Crosstalk of oxidative damage, apoptosis, and autophagy under endoplasmic reticulum (ER) stress involved in thifluzamide-induced liver damage in zebrafish (Danio rerio) Полный текст

Although the hepatotoxicity of thifluzamide in zebrafish has been characterized, its toxic mechanisms have not been fully explored. The present study demonstrated that thifluzamide damaged the zebrafish liver and endoplasmic reticulum (ER). In addition, thifluzamide significantly changed the expression of genes encoding antioxidant proteins and increased the malondialdehyde (MDA) content, leading to oxidative damage in zebrafish liver. Additionally, the autophagic ultrastructure was observed by transmission electron microscopy (TEM), and LC3-I/LC3-II conversion was obviously upregulated under western blotting (WB) measurements, verifying that autophagy was induced by thifluzamide. Moreover, the activities of Caspase-3 and Caspase-9 were obviously decreased, indicating that apoptosis was inhibited in adult zebrafish exposed to a higher concentration of thifluzamide. In summary, oxidative damage and autophagy but not apoptosis under ER injury might lead to the hepatotoxicity of thifluzamide in zebrafish. Our findings provide a new mechanistic insight into the toxicity of thifluzamide in zebrafish.

Soil particulate organic matter (POM) has rapid turnover and metal enrichment, but the interactions between organic matter (OM) and metals have not been well studied. The present study aimed to investigate changes in the OM concentration and composition of the POM fraction and their corresponding effects on metal distribution and extractability in long-term polluted paddy soils. Soil 2000–53 μm POM size fractions had higher contents of C–H and C=O bonds, C–H/C=O ratios and concentrations of fulvic acid (FA), humic acid (HA), cadmium (Cd) and zinc (Zn) than the bulk soils. Cadmium and Zn stocks in soil POM fractions were 24.5–27.9% and 7.12–16.7%, respectively, and were more readily EDTA-extractable. Compared with the control soil, the 2000–250 μm POM size fractions had higher organic carbon concentrations and C/N ratios in the polluted soils. However, there were no significant differences in the contents in C–H and C=O bonds or C–H/C=O ratios of POM fractions among the control, slightly and highly polluted soils. In accordance with the lower contents of C=O bonds and FA and HA concentrations, the Cd and Zn concentrations in 250–53 μm POM size fractions were lower than those in 2000–250 μm POM size fractions. Enrichment of Cd in POM fractions increased with increasing soil pollution level. These results support the view that changes in the OM concentration and the size and composition of POM fractions play a key role in determining the distribution of Cd and Zn in paddy soils.

In recent years, air pollution events have occurred frequently in China during the winter. Most studies have focused on the physical and chemical composition of polluted air. Some studies have examined the bacterial bioaerosols both indoors and outdoors. But few studies have focused on the relationship between air pollution and bacteria, especially pathogenic bacteria. Airborne PM samples with different diameters and different air quality index values were collected in Hangzhou, China from December 2014 to January 2015. High-throughput sequencing of 16S rRNA was used to categorize the airborne bacteria. Based on the NCBI database, the “Human Pathogen Database” was established, which is related to human health. Among all the PM samples, the diversity and concentration of total bacteria were lowest in the moderately or heavily polluted air. However, in the PM2.5 and PM10 samples, the relative abundances of pathogenic bacteria were highest in the heavily and moderately polluted air respectively. Considering the PM samples with different particle sizes, the diversities of total bacteria and the proportion of pathogenic bacteria in the PM10 samples were different from those in the PM2.5 and TSP samples. The composition of PM samples with different sizes range may be responsible for the variances. The relative humidity, carbon monoxide and ozone concentrations were the main factors, which affected the diversity of total bacteria and the proportion of pathogenic bacteria. Among the different environmental samples, the compositions of the total bacteria were very similar in all the airborne PM samples, but different from those in the water, surface soil, and ground dust samples. Which may be attributed to that the long-distance transport of the airflow may influence the composition of the airborne bacteria. This study of the pathogenic bacteria in airborne PM samples can provide a reference for environmental and public health researchers.

Microplastic is an emerging contaminant of concern in soils globally, probably gradually increasing in soil due to slow degradation. Few studies on microplastic effects on soil biota are available, and no study in a microplastic contamination context has specifically addressed soil protists. Soil protists, a phylogenetically and functionally diverse group of eukaryotic, unicellular soil organisms, are major consumers of bacteria in soils and are potentially important vehicles for the delivery of microplastics into the soil food chain. Here we build a case for focusing research on soil protists by drawing on data from previous, older studies of phagocytosis in protist taxa, which have long made use of polystyrene latex beads (microspheres). Various soil-borne taxa, including ciliates, flagellates and amoebae take up microplastic beads in the size range of a few micrometers. This included filter feeders as well as amoebae which engulf their prey. Discrimination in microplastic particle uptake depended on species, physiological state as well as particle size. Based on the results of the studies we review here, there is now a need to study microplastic effects in a pollution ecology context: this means considering a broad range of particle types under realistic conditions in the soil, and exploring longer-term effects on soil protist communities and functions.

Biochar, a natural sourced carbon-rich material, has been used commonly in particle shape for carbon sequestration, soil fertility and environmental remediation. Here, we report a facile approach to fabricate freestanding biochar composite membranes for the first time. Wood biochars pyrolyzed at 300 °C and 700 °C were blended with polyvinylidene fluoride (PVdF) in three percentages (10%, 30% and 50%) to construct membranes through thermal phase inversion process. The resultant biochar composite membranes possess high mechanical strength and porous structure with uniform distribution of biochar particles throughout the membrane surface and cross-section. The membrane pure water flux was increased with B300 content (4825–5411 ± 21 L m⁻² h⁻¹) and B700 content (5823–6895 ± 72 L m⁻² h⁻¹). The membranes with B300 were more hydrophilic with higher surface free energy (58.84–60.31 mJ m⁻²) in comparison to B700 (56.32–51.91 mJ m⁻²). The biochar composite membranes indicated promising adsorption capacities (47–187 mg g⁻¹) to Rhodamine B (RhB) dye. The biochar membranes also exhibited high retention (74–93%) for E. coli bacterial suspensions through filtration. After simple physical cleaning, both the adsorption and sieving capabilities of the biochar composite membranes could be effectively recovered. Synergistic mechanisms of biochar/PVdF in the composite membrane are proposed to elucidate the high performance of the membrane in pollutant management. The multifunctional biochar composite membrane not only effectively prevent the problems caused by directly using biochar particle as sorbent but also can be produced in large scale, indicating great potential for practical applications.

Triclosan (TCS) is a broad-spectrum antimicrobial used in a variety of consumer products. While it was recently banned from hand soaps in the US, it is still a key ingredient in a top-selling toothpaste. TCS is a hydrophobic micropollutant that is recalcitrant under anaerobic digestion thereby resulting in high TCS concentrations in biosolids. The objective of this study was to determine the impact of TCS on the antibiotic resistome and potential cross-protection in lab-scale anaerobic digesters using shotgun metagenomics. It was hypothesized that metagenomics would reveal selection for antibiotic resistance genes (ARGs) not previously found in pure culture studies or mixed-culture studies using targeted qPCR. In this study, four different levels of TCS were continuously fed to triplicate lab-scale anaerobic digesters to assess the effect of TCS levels on the antibiotic resistance gene profiles (resistome). Blasting metagenomic reads against antibiotic/metal resistance gene database (BacMet) revealed that ARG diversity and abundance changed along the TCS concentration gradient. While loss of bacterial diversity and digester function were observed in the digester treated with the highest TCS concentration, FabV, which is a known TCS resistance gene, increased in this extremely high TCS environment. The abundance of several other known ARG or metal resistance genes (MRGs), including corA and arsB, also increased as the concentrations of TCS increased. Analysis of other functional genes using SEED database revealed the increase of potentially key genes for resistance including different types of transporters and transposons. These results indicate that antimicrobials can alter the abundance of multiple resistance genes in anaerobic digesters even when function (i.e. methane production) is maintained. This study also suggests that enriched ARGs could be released into environments with biosolids land application.

Silver nanomaterials (AgNMs) are released into sewers and consequently find their way to sewage treatment plants (STPs). The AgNMs are transformed en route, mainly into silver sulfide (Ag₂S), which is only sparingly soluble in water and therefore potentially less harmful than the original AgNMs. Here we investigated the toxicity and fate of different sulfidized AgNMs using an exposure scenario involving the application of five different test materials (NM-300K, AgNO₃, Ag₂S NM-300K, Ag₂S NM and bulk Ag₂S) into a simulated STP for 10 days. The sewage sludge from each treatment was either dewatered or anaerobically digested for 35 days and then mixed into soil. We then assessed the effect on soil microorganisms over the next 180 days. After 60 days, a subsample of each test soil was used to assess chronic toxicity in oat plants (Avena sativa L) and a potential uptake into the plants. The effect of each AgNM on the most sensitive test organism was also tested without the application of sewage sludge. Although Ag sulfidized species are considered poorly soluble and barely bioavailable, we observed toxic effects on soil microorganisms. Furthermore, whether or not the AgNM was sulfidized before or during the passage through the STP, comparable effects were observed on ammonium oxidizing bacteria after sewage sludge application and incubation for 180 days. We observed the uptake of Ag into oat roots following the application of all test substances, confirming their bioavailability. The oat shoots generally containing less Ag than the roots.

To characterize major trace elements in PM₂.₅ and associated sources in two megacities, Chengdu (CD) and Chongqing (CQ), in Sichuan Basin of southwest China, daily PM₂.₅ samples were collected at one urban site in each city from October 2014 to July 2015 and were analyzed for their contents of thirteen trace elements including four crustal elements (Al, Ca, Fe, and Ti), eight trace metals (K, Cr, Zn, Cu, Mn, Pb, Ni, and V), and As. Multiple approaches including correlation analysis, enrichment factor, principal component analysis, and conditional probability function (CPF) were applied to identify potential sources of these elements. Most of the measured trace elements in Sichuan Basin were found to have lower concentrations than in the other regions of China. K and Fe were the most abundant elements at CD with an annual mean concentrations of 720 ± 357 and 456 ± 248 ng m⁻³, accounting for 34.6% and 21.9% of the total analyzed trace elements, respectively. Ca presented the highest concentration among all of the elements at CQ with annual mean of 824 ± 633 ng m⁻³ (29.1% of the total). Crustal elements had the highest concentrations in spring while heavy metals had distinct seasonal variations typically with the highest concentrations in winter and the lowest in summer. Ti and Al were identified to be primarily from soil while most of the analyzed heavy metals (Cr, Mn, Cu, Zn, Pb, Ni) and As were from anthropogenic sources associated with coal combustion, industrial emission from glassmaking production and iron/steel manufacturing, and non-exhaust vehicle emission.

It is acknowledged that organohalide-respiring bacteria (OHRB) can degrade polybrominated diphenyl ethers (PBDEs); however, very little is known about the distribution of OHRB or their response to PBDE contamination in natural sediments. We collected sediments from 28 sampling sites in Taihu Lake, China, and investigated the spatial distribution and diversity of OHRB, and the relationships between the PBDE contamination levels and the PBDE removal potential. The abundances of five typical OHRB genera, namely Dehalobacter, Dehalococcoides, Dehalogenimonas, Desulfitobacterium, and Geobacter, ranged from 0.34 × 10⁴ to 19.4 × 10⁷ gene copies g⁻¹ dry sediment, and varied significantly among different areas of Taihu Lake. OHRB were more abundant in sediments from Meiliang and Zhushan Bay, where the PBDE concentrations were higher, and the phylotype diversity of the OHRB belonging to the family Dehalococcoidaceae was lower, than reported for other areas. While the sulfate concentrations explained much of the spatial distribution of OHRB, PBDE concentrations were also a strong influence on the abundance and diversity of OHRB in the sediments. For Dehalococcoides, Dehalogenimonas and Geobacter, the abundance of each genus was positively related to its own potential to remove PBDEs. The dominant OHRB genus, Dehalogenimonas, may contribute most to in situ bioremediation of PBDEs in Taihu Lake.

In this study, we conducted an assessment of polyurethane foam (PUF) passive sampling for metals combining active sampling. Remarkably, we found that the metals collected in the passive samples differed greatly from those collected in active samples. By composition, Cu and Ni accounted for significantly higher proportions in passive samples than in active samples, leading to significantly higher uptake rates of Cu and Ni. In assessing seasonal variation, metals in passive samples had higher concentrations in summer (excluding Heshan), which differed greatly from the pattern of active samples (winter > summer), indicating that the uptake rates of most metals were higher in summer than in winter. Overall, due to the stable passive uptake rates, we considered that PUF passive samplers can be applied to collect atmospheric metals. Additionally, we created a snapshot of the metal pollution in the Pearl River Delta using principal component analysis of PUF samples and their source apportionment.
