Paraquat (PQ) is one of the most widely used herbicides in the world due to its excellent weed control effects. Accumulating evidence has revealed that long-term exposure to PQ can significantly increase the risk of Parkinson’s disease (PD). However, the underlying molecular mechanisms are yet to be fully understood. Hence, we investigated the potential role of reactive oxygen species (ROS) and dynamin-related protein 1 (DRP1) in PQ-induced mitophagy, aiming to elaborate on possible molecular mechanisms involved in PQ-triggered neurotoxicity. Our results showed that ROS were increased, mitochondrial membrane potential was decreased at 100, 200, and 300 μM PQ concentrations, and autophagy pathways were activated at a concentration of 100 μM in neuronal cells. In addition, excessive mitophagy was observed in neurons exposed to 300 μM PQ for 24 h. Then, ROS-mediated mitochondrial fission was found to contribute to PQ-induced excessive mitophagy. Moreover, all aforementioned changes were significantly ameliorated by mdivi-1. Thus, our findings provide a novel neurotoxic mechanism and reveal the DRP1-mitochondrial fission pathway as a potential target for treatments of PQ-induced excessive mitophagy, serving as an alternative target for the prevention and treatment of Parkinson’s disease. Because harmful substances are transmitted and enriched in the food chain, the toxic effect of environmental paraquat is nonnegligible, and more investigations are needed.

Fomesafen (FSA) is widely used in soybean fields for weed control. However, the persisting characteristics of FSA in the agricultural soil or water may become a hidden danger causing environmental pollution and phytotoxicity to succession crops. In this study, the growth and physiological responses of rice to FSA were investigated. It was found that the growth of rice seedlings was obviously inhibited by FSA exposure especially at over 0.1 mg L⁻¹. To gain an insight into the molecular mechanisms for the potential ecotoxicology, four libraries of rice roots and shoots exposed to FSA were created and subjected to the global RNA-sequencing (RNA-Seq) combined with HRLC-Q-TOF-MS/MS analytical technologies to comprehensively characterize the biochemical processes and catalytic reactions involved in FSA metabolism in rice. Compared with those without FSA, 499 and 450 up-regulated genes in roots and shoots with FSA were detected. Many of them were closely correlated with the tolerance to environmental stress, detoxification of xenobiotics and molecular metabolism process including cytochrome P450, glutathione S-transferases and acetyltransferase. A total of eight metabolites and fourteen conjugates in the reactive pathways of hydrolysis, substitution, reduction, methylation, glycosylation, acetylation, and malonylation were characterized by HRLC-Q-TOF-MS/MS. The relationship between the metabolized derivatives of FSA and enhanced expression the corresponding enzymatic regulators was established. This study will help understand the mechanisms and pathways of FSA metabolism and inspire the further research on FSA degradation in the paddy crops and environmental or health risks.

Glyphosate-based herbicides (GBHs) are the most widely used pesticides for weed control. In parallel with the renewal of the active ingredient, polyethoxylated POE(15) containing GBHs were banned in the EU in 2016. Since then, co-formulants were changed and numerous GBHs are marketed with different excipients declared as inert substances. In our study, we focused to determine acute and chronic cytotoxicity (by Aliivibrio fischeri assay) and direct hormonal activity (estrogenic and androgenic effects measured by Saccharomyces cerevisiae BLYES/BLYAS strains, respectively) of glyphosate, AMPA, polyethoxylated POE(15) and 13 GBHs from which 11 formulations do not contain polyethoxylated POE(15). Among the pure substances, neither glyphosate nor AMPA had any effects, while polyethoxylated POE(15) exhibited pronounced toxicity and was also estrogenic but not androgenic. Regarding the acute and chronic cytotoxicity and hormonal activity of GBHs, dilution percentages calculated from EC₅₀ values were in the most cases by one or two order of magnitude lower than the minimum recommended dilution for agricultural and household use. Relation could not be observed between the biological effects and type of glyphosate-salts; hence toxicity could be linked to the co-formulants, which are not even declared in 3 GBHs. Toxicological evaluation must focus on these substances and free accessibility of GBHs should be reconsidered.

In the Pampas, public concern has strongly risen because of the intensive use of glyphosate for weed control and fallow associated with biotech crops. The present study was aimed to evaluate the occurrence and concentration of the herbicide and its main metabolite (AMPA) in soil and other environmental compartments of the mentioned agroecosystem, including groundwater, in relation to real-world agricultural management practices in the region. Occurrence was almost ubiquitous in solid matrices (83–100%) with maximum concentrations among the higher reported in the world (soil: 8105 and 38939; sediment: 3294 and 7219; suspended particulate matter (SPM): 584 and 475 μg/kg of glyphosate and AMPA). Lower detection frequency was observed in surface water (27–55%) with maximum concentrations in whole water of 1.80 and 1.90 μg/L of glyphosate and AMPA, indicating that SPM analysis would be more sensitive for detection in the aquatic ecosystem. No detectable concentrations of glyphosate or AMPA were observed in groundwater. Glyphosate soil concentrations were better correlated with the total cumulative dose and total number of applications than the last spraying event dose, and an increment of 1 mg glyphosate/kg soil every 5 spraying events was estimated. Findings allow to infer that, under current practices, application rates are higher than dissipation rates. Hence, glyphosate and AMPA should be considered “pseudo-persistent” pollutants and a revisions of management procedures, monitoring programs, and ecological risk for soil and sediments should be also recommended.

Herbicide application is a practice commonly used in agricultural systems because it is an efficient method of weed control. An inherent characteristic of some herbicides used in mountain agriculture, such as oxyfluorfen, is high adsorption to soil organic matter (SOM). Thus, intensive management that changes the quantity and quality of SOM, such as soil tillage and the massive application of organic fertilizers such as poultry litter, may favor soil contamination by this herbicide and alter its dynamics in the environment. Therefore, this study aimed to characterize the structures of humic substances (HSs) in the soil of forest areas and areas with intensive production of vegetables, relating them to the accumulation of the herbicide oxyfluorfen in tropical mountain agroecosystems. Organic carbon content was quantified in HSs, humic acid (HAs) were structurally characterized by CP/MAS ¹³C-NMR spectroscopy, and the oxyfluorfen molecule was detected and quantified using the QuEChERS residue detection method with subsequent analysis by LC-MS/MS. Oxyfluorfen was not detected in the forest areas, but it was detected in the vegetable growing areas at points with the lowest slope and high contents of organic matter and clay, with values of up to 0.13 mg kg⁻¹. The intensification in the SOM mineralization process, promoted by the intensive management adopted in the vegetable growing areas, resulted in a 16.46% reduction in COT, a 58.84% reduction in the carbon content in the form of SH and a reduction in the structures that give recalcitrance to the HA molecule (CAₗₖyₗ-H,R, CCOO–H,R, CAᵣₒₘₐₜᵢc-H,R, and CAᵣₒₘₐₜᵢc-O) when compared to those values in the forest area, presenting HAs with more aliphatic and labile properties. Thus, due to the structural characteristics of the HAs in the vegetable production areas, the herbicide oxyfluorfen showed a close relationship with the more aliphatic oxygenated structures, namely, CAₗₖyₗ-O,N, CAₗₖyₗ-O and CAₗₖyₗ-di-O.

The phenylurea herbicide, linuron (LIN), is used to control various types of weeds. Despite its efficient role in controlling weeds, it presents a persistent problem to the environment. In the current study, phytoremediation properties of transgenic CYP1A2 Arabidopsis thaliana plants to LIN were assessed. CYP1A2 gene was firstly cloned and expressed in bacteria before proceeding to plants. In presence of LIN, The growth of CYP1A2 expressing bacteria was superior compared to control bacteria transformed with the empty bacterial expression vector pET22b(+). No clear morphological changes were detected on CYP1A2 transgenic plants. However, significant resistance to LIN herbicide application either via spraying the foliar parts of the plant or via supplementation of the herbicide in the growth medium was observed for CYP1A2 transformants. Plant growth assays under LIN stress provide strong evidence for the enhanced capacity of transgenic lines to grow and to tolerate high concentrations of LIN compared to control plants. HPLC analyses showed that detoxification of LIN by bacterial extracts and/or transgenic plant leaves is improved as compared to the corresponding controls. Our data indicate that over expression of the human CYP1A2 gene increases the phytoremediation capacity and tolerance of Arabidopsis thaliana plants to the phenylurea herbicide linuron.

Triafamone, a sulfamide herbicide, has been extensively utilized for weed control in rice paddies in Asia. However, its fate and transformation in the environment have not been established. Through a rice paddy microcosm-based simulation trial combined with multiple spectroscopic analyses, we isolated and identified three novel metabolites of triafamone, including hydroxyl triafamone (HTA), hydroxyl triafamone glycoside (HTAG), and oxazolidinedione triafamone (OTA). When triafamone was applied to rice paddies at a concentration of 34.2 g active ingredient/ha, this was predominantly distributed in the paddy soil and water, and then rapidly dissipated in accordance with the first-order rate model, with half-lives of 4.3–11.0 days. As the main transformation pathway, triafamone was assimilated by the rice plants and was detoxified into HTAG, whereas the rest was reduced into HTA with subsequent formation of OTA. At the senescence stage, brown rice had incurred triafamone at a concentration of 0.0016 mg/kg, but the hazard quotient was <1, suggesting that long-term consumption of the triafamone-containing brown rice is relatively safe. The findings of the present study indicate that triafamone is actively metabolized in the agricultural environment, and elucidation of the link between environmental exposure to these triazine or oxazolidinedione moieties that contain metabolites and their potential impacts is warranted.

As the two most commonly used organophosphorus herbicides, glyphosate (Gly) and glufosinate-ammonium (Glu) have unique properties for weed control and algae removal in aquaculture. However, the occurrences and health risks of Gly and Glu in aquaculture ponds are rare known. This study aimed to investigate the occurrences of Gly, AMPA (primary metabolity of Gly) and Glu in surface water, sediment and aquatic products from the grass carp (ctenopharyngodon idella), crayfish (procambarus clarkii) and crab (eriocheir sinensis) ponds around Lake Honghu, the largest freshwater lake in Hubei province, China where aquaculture has become the local pillar industry. Three age groups (children, young adults, middle-aged and elderly) exposure to these compounds through edible aquatic products (muscle) consumption were also assessed by target hazard quotient (THQ) method. The results indicated that Gly, AMPA and Glu were widely occurred in surface water, sediment and organisms in the fish, crayfish and crab ponds. AMPA was more likely to accumulate in the intestine of aquatic products than Gly and Glu. According to the total THQ value (1.04＞1), muscle consumption of grass carp may pose potential risk to children.

Glyphosate herbicide is ubiquitously used in agriculture and weed control. It has now been identified in aquatic ecosystems worldwide, where numerous studies have suggested that it may have both suppressive and stimulatory effects on diverse non-target organisms. We cultured natural biofilms from a hypereutrophic environment to test the effects on periphytic diatoms of exposure to a glyphosate-based herbicide formulation at concentrations from 0 to 10 mg L⁻¹ of active ingredient. There were clear and significant differences between treatments in diatom community structure after the 15-day experiments. Diversity increased more in low glyphosate treatments relative to higher concentrations, and compositional analyses indicated statistically significant differences between glyphosate treatments. The magnitude of change observed was significantly correlated with glyphosate-based herbicide concentration. Our results show that glyphosate-based herbicides have species-selective effects on benthic diatoms that may significantly alter trajectories of community development and therefore may affect benthic habitats and whole ecosystem function.

Fomesafen, a long-lived protoporphyrinogen-oxidase inhibitor, specially developed for post-emergence control of broad-leaf weeds, is used widely in soybean fields in northern China (Dayan and Duke, 2010). The impact of fomesafen on microbial communities in rhizosphere soils, however, is unknown. In this study we examined fomesafen degradation as well as its effects in the rhizosphere of soybean plants grown in a greenhouse. Fomesafen had shorter half-life in rhizosphere soil than previously reported for bulk soil from the same location (87 vs 120 days). The enzyme activity of soil extracts and the microbial community composition of 16S rRNA genes (16S) amplified from soil DNA were also investigated. Although not immediately apparent, both the high (37.5 mg kg⁻¹) and low (18.75 mg kg⁻¹) doses of fomesafen significantly decreased urease and invertase activities in the rhizosphere soil from days 30 and 45 respectively until the end of the experiment (90 days). Analysis of 16S amplicons demonstrated that fomesafen had a dose dependent effect, decreasing alpha diversity and altering beta diversity. Significant phylum level decreases were observed in five of the ten phyla that were most abundant in the control. Proteobacteria was the only phylum whose relative abundance increased in the presence of fomesafen, driven by increases in the genera Methylophilacaea, Dyella, and Sphingomonas. The functional implications of changes in 16S abundance as predicted using PICRUSt suggested that fomesafen enriched for enzymes involved in xenobiotic metabolism and detoxification (cytochrome P450s and glutathione metabolism). Our data suggest that, despite being degraded more rapidly in the rhizosphere than in bulk soil, fomesafen had long-lasting functional impacts on the soil microbial community.
