Enhanced treatment of organic matter in slaughter wastewater through live Bacillus velezensis strain using nano zinc oxide microsphere

Slaughter wastewater is an important and wide range of environmental issues, and even threaten human health through meat production. A high efficiency and stability microsphere-immobilized Bacillus velezensis strain was designed to remove organic matter and inhibit the growth of harmful bacteria in process of slaughter wastewater. Bacillus velezensis was immobilized on the surface of sodium alginate (SA)/Polyvinyl alcohol (PVA)/Nano Zinc Oxide (Nano-ZnO) microsphere with the adhesion to bio-carrier through direct physical adsorption. Results indicated that SA/PVA/ZnO and SA/ZnO microspheres could inhibit E.coli growth with adding 0.15 g/L nano-ZnO and not affect Bacillus velezensis strain, and the removal the chemical oxygen demand (COD) rates of SA/PVA/ZnO microsphere immobilized cells are 16.99%, followed by SA/ZnO (13.69%) and free bacteria (7.61%) from 50% concentration slaughter wastewater within 24 h at 37 °C, pH 7.0, and 120 rpm, a significant difference was found between the microsphere and control group. Moreover, when the processing time reaches 36 h, COD degradation of SA/PVA/ZnO microsphere is obviously higher than other groups (SA/PVA/ZnO:SA/ZnO:control vs 18.535 : 15.446: 10.812). Similar results were obtained from 30% concentration slaughter wastewater. Moreover, protein degradation assay was detected, and there are no significant difference (SA/PVA/ZnO:SA/ZnO:control vs 35.4 : 34.4: 36.0). The design of this strategy could greatly enhance the degradation efficiency, inhibit the growth of other bacteria and no effect on the activity of protease in slaughter wastewater. These findings suggested that the nano-ZnO hydrogel immobilization Bacillus velezensis system wastewater treatment is a valuable alternative method for the remediation of pollutants from slaughter wastewater with a novel and eco-friendly with low-cost investment as an advantage.

Micro and nanoplastics are new generation contaminants of global concern. It is important to evaluate the effects on edible products due to the presence of micro- and nano-sized plastics in the treated wastewater. A hydroponic experiment was carried out to explore the effect of polsytrene nanoplastics (PS-NPs; 20 nm) at different concentrations (0, 12.5, 25, and 50 mg L⁻¹) on Glycine max L. (soybean) seedlings for 7-days. In the current study, firstly the uptake of PS-NPs by Glycine max L. (soybean) roots were confirmed by laser confocal scanning microscope. Exposure to PS-NPs, negatively affected growth parameters and increased Fe, Zn and Mn contents in roots and leaves of soybean seedlings. PS-NPs treatments caused oxidative stress in soybean seedlings. The hydrogen peroxide and malondialdehyde contents, showed similar increase pattern in seedlings exposed to PS-NPs. Response to PS-NPs, the level of antioxidant enzymes (superoxide dismutase, catalase, ascorbate peroxidase, and guaiacol peroxidase) and proline content were generally enhanced in roots and leaves of soybean. The expression level of stress-related genes examined in the study included CSD5, FSD3, APX1, and POD up-regulated in PS-NPs treated-soybean seedlings in a tissue specific manner. The results of the present study showed the adverse effects of PS-NPs on soybean seedlings, which may have important implications for the risk assessment of NPs on crop production and environmental safety.

Targeting the removal of Pb²⁺ in wastewater, cellulosic materials were carbonized in an aerobic environment and activated via ion exchange. The maximum adsorption capacity reached 243.5 mg/g on an MCC-derived adsorbent activated with sodium acetate. The modified porous properties improved the adsorption capacity. The capacity could be completely recovered five times through elution with EDTA. Because of the negative effects of Ni, Mg, and Ca elements, the adsorption capacities of activated carbonized natural materials were lower than that of pure cellulose. N₂ adsorption measurement showed that the adsorbent had a large specific surface area as well as abundant micropores and 4-nm-sized mesopores. FTIR and surface potential results proved that carboxyl group was generated in the aerobic carbonization, and was deprotonated during ion exchange. This adsorbent consisted of C–C bonds as the building blocks and hydrophilic groups on the surface. XPS results demonstrated that the Pb 4f binding energies were reduced by 0.7–0.8 eV due to the interaction between Pb²⁺ and the activated adsorbent, indicating that the carboxylate groups bonded with Pb²⁺ through coordination interactions. Pseudo-second-order and Elovich kinetic models were well fitted with the adsorption processes on the pristine and activated carbonized adsorbents, indicative of chemisorption on heterogeneous surfaces. The Freundlich expression agreed well with the data measured, and the pristine and activated adsorbents had weak and strong affinities for Pb²⁺, respectively. The Pb²⁺ adsorption process was exothermic and spontaneous, and heat release determined the spontaneity. The adsorption capacity is attributed to the carboxylate groups and pores generated in the aerobic oxidation and ion exchange procedures.

Wastewater-based epidemiology (WBE) has emerged as a valuable approach for forecasting disease outbreaks in developed countries with a centralized sewage infrastructure. On the other hand, due to the absence of well-defined and systematic sewage networks, WBE is challenging to implement in developing countries like Bangladesh where most people live in rural areas. Identification of appropriate locations for rural Hotspot Based Sampling (HBS) and urban Drain Based Sampling (DBS) are critical to enable WBE based monitoring system. We investigated the best sampling locations from both urban and rural areas in Bangladesh after evaluating the sanitation infrastructure for forecasting COVID-19 prevalence. A total of 168 wastewater samples were collected from 14 districts of Bangladesh during each of the two peak pandemic seasons. RT-qPCR commercial kits were used to target ORF1ab and N genes. The presence of SARS-CoV-2 genetic materials was found in 98% (165/168) and 95% (160/168) wastewater samples in the first and second round sampling, respectively. Although wastewater effluents from both the marketplace and isolation center drains were found with the highest amount of genetic materials according to the mixed model, quantifiable SARS-CoV-2 RNAs were also identified in the other four sampling sites. Hence, wastewater samples of the marketplace in rural areas and isolation centers in urban areas can be considered the appropriate sampling sites to detect contagion hotspots. This is the first complete study to detect SARS-CoV-2 genetic components in wastewater samples collected from rural and urban areas for monitoring the COVID-19 pandemic. The results based on the study revealed a correlation between viral copy numbers in wastewater samples and SARS-CoV-2 positive cases reported by the Directorate General of Health Services (DGHS) as part of the national surveillance program for COVID-19 prevention. The findings of this study will help in setting strategies and guidelines for the selection of appropriate sampling sites, which will facilitate in development of comprehensive wastewater-based epidemiological systems for surveillance of rural and urban areas of low-income countries with inadequate sewage infrastructure.

Amyl salicylate (AS) is a fragrance massively used as a personal care product and following the discharged in wastewaters may end up in the aquatic environment representing a potential threat for the ecosystem and living organisms. AS was recently detected in water of the Venice Lagoon, a vulnerable area continuously subjected to the income of anthropogenic chemicals. The lagoon is a relevant area for mollusc farming, including the Mediterranean mussels (Mytilus galloprovincialis) having an important economic and ecological role. Despite high levels of AS occurred in water of the Lagoon of Venice, no studies investigated the possible consequences of AS exposures on species inhabiting this ecosystem to date. For the first time, we applied a multidisciplinary approach to investigate the potential effects of the fragrance AS on Mediterranean mussels. To reach such a goal, bioaccumulation, cellular, biochemical, and molecular analyses (RNA-seq and microbiota characterization) were measured in mussels treated for 7 and 14 days with different AS Venice lagoon environmental levels (0.1 and 0.5 μg L⁻¹). Despite chemical investigations suggested low AS bioaccumulation capability, cellular and molecular analyses highlighted the disruption of several key cellular processes after the prolonged exposures to the high AS concentration. Among them, potential immunotoxicity and changes in transcriptional regulation of pathways involved in energy metabolism, stress response, apoptosis and cell death regulations have been observed. Conversely, exposure to the low AS concentration demonstrated weak transcriptional changes and transient increased representation of opportunistic pathogens, as Arcobacter genus and Vibrio aestuarianus. Summarizing, this study provides the first overview on the effects of AS on one of the most widely farmed mollusk species.

Antibiotics usage in animal production is considered a primary driver of the occurrence, supply and spread of antibiotic resistance genes (ARGs) in the environment. Pig farms and fish ponds are important breeding systems in food animal production. In this study, we compared and analyzed broad ARGs profiles, mobile genetic elements (MGEs) and bacterial communities in a representative pig farm and neighboring fish ponds around Poyang Lake, the largest freshwater lake in China. The factors influencing the distribution of ARGs were also explored. The results showed widespread detection of ARGs (from 57 to 110) among 283 targeted ARGs in the collected water samples. The differences in the number and relative abundance of ARGs observed from the pig farm and neighboring fish ponds revealed that ARG contamination was more serious on the pig farm than in the fish ponds and that the water treatment plant on the pig farm was not very effective. Based on the variance partition analysis (VPA), MGEs, bacterial communities and water quality indicators (WIs) codrive the relative abundance of ARGs. Based on network analysis, we found that total phosphorus and Tp614 were the most important WIs and MGEs affecting ARG abundance, respectively. Our findings provide fundamental data on farms in lakeside districts and provide insights into establishing standards for the discharge of aquaculture wastewater.

Synthetic microfibers have been identified as the most prevalent type of microplastic in samples from aquatic, atmospheric, and terrestrial environments across the globe. Apparel washing has shown to be a major source of microfiber pollution. We used California as a case study to estimate the magnitude and fate of microfiber emissions, and to evaluate potential mitigation approaches. First, we quantified synthetic microfiber emissions and fate from apparel washing in California by developing a material flow model which connects California-specific data on synthetic fiber consumption, apparel washing, microfiber generation, and wastewater and biosolid management practices. Next, we used the model to assess the effectiveness of different interventions to reduce microfiber emissions to natural environments. We estimate that in 2019 as much as 2.2 kilotons (kt) of synthetic microfibers were generated by apparel washing in California, a 26% increase since 2008. The majority entered terrestrial environments (1.6 kt), followed by landfills (0.4 kt), waterbodies (0.1 kt), and incineration (0.1 kt). California's wastewater treatment network was estimated to divert 95% of microfibers from waterbodies, mainly to terrestrial environments and primarily via land application of biosolids. Our analysis also reveals that application of biosolids on agricultural lands facilitates a directional flow of microfibers from higher-income urban counties to lower-income rural communities. Without interventions, annual synthetic microfiber emissions to California's natural environments are expected to increase by 17% to 2.1 kt by 2026. Further increasing the microfiber retention efficiency at the wastewater treatment plant would increase emissions to terrestrial environments, which suggests that microfibers should be removed before entering the wastewater system. In our model, full adoption of in-line filters in washing machines decreased annual synthetic microfiber emissions to natural environments by 79% to 0.5 kt and offered the largest reduction of all modeled scenarios.

The treatment of contaminants from lignocellulosic biorefinery effluent has recently been identified as a unique challenge. This study focuses on removing phenolic contaminants and polycyclic aromatic hydrocarbons (PAHs) from lignocellulosic biorefinery wastewater (BRW) applying a laccase-assisted approach. Cassava waste was used as a substrate to produce the maximum yield of laccase enzyme (3.9 U/g) from Pleurotus ostreatus. Among the different inducers supplemented, CuSO₄ (0.5 mM) showed an eight-fold increase in enzyme production (30.8 U/g) after 240 h of incubation. The catalytic efficiency of laccase was observed as 128.7 ± 8.47 S⁻¹mM⁻¹ for syringaldazine oxidation at optimum pH 4.0 and 40 °C. Laccase activity was completely inhibited by lead (II) ion, mercury (II) ion, sodium dodecyl sulphate, sodium azide and 1,4 dithiothretiol and induced significantly by manganese (II) ion and rhamnolipid. After treating BRW with laccase, the concentrations of PAHs and phenolic contaminants of 1144 μg/L and 46160 μg/L were reduced to 96 μg/L and 16100 μg/L, respectively. The ability of laccase to effectively degrade PAHs in the presence of different phenolic compounds implies that phenolic contaminants may play a role in PAHs degradation. After 240 h, organic contaminants were removed from BRW in the following order: phenol >2,4-dinitrophenol > 2-methyl-4,6-dinitrophenol > 2,3,4,6-tetrachlorophenol > acenaphthene > fluorine > phenanthrene > fluoranthene > pyrene > anthracene > chrysene > naphthalene > benzo(a)anthracene > benzo(a)pyrene > benzo(b)fluoranthene > pentachlorophenol > indeno(1,2,3-cd)pyrene > benzo(j) fluoranthene > benzo[k]fluoranthène. The multiple contaminant remediation from the BRW by enzymatic method, clearly suggests that the laccase can be used as a bioremediation tool for the treatment of wastewater from various industries.

Wastewater and stormwater are both considered as critical pathways contributing microplastics (MPs) to the aquatic environment. However, there is little information in the literature about the potential influence of constructed wetlands (CWs), a commonly used wastewater and stormwater treatment system. This study was conducted to investigate the abundance and distribution of MPs in water and sediment at five CWs with different influent sources, namely stormwater and wastewater. The MP abundance in the water samples ranged between 0.4 ± 0.3 and 3.8 ± 2.3 MP/L at the inlet and from 0.1 ± 0.0 to 1.3 ± 1.0 MP/L at the outlet. In the sediment, abundance of MPs was generally higher at the inlet, ranging from 736 ± 335 to 3480 ± 4330 MP/kg dry sediment and decreased to between 19.0 ± 16.4 and 1060 ± 326 MP/kg dry sediment at the outlet. Although no significant differences were observed in sediment cores at different depth across the five CWs, more MPs were recorded in silt compared to sandy sediment which indicated sediment grain size could be an environmental factor contributing to the distribution of MPs. Polyethylene terephthalate (PET) fibres were the dominant polymer type found in the water samples while polyethylene (PE) and polypropylene (PP) fragments were predominantly recorded in the sediment. While the size of MPs in water varied across the studied CWs, between 51% and 64% of MPs in the sediment were smaller than 300 μm, which raises concerns about the bioavailability of MPs to a wider range of wetland biota and their potential ecotoxicological effects. This study shows that CWs can not only retain MPs in the treated water, but also become sinks accumulating MPs over time.

Recently, hetero junction materials (p-n-p and n-p-n) have been developed for uplifting the visible light activity to destroy the harmful pollutants in wastewater. This manuscript presents a vivid description of novel n-p-n junction materials namely CeO₂-PPy-ZnO. This novel n-p-n junction was applied as the photocatalyst in drifting the mobility of charge carriers and hence obtaining the better photocatalytic activity when compared with p-n and pure system. Such catalyst's syntheses were successful via the copolymerization method. The structural, morphological and optical characterization techniques were applied to identify the physio-chemical properties of the prepared materials. Additionally, the superior performance of this n-p-n nanostructured material was demonstrated in the destruction of micro organic (chlorophenol) toxic wastes under visible light. The accomplished ability of the prepared catalysts (up to 92% degradation of chlorophenol after 180 min of irradiation) and their profound degradation mechanism was explained in detail.
