Testicular junctions are pivotal to male fertility and regulated by constituent proteins. Increasing evidence suggests that environmental chemicals, including bisphenol A (BPA), may impact these proteins, but whether the impacts persist for generations is not yet known. Here, we investigate the effect of BPA (a ubiquitous endocrine-disrupting chemical) on testis and sperm functions and whether the effects are transferred to subsequent generations. Male mice (F0) were exposed to corn oil (Control) or 5 or 50 mg BPA/kg body weight/day from 6 to 12 weeks of age. The F0 were mated with wild-type females to produce the first filial (F1) generation. F2 and F3 were produced using similar procedures. Our results showed that BPA doses decreased the levels of some junctional proteins partly via binding with estrogen receptors (ERα and Erβ), upregulation of p-ERK1/2, P85, p-JNK and activation of p38 mitogen-activated protein kinase signaling. Consequently, testicular histological abnormalities, disrupted spermatogenesis, decreased sperm count, and inability to fertilize eggs were observed in mice exposed to BPA. These effects were transferred to successive generations (F2), partly through DNA methylation, but mostly alleviated in F3 males. Our findings suggest that paternal exposure to chemicals promoting alteration of testicular junctional proteins and its transgenerational inheritance is a key component of the origin of male reproductive health problems.

Long-term exposure to PM₂.₅ has been linked to lung cancer incidence and mortality, but limited evidence existed for other cancers. This study aimed to assess the association between PM₂.₅ on cancer specific mortality. An ecological study based on the cancer mortality data collected from 5,565 Brazilian cities during 2010–2018 using a difference-in-differences approach with quasi-Poisson regression, was applied to examine PM₂.₅-cancer mortality associations. Globally gridded annual average surface PM₂.₅ concentration was extracted and linked with the residential municipality of participants in this study. Sex, age stratified and exposure-response estimations were also conducted. Totalling 1,768,668 adult cancer deaths records of about 208 million population living across 5,565 municipalities were included in this study. The average PM₂.₅ concentration was 7.63 μg/m³ (standard deviation 3.32) with range from 2.95 μg/m³ to 28.5 μg/m³. With each 10 μg/m³ increase in three-year-average (current year and previous two years) concentrations of PM₂.₅, the relative risks (RR) of cancer mortality were 1.16 (95% confidence interval [CI]: 1.11–1.20) for all-site cancers. The PM₂.₅ exposure was significantly associated with several cancer-specific mortalities including oral, nasopharynx, oesophagus, and stomach, colon rectum, liver, gallbladder, larynx, lung, bone, skin, female breast, cervix, prostate, brain and leukaemia. No safe level of PM₂.₅ exposure was observed in the exposure-response curve for all types of cancer. In conclusion, with nationwide cancer death records in Brazil, we found that long-term exposure to ambient PM₂.₅ increased risks of mortality for many cancer types. Even low level PM₂.₅ concentrations had significant impacts on cancer mortality.

Human-induced nitrogen (N) and phosphorus (P) enrichment have profound effects on grassland net primary production (NPP) and species richness. However, a comprehensive understanding of the relative contribution of N vs. P addition and their interaction on grassland NPP increase and species loss remains elusive. We compiled data from 80 field manipulative studies and conducted a meta-analysis (2107 observations world-wide) to evaluate the individual and combined effects of N and P addition on grassland NPP and species richness. We found that both N addition and P addition significantly enhanced grassland above-ground NPP (ANPP; 33.2% and 14.2%, respectively), but did not affect total NPP, below-ground NPP (BNPP), and species evenness. Species richness significantly decreased with N addition (11.7%; by decreasing forbs) probably due to strong decreased soil pH, but not with P addition. The combined effects of N and P addition were generally stronger than the individual effects of N or P addition, and we found the synergistic effects on ANPP, and additive effects on total NPP, BNPP, species richness, and evenness within the combinations of N and P addition. In addition, N and P addition effects were strongly affected by moderator variables (e.g. climate and fertilization type, duration and amount of fertilizer addition). These results demonstrate a higher relative contribution of N than P addition to grassland NPP increase and species loss, although the effects varied across climate and fertilization types. The existing data also reveals that more long-term (≥5 years) experimental studies that combine N and P and test multifactor effects in different climate zones (particularly in boreal grasslands) are needed to provide a more solid basis for forecasting grassland community response and C sequestration response to nutrient enrichment at the global scale.

Persulfate-based advanced oxidation process is considered as a promising technology for the degradation of phenol, where efficient, cost effective, and green methods with high peroxydisulfate (PS) activation capacity is of increasing demand. In this work, an in-situ liquid phase precipitation combined with ball milling method was applied for the synthesized of α-FeOOH/biochar, as be the PS activator for phenol degradation. Results showed that the ball-milled α-FeOOH and red pine wood biochar prepared at 700 °C (BM-α-FeOOH/PBC700) exhibited the highest catalytic property with PS for phenol oxidation (a phenol removal rate of 100%), compared with the BM-α-FeOOH (16.0%) and BMPBC700 (66.3%). The presence of intermediate products such as hydroquinone and catechol, and total organic carbon (TOC) removal rate (88.9%) proved the oxidation of phenol in the BM-α-FeOOH/PBC700+PS system. The characterization results showed that the functional groups (e.g., CO, C–O, Fe–O, and Si–O), the dissolved organic matter (DOM) in biochar, the loading of Fe element, and higher degree of graphitization and defect structures, contributed to the activation of PS to form free radicals (i.e., SO₄·⁻, ·OH, ·O₂⁻, and hVB⁺) for phenol oxidation, of which, SO₄·⁻ and ·OH account for 72.1% of the phenol removal rate. The specific contribution to the PS activation for phenol oxidation by each part of the materials was calculated based on the “whole to part” experiment. The contribution of DOM, acid-soluble substance, and carbon matrix and basal part in BM-α-FeOOH/PBC700 were 6.0%, 40.9%, and 53.1%, respectively. The reusability experiments of BM-α-FeOOH/PBC700 demonstrated that the composite was relatively stable after four cycles of reuse. Among three co-existing anions (NO₃⁻, Cl⁻, and HCO₃⁻), HCO₃⁻ played the most significant inhibition effects on phenol removal through reducing the phenol removal rate from 89.6% to 77.9%. This work provides guidance for the design of high active and stable carbon materials that activate PS to remove phenol.

In spring, ozone (O₃) pollution frequently occurrs in eastern China, but key drivers remain uncertain. In this study, interannual variations in springtime ozone in Shanghai, China, from 2013 to 2021, were investigated to assess the health impacts and the effectiveness of recent air pollution control measures. A combination of ground-level measurements of regulated air pollutants, lidar observations, and backward trajectories of air masses was used to identify the key drivers for enhancing springtime O₃. The results show that external imports of O₃ driven by atmospheric circulation are notable sources of springtime surface O₃. For example, the downward transport from the free troposphere could contribute to over 50% of surface O₃ in the morning. The surface O₃ mixing ratios in spring exhibited an upward trend of 0.93 ppb yr⁻¹ (p < 0.05) from 2013 to 2021. The change in meteorological variables, particularly the increase in air temperature, could explain nearly 87% of the springtime O₃ upward trend. The change in anthropogenic emissions of precursors only contributed to a small fraction (<13%) of the increase in springtime O₃. The cumulative exposure of urban residents to O₃ in spring also exhibited a significant upward trend (111 ppb yr⁻¹, p < 0.05). With the rapid increase in surface O₃, premature respiratory mortality attributable to O₃ exposure has fluctuated at approximately 2933 deaths per year since 2016, even though the total deaths from respiratory diseases have significantly declined. Long-term exposure to high O₃ concentrations is a significant contributor to premature respiratory mortality.

Microplastics (MPs) have become an emerging threat for organisms. However, the toxicity mechanisms on biota, especially soil biota remain largely unclear. This study distinguished the effects of five types of MPs and their extractable additives on a typical soil oligochaete Enchytraeus crypticus using a traditional ecotoxicological approach combined with gut microbiota analysis. A variety of inorganic and organic compounds were screened in extractable solutions. Both MPs and their extractable additives decreased the growth and survival rates of the worms and shifted the gut microbiota, and the effects were type-specific. The differences between the effects of MPs and their extractable additives on traditional ecotoxicological parameters were insignificant, suggesting that extractable additives were the main toxicity pathways on soil fauna. The type-specific effects of MPs were attributed to the varied chemical compositions of extractable additives, and the compounds responsible for the shift of gut microbiota were further identified. The distinguishable effects on gut microbiota between MPs and their extractable additives together with the significant regressions between gut microbiota and traditional ecotoxicological parameters confirmed that gut microbiota could be a more sensitive indicator of organism's health conditions. Combined, the study provided an important insight into the toxicity mechanisms of MPs on soil fauna and extractable additives of MPs may be a hidden threat.

Zinc oxide nanoparticles (ZnO NPs) and its sulfidized form (ZnS NPs) are increasingly entering into freshwater systems through multiple pathways. However, their impacts on the composition and function of sedimentary microbial communities are still largely unknown. Here, two kinds of lake-derived microcosms were constructed and incubated with ZnO NPs, or ZnS NPs to investigate the short-term (7 days) and long-term (50 days) impacts on sedimentary microbial communities and nitrogen cycling. After 7 days, both ZnO NPs and ZnS NPs dosed microbial communities experienced distinct alterations as compared to the undosed controls. By day 50, the structural shifts of microbial communities caused by ZnO NPs were significantly enlarged, while the microbial shifts induced by ZnS NPs were largely resolved. Additionally, ZnO NPs and ZnS NPs could significantly alter nitrogen species and nitrogen cycling genes in sediments, revealing their non-negligible impacts on nitrogen cycling processes. Furthermore, our data clearly indicated that the impacts of ZnO NPs and ZnS NPs on nitrogen cycling differed distinctly in different lake-derived microcosms, and the impacts were significantly correlated with microbial community structure. Overall, this research suggests that the entrance of pristine or sulfidized ZnO NPs into freshwater systems may significantly impact the sedimentary microbial community structure and nitrogen cycling.

The exposure of Caenorhabditis elegans to polystyrene (PS) beads of a wide range of sizes impedes feeding, by reducing food consumption, and has been linked to inhibitory effects on the reproductive capacity of this nematode, as determined in standardized toxicity tests. Lipid storage provides energy for longevity, growth, and reproduction and may influence the organismal response to stress, including the food deprivation resulting from microplastics exposure. However, the effects of microplastics on energy storage have not been investigated in detail. In this study, C. elegans was exposed to ingestible sizes of PS beads in a standardized toxicity test (96 h) and in a multigeneration test (∼21 days), after which lipid storage was quantitatively analyzed in individual adults using coherent anti-Stokes Raman scattering (CARS) microscopy. The results showed that lipid storage distribution in C. elegans was altered when worms were exposed to microplastics in form of PS beads. For example, when exposed to 0.1-μm PS beads, the lipid droplet count was 93% higher, the droplets were up to 56% larger, and the area of the nematode body covered by lipids was up to 79% higher than in unexposed nematodes. The measured values tended to increase as PS bead sizes decreased. Cultivating the nematodes for 96 h under restricted food conditions in the absence of beads reproduced the altered lipid storage and suggested that it was triggered by food deprivation, including that induced by the dilutional effects of PS bead exposure. Our study demonstrates the utility of CARS microscopy to comprehensively image the smaller microplastics (<10 μm) ingested by nematodes and possibly other biota in investigations of the effects at the level of the individual organism.

GABA, a four-carbon non-protein amino acid, plays an important role in animals and plants. We previously found GABA could alleviate alkali stress in apple seedlings. However, its physiological mechanism under heavy metal cadmium (Cd) stress need to be further studied. Thus, we explored its biological role in response to Cd stress. It was verified that 0.5 mM GABA could effectively alleviate Cd toxicity. Using NMT technique, we found that exogenous GABA could significantly reduce the net Cd²⁺ fluxes in apple roots, and Cd content was significantly lower than that in roots under Cd stress. Further analysis indicated exogenous GABA could significantly reduce the expression of genes related to the uptake and transport of Cd in apples under Cd stress. In addition, exogenous GABA could significantly increase the content of amino acids in apple roots under Cd stress. GAD is a key enzyme in GABA synthesis, we obtained transgenic apple roots of overexpression MdGAD1. Compared with the control, transgenic roots accumulated less Cd, maintained lower Cd uptake by roots, and lower expression of related transport genes. These results showed that GABA could effectively alleviate Cd toxicity in apple seedlings and provide a new perspective of GABA to alleviate Cd stress.

The first investigation of microplastics in G. affinis from the Brantas River was carried out in this study. Microplastics were found at higher concentrations in gambusia fish captured downstream (209.18 ± 48.85 particles/gram) than upstream (24.44 ± 0.14 particles/gram). Microplastic particle concentrations in G. affinis have a positive linear relationship with fish length. The fiber was the most prominent shape at Sites 1 and 2, whereas the fragment was dominant at Sites 3 and 4. With a value of 45–48%, black dominates the entire site, followed by blue (29–38%), transparent colors (7–11%), red (2–4%), purple (1–3%), and other colors (5–7%). Microplastics measuring <0.1 mm are commonly found in fish bodies. Cyclohexylmethyl octyl ester (phthalic acid) is the most abundant component found in microplastics, accounting for 30.11% of the total. This study provides evidence that G. affinis can be used to monitor the presence of microplastic pollution in the Brantas River but further studies are needed regarding the effects of microplastics and their health hazards on fish.
