Cytotoxic response in human lung epithelial cells and ion characteristics of urban-air particles from Torino, a northern Italian city Полный текст

Recently, much attention has been devoted to urban air pollution because epidemiological studies have reported health impacts related to particulate matter (PM). PM10 and PM2.5 were collected during different seasons in Torino, a northern Italian city, and were characterised by inorganic chemical species (secondary particulates and bio-available iron). The biological effects of aqueous and organic solvent PM extracts on human epithelial lung A549 were evaluated, and the effects on cell proliferation and lactate dehydrogenase (LDH) release were assayed. The average PM10 concentration during the sampling period was 47.9 ± 18.0 μg/m³; the secondary particles accounted for 49 % ± 9 % of the PM10 total mass, and the bio-available iron concentration was 0.067 ± 0.045 μg/m³. The PM2.5/PM10 ratio in Torino ranged from 0.47 to 0.90 and was higher in cold months than in warm months. The PM10 and PM2.5 extracts inhibited cell proliferation and induced LDH release in a dose-dependent manner with a seasonal trend. The PM10 extract had a stronger effect on LDH release, whereas the PM2.5 extract more strongly inhibited cell proliferation. No significant differences were observed in the effects induced by the two extracts, and no significant correlations were found between the biological effects and the PM components evaluated in this study, thus emphasising the importance of the entire mixture in inducing a cytotoxic response.

Copper (Cu) is one of the essential elements for plant growth, while excessive Cu in soils has potential environmental risks. There is little information on spatial variation of Cu in practical paddy fields. This is now important for appropriate agricultural management. The spatial patterns of Cu, its fractions in soils, and its concentrations in rice were investigated in a typical rice production region—Wenling of southeastern China. A total of 96 pairs of rice grain and soil samples (0–15 cm) were collected. The total concentration of Cu and its fractions were very variable, with large skewness, kurtosis, and coefficient of variation (CV) values. Compared to the guideline value (50 mg kg⁻¹), Cu pollution in paddy fields was observed in the study area. All the measured Cu concentrations in rice were lower than 10 mg kg⁻¹, suggesting that they remained at a safe level. Spatial analyses including Moran’s I index and geostatistics results indicated that high-high spatial patterns for both Cu in soils and rice were found in the northwest part, which was mainly related to industrial and E-waste dismantling activities. The low-low spatial patterns of Cu in the soil-rice system were located in the south part of study area. The cross-correlogram results indicated that Cu concentration in rice was significantly spatially correlated with total Cu in soils, its fractions, and soil organic matter (SOM), but significantly negatively correlated with pH and electrical conductivity (EC). Most of the selected variables had a clear spatial correlation range with Cu in rice. The ranges of significant spatial correlation (p < 0.05) could be obtained and further used for dividing agricultural management zones.

Anticoagulants are biocides widely used as pest control agents in agriculture, urban infrastructures, and domestic applications for the control of rodents. Other anticoagulants such as warfarin and acenocoumarol are also used as drugs against thrombosis. After use, anticoagulants are discharged to sewage grids and enter wastewater treatment plants (WWTPs). Our hypothesis is that WWTP effluents can be a source of anticoagulants to receiving waters and that these can affect aquatic organisms and other nontarget species. Therefore, the objective of the present study was to determine the occurrence of 11 anticoagulants in WWTPs receiving urban and agricultural wastewaters. Warfarin was the most ubiquitous compound detected in influent waters and was partially eliminated during the activated sludge treatment, and low nanograms per liter concentration were found in the effluents. Other detected compounds were coumatetralyl, ferulenol, acenocoumarol, flocoumafen, brodifacoum, bromadiolone, and difenacoum at concentrations of 0.86–87.0 ng L⁻¹. Considering water volumes of each WWTP, daily emissions were estimated to be 0.02 to 21.8 g day⁻¹, and thus, WWTPs contribute to the loads of anticoagulants to receiving waters. However, low aquatic toxicity was observed using Daphnia magna as a model aquatic organism.

This study evaluated the variability of GSTM1 and GSTT1 polymorphisms in individuals occupationally exposed to pesticides in ten Goias municipalities that present intense agricultural activity. We evaluated blood samples of 235 individuals, which 120 were rural workers occupationally exposed to pesticides and 115 formed the control group, analyzing GST polymorphisms by quantitative polymerase chain reaction (qPCR).The exposed group consisted of 111 men and nine women only getting an average of 39 ± 9 years. These workers were from ten rural municipalities situated at Goias state. It was found that 18 % of the exposed individuals had the GSTT1 null genotype and 49 % had the GSTM1 null genotype, and 10 % had both null genotypes. Data as intoxication (42 %), use of Personal Protection Equipment (PPE; 52 %) and if the worker prepared the pesticide (7 %), or if just applied the pesticide (22 %) or if the worker prepared and applied (71 %) have all been correlated with genetic polymorphisms. There were no statistically significant differences between the GSTM1 and GSTT1 polymorphisms between control and exposed groups. Finally, we could not associate a null GSTT1 or null GSTM1 polymorphisms or both to intoxication events caused by pesticides, but instead we presented the importance to use PPE to prevent such harm, once we found a statistically significant association between the use of PPE and events of intoxication (p ≤ 0.001).

This research has been conducted continuously since 2009 as part of a cohort of studies examining relationships between asthma and genetic factors, dietary habits, and environmental factors. Based on data from environmental research on house dust mites and endotoxins, which are widely known as pollutants in bedding that cause asthma in pregnant women and children, this work was conducted to obtain basic data that can be used in future cohort studies that analyze links between distribution of biological hazards and physical features of residential environments. The detection rates of house dust mite allergens, Der p1 and Der f1, were 52.7 and 86.5 %, respectively, indicating that Der f1 is a dominant species in domestic indoor environments. According to comparisons between concentrations of house dust mites and endotoxins in bedding of pregnant women and children, Der p1 and endotoxins showed significantly lower concentrations in bedding of pregnant women compared with those in bedding of children, whereas Der f1 showed no significant difference in concentration according to bedding.

The aim of this study was to evaluate the antimutagenic and antigenotoxic potential of grape juice concentrate in rodent organs exposed to cadmium chloride intoxication. A total of 15 Wistar rats were distributed into three groups (n = 5), as follows: control group (CTRL; nontreated group), cadmium group (Cd), and cadmium-grape juice group (Cd + GJ). Exposed animals received intraperitoneal injection of cadmium chloride (1.2 mg/kg body weight) diluted in water and, after 15 days, Cd + GJ group received grape juice concentrate for 15 days, by gavage (0.8 mL, 1.18 mg of polyphenols kg⁻¹ day⁻¹). Grape juice concentrate was able to decrease genotoxic effects induced by cadmium in peripheral blood and liver cells as depicted by single cell gel (comet) and micronucleus assays. A decrease for anti-8-hydroxy-20-deoxyguanosine (8OHdG) expression in hepatocytes of animals exposed to cadmium and treated with grape juice concentrate was also detected. Higher CuZn-SOD activity was observed in liver cells of the Cd + GJ group. No remarkable differences were seen regarding Mn-SOD activity among groups. Taken together, our results demonstrate that grape juice concentrate was able to exert antimutagenic and antigenotoxic activities in blood and liver cells of rats exposed to cadmium.

The study covers a thorough assessment of the overall degradation of diclofenac-Na (DCF) by high-frequency ultrasound, focusing particularly on identification, interpretation, and characterization of the oxidation byproducts and their reaction mechanisms. It was found that sonication of 5 mg L⁻¹DCF at near neutral pH rendered complete conversion of the compound, 45 % carbon, 30 % chlorine, and 25 % nitrogen mineralization. Density functional theory (DFT) calculations confirmed the experimentally detected major byproduct 2,6-dichloroaniline, the formation of which was explained by OH• addition to the ipso-position of the amino group. The stability of UV absorption at around 276–280 nm throughout reaction was in agreement with the detected byproduct structures, i.e., the presence of amino/amine groups and phenolic, aniline, benzene, and quinine-type derivatives, which all absorbed at around the same band. Microtox toxicity of the reactor aliquots at early reaction showed that initially the reaction products, specifically 1-(2,6-dichlorophenyl)-2-indoline-one, were very toxic; subsequently toxicity exhibited a fluctuating pattern, and a steady declination towards the “non-toxic” level was observed only after 90 min. Oxygen uptake analysis also revealed the formation of harmful products at early reaction, but the reactor was totally biodegradable upon 1-h sonication.

There is an increasing attempt in the world to determine the exposures of children to environmental chemicals. To analyze the genotoxic effect of air pollution, micronucleus (MN) assay was carried out in buccal epithelial cells (BECs) of children living in an urban city of Turkey. Children from two schools at urban-traffic and suburban sites were investigated in summer and winter seasons for the determination of BEC-MN frequency (per mille) and frequency of BEC with MN (per mille). The same children were also recruited for lung function measurements within a MATRA project (“Together Towards Clean Air in Eskisehir and Iskenderun”) Measured NO₂and SO₂concentrations did not exceed the European Union (EU) limit levels either in urban-traffic or suburban regions. Higher O₃concentrations were measured in the suburban site especially in the summer period. Particulate matter (PM₂.₅and PM₁₀) levels which did not differ statistically between two regions were above the EU limits in general. Although BEC-MN frequencies of children living in the suburban sites were higher in general, the difference between two regions was not significant either in the summer or winter periods. BEC-MN frequencies of the urban-traffic children were found to be significantly higher in summer period (mean ± SD, 2.68 ± 1.99) when compared to winter period (1.64 ± 1.59; p = 0.004). On the other hand, no seasonality was observed for the suburban children. Similar results have been obtained in the BEC frequency with MN in our study. In summer, BEC-MN frequencies were significantly increased with the decrease in pulmonary function levels based on forced expiratory flow between 25 and 75 % of vital capacity (FEF₂₅–₇₅ %) levels (p < 0.05). As a conclusion, children living in urban-traffic and suburban areas in the city of Eskişehir exhibited similar genotoxicity. Seasonal variation in genotoxicity may be interpreted as relatively high ozone levels and increasing time spent at outdoors in the summer.

As pollution becomes one of the biggest environmental challenges of the twenty-first century, pollution of water threatens the very existence of humanity, making immediate action a priority. The most persistent and hazardous pollutants come from industrial and agricultural activities; therefore, effective treatment of this wastewater prior to discharge into the natural environment is the solution. Advanced oxidation processes (AOPs) have caused increased interest due to their ability to degrade hazardous substances in contrast to other methods, which mainly only transfer pollution from wastewater to sludge, a membrane filter, or an adsorbent. Among a great variety of different AOPs, a group of electrochemical advanced oxidation processes (EAOPs), including electro-Fenton, is emerging as an environmental-friendly and effective treatment process for the destruction of persistent hazardous contaminants. The only concern that slows down a large-scale implementation is energy consumption and related investment and operational costs. A combination of EAOPs with biological treatment is an interesting solution. In such a synergetic way, removal efficiency is maximized, while minimizing operational costs. The goal of this review is to present cutting-edge research for treatment of three common and problematic pollutants and effluents: dyes and textile wastewater, olive processing wastewater, and pharmaceuticals and hospital wastewater. Each of these types is regarded in terms of recent scientific research on individual electrochemical, individual biological and a combined synergetic treatment.

Peroxicoagulation treatment of aqueous solution containing hazardous dye, Rhodamine B, with commercially available graphite as cathode and iron as anode has been studied. The effect of various operational parameters such as solution pH, applied voltage, electrode area, other ions, etc. on the dye removal was investigated. The experimental result showed that pH-regulated peroxicoagulation system is an efficient process for the dye removal. Ninety-five percent of the dye was removed after 180 min of electrolysis. Anions such as carbonate, bicarbonate, chloride and sulphate negatively affected the efficiency of peroxicoagulation system. From the present study, it can be concluded that peroxicoagulation process is an efficient tool for dye removal from aqueous solution.
