Lead tolerance and physiological adaptation mechanism in roots of accumulating and non-accumulating ecotypes of Sedum alfredii Полный текст

BACKGROUND, AIM AND SCOPE: Lead (Pb) accumulation in soils affects plants primarily through their root systems. The aim of this study was to investigate early symptoms of the loss of membrane integrity and lipid peroxidation in root tissues and physiological adaptation mechanism to Pb in accumulating ecotypes (AE) and non-accumulating ecotypes (NAE) of Sedum alfredii under Pb stress in hydroponics. METHODS AND RESULTS: Histochemical in situ analyses, fluorescence imaging, and normal physiological analysis were used in this study. Pb accumulation in roots of both AE and NAE increased linearly with increasing Pb levels (0–200 μM), and a significant difference between both ecotypes was noted. Both loss of plasma membrane integrity and lipid peroxidation in root tissues became serious with increasing Pb levels, maximum tolerable Pb level was 25 and 100 μM for NAE and AE, respectively. Pb supplied at a toxic level caused a burst of reactive oxygen species (ROS) in root cells in both ecotypes. However, the root cells of AE had inherently higher activities of superoxide dismutase (SOD), guaiacol peroxidase (POD), and lipoxygenase (LOX) in control plants, and the induction response of these antioxidant enzymes occurred at lower Pb level in AE than NAE. AE plants maintained higher ascorbic acid and H2O2 concentrations in root cells than NAE when exposed to different Pb levels, and Pb induced more increase in dehydroascorbate (DHA), catalase (CAT), and ascorbate peroxidase (APX) in AE than NAE roots. DISCUSSION AND CONCLUSION: Results indicate that histochemical in situ analyses of root cell death and lipid peroxidation under Pb short-term stress was sensitive, reliable, and fast. Higher tolerance in roots of accumulating ecotype under Pb stress did depend on effective free oxygen scavenging by making complex function of both constitutively higher activities and sensitive induction of key antioxidant enzymes in root cells of S. alfredii.

The aim of this study was to demonstrate that zebrafish embryos subjected to a priming exposure provided by one environmental stressor (low-dose alpha particles) can induce an adaptive response against a subsequent challenging exposure provided by another environmental stressor (heavy metal Cd). The effect thus identified would be an antagonistic multiple stressor effect. The effects of alpha particle radiation and/or Cd on whole embryos were studied through quantification of apoptotic signals at 24 h post-fertilization (hpf). Embryos were stained with the vital dye acridine orange, followed by counting the stained cells. For each set of experiments, 30 dechorionated embryos were divided into three groups, each having ten embryos. The three groups of embryos were referred to as (A) the control group, which received no more further treatments after dechorionation, (B) Cd-treated group, which did not receive any priming exposure and would receive a challenging exposure at 10 hpf and (C) (alpha + Cd)-treated group, which would receive both priming and challenging exposures. We defined the normalized net number of apoptotic signals in the (alpha + Cd)-treated group as N C * = [(apoptotic signals for (alpha + Cd)-treated group − average apoptotic signals for the corresponding control group)/average apoptotic signals for the corresponding control group] and that in the Cd-treated group as N B* = [(apoptotic signals for Cd-treated group − average apoptotic signals for the corresponding control group)/ average apoptotic signals for the corresponding control group]. By using the non-parametric Mann–Whitney U statistic, we were able to show that N C * was significantly smaller than N B *(p = 0.006). These demonstrated an antagonistic multiple stressor effect between ionizing radiation and Cd through the induction of an adaptive response by the ionizing radiation against subsequent exposures to Cd.

BACKGROUND, AIM, AND SCOPE: The purpose of this study was to monitor and present the heavy metal concentrations in the blood of residents of areas near municipal waste incinerators (MWIs), who are more prone to environmental pollution. We also sought to compare and analyze the residents’ perception of environmental pollution as one of the factors affecting heavy metal concentrations in the blood using a survey about the perceived damage caused by the facilities. Since heavy metal levels in the blood can be affected not only by local environmental pollution but also by personal and occupational factors, heavy metal levels in the blood need to be verified and consistently monitored. METHODS: Residents who live within 300 m of MWIs in Seoul are acknowledged to be under indirect influence according to the Waste Disposal Act. A survey was given to 841 residents living within 300 m of a MWI from 2006 to 2009. The concentrations of heavy metals (lead, cadmium, and mercury) in the blood were measured in the 841 surveyed residents and in 105 residents in reference areas. Additionally, the perception of the damage caused by municipal waste incinerators was investigated using scores from 1 to 5 on a Likert scale. RESULTS: The measurements of the heavy metal concentrations in the blood showed that the mean concentrations of lead, cadmium, and mercury were 43.1, 1.7, and 1.3 ug/L, respectively. The blood levels of lead and cadmium were slightly higher in the group of the subjects who had resided the longest near the municipal waste incinerators. When compared with the domestic investigation by the Ministry of Environment, the concentrations of lead and cadmium were a little higher, while that of mercury was a little lower. Overall, there was no significant difference in the distribution of heavy metal levels in the blood among age groups. BACKGROUND, AIM AND SCOPE: The breast milk has been recommended to carry out as a monitoring tool for effectiveness evaluation of the Stockholm Convention on Persistent Organic Pollutants (POPs). Polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloro-ethane (DDT) and its metabolites (DDX), hexachlorbenzene (HCB) and isomers of hexachlocyklohexane (HCHs) have been monitored in the breast milk of nursing mothers in the Czech Republic since 1994 as a part of The Environmental Health Monitoring System. Knowledge about long-term POPs distribution and accumulation in the human body is crucial to understanding uptake, degradation and subsequent effects as well as to conduct risk assessments. The main aim of this study is to evaluate 15-years long-term trends of selected POPs in human milk in the Czech Republic and to elucidate the questionnaire information about the age, parity and social habits, to the final concentrations. This effectiveness evaluation of POPs restriction is quite precisely after 15-years monitoring campaigns. MATERIALS, METHODS AND RESULTS: The human milk samples (4,753 samples) were analysed for a number of chlorinated organic chemicals including PCBs and selected chlorinated pesticides (OCPs, HCB, HCHs, DDX). The relative change of concentration per year for all chemicals was analysed. The remaining percentages of POPs in breast milk in comparison to 1994 are also expressed. Czech population half-lives of POPs in breast milk, derived from either linear or exponential models were computed. CONCLUSIONS AND PERSPECTIVES: The long-term data indicates a continuation of a decreasing trend of POPs concentrations on breast milk. Our study did not confirm lactation and parity as an important outflux resulting in the decrease in concentrations in mothers, which is in the antagonism with most of the studies. The higher BMI was associated with higher amounts of HCB and lower amounts of higher chlorinated PCBs. The results confirm the effectiveness of restrictions of POPs usage in the Czech Republic. This ongoing long-term study is very useful tool for parametric effectiveness evaluation of Stockholm Convention.

BACKGROUND: This work focuses on the accumulation and mobility properties of arsenic (As) and the effects of phosphate (P) on its movement in Pennisetum clandestinum Hochst (kikuyu grass), grown hydroponically under increasing arsenate (As(V)) concentrations. The uptake of both ions and the relative kinetics show that phosphate is an efficient competitive inhibitor of As(V) uptake. The P/As uptake rate ratios in roots indicate that P is taken up preferentially by P/As transporters. An arsenite (As(III)) efflux from roots was also found, but this decreased when the arsenate concentration in the solution exceeded 5 μM. METHODS: Increases in both arsenite and arsenate concentrations in roots were observed when the arsenate concentration in the solution was increased, and the highest accumulation of As(III) in roots was found when plants were grown at 5 μM As(V). The low ratios of As accumulated in shoots compared to roots suggest limited mobility of the metalloid within Kikuyu plants. RESULTS: The results indicate that arsenic resistance in kikuyu grass in conditions of moderate exposure is mainly dependent on the following factors: 1) phosphate nutrition: P is an efficient competitive inhibitor of As(V) uptake because of the higher selectivity of membrane transporters with respect to phosphate rather than arsenate; and 2) a detoxification mechanism including a reduction in both arsenate and arsenite root efflux. CONCLUSIONS: The As tolerance strategy of Kikuyu limits arsenate uptake and As translocation from roots to shoots; therefore, this plant cannot be considered a viable candidate for use in the phytoextraction of arsenic from contaminated soils or water.

INTRODUCTION: The accelerated biodegradation of 3-nitrophenol (3-NP) in the rhizosphere of giant duckweed (Spirodela polyrrhiza) was investigated. MATERIALS AND METHODS: Biodegradation of 3-nitrophenol in the rhizosphere of a floating aquatic plant, S. polyrrhiza, was investigated by using three river water samples supplemented with 10 mg l−1 of 3-NP. Isolation and enrichment culture of 3-NP-degrading bacteria were performed in basal salts medium containing 3-NP (50 mg l−1). The isolated strains were physiologically and phylogenetically characterized by using an API20NE kit and 16S rRNA gene sequencing. RESULTS AND DISCUSSION: Accelerated removal of 3-NP (100%) was observed in river water samples with S. polyrrhiza compared with their removal in plant-free river water. Also, 3-NP persisted in an autoclaved solution with aseptic plants, suggesting that the accelerated 3-NP removal resulted largely from degradation by bacteria inhabiting the plant rather than from adsorption and uptake by the plant. We successfully isolated six and four strains of 3-NP-degrading bacteria from the roots of S. polyrrhiza and plant-free river water, respectively. Phylogenetic analysis based on 16S rRNA gene divided the 3-NP-degrading bacteria into two taxonomic groups: the genera Pseudomonas and Cupriavidus. The strains belonging to the genus Cupriavidus were only isolated from the roots of duckweed. All strains isolated from the roots utilized 3-NP (0.5 mM) as a sole carbon and energy source, indicating that they could have contributed to the accelerated degradation of 3-NP in the rhizosphere of S. polyrrhiza. CONCLUSIONS: The rhizoremediation using S. polyrrhiza and its rhizosphere bacteria can be an effective strategy for cleaning up the 3-NP-contaminated surface waters.

PURPOSE: Increases in dissolved organic carbon (DOC) concentrations have been reported in surface waters worldwide in the last 10 to 20 years. The causes behind these increases have been attributed to many factors, including climate change and decreasing depositions of atmospheric sulphate ([Formula: see text]). Trends in DOC concentrations and their potential causal factors were examined in a network of 30 lakes lying in undisturbed temperate and boreal catchments in the province of Quebec, Canada. METHODS: Temporal trends in lake DOC concentrations were analysed with the seasonal Kendall test. For each lake, the variation in DOC concentration over time was assessed in light of the variation in [Formula: see text] concentration in precipitation, air temperature, precipitation and solar radiation using the forward stepwise multiple regression. RESULTS: Between 1989 and 2006, significant increases in DOC were observed in most of the lakes, the mean rate of change being 0.05 mg L−1 year−1. Lake DOC concentrations were significantly explained by different models that yielded a variance explanation ranging from 13% to 77%. The models included long-term temperature variables (i.e. averaged over intervals of 10 years), short-term precipitation variables (i.e. summed over intervals 6 months), radiation (i.e. ice-free period prior to the DOC observation) and precipitation [Formula: see text] concentration as explanatory variables. CONCLUSION: Temporal changes in DOC concentrations seem more consistent with the evolution of climate parameters rather than [Formula: see text] concentrations despite the fact that most lakes were in the process of recovery, showing increases in pH.

INTRODUCTION: From December 2008 to November 2009, an investigation of water quality was performed in the 70-km long downstream from Gezhouba Dam in Yangtze River. METHODS: Twelve sites in all were chosen. Nine parameters of water quality including the total phosphorus, the total nitrogen, chlorophyll a (Chl.a), nitrite, nitrate, ammonia, water temperature, DO, and pH were monitored almost monthly. The multivariate statistical technique (cluster analysis) and the nonparametric method (Kruskal–Wallis Test and Spearman’s rank correlation) were applied to evaluate the spatiotemporal variations of water quality data sets. RESULTS AND DISCUSSION: According to the Chinese environmental quality standards for surface water (GB3838-2002), the water quality in the river section investigated can attain to the III water standards except total nitrogen. Further analysis indicated that there were no significant spatial differences in these parameters of water quality, but the sampling date had a significant effect. The temporal variation of water quality can be related to the discharge of Gezhouba Dam and moreover be affected by the reservoir regulation. During the discharge, the discharge increased the concentration of total phosphorus and then decreased the N:P ratio, which helps to the occurrence of algae blooms. The high consternation of phosphorus and the low N:P ratio show that the water body can be in the process of eutrophication during the discharge of Gezhouba Dam. In fact, Chl.a had begun to rise in the same period.

Perfluorinated compounds (PFCs) are persistent and bioaccumulative organic compounds used as additives in many industrial products. After use, these compounds enter wastewater treatment plants (WWTP) and long-chain PFCs are primarily accumulated in sludge. The aim of this study was to determine the occurrence and behavior of five PFCs in sludge from 15 WWTP from Spain and Germany that receive both urban and industrial wastes. The PFCs studied were perfluorooctanesulfonate (PFOS), perfluorohexanesulfonate (PFHxS), perfluorobutanesulfonate (PFBS), perfluorooctanoic acid (PFOA), and perfluorononanoic acid (PFNA). One gram of freeze-dried, sieved, and homogenized sludge was extracted using an ultrasonic bath with methanol and glacial acetic acid. After that, the extract was recovered and evaporated to dryness with a TurboVap and then 1 mL of acetonitrile was added and the extract was cleaned up with black carbon. Liquid chromatography coupled to mass spectrometry operated in selected reaction monitoring was used to determine target compounds. Quality parameters are provided for the set of compounds studied. PFCs were detected in all samples. In Spanish sludge, ∑PFC ranged from 0.28 to 5.20 ng/g dry weight (dw) with prevalence of PFOS, while in German sludge, ∑PFC ranged from 20.7 to 38.6 ng/g dw and PFBS was the dominant compound. As a next step, the evolution of PFC concentrations within the sludge treatment steps (primary sludge, anaerobic digested sludge, and centrifuged sludge) was evaluated and differences among levels and patterns were observed and were attributed to the influent water quality and treatment used. Finally, we estimated the amount of PFCs discharged via sludge in order to determine the potential impact to the environment according to different sludge usage practices in the two regions investigated. This manuscript provided an intra-European overview of PFC distribution in sludge. Levels and compound distribution depend on the WWTP sampled. This study demonstrates that PFCs are persistent to sludge treatment and the loads in sludge may pose a future environmental risk, if not controlled.

PURPOSE: This study aims to examine whether addition of immobilising agents to a sandy, alkaline (pH = 8.1) soil, which had been contaminated with Pb and Zn by airborne particles from a Pb/Zn smelter, would substantially reduce metal bioavailability. METHODS: The effectiveness of five waste materials (blast furnace (BF) slag, alum water treatment (WT) sludge, red mud, sugar mill mud and green waste compost) as metal immobilising agents was evaluated by incubating them with a contaminated soil for a period of 12 months at rates of 5% and 10% (w/w), after which, Rhodes grass was grown in the soils in a greenhouse study. RESULTS: Additions of WT sludge, BF slag and red mud reduced CaCl2, CH3COOH, HCl and EDTA-extractable Zn but compost and mill mud had no appreciable immobilising effects. Additions of all amendments reduced levels of CaCl2, CH3COOH and HCl-extractable Pb although concentrations of EDTA-extractable Pb remained unchanged. A sequential extraction procedure showed that additions of mill mud and compost increased the percentage of total Pb and Zn present in the oxidisable fraction whilst additions of the other materials increased the percentage present in the residual fraction. Rhodes grass yields were promoted greatly by additions of red mud, compost and particularly mill mud, and yields were negatively correlated with tissue Pb concentrations and extractable Pb. CONCLUSIONS: Red mud was the most effective material for lowering extractable Pb and Zn levels simultaneously while mill mud and compost were notably effective for Pb. A field evaluation in the study area is justified.
