Effects of soil texture and drought stress on the uptake of antibiotics and the internalization of Salmonella in lettuce following wastewater irrigation

Treated wastewater is expected to be increasingly used as an alternative source of irrigation water in areas facing fresh water scarcity. Understanding the behaviors of contaminants from wastewater in soil and plants following irrigation is critical to assess and manage the risks associated with wastewater irrigation. The objective of this study was to evaluate the effects of soil texture and drought stress on the uptake of antibiotics and the internalization of human pathogens into lettuce through root uptake following wastewater irrigation. Lettuce grown in three soils with variability in soil texture (loam, sandy loam, and sand) and under different levels of water stress (no drought control, mild drought, and severe drought) were irrigated with synthetic wastewater containing three antibiotics (sulfamethoxazole, lincomycin and oxytetracycline) and one Salmonella strain a single time prior to harvest. Antibiotic uptake in lettuce was compound-specific and generally low. Only sulfamethoxazole was detected in lettuce with increasing uptake corresponding to increasing sand content in soil. Increased drought stress resulted in increased uptake of lincomycin and decreased uptake of oxytetracycline and sulfamethoxazole. The internalization of Salmonella was highly dependent on the concentration of the pathogen in irrigation water. Irrigation water containing 5 Log CFU/mL Salmonella resulted in limited incidence of internalization. When irrigation water contained 8 Log CFU/mL Salmonella, the internalization frequency was significantly higher in lettuce grown in sand than in loam (p = 0.009), and was significantly higher in lettuce exposed to severe drought than in unstressed lettuce (p = 0.049). This work demonstrated how environmental factors affected the risk of contaminant uptake by food crops following wastewater irrigation.

A thorough understanding of the labile status and dynamics of phosphorus (P) and iron (Fe) across the sediment-water interface (SWI) is essential for managing internal P release in eutrophic lakes. Fe-coupled inactivation of P in sediments is an important factor which affects internal P release in freshwater lakes. In this study, two in-situ high-resolution diffusive gradients in thin films (DGT) techniques, Zr-Oxide DGT and ZrO-Chelex DGT, were used to investigate the release characteristics of P from sediments in a large freshwater lake (Dongting Lake, China; area of 2691 km2) experiencing a regional summer algal bloom. Two-dimensional distributions of labile P in sediments were imaged with the Zr-Oxide DGT without destruction of the original structure of the sediment layer at four sites of the lake. The concentration of DGT-labile P in the sediments, ranging from 0.007 to 0.206 mg L−1, was highly heterogeneous across the profiles. The values of apparent diffusion flux (Fd) and release flux (Fr) of P varied between −0.027–0.197 mg m−2 d−1 and 0.037–0.332 mg m−2 d−1, respectively. Labile P showed a high and positive correlation (p < 0.01) with labile Fe(II) in the profiles, providing high-resolution evidence for the key role of Fe-redox cycling in labile P variation in sediments.

A pot experiment was conducted to investigate the effects of biochars on the availability of heavy metals (Cd, Cu, Mn, Ni, Pb, and Zn) to ryegrass in an alkaline contaminated soil. Biochars only slightly decreased or even increased the availability of heavy metals assesses by chemical extractant (a mixture of 0.05 mol L−1 ethylenediaminetetraacetic acid disodium, 0.01 mol L−1 CaCl2, and 0.1 mol L−1 triethanolamine). The significantly positive correlation between most chemical-extractable heavy metals and the ash content in biochars indicated the positive role of ash in this extraction. Biochars significantly reduced the plant uptake of heavy metals, excluding Mn. The absence of a positive correlation between the chemical-extractable heavy metals and the plant uptake counterparts (except for Mn) indicates that chemical extractability is probably not a reliable indicator to predict the phytoavailability of most heavy metals in alkaline soils treated with biochars. The obviously negative correlation between the plant uptake of heavy metals (except for Mn) and the (O + N)/C and H/C indicates that biochars with more polar groups, which were produced at lower temperatures, had higher efficiency for reducing the phytoavailability of heavy metals. The significantly negative correlations between the plant uptake of Mn and ryegrass biomass indicated the “dilution effect” caused by the improvement of biomass. These observations will be helpful for designing biochars as soil amendments to reduce the availability of heavy metals to plants in soils, especially in alkaline soils.

This study is a systematic assessment of different houses and apartments, their ages and renovation status, indoors and outdoors, and in summer vs. winter, with a goal of bringing some insight into the major sources of semivolatile organic compounds (SVOCs) and their variability. Indoor and outdoor air concentrations of polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs) and novel flame retardants (NFRs) were determined at 17–20 homes in Czech Republic in winter and summer.Indoor concentrations were consistently higher than outdoor concentrations for all compounds; indoor/outdoor ratios ranged from 2–20, with larger differences for the current use NFRs than for legacy PCBs. Seasonal trends differed according to the use status of the compounds: the PCBs had higher summer concentrations both indoors and outdoors, suggesting volatilization as a source of PCBs to air. PBDEs had no seasonal trends indoors, but higher summer concentrations outdoors. Several NFRs (TBX, PBT, PBEB) had higher indoor concentrations in winter relative to summer. The seasonal trends in the flame retardants suggest differences in air exchange rates due to lower building ventilation in winter could be driving the concentration differences.Weak relationships were found with building age for PCBs, with higher concentrations indoors in buildings built before 1984, and with the number of electronics for PBDEs, with higher concentrations in rooms with three or more electronic items. Indoor environments are the primary contributor to human inhalation exposure to these SVOCs, due to the high percentage of time spent indoors (>90%) combined with the higher indoors levels for all the studied compounds. Exposure via the indoor environment contributed ∼96% of the total chronic daily intake via inhalation in summer and ∼98% in winter.

Intense biomass burning (BB) events are widespread in tropical and subtropical Asia. However, the impact of BB aerosols on the Tibetan Plateau (TP), especially on Tibetan glaciers, is poorly understood. In this study, BB signals are revealed using the specific molecular tracer levoglucosan in snow and ice samples from different Tibetan glaciers. Tibetan glaciers mainly act as receptors of BB emissions from surrounding regions. Significant differences in levoglucosan concentrations in glacier samples collected from two slopes on the same mountain range indicate that high mountains can act as natural barriers to block the transport of smoke aerosols to the TP. Levoglucosan concentrations show a decreasing trend from west to east on glaciers impacted by the Indian summer monsoon on the southern edge of the TP, while the opposite pattern was observed on glaciers under the prevailing westerlies along the northern edge. The emission sources, the controlling climate system, as well as deposition and degradation during transport determined the spatial distribution regimes of levoglucosan concentration on Tibetan glaciers.

Mangroves are complex and dynamic ecosystems highly dependent on diverse microbial activities. In the last decades, these ecosystems have been exposed to and affected by diverse human activities, such as waste disposal and accidental oil spills. Complex microbial communities inhabiting the soil and sediment of mangroves comprise microorganisms that have developed mechanisms to adapt to organic and inorganic contaminants. The resistance of these microbes to contaminants is an attractive property and also the reason why soil and sediment living microorganisms and their enzymes have been considered promising for environmental detoxification. The aim of the present study was to identify active microbial genes in heavy metals, i.e., Cu, Zn, Cd, Pb and Hg, and antibiotic resistomes of polluted and pristine mangrove sediments through the comparative analysis of metatranscriptome data. The concentration of the heavy metals Zn, Cr, Pb, Cu, Ni, Cd, and Hg and abundance of genes and transcripts involved in resistance to toxic compounds (the cobalt-zinc-cadmium resistance protein complex; the cobalt-zinc-cadmium resistance protein CzcA and the cation efflux system protein CusA) have been closely associated with sites impacted with petroleum, sludge and other urban waste. The taxonomic profiling of metatranscriptome sequences suggests that members of Gammaproteobacteria and Deltaproteobacteria classes contribute to the detoxification of the polluted soil. Desulfobacterium autotrophicum was the most abundant microorganism in the oil-impacted site and displayed specific functions related to heavy metal resistance, potentially playing a key role in the successful persistence of the microbial community of this site.

Respiratory diseases, exacerbated through point source pollution, are currently among the leading causes of hospitalization of children in the United States. This paper investigates the relationship between the proximity of hazardous air pollutants (HAPs) emitted from Toxic Release Inventory (TRI) facilities and the number of children diagnosed in hospitals with a respiratory disease in Tennessee. The importance of controlling for toxicity of those HAPs is of particular interest. Hospital discharge, socioeconomic, TRI emission, and HAP toxicity data are used to estimate, via Generalized Linear Methods, a logistic regression model describing the relationship between the percent of children living in a zip code area treated for respiratory illness and the average annual emissions over the previous 10 years of HAPs from TRI sites in that area. Controlling for area socioeconomic characteristics, we find that accounting for toxicity is important in uncovering the relationship between HAP emissions and respiratory health of children. A one standard deviation increase in toxicity-weighted emissions per 100 square miles is associated with an increase in the number of children diagnosed with asthma (chronic bronchitis) by about 1205 (260). The evidence suggests that, with a goal to improving children’s respiratory health, monitoring the toxicity of chemicals being emitted is at least as important as simply monitoring total emission levels. This suggests that the EPA should consider making efforts toward establishing toxicity adjusted emission guidelines.

This work develops a new approach for delineating sites that are contaminated by multiple soil heavy metals and applies it to a case study. First a number of contaminant sample data are transformed into multiple spatially un-correlated factors using Uniformly Weighted Exhaustive Diagonalization with Gauss iterations (U-WEDGE). Sequential Gaussian simulation (sGs) is then used to generate sets of realizations of each resultant factor. These are then transformed into sets of sGs contaminant distribution realizations, which are then used to analyze the local and spatial (global) uncertainties in the distribution and concentration of contaminants via joint simulation. Finally, Info-Gap Decision Theory (IGDT) is used to consider different monitoring and or remediation regimes based on the analysis of contaminant realization spatial uncertainty. In our case study each heavy metal contaminant was considered individually and together with all other heavy metals; as the number of heavy metals considered increased, higher critical proportion values of local probability were chosen to obtain a low global uncertainty (to provide high reliability). Info-Gap Decision Theory (IGDT) yielded the most appropriate critical proportion values which minimized information loss in terms of specific goals. When the false negative rate is set to zero, meaning that it is necessary to monitor all potentially polluted areas, the corresponding false positive rates are at least 63%, 65%, 66%, 68%, 70%, and 78% to yield robustness levels of 0.50, 0.60, 0.70, 0.80, 0.90, and 1.00 respectively. However, when the false negative rate tolerance threshold is raised to 50%, the false positive rate tolerance which yields robustness levels of 0.50, 0.60, 0.70, 0.80, 0.90 and 1.00 drop to 12%, 14%, 15%, 18%, 20%, and 39%. The case study demonstrates the effectiveness of the developed approach at making robust decisions concerning the delineation of sites contaminated by multiple heavy metals.

The effluents from wastewater treatment plants have been recognized as a significant environmental reservoir of antibiotics and antibiotic resistance genes (ARGs). Reclaimed water irrigation (RWI) is increasingly used as a practical solution for combating water scarcity in arid and semiarid regions, however, impacts of RWI on the patterns of ARGs and the soil bacterial community remain unclear. Here, we used high-throughput quantitative PCR and terminal restriction fragment length polymorphism techniques to compare the diversity, abundance and composition of a broad-spectrum of ARGs and total bacteria in 12 urban parks with and without RWI in Victoria, Australia. A total of 40 unique ARGs were detected across all park soils, with genes conferring resistance to β-lactam being the most prevalent ARG type. The total numbers and the fold changes of the detected ARGs were significantly increased by RWI, and marked shifts in ARG patterns were also observed in urban parks with RWI compared to those without RWI. The changes in ARG patterns were paralleled by a significant effect of RWI on the bacterial community structure and a co-occurrence pattern of the detected ARG types. There were significant and positive correlations between the fold changes of the integrase intI1 gene and two β-lactam resistance genes (KPC and IMP-2 groups), but no significant impacts of RWI on the abundances of intI1 and the transposase tnpA gene were found, indicating that RWI did not improve the potential for horizontal gene transfer of soil ARGs. Taken together, our findings suggested that irrigation of urban parks with reclaimed water could influence the abundance, diversity, and compositions of a wide variety of soil ARGs of clinical relevance.Irrigation of urban parks with treated wastewater significantly increased the abundance and diversity of various antibiotic resistance genes, but did not significantly enhance their potential for horizontal gene transfer.

This study investigates, for the first time, the intestinal responses of European sea bass Dicentrarchus labrax chronically exposed to microplastics through ingestion. Fish (n = 162) were fed with 3 different treatment diets for 90 days: control, native polyvinyl chloride (PVC) and polluted polyvinyl chloride (PVC) pellets. Intestines were fixed and processed for histological analysis using standard techniques. Histopathological alterations were examined using a score value (from 0 to 4). The distal part of intestine in all samples proved to be the most affected by pathological alterations, showing a gradual change varying from moderate to severe related to exposure times. The histological picture that characterizes both groups especially after 90 days of exposure, suggests that the intestinal functions can be in some cases totally compromised.The worst condition is increasingly evident in the distal intestine of fish fed with polluted PVC pellets respect to control groups (p < 0.05) to different exposure times.These first results underline the need to assess the impact of increasing microplastics pollution on the marine trophic web.
