Differential Legionella spp. survival between intracellular and extracellular forms in thermal spring environments Полный текст

Legionella are commonly found in natural and man-made aquatic environments and are able to inhabit various species of protozoa. The relationship between the occurrence of Legionella spp. within protozoa and human legionellosis has been demonstrated; however, the proportions of intracellular and extracellular Legionella spp. in the aquatic environment were rarely reported. In this study, we developed a new method to differentiate intracellular and extracellular Legionella spp. in the aquatic environment. Water samples from three thermal spring recreational areas in southeastern Taiwan were collected and analyzed. For each water sample, concurrent measurements were performed for Legionella spp. and their free-living amoebae hosts. The overall detection rate was 32 % (16/50) for intracellular Legionella spp. and 12 % (6/50) for extracellular Legionella spp. The most prevalent host of Legionella spp. was Hartmannella vermiformis. The identified Legionella spp. differed substantially between intracellular and extracellular forms. The results showed that it may be necessary to differentiate intracellular and extracellular forms of Legionella spp.

The CROMIS AhR kit, a simple and rapid yeast bioassay kit, was developed and used to detect dioxins and dioxin-like compounds in 20 gas and solid samples collected from refuse incineration plants in Japan. The World Health Organization toxic equivalent (WHO-TEQ) values of the samples were also calculated using high-resolution gas chromatography/high--resolution mass spectrometry. The WHO-TEQ values of the samples varied greatly, ranging from 0.0021–6.3 ng/g to 0.00013–16 ng/m³N (normal cubic meter) in the solid and gas samples, respectively. 2,3,4,7,8-Pentachlorodibenzofuran (23478-PeCDF) and 1,2,3,7,8-pentachlorodibenzo-p-dioxin (12378-PeCDD) were the major contributors to the samples’ WHO-TEQ values. The yeast in the bioassay responded to these congeners, and the EC₅₀ values of 2,3,7,8-tetrachlorodibenzo-p-dioxin (2378-TeCDD), 12378-PeCDD, and 2,3,4,7,8-PeCDF were 490, 560, and 590 nM, respectively. The incinerator samples were subjected to the bioassay to obtain 2378-TeCDD equivalent values (CROMIS-TEQ values). The CROMIS-TEQ values of the solid and gas samples ranged from 0.0019 to 5.64 ng/g and from 0.14 to 20 ng/m³N, respectively. The CROMIS-TEQ and WHO-TEQ values displayed good correlations (r ² = 0.94 and 0.95 in the solid and gas samples, respectively), except for those of the samples with low dioxin concentrations (below the Japanese emission standards). Therefore, the CROMIS AhR kit is a useful tool for the initial screening of samples containing dioxin-like compounds.

The aim of the present work was to study the response of two sunflower genotypes (cultivated sunflower Helianthus annuus cv. 1114 and newly developed genotype H. annuus × Helianthus argophyllus) to Pb medium-term stress and the role of exogenously applied EDTA in alleviating Pb toxicity in hydroponics. Plant growth, morpho-anatomical characteristics of the leaf tissues, electrolyte leakage, total antioxidant activity, free radical scavenging capacity, total flavonoid content, and superoxide dismutase isoenzyme profile were studied by conventional methods. Differential responses of both genotypes to Pb supplied in the nutrient solution were recorded. Pb treatment induced a decrease in the relative growth rate, disturbance of plasma membrane integrity, and changes in the morpho-anatomical characteristics of the leaf tissues and in the antioxidant capacity, which were more pronounced in the cultivated sunflower H. annuus cv. 1114. The new genotype demonstrated higher tolerance to Pb when compared with the cultivar. This was mainly due to increased photosynthetically active area, maintenance of plasma membrane integrity, permanently high total antioxidant activity, and free radical scavenging capacity as well as total flavonoid content. The addition of EDTA into the nutrient solution led to limitation of the negative impact of Pb ions on the above parameters in both genotypes. This could be related to the reduced content of Pb in the roots, stems, and leaves, suggesting that the presence of EDTA limited the uptake of Pb. The comparative analysis of the responses to Pb treatment showed that the deleterious effect of Pb was more pronounced in the cultivated sunflower H. annuus cv. 1114. The new genotype H. annuus × H. argophyllus was more productive and demonstrated higher tolerance to Pb medium-term stress, which could indicate that it may possess certain mechanisms to tolerate high Pb concentrations. This character could be inherited from the wild parent used in the interspecific hybridization. The ability of EDTA to prevent Pb absorption by the plants could underly the mechanism of limiting of the negative impact of Pb ions. Hence, EDTA cannot be used to enhance Pb absorption from nutrient solution by sunflower plants for phytoremediation purposes.

In densely populated countries like China, clean water is one of the most challenging issues of prospective politics and environmental planning. Water pollution and eutrophication by excessive input of nitrogen and phosphorous from nonpoint sources is mostly linked to soil erosion from agricultural land. In order to prevent such water pollution by diffuse matter fluxes, knowledge about the extent of soil loss and the spatial distribution of hot spots of soil erosion is essential. In remote areas such as the mountainous regions of the upper and middle reaches of the Yangtze River, rainfall data are scarce. Since rainfall erosivity is one of the key factors in soil erosion modeling, e.g., expressed as R factor in the Revised Universal Soil Loss Equation model, a methodology is needed to spatially determine rainfall erosivity. Our study aims at the approximation and spatial regionalization of rainfall erosivity from sparse data in the large (3,200 km(2)) and strongly mountainous catchment of the Xiangxi River, a first order tributary to the Yangtze River close to the Three Gorges Dam. As data on rainfall were only obtainable in daily records for one climate station in the central part of the catchment and five stations in its surrounding area, we approximated rainfall erosivity as R factors using regression analysis combined with elevation bands derived from a digital elevation model. The mean annual R factor (R a) amounts for approximately 5,222 MJ mm ha(-1) h(-1) a(-1). With increasing altitudes, R a rises up to maximum 7,547 MJ mm ha(-1) h(-1) a(-1) at an altitude of 3,078 m a.s.l. At the outlet of the Xiangxi catchment erosivity is at minimum with approximate R a = 1,986 MJ mm ha(-1) h(-1) a(-1). The comparison of our results with R factors from high-resolution measurements at comparable study sites close to the Xiangxi catchment shows good consistance and allows us to calculate grid-based R a as input for a spatially high-resolution and area-specific assessment of soil erosion risk.

A glasshouse study of the coastal shrub Limoniastrum monopetalum was carried out to evaluate its tolerance and capacity to accumulate copper. We investigate the effects of Cu from 0 to 60 mmol l(-1) on the growth, photosynthetic apparatus, and nutrient uptake of L. monopetalum, by measuring gas exchange, chlorophyll fluorescence parameters, photosynthetic pigments, and total copper, nitrogen, phosphorus, sulfur, calcium, and magnesium content in the plant tissues. Although L. monopetalum did not survive at 60 mmol l(-1) Cu, the species demonstrated a high tolerance to Cu-induced stress, since all plants survived external Cu concentrations of up to 35 mmol l(-1) and displayed similar growth in the Cu-enriched medium as in the control treatment of up to the external level of 15 mmol Cu l(-1) (1,000 mg Cu l(-1)). The reduced growth registered in plants exposed to 35 mmol Cu l(-1) can be attributed to reduced photosynthetic carbon assimilation associated with the adverse effect of the metal on the photochemical apparatus and a reduction in the absorption of essential nutrients. Copper tolerance was associated with the capacity of the plant to accumulate the metal in its roots and effectively prevent its translocation to photosynthetic tissues. L. monopetalum has the characteristics of a Cu-excluder plant and could be used in the revegetation of Cu-contaminated soils.

This paper analyzes the characterization of energy consumption and contaminant emissions from a municipal solid waste (MSW) treatment system that comprises transfer station, landfill site, combustion plant, composting plant, dejecta treatment station, and an integrated MSW treatment plant. The consumed energy and energy medium materials were integrated under comprehensive energy consumption (CEC) for comparison. Among typical MSW disposal methods such as combustion, composting, and landfilling, landfilling has the minimum CEC value. Installing an integrated treatment plant is the recommended MSW management method because of its lower CEC. Furthermore, this method is used to ensure process centralization. In landfill sites, a positive linear correlation was observed between the CEC and contaminant removal ratios when emitted pollutants have a certain weight coefficient. The process should utilize the minimum CEC value of 5.3702 kgce/t MSW and consider energy consumption, energy recovery, MSW components, and the equivalent of carbon dioxide emissions.

Personal exposure measurement can serve as an effective tool to understand the effect of exposure to air pollutants. Alternatively, exposure assessment using pollutant concentrations in different microenvironments and accurate time–activity information for the subjects can provide good information regarding human integrated exposure. A panel of 18 healthy students of Indian Institute of Technology (IIT) Kanpur in the age group of 18 to 30 years participated in the personal exposure measurements for particulate matter, CO, NO₂ and VOC during post-monsoon and pre-monsoon seasons. Overall, 432 h person exposure data was collected in this study. The major sources of particulate and gaseous co-pollutants were identified. These directly obtained personal exposure values were then compared to the indirectly estimated integrated exposure values. Personal and integrated exposures gave statistically similar results. Through this study, we have shown that integrated exposure values could closely estimate the personal exposure values for particulate matter that can significantly reduce time and cost involved in personal exposure studies. The lung parameters for all the subjects measured during the pre-monsoon and post-monsoon seasons showed statistically significant reduction during pre-monsoon. This was attributed to the high levels of coarse particles during pre-monsoon.

A scale-free network model with surface and vertical field measurements was used to identify the connectivity distribution of the scale-free network behavior of ambient volatile organic compounds (VOCs). The results show that the carbon number (C ₙ ) with the total amount of C ₙ compounds (P(C ₙ )) possesses an explicit relationship with the scale-free network behavior. The proportionate coefficient (α) and exponent ( γ) of the scale-free network model with spatial and temporal variations are estimated and discussed. The analytical results demonstrate that although photochemical reactions cause the VOCs fraction variation, they do not alter the fraction of C ₙ compounds observably. Therefore, the values of α and of γ did not vary with time, but with local regional characteristics. The results indicate that the influence of local VOCs emissions occurs at a height of 100 m, but becomes insufficient at a height of 300 m. Air mass mixing increases with greater height; thus, the influence of regional characteristics at a height of 700 m is low. Finally, a successful empirical model was established to evaluate the distribution of surface VOCs in various regions.

Laboratory experiments were conducted to investigate copper (Cu) subcellular distribution and toxicity in Hydrilla verticillata. Fronds were subjected to different concentrations (15, 75, and 150 μM) of Cu for 7 days. Cu grains were found in cell walls, plasmodesmata, and within the nuclei and chloroplasts using the autometallographic technique. Subcellular fractionation of Cu-containing tissues indicated that in leaves subjected to high Cu concentrations, 59-65 % of the element was located in the cell wall fraction, followed by cell organelles (21-30 %) and the soluble fraction (10-14 %). The levels of K, P, Zn, and Mg declined under all Cu concentrations, but Ca, Mn, and Fe contents reached their peak at 15 μM Cu and decreased thereafter. F v/F m, F 0, and F m fell significantly in line with the decrease in pigment content. Cu exposure also caused significant damage to the chloroplasts, mitochondria, and nuclei, including disintegration of the chloroplasts and vacuolization of the mitochondria and nuclei, all of which suggested that Cu hastened plant senescence. The Cu maximum permissible concentration for H. verticillata was 10 μM, which was less than the existing general water quality standard. This suggested that H. verticillata could be used to assess Cu phytotoxicity.

Different batches of valued mussel shell and waste mussel shell ash are characterised. Shell ash has pH > 12 and high electrical conductivities (between 16.01 and 27.27 dS m⁻¹), while calcined shell shows pH values up to 10.7 and electrical conductivities between 1.19 and 3.55 dS m⁻¹. X-ray fluorescence, nitric acid digestion and water extractions show higher concentrations in shell ash for most parameters. Calcite is the dominant crystalline compound in this ash (95.6 %), followed by aragonite. Adsorption/desorption trials were performed for mussel shell ash and for a waste mixture including shell ash, sewage sludge and wood ash, showing the following percentage adsorptions: Hg(II) >94 %, As(V) >96 % and Cr(VI) between 11 and 30 % for shell ash; Hg(II) >98 %, As(V) >88 % and Cr(VI) between 30 and 88 % for the waste mixture. Hg and As desorption was <5 % for both shell ash and the waste mixture, while Cr desorption was between 92 and 45 % for shell ash, and between 19 and 0 % for the mixture. In view of that, mussel shell ash and the mixture including shell ash, sewage sludge and wood ash could be useful for Hg(II) and As(V) removal.
