Evaluation of phenol detoxification by Brassica napus hairy roots, using Allium cepa test

INTRODUCTION: Meristematic mitotic cells of Allium cepa constitute an adequate material for cytotoxicity and genotoxicity evaluation of environmental pollutants, such as phenol, which is a contaminant frequently found in several industrial effluents. RESULTS AND DISCUSSION: In the present work, Brassica napus hairy roots (HR) were used for phenol removal assays. The toxicity of post-removal solutions (PRS) and phenol solutions was analyzed. These HR removed the contaminant with high efficiency (100–80% for phenol solutions containing 10–250 mg/L, respectively). Phenol solutions treated with B. napus HR showed a significant reduction of general toxicity compared to untreated phenol solutions, since the IC50 values were 318.39 and 229.02 mg/L, respectively. Moreover, PRS presented lower cytotoxicity and genotoxicity than that found in phenol solutions untreated. The mitotic index (MI) observed in meristematic cells treated with PRS (100 and 250 mg/L of phenol) showed an increase of 35% and 42%, whereas the chromosome aberrations showed a significant decrease. According to these results, B. napus HR cultures could be used for the treatment of solutions contaminated with phenol, since we observed not only high removal efficiency, but also an important reduction of the general toxicity, cytotoxicity, and genotoxicity.

PURPOSE: Perfluorooctane sulfonate (PFOS) belongs to a group of chemicals called perfluoroalkyl acids that have been extensively used in various applications such as stain and oil resistant treatments for fabrics, fire-fighting foams, and insecticides. These chemicals present an environmental and health risk being present in many samples both in wildlife and humans. In this study, we investigate the effect of PFOS on fatty acid β-oxidation in developing chicken embryos. METHODS: Fertilized chicken eggs were exposed in ovo to PFOS at day 4 of incubation. On day 10, the eggs were dissected and livers were incubated in vitro with 3H-palmitic acid for 2 h. The media were collected, and after clean up, the amount of tritiated water was measured with liquid scintillation counting to determine the rate of palmitic acid β-oxidation. RESULTS: PFOS was found to induce fatty acid β-oxidation at doses starting from a lowest observed effect level (LOEL) of 0.1 μg/g egg weight. Maximum induction of 77 % compared to control was seen at 0.3 μg/g. CONCLUSIONS: The administered doses in which effects are seen are around and even lower than the levels that can be found in wild populations of birds. General population human levels are a factor of two to three times lower than the LOEL value of this study. The environmental contamination of PFOS therefore presents a possibility of effects in wild populations of birds.

PURPOSE: In the assessment of health risks of environmental pollutants, the method of dose addition and the method of independent action are used to assess mixture effects when no synergistic and/or antagonistic effects are present. Currently, no method exists to quantify synergistic and/or antagonistic effects for mixtures. The purpose of this paper is to develop the theoretical concepts of an overall risk probability (ORP)-based method to quantify the synergistic and antagonistic effects in health risk assessment for mixtures. METHOD: The ORP for health effects of environmental chemicals was determined from the cumulative probabilities of exposure and effects. This method was used to calculate the ORP for independent mixtures and for mixtures with synergistic and antagonistic effects. RESULTS: For the independent mixtures, a mixture ORP can be calculated from the product of the ORPs of individual components. For systems of interacting mixtures, a synergistic coefficient and an antagonistic coefficient were defined respectively to quantify the ORPs of each individual component in the mixture. The component ORPs with synergistic and/or antagonistic effects were then used to calculate the total ORP for the mixture. CONCLUSIONS: An ORP-based method was developed to quantify synergistic and antagonistic effects in health risk assessment for mixtures. This represents a first method to generally quantify mixture effects of interacting toxicants.

PURPOSE: Heavy metals have been detected in water and sediments from the Embalse La Florida, an artificial lake in the arid region of San Luis province, Argentina, representing one of the few sources of permanent water for reproduction of native anuran species. This study assesses lead (Pb) and cadmium (Cd) concentrations in the anuran species found in this water reservoir as well as differences between compounds, species and sites of collection. METHODS: Adult anuran amphibians were collected on the north and south shores of the Embalse La Florida and Pb and Cd concentrations were measured in whole body homogenates digested using wet ashing techniques. RESULTS: All individuals of the six species assayed had detectable levels of Pb and Cd that ranged from 1.19 to 5.57 μg/g dry mass and from 1.09 to 6.86 μg/g dry mass, respectively. Anuran amphibians collected in the more contaminated south shore accumulated 21% more Cd and 40% more Pb than individuals from the less altered north shore. Cd and Pb accumulation was not significantly correlated with the concentration in water at the site of collection. CONCLUSIONS: Amphibians of the Embalse La Florida accumulate Cd and Pb. Between and within species, differences were detected in Cd and Pb concentrations. Differences in metal concentrations between species, metals, and individuals collected on shores of the Embalse La Florida with different contamination, were detected. Therefore, it is crucial to implement adequate policies to protect amphibians from the accelerated urban development experienced in this location.

BACKGROUND, AIM AND SCOPE: There is growing evidence to show that dissolved humic substances, HSs, can directly interact with freshwater organisms, such as phototrophic organisms, cladocerans, amphipods and fish. The responses are—at least in part—transcriptionally controlled. These interactions can lead to stress symptoms in the exposed organisms. In phototrophs, stress symptoms include a reduction in photosynthetic oxygen release and antioxidative stress. Besides the direct effects, HSs also cause indirect effects that provoke different physiological adaptations in the phototrophs. MATERIALS AND METHODS: The HS-influenced photosynthetic performance and stress response of two different green algae, Pseudokirchneriella subcapitata (Koršikov) Hindák and Monoraphidium braunii (Nägeli in Kützing) Komárková-Legnerová, and two cyanobacterial species, Synechocystis sp. (PCC 6803, Institut Pasteur) and Microcystis aeruginosa (PCC 7806, Institut Pasteur), were tested. Two humic preparations were applied, the synthetic HS1500 and HuminFeed®, HF, which had previously been proven effective in bioassays with invertebrates and a water mould. RESULTS AND DISCUSSION: When the algae were grown near light saturation, most of the tested species were positively affected by HSs in growth rate or chlorophyll content. Cell sizes decreased with increasing HS concentrations for all eukaryotic phototrophs, except for the cyanobacteria. After 4 to 5 days of cultivation at the highest HS exposure, there was a decrease in total dry weight due to reduced cell sizes in contrast to an increase in cell numbers. With the exception of Synechocystis, the dry weight per cell ratio decreased with increasing HS concentration. The efficiency of utilizing absorbed light quanta increased with increasing HS concentrations; the maximum quantum yield of photosystem II (ΦPSIImax) was higher in all of the tested species, with the exception of M. aeruginosa, after exposure to HS. CONCLUSION: The applied humic preparations did not interact directly with PSII, but changed the physiological state of the algae, especially the photosynthetic performance. Neither the green algae nor the cyanobacteria were inhibited in growth or negatively affected in their photosynthetic performance. The exposure to lower concentrations of HS stimulated better growth of the phototrophs. The tested humic preparations obviously did not have the potency to act as xenobiotic stressors; furthermore, there was no sign of herbicide potency.

Increasing amounts of lanthanum (La) is released into aquatic environments. However, little information is available on the influence of La on aquatic plants. In this study, physiological and ultrastructural responses of Hydrocharis dubia (Bl.) Backer leaves to elevated concentrations of La (up to 160 μM) were investigated. The accumulation of La was found to be increased in a concentration-dependent manner. La disturbed the intrinsic balance of nutrient elements (P, Mg, Ca, Fe, K, and Zn). Pigment content decreased with the rise of the La concentrations and the EC50 value for chlorophyll was 20 μM on day 7. The antioxidants (superoxide dismutase, peroxidase, catalase, reduced ascorbate, and reduced glutathione) exhibited varied response to the La treatments. Malondialdehyde content enhanced gradually at all La concentrations. The enhancement in proline content was found in a concentration-dependent manner. The amounts of three polypeptides with apparent molecular weights of 61.9, 51.5, and 16.7 kDa, respectively, were gradually diminished, as well as one existing polypeptides with apparent molecular weight of 22.3 kDa, elevating in response to increasing La concentrations. Significant damage to the chloroplast, mitochondrion, and nucleus was imposed by La indicated a general disarray in the cellular functions. The negative effects of La on H. dubia unequivocally indicate that La could exert an adverse influence on aquatic ecosystem and should lead to a more careful discharge of such elements into water environment.

PURPOSE: The main goals of this study were (1) to examine the effects of zinc on the microbial community structure of anthropogenically impacted sediments in a tropical coastal ecosystem and (2) to determine whether these microbial benthic communities may enhance the adsorption of zinc. METHODS: The interactions between zinc and bacteria in tropical sediments were studied in sediment microcosms amended with 2.5 mg L−1 of Zn in the water phase and incubated for 8 days under different environmental conditions, oxic/anoxic and glucose addition. At the end of incubation, microbial structure was assessed by molecular fingerprints (T-RFLP) analysis and Zn speciation in the sediment was determined by sequential extraction. RESULTS: In the three studied sediments, Zn spiking resulted in only slight changes in bacterial community structure. In contrast, the addition of low concentrations of glucose (5 mM) strongly modified the bacterial community structure: <20% of similarity with the initial structure concomitant with a strong diminution of the specific richness. Overall, these results suggest that highly labile organic matter has a larger impact on microbial structure than heavy metal. These weak impacts of Zn on bacteria diversity might be partly explained by (1) the strong adsorption of Zn in the presence of bacteria and/or (2) the incorporation of Zn into a nonbioavailable fraction. Nevertheless, Zn spiking resulted in significant changes in nutrient cycles, suggesting that bacterial metabolisms were impacted by the heavy metal. This led to an increase in nutrient supplies to the water column, potentially enhancing eutrophication in a nutrient-limited, oligotrophic ecosystem.

The environmental fate of transformation products from organic pollutants such as drugs has become a new research area of increasing interest over the last few years. Whereas in the past mainly parent compounds or their major human metabolites were studied, new questions have arisen what compounds could be formed during incomplete degradation in the aquatic environment and what effects the resulting transformation products might have on nature and mankind. Psychiatric drugs are among the most important prescription drugs worldwide, but so far only little data is provided upon their degradation behavior. This especially accounts for tricyclic antipsychotic drugs of the phenothiazine class. Therefore, the degradation of such drugs was investigated in this study. In this study the aerobic Closed Bottle test (The Organisation for Economic Co-operation and Development (OECD) 301D) was used to assess the ready biodegradability of three trifluoromethylated phenothiazine drugs: fluphenazine, triflupromazine, and trifluoperazine. As it is known from literature that phenothiazine drugs can easily form various photolytic transformation products under light exposure, photochemical transformation was also investigated. Since transformation products are usually not available commercially, the calculation of environmental parameters with the aid of quantitative structure activity relationship (QSAR) software was used for first evaluation of these compounds. According to the OECD test guideline, all trifluoromethylated phenothiazines had to be classified as not readily biodegradable. Chromatographic data revealed the formation of some transformation products. Comparing retention time and mass spectrometric data with the analytical results of the light exposure experiments, we found peaks with the same retention time and mass spectra. So these transformation products were not of bacterial, but photolytic, origin and are formed very quickly even under low light doses. A special chromatographic column and solvent gradient along with multiple stage mass spectrometric fragmentation experiments uncovered the presence of, in total, nine photolytic transformation products and allowed for their structural elucidation. Typical modifications of the molecules were sulfoxidation, exocyclic N-oxidation, and transformation of the trifluoromethyl to a carboxylic moiety. The obtained results of the QSAR calculations show that all transformation products are highly mobile in the aquatic environment and elimination through biotic or abiotic pathways cannot be expected. Transformation products of trifluoromethylated phenothiazine drugs have to be expected in the aquatic environment, yet nothing is known about their toxicological properties. Therefore, further risk assessment upon these drugs and their fate is strongly recommended.

INTRODUCTION: Dissolved organic matter (DOM) is the most active component in environmental system and its chemical and structural characteristics most likely influence its biodegradation. Four surface soil (0–20 cm) and three core sediment samples (0–10 cm) were collected from Wuliangsuhai Lake. The objectives of this study were to investigate the spectral properties and humification degree of DOM and to determine and discuss comparatively the complexing capacities and stability constants of DOM by Cu (II) in the Hetao region. MATERIALS AND METHODS: In this study, fluorescence spectra and fluorescence quenching methods were used to evaluate the humification degree of DOM and calculate the complexing capacities and the stability constants between DOM and Cu (II). RESULTS AND DISCUSSION: Two defined peaks, at wavelengths of 260∼300 nm (peak I) and 300∼350 nm (peak II), could be identified for soil DOM at a Δλ value of 30 nm. In sediment DOM extracts, a third peak (III) was observed near 364 nm. The results show that there is a significant difference in the structure of DOM because of different sources. The humification degree is significantly higher for soil samples than those of sediment samples. The FT-IR spectra of DOM show that structure in sediment DOM is more functional groups than those in soil DOM. DOM has a stronger Cu binding affinity in soils than in sediment in the Hetao region, which may lead to potentially significant influence on the migration and transformation of Cu (II).

Two emergent macrophytes, Arundo donax and Phragmites australis, were established in experimental horizontal subsurface flow (HSSF), gravel-based constructed wetlands (CWs) and challenged by treated dairy processing factory wastewater with a median electrical conductivity of 8.9 mS cm−1. The hydraulic loading rate was tested at 3.75 cm day−1. In general, the plants grew well during the 7-month study period, with no obvious signs of salt stress. The major water quality parameters monitored (biological oxygen demand (BOD), suspended solids (SS) and total nitrogen (TN) but not total phosphorus) were generally improved after the effluent had passed through the CWs. There was no significance different in removal efficiencies between the planted beds and unplanted gravel beds (p > 0.007), nor was there any significant difference in removal efficiencies between the A. donax and P. australis beds for most parameters. BOD, SS and TN removal in the A. donax and P. australis CWs was 69, 95 and 26 % and 62, 97 and 26 %, respectively. Bacterial removal was observed but only to levels that would allow reuse of the effluent for use on non-food crops under Victorian state regulations. As expected, the A. donax CWs produced considerably more biomass (37 ± 7.2 kg wet weight) than the P. australis CWs (11 ± 1.4 kg wet weight). This standing crop equates to approximately 179 and 68 tonnes ha−1 year−1 biomass (dry weight) for A. donax and P. australis, respectively (assuming a 250-day growing season and single-cut harvest). The performance similarity of the A. donax and P. australis planted CWs indicates that either may be used in HSSF wetlands treating dairy factory wastewater, although the planting of A. donax provides additional opportunities for secondary income streams through utilisation of the biomass produced.
