The study investigated the effect of acetate/starch mixture on the formation of storage biopolymers as compared with the storage mechanism in systems fed with these compounds as single substrates. Experiments involved two sequencing batch reactor sets operated at steady state, at sludge ages of 8 and 2 days, respectively. Each set included three different runs, one fed with acetate, the other with starch and the last one with the acetate/starch mixture. In single substrate systems with pulse feeding, starch was fully converted to glycogen, whereas 25 % of acetate was utilized for direct microbial growth at sludge age of 8 days, together with polyhydroxybutyric acid (PHB) storage. The lower sludge age slightly increased this fraction to 30 %. Feeding of acetate/starch mixture induced a significant increase in acetate utilization for direct microbial growth; acetate fraction converted to PHB dropped down to 58 and 50 % at sludge ages of 8 and 2 days respectively, while starch remained fully converted to glycogen for both operating conditions. Parallel microbiological analyses based on FISH methodology confirmed that the biomass acclimated to the substrate mixture sustained microbial fractions capable of performing both glycogen and PHB storage.

The synthetic growth-promoting hormones trenbolone and melengestrol acetate have been detected in the environment near beef cattle feedlots and are reportedly transported via wind-borne particulate matter. Therefore, movement of synthetic hormones from beef cattle feedlots to water bodies via particulate matter is possible. Our objective was to evaluate potential effects of 17α-trenbolone (17α-TB), melengestrol acetate (MGA), and combinations of both on growth, development, and survival of Xenopus laevis larvae. On post-hatch day 2 (stage 33/34), X. laevis larvae were exposed to three nominal concentrations of 17α-TB (10, 100, and 500 ng/L), MGA (1, 10, and 100 ng/L), a combination of both (1/10, 10/100, and 100/500 ng/L MGA/17α-TB), frog embryo teratogenesis assay-Xenopus medium, or a solvent control. Significant increases in all X. laevis growth metrics were observed among larvae in the 1 ng/L MGA + 10 ng/L 17α-TB and 10 ng/L MGA + 100 ng/L 17α-TB treatments. Stage of development was increased among larvae in the 1 ng/L MGA + 10 ng/L 17α-TB treatment group and significantly decreased among those in the 500 ng/L 17α-TB treatment. Total body mass and snout–vent length of X. laevis larvae were significantly reduced in the 100 ng/L MGA and 100 ng/L MGA + 500 ng/L 17α-TB treatment groups. Larvae exposed to 500 ng/L 17α-TB had decreased total body mass, snout–vent length, and total length. In general, growth measurements decreased with increasing concentration of MGA, 17α-TB, or a combination of both. Survival among all treatments was not significantly different from controls. Amphibians exposed to MGA and 17α-TB in the environment may experience alterations in growth and development.

Health effects of ambient air pollution were studied in three groups of schoolchildren living in areas (suburban, urban and urban-traffic) with different air pollution levels in Eskişehir, Turkey. This study involved 1,880 students aged between 9 and 13 years from 16 public primary schools. This two-season study was conducted from January 2008 through March 2009. Symptoms of asthma, rhinitis and eczema were determined by the International Study of Asthma and Allergies in Childhood questionnaire in 2008. Two lung function tests were performed by each child for summer and winter seasons with simultaneous ambient air measurements of ozone (O3), nitrogen dioxide (NO2) and sulfur dioxide (SO2) by passive sampling. Effects of air pollution on impaired lung function and symptoms in schoolchildren were estimated by multivariate logistic regression analyses. Girls with impaired lung function (only for the summer season evaluation) were more observed in suburban and urban areas when compared to urban-traffic area ([odds ratio (OR) = 1.49; 95 % confidence interval (CI) 1.04-2.14] and [OR = 1.69 (95 % CI 1.06-2.71)] for suburban vs. urban-traffic and urban vs. urban-traffic, respectively). Significant association between ambient ozone concentrations and impaired lung function (for an increase of 10 μg m(-3)) was found only for girls for the summer season evaluation [OR = 1.11 (95 % CI 1.03-1.19)]. No association was found for boys and for the winter season evaluation. No association was found between any of the measured air pollutants and symptoms of current wheeze, current rhinoconjunctivitis and current itchy rash. The results of this study showed that increasing ozone concentrations may cause a sub-acute impairment in lung function of school aged children.

Association of selected essential (Co, Cu, Fe, Mn, Mo, Se, and Zn) and nonessential (Cd, Pb) trace elements with cytosolic proteins of different molecular masses was described for the liver of European chub (Squalius cephalus) from weakly contaminated Sutla River in Croatia. The principal aim was to establish basic trace element distributions among protein fractions characteristic for the fish living in the conditions of low metal exposure in the water. The fractionation of chub hepatic cytosols was carried out by size exclusion high performance liquid chromatography (SE-HPLC; Superdex™ 200 10/300 GL column), and measurements were performed by high resolution inductively coupled plasma mass spectrometry (HR ICP-MS). Elution profiles of essential elements were mostly characterized by broad peaks covering wide range of molecular masses, as a sign of incorporation of essential elements in various proteins within hepatic cytosol. Exceptions were Cu and Fe, with elution profiles characterized by sharp, narrow peaks indicating their probable association with specific proteins, metallothionein (MT), and ferritin, respectively. The main feature of the elution profile of nonessential metal Cd was also single sharp, narrow peak, coinciding with MT elution time, and indicating almost complete Cd detoxification by MT under the conditions of weak metal exposure in the water (dissolved Cd concentration ≤0.3 μg L⁻¹). Contrary, nonessential metal Pb was observed to bind to wide spectrum of proteins, mostly of medium molecular masses (30–100 kDa), after exposure to dissolved Pb concentration of ~1 μg L⁻¹. The obtained information within this study presents the starting point for identification and characterization of specific metal/metalloid-binding proteins in chub hepatic cytosol, which could be further used as markers of metal/metalloid exposure or effect on fish.

The Gulf War brought about to the State of Kuwait some of the worst environmental pollution as a result of oil spill. Since 1995, research programs have been initiated to avoid further damage to the Kuwaiti desert and marine environment and to restore and rehabilitate the polluted land, water, and air ecosystems. During the following 15 years, different bioremediation methods both on laboratory and small field scales were tested and evaluated. The findings of these studies were implemented to establish a bio-park in which ornamental shrubs and trees were grown in bioremediated soil. This review will focus on Kuwait’s experience in rhizoremediation and its positive impacts on oil-contaminated sites.

A permeable reactive barrier (PRB) was installed in Aznalcóllar (Spain) in order to rehabilitate the Agrio aquifer groundwater severely contaminated with acid mine drainage after a serious mining accident. The filling material of the PRB consisted of a mixture of calcite, vegetal compost and, locally, Fe(0) and sewage sludge. Among the successes of the PRB are the continuous neutralisation of pH and the removal of metals from groundwater within the PRB (removals of >95 %). Among the shortcomings are the improper PRB design due to the complexity of the internal structure of the Agrio alluvial deposits (which resulted in an inefficient capture of the contaminated plume), the poor degradability of the compost used and the short residence time within the PRB (which hindered a complete sulphate reduction), the clogging of a section of the PRB and the heterogeneities of the filling material (which resulted in preferential flows within the PRB). Undoubtedly, it is only through accumulated experience at field-scale systems that the potentials and limits of the PRB technology can be determined.

Monitoring of marine microalgae is important to predict and manage harmful algae blooms. It currently relies mainly on light-microscopic identification and enumeration of algal cells, yet several molecular tools are currently being developed to complement traditional methods. MIcroarray Detection of Toxic ALgae (MIDTAL) is an FP7-funded EU project aiming to establish a hierarchical multispecies microarray as one of these tools. Prototype arrays are currently being tested with field samples, yet the analysis of the large quantities of data generated by these arrays presents a challenge as suitable analysis tools or protocols are scarce. This paper proposes a two-part protocol for the analysis of the MIDTAL and other hierarchical multispecies arrays: Signal-to-noise ratios can be used to determine the presence or absence of signals and to identify potential false-positives considering parallel and hierarchical probes. In addition, normalized total signal intensities are recommended for comparisons between microarrays and in order to relate signals for specific probes to cell concentrations using external calibration curves. Hybridization- and probe-specific detection limits can be calculated to help evaluate negative results. The suggested analyses were implemented in “GPR-Analyzer”, a platform-independent and graphical user interface-based application, enabling non-specialist users to quickly and quantitatively analyze hierarchical multispecies microarrays. Diatoms have been regularly used as bioindicators to assess water quality of surface waters. However, diatom-based indices developed for a specific geographic region may not be appropriate elsewhere. We sampled benthic diatom assemblages in the upper Han River, a subtropical river in China, to evaluate applicability of 14 diatom-based indices used worldwide for water quality assessment. A total of 194 taxa from 31 genera were identified in the dry season and 139 taxa from 23 genera in the wet season. During the dry season, significant relationships were found for all but one of the diatom-based indices (Index Diatom Artois–Picardie) with one or more physical and chemical variables including nutrients and ion concentrations in river waters. The Biological Diatom Index (IBD) and diatom-based eutrophication/pollution index (EPI-D) were strongly related to trophic status and ionic content, while Watanabe’s Index was related to organic pollution and conductivity. Yet, the diatom indices showed weak relationships with physical and chemical variables during the wet season. It suggests that diatom-based indices developed in Europe can be applied with confidence as bioindicators of water quality in subtropical rivers of China, at least during base-flow conditions.

Prior investigations identified an association between low-level blood arsenic (As) and bladder cancer risk among Tunisian men but questions remain regarding confounding by cadmium (Cd), a well-established bladder carcinogen. A case-control study of Tunisian men was re-examined to assess the levels of cadmium in blood and reparse the association between the simultaneous exposure to these metals and bladder cancer risk. Levels of blood Cd were significantly twice higher among cases than in controls (P < 0.05) and were positively correlated with smoking and age. Additionally, analysis of metal levels among non-smokers according to the region of residence showed very high blood Cd and As levels for the coastal regions of Sfax and central Tunisia. After controlling for potential confounders, for low blood As levels (<0.67 μg/L), the OR for blood Cd was 4.10 (95 % CI 1.64-10.81), while for higher levels (>0.67 μg/L), it was reduced to 2.10 (CI, 1.06-4.17). Adjustment for Cd exposure did not alter the risk associated to As exposure. This study is the first to report the relationship between Cd exposure and risk of bladder cancer occurrence in interaction with smoking and As exposure. Smoking is shown to be the main exposure source to Cd in the Tunisian population but also environmental pollution seems to be responsible of Cd exposure among non-smokers. Exposure assessment studies encompassing a wider population are needed.

Concentrations of Hg, Pb, Cd, and Cr in 240 shellfish including oyster, short-necked clam, razor clam, and mud clam collected from six administrative regions in Xiamen of China were measured. The daily intakes of heavy metals through the consumption of shellfish were estimated based on both of the metal concentrations in shellfish and the consuming amounts of shellfish. In addition, the target hazard quotients (THQ) were used to evaluate the potential risk of heavy metals in shellfish on human body. Results showed that the concentrations of heavy metals in shellfish ranged at the following sequence: Cr > Cd > Pb > Hg. The concentrations of Hg and Pb in most samples were below the limits (0.3 mg kg⁻¹ for Hg and 0.5 mg kg⁻¹ for Pb) of national standard (GB 18406.4-2001) set in China. About 57 % of samples were found to contain more than 0.1 mg kg⁻¹ of Cd, in which the highest level was found in oyster from Xiangan with a value of 1.21 mg kg⁻¹. The average concentrations of Cd in oyster and mud clam samples were 0.338 and 0.369 mg kg⁻¹, respectively, which were significantly higher (p < 0.05) than those in the samples of short-necked clam and razor clam. The highest concentration of Cr was found to present in short-necked clam from Jimei with a value of 10.4 mg kg⁻¹, but a mean value of 1.95 mg kg⁻¹ in all the shellfish was observed, and no significant difference was found among the different sampling regions. The calculated daily intakes of Hg, Pb, Cd, and Cr through consuming the shellfish were 0.005, 0.122, 0.137, and 1.20 μg kg⁻¹ day⁻¹, respectively, which accounted for 2.19, 3.42, 13.7, and 40.1 % of the corresponding tolerable limits suggested by the Joint FAO/WHO Expert Committee on Food Additives. The THQ values of the four metals were far below 1 for most samples, except for those of Cd and Cr in the four shellfish species with the mean values of 0.132 and 0.385, respectively. The highest THQ values of Cd were observed in the species of oyster (0.719) and mud clam (0.568). But the high THQ values of Cr observed in all the four species were derived from the applied reference dose (RfD) data of Cr(VI) due to the unavailable RfD value of total Cr. The results indicate that the intakes of heavy metals by consuming shellfish collected from Xiamen of China do not present an appreciable hazard risk on human health, but attention should be paid to consuming those with relatively high THQ values, such as oyster, mud clam, and short-necked clam.
