibition impacts of the BSA-HA mixture (1:1) were lower than BSA alone. The results of Fourier transform infrared (FT-IR) spectroscopy and excitation-emission matrix (EEM) fluorescence spectroscopy suggested that HA could bind with the fluorescent tryptophan residues in BSA greatly, competing the binding sites with PFASs and forming a cover on the surface of BSA. As a result, the binding of PFASs with BSA-HA complex was much lower than that with BSA, but close to HA. The results of this study shed light on the impacts of DOM in soil on the bioaccumulation and translocation of PFASs in plants.

Microplastic pollution is a recognized hazard in aquatic systems, but in the past decade has emerged as a pollutant of interest in terrestrial ecosystems. This paper is the first formal meta-analysis to examine the phytotoxic effects of microplastics and their impact on soil functions in the plant-soil system. Our specific aims were to: 1) determine how the type and size of microplastics affect plant and soil health, 2) identify which agricultural plants are more sensitive to microplastics, and 3) investigate how the frequency and amount of microplastic pollution affect soil functions. Plant morphology, antioxidant production and photosynthesis capacity were impacted by the composition of polymers in microplastics, and the responses could be negative, positive or neutral depending on the polymer type. Phytotoxicity testing revealed that maize (Zea mays) was more sensitive than rice (Oryza sativa) and wheat (Triticum aestivum) within the Poaceae family, while wheat and lettuce (Lactuca sativa) were less sensitive to microplastics exposure. Microplastics-impacted soils tend to be more porous and retain more water, but this did not improve soil stability or increase soil microbial diversity, suggesting that microplastics occupied physical space but were not integrated into the soil biophysical matrix. The meta-data revealed that microplastics enhanced soil evapotranspiration, organic carbon, soil porosity, CO₂ flux, water saturation, nitrogen content and soil microbial biomass, but decreased soil N₂O flux, water stable aggregates, water use efficiency, soil bulk density and soil microbial diversity.

High cadmium (Cd) concentration in common wheat (Triticum aestivum L.) grains poses potential health risks. Several management strategies have been used to reduce grain Cd concentration. However, limited information is available on the use of ammonium-nitrogen (NH₄⁺-N) as a strategy to manage Cd concentration in wheat grains. In this study, NH₄⁺-N addition at the seedling stage unchanged the grain Cd concentration in the high-Cd accumulator, Zhoumai 18 (ZM18), but dramatically increased that in the low-Cd accumulator, Yunmai 51 (YM51). Further analysis revealed that the effects of NH₄⁺-N addition on whole-plant Cd absorption, root-to-shoot Cd translocation, and shoot-to-grain Cd remobilization were different between the two wheat cultivars. In ZM18, NH₄⁺-N addition did not change whole-plant Cd absorption, but inhibited root-to-shoot Cd translocation and Cd remobilization from lower internodes, lower leaves, node 1, and internode 1 to grains via the down-regulation of yellow stripe-like transporters (YSL), zinc transporters (ZIP5, ZIP7, and ZIP10), and heavy-metal transporting ATPases (HMA2). This inhibition decreased the grain Cd content by 29.62%, which was consistent with the decrease of the grain dry weight by 23.26%, leading to unchanged grain Cd concentration in ZM18. However, in YM51, NH₄⁺-N addition promoted continuous Cd absorption during grain filling, root-to-shoot Cd translocation and whole-plant Cd absorption. The absorbed Cd was directly transported to internode 1 via the xylem and then re-transported to grains via the phloem by up-regulated YSL, ZIP5, and copper transporters (COPT4). This promotion increased the grain Cd content by 245.35%, which was higher than the increased grain dry weight by 132.89%, leading to increased grain Cd concentration in YM51. Our findings concluded that the addition of NH₄⁺-N fertilizer at the seedling stage is not suitable for reducing grain Cd concentration in common wheat cultivars.

Titanium dioxide nanoparticles (TiO₂NPs) application in variety of commercial products would likely release these NPs into the environment. The interaction of TiO₂NPs with terrestrial plants upon uptake can disturb plants functional traits and can also transfer to the food chain members. In this study, we investigated the impact of TiO₂NPs on wheat (Triticum aestivum L.) plants functional traits, primary macronutrients assimilation, and change in the profile of bio-macromolecule. Moreover, the mechanism of biochar-TiO₂NPs interaction, immobilization, and tissue accumulation to cell translocation of NPs in plants was also explored. The results indicated that the contents of Ti in wheat tissues was reduced about 3-fold and the Ti transfer rate (per day) was reduced about 2 fold at the 1000 mg L⁻¹ exposure level of TiO₂NPs in biochar amended exposure medium. Transmission electron microscopy (TEM) with elemental mapping confirmed that Ti concentrated in plant tissues in nano-form. The interactive effect of TiO₂NPs + biochar amendment on photosynthesis related and gas exchange traits was observed at relatively low TiO₂NPs exposure level (200 mg L⁻¹), which induced the positive impact on wheat plants proliferation. TiO₂NPs alone exposure to wheat also modified the plant’s bio-macromolecules profile with the reduction in the assimilation of primary macronutrients, which could affect the food crop nutritional value and quality. X-ray photoelectron spectroscopy (XPS) chemical analysis of biochar + TiO₂NPs showed an additional peak, which indicated the binding interaction of NPs with biochar. Moreover, Fourier-transform infrared (FTIR) spectroscopy confirmed that the biochar carboxyl group is the main functionality involved in the bonding process with TiO₂NPs. These findings will help for a mechanistic understanding of the role of biochar in the reduction of NPs bioavailability to primary producers of the terrestrial environment.

Herein, we investigated the effects of di-n-butyl phthalate (DBP) on photosynthesis, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) content, oxidative damage, and biomass accumulation of different tissues in wheat (Triticum aestivum L) planted in cinnamon soils. The photosynthetic or fluorescence parameters (except for the intercellular carbon dioxide concentration), chlorophyll content, RuBisCO content, and biomass of roots, stems, and leaves decreased at the seedling, jointing, and booting stages under the stress of DBP. Compared with the control, the content of superoxide anions and hydrogen peroxide in the roots, stems, and leaves increased with increasing DBP concentrations at the seedling, jointing, and booting stages. The activities of superoxide dismutase (SOD) and catalase (CAT) in the roots, stems, and leaves increased under the 10 and 20 mg kg−1 DBP treatments; however, no significant changes were observed under the 40 mg kg−1 DBP treatment at the seedling stage (except for the SOD activity in roots). The increase in SOD and CAT activities in the roots, stems, and leaves with increasing DBP concentration at the jointing and booting stages suggested that an increase in the activities of these antioxidant enzymes may play an important role in defending against excess reactive oxygen species under DBP stress. The biomass of wheat roots, stems, and leaves decreased with an increase in DBP concentration, which was presumably caused by a decrease in photosynthesis and RuBisCO. The effect of DBP on wheat roots, stems, and leaves decreased with wheat growth.

Rare earth elements (REEs) are typically present as mixtures in the environment, but a quantitative understanding of mixture toxicity and interactions of REEs is still lacking. Here, we examined the toxicity to wheat (Triticum aestivum L.) of Y, La, and Ce when applied individually and in combination. Both concentration addition (CA) and independent action (IA) reference models were used for mixture toxicity analysis because the toxicity mechanisms of REEs remain obscure. Upon single exposure, the EC50s of Y, La, and Ce, expressed as dissolved concentrations, were 1.73 ± 0.24 μM, 2.59 ± 0.23 μM, and 1.50 ± 0.22 μM, respectively. The toxicity measured with relative root elongation followed La < Y ≈ Ce, irrespective of the dose descriptors. The use of CA and IA provided similar estimates of REE mixture interactions and toxicity. When expressed as dissolved metal concentrations, nearly additive effects were observed in Y-La and La-Ce mixtures, while antagonistic interactions were seen in Y-Ce mixtures. When expressed as free metal activities, antagonistic interactions were found for all three binary mixtures. This can be explained by a competitive effect of REEs ions for binding to the active sites of plant roots. The application of a more elaborate MIXTOX model in conjunction with the free ion activities, which incorporates the non-additive interactions and bioavailability-modifying factors, well predicted the mixture toxicity (with >92% of toxicity variations explained). Our results highlighted the importance of considering mixture interactions and subsequent bioavailability in assessing the joint toxicity of REEs.

N-ethyl perfluorooctane sulfonamide (N-EtFOSA) is an important perfluorooctanesulfonate (PFOS) precursor (PreFOS) which is used in sulfluramid. The present work studied the uptake, translocation and metabolism of N-EtFOSA in wheat (Triticum aestivum L.), soybean (Glycine max L. Merrill) and pumpkin (Cucurbita maxima L.) by hydroponic exposure. Except for parent N-EtFOSA, its metabolites of perfluorooctane sulfonamide acetate (FOSAA), perfluorooctane sulfonamide (PFOSA), PFOS, perfluorohexane sulfonate (PFHxS) and perfluorobutane sulfonate (PFBS) were detected in the roots and shoots of all the three plant species examined. This suggested that plant roots could take up N-EtFOSA from solutions efficiently, and translocate to shoots. A positive correlation was found between root concentration factors (RCFs) of N-EtFOSA and root lipid content. Much higher proportion of N-EtFOSA transformation products in plant tissues than in the solutions suggested that N-EtFOSA could be in vivo metabolized in plant roots and shoots to FOSAA, PFOSA and PFOS, and other additional shorter-chain perfluoroalkane sulfonates (PFSAs), including PFHxS and PFBS. The results suggested that plants had biotransformation pathways to N-EtFOSA that were different than those from microorganisms and animals. This study provides important information about the uptake and metabolism of PreFOSs in plants.

Copper nanoparticles (nCu) are widely used in industry and in daily life, due to their unique physical, chemical, and biological properties. Few studies have focused on nCu phytotoxicity, especially with regard to toxicity mechanisms in crop plants. The present study examined the effect of 15.6 μM nCu exposure on the root morphology, physiology, and gene transcription levels of wheat (Triticum aestivum L.), a major crop cultivated worldwide. The results obtained were compared with the effects of exposing wheat to an equivalent molar concentration of ionic Cu (Cu²⁺ released from CuSO₄) and to control plants. The relative growth rate of roots decreased to approximately 60% and the formation of lateral roots was stimulated under nCu exposure, possibly due to the enhancement of nitrogen uptake and accumulation of auxin in lateral roots. The expression of four of the genes involved in the positive regulation of cell proliferation and negative regulation of programmed cell death decreased to 50% in the Cu²⁺ treatment compared to that of the control, while only one gene was down-regulated to about half of the control in nCu treatment. This explained the decreased root cell proliferation and higher extent of induced cell death in Cu²⁺- than in nCu-exposed plants. The increased methane dicarboxylic aldehyde accumulation (2.17-fold increase compared with the control) and decreased antioxidant enzyme activities (more than 50% decrease compared with the control) observed in the Cu²⁺ treatment in relation to the nCu treatment indicated higher oxidative stress in Cu²⁺- than in nCu-exposed plants. Antioxidant (e.g., proline) synthesis was pronouncedly induced by nCu to scavenge excess reactive oxygen species, alleviating phytotoxicity to wheat exposed to this form of Cu. Overall, oxidative stress and root growth inhibition were the main causes of nCu toxicity.
