Accumulation characteristics and biological response of ginger to sulfamethoxazole and ofloxacin

The potential risk to human health of antibiotics that pass through the food chain has become an important global issue, but there are few reports on the response of ginger (Zingiber officinale) to antibiotic pollution. In this study, we investigated the enrichment characteristics and biological response of ginger to sulfamethoxazole (SMZ) and ofloxacin (OFL) residues, which are common in the environment. Lower levels of SMZ, OFL and their combined duplex treatment (SMZ+OFL) promoted the growth of ginger, but the critical doses necessary to stimulate growth differed among treatments: 10 mg L⁻¹ SMZ, 1 mg L⁻¹ OFL and 1 mg L⁻¹ (SMZ+OFL) had the strongest stimulating effects. At higher dosages, the root growth and light energy utilization efficiency of ginger were impaired, and (SMZ+OFL) had the strongest inhibitory effect. Treatments with lower levels of antibiotics had no significant effect on reactive oxygen species and antioxidant enzyme activities. However, when SMZ, OFL and SMZ+OFL concentrations exceeded 10 mg L⁻¹, the contents of H₂O₂, O₂⁻ and MDA continued to increase, while the activities of SOD, POD, CAT first increased and then decreased, especially in SMZ+OFL. Ginger accumulated more SMZ and OFL in rhizomes and less in leaves, and accumulation increased significantly as antibiotic concentration increased. When SMZ concentration was 1 mg L⁻¹, the SMZ concentrations in rhizomes, roots, and leaves were 0.23, 0.15, and 0.05 mg kg⁻¹, respectively, and the residual SMZ in the rhizome was 2.3 times higher than the maximum residue limit. The abundance of the resistance genes sul1, sul2, qnrS, and intI1 increased with increasing antibiotic concentrations, and intI1 abundance was the highest. OFL induced higher levels of intI1 expression than did SMZ.

Improving digestibility, fermentation characteristics, and reducing greenhouse biogases to protect the environment without the use of synthetic materials is an important goal of modern-day farming and nutritionist. Plant extracts are capable of solving these. This is due to the digestive enzymes and the bioactive components capable of performing antimicrobial functions inherent in these plants. This study was aimed to investigate the effect of standard maize substrate treated with selected herbs and spices extracts on ruminal environmental biogas production and pressure during fermentation via biogas production technique. Herbs (Azadirachta indica leaves (T1), Moringa oleifera leaves (T2), Ocimum gratissimum leaves (T3) and spices (Allium sativum bulb (T4), Zingiber officinale rhizome (T5)) were harvested, air dried, and milled using standard procedures. Methanolic extracts of the herbs and spices were prepared and used as additives at different concentrations (50, 100, and 150 μL) to the maize substrate for in vitro biogas production. Data were analyzed using regression analysis. There were significant (P < 0.05) differences across all the treatments on the volume and pressure of biogas. The pressure and volume of biogas when compared with the levels tested showed differences (P < 0.05) across all the treatments for the prediction of volume from pressure of biogas. The pressure and volume of gas produced in vitro increased (P < 0.05) and biogases decreased (P < 0.05) by the substrate treated with herbs and spices but for the drum stick leaves which was similar for the levels of concentration tested. This means that the level tested had a pronounced mitigation effect on pressure of biogas and volume of biogas produced. It was concluded that the herb and spice extracts have the potential to improve rumen fermentation and reduce the production of biogases in ruminant diet.

Occupational exposure to low-level ionizing radiation (<1 Gy) was shown to enhance cell protection via attenuating an established inflammatory process. Nicotine, a major toxic component of cigarette smoke, is responsible for smoking-mediated renal dysfunction. The present study was therefore aimed to investigate the protective impact of ginger Zingiber officinale selenium nanoparticles (SeNPs) with whole-body low-dose gamma radiation (γ-R) against nicotine-induced nephrotoxicity in male albino rats. Nicotine intoxication was induced with 0.5 mg/kg BW. Rats received 0.1 mg SeNPs/kg BW by gastric gavage concomitant with 0.5 Gy γ-R over 4 weeks. Characterization studies showed the formation of spherical SeNPs with a size ranged from 10 to 30 nm in diameter with a thin film encapsulating the nanoballs. Our data revealed that nicotine induced renal dysfunction manifested by significant abnormal levels of kidney function markers (creatinine, urea, sodium and potassium) accompanied by increased levels of malondialdehyde along with a reduction in glutathione level, glutathione peroxidase, and glutathione S-transferase activities. It is worthy to note that nicotine toxicity induced significant increments in serum inflammatory markers: tumor necrosis factor-α and vascular cell adhesion protein 1. Western blotting showed marked significant elevation in caspase-3 activities against nicotine. The mRNA gene expression of inducible cyclooxygenase-2 gene was highly increased with nicotine intoxication while that of cyclooxygenase-1 did not show any changes. Interestingly, our data demonstrated that SeNPs in synergistic interaction with γ-R are efficacious control against nicotine-induced nephrotoxicity via anti-oxidant-mediated anti-inflammatory activities. Thus, it is tempting to recommend dietary approaches with ginger SeNPs for smokers at workplaces exposed occupationally and regularly to low-level ionizing radiation.

Zingerone (ZO), one of the active components of ginger (Zingiber officinale), is a phenolic alkanone with antioxidant, antiapoptotic, and anti-inflammatory properties. Cisplatin (CP) is a widely used chemotherapeutic drug for solid tumors, but its therapeutic use is limited due to dose-dependent nephrotoxicity. In the present study, we investigated the ameliorative effect of ZO against CP-induced nephrotoxicity. Intraperitoneal administration of single-dose CP (7 mg/kg body weight) on the first day enhanced kidney lipid peroxidation and reduced antioxidant enzyme activities such as catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and glutathione (GSH). CP increased serum urea and creatinine levels and disrupted histological integrity while causing a decrease aquaporin 1 (AQP1) level in the kidney tissues. CP induced inflammatory responses by elevating the levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), interleukin-6 (IL-6), interleukin-33 (IL-33) and nuclear factor kappa B (NF-κB), and activities of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). Moreover, it also caused oxidative DNA damage and activation of apoptotic pathway by increasing of 8-hydroxy-2′-deoxyguanosine (8-OHdG), p53, cysteine aspartate-specific protease-3 (caspase-3), and Bcl-2-associated x protein (bax) while decreasing B cell lymphoma-2 (Bcl-2). However, treatment with ZO at a dose of 25 and 50 mg/kg b.wt. for 7 days significantly decreased oxidative stress, apoptosis, inflammation, and histopathological alterations while increased AQP1 levels in the kidney tissue. The results of the current study suggested that ZO as an effective natural product attenuates CP-induced nephrotoxicity.
