Protection from metal toxicity by Hsp40-like protein isolated from contaminated soil using functional metagenomic approach

International audience | Pollution in the environment due to accumulation of potentially toxic metals results in deterioration of soil and water quality, thus impacting health of all living organisms including microbes. In the present investigation, a functional metagenomics approach was adopted to mine functional genes involved in metal tolerance from potentially toxic metal contaminated site. Eukaryotic cDNA library (1.0-4.0 kb) was screened for the genes providing tolerance to cadmium (Cd) toxicity through a functional complementation assay using Cd-sensitive Saccharomyces cerevisiae mutant ycf1(Delta). Out of the 98 clones able to recover growth on Cd-supplemented selective medium, one clone designated as PLCc43 showed more tolerance to Cd along with some other clones. Sequence analysis revealed that cDNA PLCc43 encodes a 284 amino acid protein harbouring four characteristic zinc finger motif repeats (CXXCXGXG) and showing partial homology with heat shock protein (Hsp40) of Acanthamoeba castellanii. qPCR analysis revealed the induction of PLCc43 in the presence of Cd, which was further supported by accumulation of Cd in ycf1(Delta)/PLCc43 mutant. Cu-sensitive (cup1(Delta)), Zn-sensitive (zrc1(Delta)) and Co-sensitive (cot1(Delta)) yeast mutant strains were rescued from sensitivity when transformed with cDNA PLCc43 indicating its ability to confer tolerance to various potentially toxic metals. Oxidative stress tolerance potential of PLCc43 was also confirmed in the presence of H2O2. Present study results suggest that PLCc43 originating from a functional eukaryotic gene of soil community play an important role in detoxification of potentially toxic metals and may be used as biomarker in various contaminated sites.

International audience | The bioaccumulation of metals (As, Cd, Co, Cr, Cu, Ni, Pb, Sb, V, Zn, Al, Fe) and organochlorine compounds (PCDD-Fs and PCBs) was assessed in soils and vegetables of 3 sites of contrasted anthropogenic influence (rural and industrial-urban areas). Cultivated soils in industrial areas exhibited diffuse pollution in organochlorine pollutants (PCBs and PCDD-Fs). The pollutant levels encountered in vegetables were always lower than the EU regulatory or recommended values. However, the contents measured in vegetables cultivated near industrialized areas were significantly higher than those observed in rural areas. This was notably the case for Co, Cd, Cr, Ni, Pb, V, NDL- and DL-PCB, PCDD, and PCDF. The leaf pathway appeared as the main absorption pathway for many contaminants. The results suggested that population exposure to pollutants was mainly caused by vegetable ingestion. In the vegetables and soils, the toxicity was mainly caused by the V, Co, Cd, and Pb contents to which can be added As and PCDD-Fs for soils. Therefore, the proximity of vegetable crops to highly anthropised areas has led to long-term exposure of vegetables and soils to air pollutants, leading to an accumulation in the food chain and thus a risk for human health.

International audience | In tropical montane South-East Asia, recent changes in land use have induced increased runoff, soil erosion and in-stream suspended sediment loads. Land use change is also contributing to increased microbial pathogen dissemination and contamination of stream waters. Escherichia coli (E. coli) is frequently used as an indicator of faecal contamination. Field rain simulations were conducted to examine how E. coli is exported from the surface of upland, agricultural soils during runoff events. The objectives were to characterize the loss dynamics of this indicator from agricultural soils contaminated with livestock waste, and to identify the effect of splash on washoff. Experiments were performed on nine 1 m2 plots, amended or not with pig or poultry manure. Each plot was divided into two 0.5 m2 sub-plots. One of the two sub-plots was protected with a mosquito net for limiting the raindrop impact effects. Runoff, soil detachment by raindrop impact and its entrainment by runoff, and E. coli loads and discharge were measured for each sub-plot. The results show that raindrop impact strongly enhances runoff generation, soil detachment and entrainment and E. coli export. When the impact of raindrops was reduced with a mosquito net, total runoff was reduced by more than 50%, soil erosion was on average reduced by 90% and E. coli export from the amended soil surface was on average 3 to 8 times lower. A coupled physics-based approach was performed using the Cast3M platform for modelling the time evolutions of runoff, solid particles detachment and transfer and bacteria transport that were measured for one of the nine plots. After estimation of the saturated hydraulic conductivity, soil erodibility and attachment rate of bacteria, model outputs were consistent with measured runoff coefficients, suspended sediment and E. coli loads. This work therefore underlines the need to maintain adequate vegetation at the soil surface to avoid the erosion and export of soil borne potential pathogens towards downstream aquatic systems.

International audience | Groundwater aquifers in Morocco’s coastal regions are under serious threat as a result of climate change. This study was conducted to evaluate and map the quality of water resources, by evaluating the level of pollution of the groundwater in the Meskala-Ouazzi sub-basin, a coastal area of Essaouira based on the physico-chemical analysis of 58 samples using a geographic information system (GIS) technique, analytical analysis, nitrate pollution index (NPI), and groundwater pollution index (GPI). The diagram piper of the study area is dominated by Cl-Ca-Mg, Cl-Na, HCO3-Ca-Mg, and SO4-Ca types. The concentrations of nitrate ranged from 2 to 175 mg/L. It was discovered that 22% of the groundwater samples had nitrate amounts greater than the World Health Organization’s recommended maximum allowable level of 50 mg/L. The NPI ranged between − 0.9 and 7.8. According to the classification of NPI, 44.8% of the total groundwater samples represent clean water, indicating that the groundwater in the study area is suitable for irrigation. GPI values ranging from 0.6 to 3.7, with an average of 1.7, identifies 37.9% of all groundwater samples as low polluted. The inverse distance weighting (IDW) approach was used to generate a spatial distribution map, which indicates that appropriate groundwater is present in the sub-upstream basin’s part. Overall, the forte concentration in groundwater samples detected in western and central areas showed that the nitrate originated from large amounts of nitrogen fertilizer used by humans in agricultural activities during periods of irrigation. The low tritium (δ3H) content shows that the aquifer recharge is stale water and excessive use of fertilizers leads to groundwater pollution faster over time.

International audience | We studied the removal of 61 emerging micropollutants, including illicit drugs, in a biofilter wastewater treatment plant located in the French Indies (Martinique). Raw wastewater concentrations were the highest for paracetamol followed by caffeine, naproxen, ibuprofen, its metabolite 2-hydroxyibuprofen, atenolol, ketoprofen, furosemide, methylparaben, cocaine, benzoylecgonine, and 11-nor-delta-9-carboxytetrahydrocannabinol (THC-COOH). The calculated removals were better than those reported in the literature, while the cumulative removal efficacy (i.e., removal of the total mass load) was estimated to be 92 ± 4%. However, this good performance may be partly explained by the removal of paracetamol (also named acetaminophen) and caffeine, which represented 86.4% of the total mass load. Our results point to the adsorption of some molecules on sludge, thus raising the question about local soil pollution from sludge spreading.

A biogeochemical model (ECO3M-Atoll) was configured to simulate the lower food web in Ahe Atoll lagoon where phytoplankton is mostly nitrogen limited. Understanding the dynamics of phytoplankton – the main food source for oysters – is crucial for the management and the allocation of new pearl farming sites. After parametrizing the model with in situ observations, we tested different hypotheses about nitrogen cycling (benthic remineralization, atmospheric N fixation, etc.) and compared the results to a large observational dataset. Model results show that simulated (pico- and nano-) phytoplankton biomass and nitrogen concentrations are close to in situ data. The simulated biogeochemical processes (uptake and primary production) are also very similar to the observed values. In the model, primary production ranged from 1.00 to 2.00 mg C m−3 h−1 for pico- and 0.40 to 1.00 mg C m−3 h−1 for nanophytoplankton; mean N uptake was 2.02 μmol N m−3 h−1 for pico- and 1.25 μmol N m−3 h−1 for nanophytoplankton.

Telomeres protect the coding sequence of chromosome ends and Telomere Length (TL) has been proposed as a biomarker of cellular aging, cumulative stress exposure and life-span in humans. With the aim to propose new biomarkers, a q-PCR protocol was adapted for the measurement of TL in the European flounder Platichthys flesus. The protocol was then applied in 2-year-old flounders from the Seine Estuary. The absolute TL in the flounder is 54 ± 13 kbp per genome (mean ± standard error). Considering relative or absolute TL, no correlation was observed with DNA damage and any of the measured contaminant concentrations (trace elements, metabolites of polycyclic aromatic hydrocarbons, polychlorobiphenyls, organochlorinated pesticides, polybrominated diphenyl ethers, perfluoroalkyl substances). Because sampling was limited, further investigations are required to state a possible impact of chemical pollution on flatfish telomeres. This is motivated by correlations observed with organochlorinated compounds when decreasing statistical significance (p ≤ 0.10).

The currents flowing across the rim of the atoll of Raroia were investigated with a 1 year-long dataset of wave, water level and currents. Offshore waves break on the edge of the reef outside the atoll's rim and drive current into the lagoon, through the shallow hoa that cut across the rim. The additional water volume generated by this wave driven flow induces an elevation of water level throughout the atoll's lagoon and is evacuated back into the open ocean through a deep reef pass. The water level inside the atoll is also driven by astronomical tides, which enter the lagoon thought the reef pass, after undergoing a ~50% decrease in amplitude and a ~4 hour lag. Using a simple parametric model with three calibrated coefficients, we show that currents across the atoll's rim can be estimated as a function of the offshore wave conditions and the water level difference between the ocean and the lagoon.

A traditional taxonomic approach coupled to a biological traits analysis was conducted in order to provide a new insight into macrobenthic communities associated with subtidal sandy environments. Results suggest that the macro-scale distribution of benthic communities is mainly driven by the migration rate of bedforms (sandbank, barchan dune and transversal dune) which changes the sediment grain size and reduces macrobenthic diversity. A classic scheme of species/traits succession was also observed from less to more physically disturbed areas. Finally, the high frequency of migration events homogenized macrobenthic communities between the troughs to the crest of bedforms. As bedforms areas are targeted for the commissioning of offshore windfarms the information provided by the present paper will be particularly useful to implement the environmental impact assessment required for such activities at sea.

The droplet size distribution (DSD) formed by gas-saturated oil jets is one of the most important characteristics of the flow to understand and model the fate of uncontrolled deep-sea oil spills. The shape of the DSD, generally modeled as a theoretical lognormal, Rosin-Rammler or non-fundamental distribution function, defines the size and the mass volume range of the droplets. Yet, the fundamental DSD shape has received much less attention than the volume median size (d50) and range of the DSD during ten years of research following the Deepwater Horizon (DWH) blowout. To better understand the importance of the distribution function of the droplet size we compare the oil rising time, surface oil mass, and sedimented and beached masses for different DSDs derived from the DWH literature in idealized and applied conditions, while keeping d50 constant. We highlight substantial differences, showing that the probability distribution function of the DSD for far-field modeling is, regardless of the d50, consequential for oil spill response.
