The integrated biomarker response revisited: optimization to avoid misuse

The growing need to evaluate the quality of aquatic ecosystems led to the development of numerous monitoring tools. Among them, the development of biomarker-based procedures, that combine precocity and relevance, is recommended. However, multi-biomarker approaches are often hard to interpret, and produce results that are not easy to integrate in the environmental policies framework. Integrative index have been developed, and one of the most used is the integrated biomarker response (IBR). However, an analysis of available literature demonstrated that the IBR suffers from a frequent misuse and a bias in its calculation. Then, we propose here a new calculation method based on both a more simple formula and a permutation procedure. Together, these improvements should rightly avoid the misuse and bias that were recorded. Additionally, a case study illustrates how the new procedure enabled to perform a reliable classification of site along a pollution gradient based on biomarker responses used in the IBR calculations.

Recent technical guidance has been published by the European Commission that outlines methodologies for the derivation of Environmental Quality Standards (EQS) in European surface waters under the Water Framework Directive (WFD). The guidance allows the derivation of a long-term EQS from a small dataset. Specifically an EQS can be derived from just three acute data points, although the safety factors built into such an EQS are large (e.g. up to a factor of 1,000). Large safety factors make such EQS uncertain, and often difficult to achieve in practice. We examine dataset requirements for the derivation of EQS and specifically the minimum number of tests needed for setting EQS for long-term chemical exposures that result in reduced relative uncertainty, as assessed simply through the reduction in standard deviation of the means of the values derived. Using ecotoxicity datasets for four example chemicals, for which EQS have been derived in many jurisdictions, we show that variation in the EQS is greatest when using the minimum dataset allowable under the WFD guidance, but decreases rapidly when seven or more datapoints are available. Increasing the minimum number of ecotoxicity data in deriving an EQS results in a greater understanding of ecotoxicological effects. With this knowledge, the mitigating effects of water chemistry can be accounted for in deriving an EQS, even with relatively limited datasets. The new guidance suggests “simplistic” approaches to account for chemical availability, but does not detail how this might be undertaken. We provide examples of ways by which water chemistry effects can be included in deriving implementable EQS for metals with relatively few reliable and relevant data.

Soil contamination with anthropogenic metals resulting from biosolid application is widespread around the world. To better predict the environmental fate and mobility of contaminants, it is critical to study the capacity of biosolid-amended soils to retain and release metals. In this paper, nickel adsorption onto a calcareous soil, a lime-stabilized biosolid, and soil–biosolid mixtures (30, 75, and 150 t biosolid/ha) was studied in batch experiments. Sorption experiments showed that (1) Ni adsorption was higher onto the biosolid than the calcareous soil, and (2) biosolid acted as an adsorbent in the biosolid–soil mixtures by increasing Ni retention capacity. The sorption tests were complemented with the estimation of Ni adsorption reversibility by successive applications of extraction solutions with water, calcium (100 mg/L), and oxalic acid (equivalent to 100 mg carbon/L). It has been shown that Ni desorption rates in soil and biosolid-amended soils were lower than 30 % whatever the chemical reagent, indicating that Ni was strongly adsorbed on the different systems. This adsorption/desorption hysteresis effect was particularly significant at the highest biosolid concentration (150 t/ha). Finally, an adsorption empirical model was used to estimate the maximum permissible biosolid application rate using French national guideline. It has been shown that desorption effects should be quantitatively considered to estimate relevant biosolid loadings.

Guangzhou is a metropolitan in south China with unique pollutants and geographic location. Unlike those in western countries and the rest of China, the appearance of haze in Guangzhou is often (about 278 days per year on average of 4 years). Little is known about the influence of these hazes on health. In this study, we investigated whether short-term exposures to haze and air pollution are associated with hospital admissions in Guangzhou. The relationships between haze, air pollution, and daily hospital admissions during 2008–2011 were assessed using generalized additive model. Studies were categorized by gender, age, season, lag, and disease category. In haze episodes, an increase in air pollutant emissions corresponded to 3.46 (95 % CI, 1.67, 5.27) increase in excessive risk (ER) of total hospital admissions at lag 1, 11.42 (95 % CI, 4.32, 18.99) and 11.57 (95 % CI, 4.38, 19.26) increases in ERs of cardiovascular illnesses at lags 2 and 4 days, respectively. As to total hospital admissions, an increase in NO₂was associated with a 0.73 (95 % CI, 0.11, 1.35) and a 0.28 (95 % CI, 0.11, 0.46) increases in ERs at lag 5 and lag 05, respectively. For respiratory illnesses, increases in NO₂was associated with a 1.94 (95 % CI, 0.50, 3.40) increase in ER at lag 0, especially among chronic obstructive pulmonary disease. Haze (at lag1) and air pollution (for NO₂at lag 5 and for SO₂at lag3) both presented more drastic effects on the 19 to 64 years old and in the females. Together, we demonstrated that haze pollution was associated with total and cardiovascular illnesses. NO₂was the sole pollutant with the largest risk of hospital admissions for total and respiratory diseases in both single- and multi-pollutant models.

The effects of wastewater from a mining and ore-dressing mill on fish in Lake Kostomukshskoe, which is used as a cesspool of circulating water and for storage of industrial wastes produced by the Kostomuksha mining and ore-dressing mill in northwest Russia, were studied. The lake is characterized by heavy mineralization, high pH, elevated levels of K⁺, Li⁺, SO₄²⁻, NO²⁻, Cl⁻, Li, Mn, and Ni, and the presence of a fine-dispersed mechanical suspension. To assess the impact of contamination on fish and determine the mechanisms of their adaptation, we investigated the biochemical indices and histology of the liver of whitefish (Coregonus lavaretus L.) and pike (Esox lucius L.) inhabiting Lake Kostomukshskoe, downstream Lake Koyvas (64° 47′ 30° 59′), and Lake Kamennoe, which is located in a nature preserve and has not been affected by anthropogenic activity (64° 28′ 30° 13′). Changes were detected in the activity of metabolic enzymes (cytochrome c oxidase (COX), lactate dehydrogenase, and glucose-6-phosphate dehydrogenase) in the liver. Specifically, the COX activity in the liver of both fish species from the contaminated lake decreased, indicating a low level of aerobic metabolism. Lipid infiltration was the most visible and widespread change observed in the liver of both fish species; therefore, it can be considered a marker of such long-term contamination. Lesions in pike liver demonstrated a wider range of severity than in those of whitefish. In summary, metabolic enzyme activity and histomorphology of the liver of whitefish and pike differed among lakes in a species-specific manner. The changes in enzyme activity and histomorphological alterations in fish that were observed can be applied for evaluation of freshwater systems that may be subjected to mineral pollution.

The effect of meteorological variables on surface ozone (O₃) concentrations was analysed based on temporal variation of linear correlation and artificial neural network (ANN) models defined by genetic algorithms (GAs). ANN models were also used to predict the daily average concentration of this air pollutant in Campo Grande, Brazil. Three methodologies were applied using GAs, two of them considering threshold models. In these models, the variables selected to define different regimes were daily average O₃ concentration, relative humidity and solar radiation. The threshold model that considers two O₃ regimes was the one that correctly describes the effect of important meteorological variables in O₃ behaviour, presenting also a good predictive performance. Solar radiation, relative humidity and rainfall were considered significant for both O₃ regimes; however, wind speed (dispersion effect) was only significant for high concentrations. According to this model, high O₃ concentrations corresponded to high solar radiation, low relative humidity and wind speed. This model showed to be a powerful tool to interpret the O₃ behaviour, being useful to define policy strategies for human health protection regarding air pollution.

Ectoparasitic arthropods are a diverse element of the Saudi fauna. Due to this, a survey of ectoparasites associated with rodents was conducted as a preliminary study in five districts of Hail region of northern Saudi Arabia for the first time. Ectoparasites extracted from 750 rodents were sampled and identified by recording their frequency of appearance. Results revealed that 1,287 ectoparasites infested 316 of the captured rodent hosts. These ectoparasites parasitized on four species of rodents including three species of rats Rattus rattus rattus, Rattus rattus frugivorus, and Rattus rattus alexandrinus and one species of mouse Acomys dimidiatus (Rodentia: Muridae). The ectoparasites belong to four different groups: ticks, fleas, lice, and mites. Ticks were the highest in the number, while fleas were the lowest among all the extracted ectoparasite groups. The collected ectoparasitic arthropods consisted of seven species. Ticks were of two species: Rhipicephalus turanicus and Rhipicephalus sanguineus (Acari: Ixodidae), fleas were of two species: Xenopsylla cheopis and Xenopsyllus conformis mycerini (Siphonaptera: Pulicidae), lice was a single species: Polyplax serrata (Anoplura: Hoplopleuridae), and mites were of two species: Laelaps nuttali and Laelaps echidninus (Mesostigmata: Laelapidae). The findings of the study showed that the intensity of infestation was varied between rodent host sexes, wherein females had the highest rate of parasitic infestation, and the parasitic index of appearance was very high for one group of parasites (i.e., ticks). The parasitic prevalence was 42.13 % on rodents, and mites were the most prevalent parasite species. Overall, this study was carried out to establish baseline data for ectoparasite-infested rodents in Hail region, Saudi Arabia, and may help for appropriate planning to control zoonotic diseases in this area.

A native Bacillus cereus isolate has been employed, for the first time, for simultaneous decolorization, dechlorination of chloroorganics, and Cr⁶⁺remediation from the real tannery effluent. Most of the physicochemical variables in 3:1 diluted effluent were well above the standard prescribed limits, which decreased substantially upon microbial treatment. The extent of bioremediation was better in diluted (3:1) as compared to undiluted effluent supplemented with nutrients and augmented with B. cereus isolate. Maximum growth, effluent decolorization (42.5 %), dechlorination (74.1 %), and Cr⁶⁺remediation (34.2 %) were attained with 4.0 % (v/v) inoculum, 0.8 % glucose, and 0.2 % ammonium chloride in 3:1 diluted effluent at natural pH (8.1) within 72 h of incubation. The efficiency of bioremediation in a bioreactor was higher as compared to a flask trial during 72 h of incubation: decolorization (47.9 %) was enhanced by 5.4 %, dechlorination (77.4 %) by 3.3 %, and Cr⁶⁺removal (41.7 %) by 7.5 % at an initial color of 286 Pt-Co units and initial concentration of 62 mg chloride ions and 108 mg l⁻¹Cr⁶⁺. Immobilized biomass of Pseudomonas putida and B. cereus coculture enhanced the extent of Cr⁶⁺remediation (51.9 %) by 10.2 % compared to the bioreactor trial. Chromate reductase activity and reduced Cr directly correlated and were mainly associated with soluble fraction of B. cereus plus effluent natural microflora. The GC-MS analyses revealed the formation of metabolites such as acetic acid and 2-butenoic acid in bacterially treated effluent. The supplementation of nutrients along with B. cereus augmentation is required for efficient effluent bioremediation.

Imidacloprid, a neonicotinoid insecticide, has been used widely in agriculture worldwide. The adverse effects of imidacloprid on exposed biota have brought it increasing attention. However, knowledge about the effects of imidacloprid on antioxidant defense systems and digestive systems in the earthworm is vague and not comprehensive. In the present study, the changes in the activity of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), cellulase, reactive oxygen species (ROS), and malondialdehyde (MDA) in the earthworm Eisenia fetida exposed to artificial soil treated with imidacloprid were examined systematically. The results showed that the activity of these biomarkers was closely related to the dose and duration of the exposure to imidacloprid. The activity of SOD was stimulated significantly at doses of 0.66 and 2 mg kg⁻¹ imidacloprid but markedly inhibited at a dose of 4 mg kg⁻¹ imidacloprid with prolonged exposure. The activities of CAT and POD increased irregularly at 0.2–4 mg kg⁻¹ imidacloprid over different exposure times. The level of ROS at a dose of 2 or 4 mg kg⁻¹ imidacloprid was significantly increased over the entire exposure period. When the concentration of imidacloprid was above 0.66 mg kg⁻¹, the balance of the activity of the antioxidant enzymes and ROS level was interrupted. The activity of cellulase decreased significantly with prolonged exposure. At the stress of 4 mg kg⁻¹ imidacloprid, the content of MDA was significantly increased with increasing exposure time. The results of the present study suggest that imidacloprid has a potentially harmful effect on E. fetida and may be helpful for assessment of the risk of imidacloprid to the soil ecosystem environment. However, to obtain more comprehensive toxicity data, it is necessary to investigate the effects of imidacloprid on earthworm using native soils in the future work.

2,4-Dichlorophenol (2,4-DCP) is widely distributed in wastewaters discharged from several industries, and it is considered as a priority pollutant due to its high toxicity. In this study, the use of different peroxidase extracts for 2,4-DCP removal from aqueous solutions was investigated. Tobacco hairy roots (HRs), wild-type (WT), and double-transgenic (DT) for tomato basic peroxidases (TPX1 and TPX2) were used to obtain different peroxidase extracts: total peroxidases (TPx), soluble peroxidases (SPx), and peroxidases ionically bound to the cell wall (IBPx). All extracts derived from DT HRs exhibited higher peroxidase activity than those obtained from WT HRs. TPx and IBPx DT extracts showed the highest catalytic efficiency values. The optimal conditions for 2,4-DCP oxidation were pH 6.5, H₂O₂0.5 mM, and 200 U mL⁻¹of enzyme, for all extracts analyzed. Although both TPx extracts were able to oxidize different 2,4-DCP concentrations, the removal efficiency was higher for TPx DT. Polyethylene glycol addition slightly improved 2,4-DCP removal efficiency, and it showed some protective effect on TPx WT after 2,4-DCP oxidation. In addition, using Lactuca sativa test, a reduction of the toxicity of post removal solutions was observed, for both TPx extracts. The results demonstrate that TPx extracts from both tobacco HRs appear to be promising candidate for future applications in removing 2,4-DCP from wastewaters. This is particularly true considering that these peroxidase sources are associated with low costs and are readily available. However, TPx DT has increased peroxidase activity, catalytic efficiency, and higher removal efficiency than TPx WT, probably due to the expression of TPX1 and TPX2 isoenzymes.
