Bacterial colonisation of plastic in the Rockall Trough, North-East Atlantic: An improved understanding of the deep-sea plastisphere

Plastic pollution has now been found within multiple ecosystems across the globe. Characterisation of microbial assemblages associated with marine plastic, or the so-called ‘plastisphere’, has focused predominantly on plastic in the epipelagic zone. Whether this community includes taxa that are consistently enriched on plastic compared to surrounding non plastic surfaces is unresolved, as are the ecological implications. The deep sea is likely a final sink for most of the plastic entering the ocean, yet there is limited information on microbial colonisation of plastic at depth. The aim of this study was to investigate deep-sea microbial communities associated with polystyrene (PS) and polyurethane (PU) with Bath stone used as a control. The substrates (n = 15) were deployed in the Rockall Trough (Atlantic), and recovered 420 days later from a depth of 1796 m. To characterise the bacterial communities, 16S rRNA genes were sequenced using the Illumina MiSeq platform. A dominant core microbiome (taxa shared across all substrates) comprised 8% of total ASVs (amplicon sequence variant) and accounted for 92% of the total community reads. This suggests that many commonly reported members of the plastisphere are simply opportunistic which freely colonise any hard surface. Transiently associated species consisted of approximately 7% of the total community. Thirty genera were enriched on plastic (P < 0.05), representing 1% of the total community. The discovery of novel deep-sea enriched taxa included Aurantivirga, Algivirga, IheB3-7, Spirosoma, HTCC5015, Ekhidna and Calorithrix on PS and Candidatus Obscuribacter, Haloferula, Marine Methylotrophic Group 3, Aliivibrio, Tibeticola and Dethiosulfatarculus on PU. This small fraction of the microbiome include taxa with unique metabolic abilities and show how bacterial communities can be shaped by plastic pollution at depth. This study outlines a novel approach in categorising the plastisphere to elucidate the ecological implications of enriched taxa that show an affinity for colonising plastic.

Neonicotinoids pollution poses a serious threat to aquatic ecosystems. However, there is currently little knowledge about how neonicotinoids are transferred from the agricultural environment to the aquatic environment. Here, we conducted in situ high-frequency monitoring of neonicotinoids in soil-water systems along the hydrological flow path during rainfall to explore the horizontal and vertical transport mechanisms of neonicotinoids. The collected samples included 240 surface runoff, 128 subsurface runoff, 60 eroded sediment, 120 soil and 144 soil solution, which were used to analyse neonicotinoids concentrations. Surface runoff, subsurface runoff and eroded sediment were the three main paths for the horizontal migration of neonicotinoids. In the CK (citrus orchards without grass cover) and grass-covered citrus orchards, there are 15.89% and 2.29% of the applied neonicotinoids were transported with surface runoff, respectively. While in the CK and grass-covered citrus orchards, there are only 1.23% and 0.19% of the applied neonicotinoids were transported with eroded sediment and subsurface runoff. Although the amount of neonicotinoids lost along with eroded sediment was small, the concentration of neonicotinoids in eroded sediment was two orders of magnitude higher than the concentration of neonicotinoids in sediments of the surface water. Meanwhile, neonicotinoids migrated vertically in soil due to water infiltration. In the CK and grass-covered citrus orchards, there are 57.64% and 24.36% of the applied neonicotinoids were retained in soil and soil solution, respectively, and their concentration decreased as soil depth increased. Another noteworthy phenomenon is that more neonicotinoids migrated to deeper soil layers under grass cover compared with no grass cover because grass roots promoted the formation of cracks and vertical preferential flow. Our results are expected to improve the accuracy of neonicotinoids pollution prediction by considering migration paths, including surface and subsurface runoff and eroded sediment.

Knowledge of the ecology and at-sea distribution of migratory species like seabirds has substantially increased over the last two decades. Furthermore, an increasing number of studies have recently focused on chemical contamination of birds over their annual cycle. However, the understanding of the combined effects of spatial movements and contamination on seabirds’ life-history traits is still scarce. During winter, seabirds can use very different areas, at the large-scale. Such overwintering strategies and distribution may expose individuals to contrasting environmental stressors, including pollutants. Here, we studied the winter distribution and contamination with mercury (Hg), and their combined effects on reproduction, in a great skua (Stercorarius skua) population breeding in Bjørnøya, Svalbard. We confirmed that individuals of this specific population overwinter in three different areas of the North Atlantic, namely Africa, Europe and northwest Atlantic. The highest Hg concentrations in feathers were measured in great skuas wintering off Europe (Linear Mixed Models - mean value ± SD = 10.47 ± 3.59 μg g ⁻¹ dw), followed by skuas wintering in northwest Atlantic (8.42 ± 3.70) and off Africa (5.52 ± 1.83). Additionally, we found that female winter distribution and accumulated Hg affected the volume of their eggs (Linear Mixed Models), but not the number of laid and hatched eggs (Kruskal-Wallis tests). This study provides new insights on the contamination risks that seabirds might face according to their overwinter distribution and the possible associated carry-over effects.

It is of great significance to explore the remediation pattern in actual heavy metal (HM) contaminated sites. The field trial was carried out to research the remediation effect of biochar near a lead-zinc smelter in Feng County, China, under the rotation condition of different crops. This kind of cultivation mode is very representative in northern of China. And the pattern of production and restoration is suitable for scarce land resources and large food demand in China. The changes of soil physiochemical properties with the biochar addition, crop growth and the accumulated HMs by crops were focused on. The results showed the biochar application was excellent in improving soil nutrient elements and crop growth. The contents of TK were more obvious than those of TN and TP, with an increase of 2.6%–28.2% compared with the controls (without the addition of biochar). The yield of first season crops, i.e., soybean and corn, increased by 30%–42% and 34%–61%, respectively, and the second season crops, i.e., rape and wheat, with the increment of 25%–41% and 9%–29%, respectively. The availability forms of Cd and Pb decreased by 1.07–10.0% and 2.92–8.35%, respectively. While the improvement on the status of the HMs accumulated by crops was disappointing. The contents of HMs accumulated by crops increased to varying degrees (e.g., Pb and Zn in root, Cu and Pb in grain, and Cd in stems and leaves). Moreover, the concentrations of HMs in seeds of crops were higher than the limited levels given by the Chinese directive. Considering the results of the study and food safety, it is suggested to change the nature of the land around the smelter into woodland or construction land to prohibit the cultivation of food crops in this area.

2,4,6-trinitrotoluene (TNT) is a highly toxic explosive that contaminates soil and water and may interfere with the degradation of co-occurring compounds, such as hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX). We proposed that TNT may influence RDX-degrading bacteria via either general toxicity or a specific effect on the |RDX degradation mechanisms. Thus, we examined the impact of TNT on RDX degradation by Rhodococcus strains YH1, T7, and YY1, which were isolated from an explosives-polluted environment. Although partly degraded, TNT did not support the growth of any of the strains when used as either sole carbon or sole nitrogen sources, or as carbon and nitrogen sources. The incubation of a mixture of TNT (25 mg/l) and RDX (20 mg/l) completely inhibited RDX degradation. The effect of TNT on the cytochrome P450, catalyzing RDX degradation, was tested in a resting cell experiment, proving that TNT inhibits XplA protein activity. A dose-response experiment showed that the IC50/trans values for YH1, T7, and YY1 were 7.272, 5.098, and 9.140 (mg/l of TNT), respectively, illustrating variable sensitivity to TNT among the strains. The expression of xplA was also strongly suppressed by TNT. Cells that were pre-grown with RDX (allowing xplA expression) and incubated with ammonium chloride, glucose, and TNT, completely transformed into their amino dinitrotoluene isomers and formed azoxy toluene isomers. The presence of oxygen-insensitive nitroreductase that enable reduction of the nitro group in the presence of O2 in the genomes of these strains suggests that they are responsible for TNT transformation in the cultures. The experimental results concluded that TNT has an adverse effect on RDX degradation by the examined strains. It inhibits RDX degradation due to the direct impact on cytochrome P450, xplA, or its expression. The tested strains can transform TNT independently of RDX. Thus, degradation of both compounds is possible if TNT concentrations are below their IC50 values.

Soil chemistry of toxic metalloids and metals differs, making their simultaneous removal difficult. Soil contaminated with As, Pb, Zn and Cd was washed with oxalic acid, Na-dithionite and EDTA solution. Toxic elements were removed from the washing solution by alkalinisation with CaO to a pH 12.5: As was co-precipitated with Fe from Fe-EDTA chelate formed after the soil washing. The toxic metals precipitated after substitution of their EDTA chelates with Ca. The novel method was scaled up on the ReSoil® platform. On average, 60, 76, 29, and 53% of As, Pb, Zn, and Cd were removed, no wastewater was generated and EDTA was recycled. Addition of zero-valent iron reduced the toxic elements’ leachability. Remediation was most effective for As: phytoaccessibility (CaCl₂ extraction), mobility (NH₄NO₃), and accessibility from human gastric and gastrointestinal phases were reduced 22, 104, 6, and 51 times, respectively. Remediation increased pH but had no effect on soil functioning assessed by fluorescein diacetate hydrolysis, dehydrogenase, β-glucosidase, urease, acid and alkaline phosphatase activities. Brassica napus produced 1.9 times more biomass on remediated soil, accumulated no As and 5.0, 2.6, and 9.0 times less Pb, Zn and Cd, respectively. We demonstrated the novel remediation technology as cost-efficient (material cost = 41.86 € t⁻¹) and sustainable.

Rice paddy fields are major sources of atmospheric methane (CH₄) and nitrous oxide (N₂O). Rice variety is an important factor affecting CH₄ and N₂O emissions. However, the interactive effects of rice metabolites and microorganisms on CH₄ and N₂O emissions in paddy fields are not clearly understood. In this study, a high greenhouse gas-emitting cultivar (YL 6) and a low greenhouse gas-emitting cultivar (YY 1540) were used as experimental materials. Metabolomics was used to examine the roots, root exudates, and bulk soil metabolites. High-throughput sequencing was used to determine the microbial community composition. YY 1540 had more secondary metabolites (flavonoids and isoflavonoids) in root exudates than YL 6. It was enriched with the uncultured members of the families Gemmatimonadanceae and Rhizobiales_Incertae_Sedis in bulk soil, and genera Burkholderia-Caballeronia-Paraburkholderia, Magnetospirillum, Aeromonas, and Anaeromyxobacter in roots, contributing to increased expression of pmoA and nosZ genes and reducing CH₄ and N₂O emissions. YL 6 roots and root exudates contained higher contents of carbohydrates [e.g., 6-O- acetylarbutin and 2-(3- hydroxyphenyl) ethanol 1′-glucoside] than those of YY 1540. They were enriched with genera RBG-16-58-14 in bulk soil and Exiguobacterium, and uncultured member of the Kineosporiaceae family in roots, which contributed to increased expression of mcrA, ammonia-oxidizing archaea, ammonia-oxidizing bacteria, nirS, and nirK genes and greenhouse gas emissions. In general, these results established a link between metabolites, microorganisms, microbial functional genes, and greenhouse gas emissions. The metabolites of root exudates and roots regulated CH₄ and N₂O emissions by influencing the microbial community composition in bulk soil and roots.

Antibiotics accumulation in the environment has given rise to multi-drug resistant 'superbugs' and antibiotics resistence genes (ARGs). Chloramphenicol (CAP), a kind of widely used antibiotics, was chosen as the model compound to investigate its degradation during electrochemical treatment process. The prepared Ti/PbO₂–La electrodes had a denser surface and a more complete PbO₂ crystal structure than Ti/PbO₂ electrode. The doping of La increased the onset potential and the overpotential, increased the current value of the oxidation peak and the reduction peak, reduced the impedance, and increased the lifetime. The reactions CAP degradation and TOC removal on Ti/PbO₂–La electrode was both primary kinetic reactions. CAP degradation rate increased with current density, and TOC obtained the highest removal at current density of 25 mA cm⁻². The electrolyte concentration had a small effect in the range of 0.050–0.150 mol L⁻¹. The effects under acidic and neutral conditions were better than under alkaline conditions. CAP was mainly directly oxidized at the electrode surface and indirect oxidation also took place via generated ·OH and SO₄·⁻. 15 intermediates and 2 degradation pathways have been postulated. The entry of CAP and CAP intermediates into the environment caused the alteration in bacterial community and ARGs, while complete degradation products had little effect on them. Redundancy analysis showed that intI1 was the dominant factor affecting ARGs, and Actinobacteria and Patescibacteria were the main factors affecting the abundances of ARGs in the microbial community.

Nowadays, the accumulation of toxic heavy metals in soil and water streams is considered a serious environmental problem that causes various harmful effects on plants and animals. Phytoremediation is an effective, green, and economical bioremediation approach by which the harmful heavy metals in the contaminated ecosystem can be detoxified and accumulated in the plant. Hyperaccumulators exude molecules called transporters that carry and translocate the heavy metals present in the soil to different plant parts. The hyperaccumulator plant genes can confine higher concentrations of toxic heavy metals in their tissues. The efficiency of phytoremediation relies on various parameters such as soil properties (pH and soil type), organic matters in soil, heavy metal type, nature of rhizosphere, characteristics of rhizosphere microflora, etc. The present review comprehensively discusses the toxicity effect of heavy metals on the environment and different phytoremediation mechanisms for the transport and accumulation of heavy metals from polluted soil. This review gave comprehensive insights into plants tolerance for the higher heavy metal concentration their responses for heavy metal accumulation and the different mechanisms involved for heavy metal tolerance. The current status and the characteristic features that need to be improved in the phytoremediation process are also reviewed in detail.

Mercury is a highly toxic and highly mobile heavy metal. It has been regarded as more toxic than other nonessential and toxic nonradioactive heavy metals. Moreover, it has a high tendency of bioaccumulation and biomagnification in the ecosystem. This study aimed to assess the environmental and health risks related to Hg. Seventy studies related to Hg in environmental media, aquatic biota, and food stuffs across Pakistan were reviewed, and their concentrations were used for ecological and human health risk assessments. High concentrations of Hg were reported in the environment, with maximum concentrations of 72 mg L⁻¹, 144 mg kg⁻¹, 887 mg kg⁻¹, and 49,807 ng m⁻³ in surface water, surface soil, surface sediments, and urban atmosphere, respectively. The possible non-carcinogenic health risk (hazard quotient) of Hg was assessed in soil, water, and fish. High risks were calculated for seafood and vegetable consumption, while low risks were estimated for soils and groundwater ingestion and exposure. Overall, children showed higher risks than adults. Last, the risk quotient analysis (RQ) revealed significant risks for aquatic species. RQs showed that multiple species, especially those with smaller resilience, could face long-term detrimental impacts. High, medium, and low risks were calculated from 66.66, 16.17, and 16.17% of the reported Hg concentrations.
