Biodegradation of low-density polyethylene and polystyrene in superworms, larvae of Zophobas atratus (Coleoptera: Tenebrionidae): Broad and limited extent depolymerization Полный текст

Larvae of Zophobas atratus (synonym as Z. morio, or Z. rugipes Kirsch, Coleoptera: Tenebrionidae) are capable of eating foams of expanded polystyrene (EPS) and low-density polyethylene (LDPE), similar to larvae of Tenebrio molitor. We evaluated biodegradation of EPS and LDPE in the larvae from Guangzhou, China (strain G) and Marion, Illinois, U.S. (strain M) at 25 °C. Within 33 days, strain G larvae ingested respective LDPE and PS foams as their sole diet with respective consumption rates of 58.7 ± 1.8 mg and 61.5 ± 1.6 mg 100 larvae⁻¹d⁻¹. Meanwhile, strain M required co-diet (bran or cabbage) with respective consumption rates of 57.1 ± 2.5 mg and 30.3 ± 7.7 mg 100 larvae⁻¹ d⁻¹. Fourier transform infrared spectroscopy, proton nuclear magnetic resonance, and thermal gravimetric analyses indicated oxidation and biodegradation of LDPE and EPS in the two strains. Gel permeation chromatography analysis revealed that strain G performed broad depolymerization of EPS, i.e., both weight-average molecular weight (Mw) and number-average molecular weight (Mₙ) of residual polymers decreased, while strain M performed limited extent depolymerization, i.e., Mw and Mₙ increased. However, both strains performed limited extent depolymerization of LDPE. After feeding antibiotic gentamicin, gut microbes were suppressed, and Mw and Mₙ of residual LDPE and EPS in frass were basically unchanged, implying a dependence on gut microbes for depolymerization/biodegradation. Our discoveries indicate that gut microbe-dependent LDPE and EPS biodegradation is present within Z. atratus in Tenebrionidae, but that the limited extent depolymerization pattern resulted in undigested polymers with high molecular weights in egested frass.

Ny-Ålesund has been significantly impacted by anthropogenic activities (e.g. coal mining, scientific research, tourist shipping) over the past 100 years. However, the studies of potential toxic elements (PTEs) contamination in Ny-Ålesund currently mainly focus on surface soil or surface fjord sediments, and little is known about the history and status of PTEs contamination over the past 100 years. In this study, we collected a palaeo-notch sediment profile YN, analyzed the contents of six typical PTEs (Cu, Pb, Cd, Hg, As, Se) in the sediments, and assessed the historical pollution status in Ny-Ålesund using the pollution load index, geo-accumulation index and enrichment factor. The results showed that the contents of PTEs over the past 100 years increased rapidly compared with those during the interval of 9400-100 BP. In addition, Pb, Cd and Hg showed a clear signal of enrichment and were the main polluters among the PTEs analyzed. The contamination was likely linked to gas-oil powered generators, coal mining, research station, tourist shipping and long-range transport of pollutants.

Technologies for cleaner production of rice in cadmium (Cd) contaminated field are being explored worldwide. In order to investigate the inhibition mechanism of phosphate on Cd transport in soil-plant system, controlled experiments were performed in this study. Experimental results showed that Cd levels in roots, flag leaves, rachises and grains of rice plants (Oryza sativa L.) were significantly reduced by supplement of 0.5–2.5 g kg⁻¹ calcium magnesium phosphate fertilizer (CMP). Path coefficient analysis revealed that phosphorous had significant negative direct effect on Cd, but positive indirect effect on essential and non-essential amino acids. Applying 2.5 g kg⁻¹ CMP made the Cd concentration decreased by 45.7% while free essential and non-essential amino acids increased by 28.0–28.6% in grains. Levels of the branched-chain amino acids in grains were much higher than other essential amino acids, and increased with the amount of CMP fertilization. After application of CMP, pH of soil solution and thickness of the iron plaque around roots increased significantly. Spectra from X-ray photoelectron spectrometer (XPS) showed that content of N, P and Fe increased apparently, C, O and Ca had no change, while S decreased by 74.2% in roots after application of 2.5 g kg⁻¹ CMP. Meanwhile, Cd concentration in protoplasts of root cells decreased by 39.5–80.1% with the increase of CMP. These results indicate that application of CMP can effectively inhibit Cd accumulation in root protoplasts by promoting iron plaque formation on the root surface, reduce Cd concentration and increase free amino acids in rice grains.

The m-aminophenol (m-AP) is a widely used industrial chemical, which enters water, soils, and sediments with waste emissions. A common soil metal oxide, birnessite (δ-MnO2), was found to mediate the transformation of m-AP with fast rates under acidic conditions. Because of the highly complexity of the m-AP transformation, mechanism-based models were taken to fit the transformation kinetic process of m-AP. The results indicated that the transformation of m-AP with δ-MnO2 could be described by precursor complex formation rate-limiting model. The oxidative transformation of m-AP on the surface of δ-MnO2 was highly dependent on reactant concentrations, pH, temperature, and other co-solutes. The UV-VIS absorbance and mass spectra analysis indicated that the pathway leading to m-AP transformation may be the polymerization through the coupling reaction. The m-AP radicals were likely to be coupled by the covalent bonding between unsubstituted C2, C4 or C6 atoms in the m-AP aromatic rings to form oligomers as revealed by the results of activation energy and mass spectra. Furthermore, the toxicity assessment of the transformation productions indicated that the toxicity of m-AP to the E. coli K-12 could be reduced by MnO2 mediated transformation. The results are helpful for understanding the environmental behavior and potential risk of m-AP in natural environment.

Antibiotics in the environment usually co-exist with their transformation products with retained toxicity, raising concerns about environmental risks of their combined exposure. Herein, we reported a novel predictive approach for evaluating the individual and combined toxicity for photodegradation products of fluoroquinolone antibiotics (FQs). Quantitative structure-activity relationship (QSAR) models with promising predictive performance were constructed and validated using experimental data obtained with 13 FQs and 78 mixtures towards E. coli. A structural descriptor reflecting the interaction among FQ molecules and the target protein was employed in the QSAR models, which was obtained through molecular docking and thus provided a rational mechanistic explanation for these models. The predicted results indicated that the degradation products displayed varying degrees of changes compared to the parent FQs, while the combined toxicity of FQs and their degradation products was mostly additive. Furthermore, following UV irradiation the degradation products displayed elevated capacity of inducing resistance mutations in E. coli, though their overall toxicity was reduced. This result highlights the implications of antibiotic degradation products on resistance development in bacteria and stresses the importance of considering such impacts during environmental risk assessments of antibiotics.

Artificial light at night (ALAN) exposes us to prolonged illumination, that adversely affects female reproduction. However, it remains to be clarified how prolonged light exposure affects oocyte meiotic maturation and quality. To this end, we exposed female mice to a constant light (CL) of 250 lux for different durations. Our findings showed that CL exposure for 7 weeks reduced the oocyte maturation rate. Meanwhile, CL exposure caused greater abnormalities in spindle assembly and chromosome alignment and a higher rate of oocyte aneuploidy than the regular light dark cycle. CL exposure also induced oxidative stress and caused mitochondrial dysfunction, which resulted in oocyte apoptosis and autophagy. Notably, our results showed that CL exposure reduced the levels of α-tubulin acetylation, DNA methylation at 5 mC, RNA methylation at m⁶A and histone methylation at H3K4me2 but increased the levels of histone methylation at H3K27me2 in oocytes. In summary, our findings demonstrate that constant bright light exposure causes oocyte meiotic defects and reduces cytoplasmic quality. These results extend the current understanding of ALAN-mediated defects in female reproduction.

Although huge interspecies differences in the response to dioxins have been acknowledged, toxic equivalency factors derived from rodent studies are often used to assess human health risk. To determine interspecies differences, we first developed a toxicokinetic model in humans by measuring dioxin concentrations in environmental and biomonitoring samples from Southern China. Significant positive correlations between dioxin concentrations in blood and age were observed for seven dioxin congeners, indicating an age-dependent elimination rate. Based on toxicokinetic models in humans, the half-lives of 15 dioxin congeners were estimated to be 1.60–28.55 years. In consideration that the highest contribution to total toxic equivalency in blood samples was by 12378-polychlorinated dibenzo-p-dioxin (P₅CDD), this study developed a physiologically based pharmacokinetic (PBPK) model of 12378-P₅CDD levels in the liver, kidney, and fat of C57/6J mice exposed to a single oral dose, and the half-life was estimated to be 26.1 days. Based on estimated half-lives in humans and mice, we determined that the interspecies difference of 12378-P₅CDD was 71, much higher than the default usually used in risk assessment. These results could reduce the uncertainty human risk assessment of 12378-P₅CDD, and our approach could be used to estimate the interspecies differences of other dioxin congeners.

Trophic magnification factor (TMF) of persistent toxic substances (PTSs: Hg, PCBs, PAHs, and styrene oligomers (SOs)) in a coastal food web (12 fish and four invertebrates) was determined in Ulsan Bay, South Korea. The nitrogen stable isotope ratios (δ¹⁵N) of amino acids [δ¹⁵NGₗᵤ₋Pₕₑ based on glutamic acid (δ¹⁵NGₗᵤ) and phenylalanine (δ¹⁵NPₕₑ)] were used to estimate the trophic position (TPGₗᵤ₋Pₕₑ) of organisms. The TPGₗᵤ₋Pₕₑ of organisms ranged from 1.64 to 3.69, which was lower than TP estimated by δ¹⁵N of bulk particulate organic matter (TPBᵤₗₖ: 2.46–4.21). Mercury and CB 138, 153, 187, and 180 were biomagnified through the whole food web (TMF > 1), while other PTSs, such as PAHs and SOs were not (biodilution of SOs firstly reported). In particular, the trophic transfer of PTSs was pronounced in the resident fish (e.g., rock bream, sea perch, Korean rockfish). Of note, CB 99, 101, 118, and 183 were additionally found to be biomagnifying PTSs in these species. Thus, fish residency appears to represent an important factor in determining the TMF of PTSs in the coastal environment. Overall, δ¹⁵NGₗᵤ₋Pₕₑ provided accurate TPs of organisms and could be applied to determine the trophic transfer of PTSs in coastal food webs.

Ecological risk assessments (ERAs) of polycyclic aromatic compounds (PACs), as single congeners or in mixtures, present technical challenges that raise concerns about their accuracy and validity for Canadian environments. Of more than 100,000 possible PAC structures, the toxicity of fewer than 1% have been tested as individual compounds, limiting the assessment of complex mixtures. Because of the diversity in modes of PAC action, the additivity of mixtures cannot be assumed, and mixture compositions change rapidly with weathering. In vertebrates, PACs are rapidly oxygenated by cytochrome P450 enzymes, often to metabolites that are more toxic than the parent compound. The ability to predict the ecological fate, distribution and effects of PACs is limited by toxicity data derived from tests of a few responses with a limited array of test species, under optimal laboratory conditions. Although several models are available to predict PAC toxicity and rank species sensitivity, they were developed with data biased by test methods, and the reported toxicities of many PACs exceed their solubility limits. As a result, Canadian Environmental Quality Guidelines for a few individual PACs provide little support for ERAs of complex mixtures in emissions and at contaminated sites. These issues are illustrated by reviews of three case studies of PAC-contaminated sites relevant to Canadian ecosystems. Interactions among ecosystem characteristics, the behaviour, fate and distribution of PACs, and non-chemical stresses on PAC-exposed species prevented clear associations between cause and effect. The uncertainties of ERAs can only be reduced by estimating the toxicity of a wider array of PACs to species typical of Canada’s diverse geography and environmental conditions. Improvements are needed to models that predict toxicity, and more field studies of contaminated sites in Canada are needed to understand the ecological effects of PAC mixtures.

Knowledge of arsenic (As) levels in gold (Au) mining areas in the Amazon is critical for determining environmental risks and the health of the local population, mainly because this region has the largest mineral potential in Brazil and one of the largest in the world. The objective of this study was to assess the environmental and human health risks of As in tailings from Au exploration in the eastern Amazon. Samples were collected from soils and tailings from different exploration forms from 25 points, and the total concentration, pollution indexes and human health risk were determined. Concentrations of As were very high in all exploration areas, especially in tailings, whose maximum value reached 10,000 mg kg⁻¹, far above the investigation value established by the Brazilian National Council of the Environment, characterizing a polluted area with high environmental risk. Exposure based on the daily intake of As demonstrated a high health risk for children and adults, whose non-carcinogenic risk indexes of 17.8, extremely above the acceptable limit (1.0) established by the United States Environmental Protection Agency. High levels of As in reactive fractions in underground, cyanidation, and colluvium mining areas, as well as extremely high gastric and intestinal bioaccessibility were found, suggesting that high levels may be absorbed by the local population. The results show that the study area is highly polluted through Au mining activities, putting the environment and population health at risk, and that there is an urgent need for intervention by the environmental control agencies for remediation.
