Fungal Generation of Organic Acids for Removal of Lead from Contaminated Soil

The use of solid-state fermentation (SFF) of low cost substrates by fungal species to generate organic acid solutions for washing of lead from a contaminated soil was evaluated in this study. SFF filtrates were generated by fermentation of four substrates (corn cobs, apple pomace, rice and hay) with three fungal species (Aspergillus niger NRRL 2001 (A. niger 1), Aspergillus niger ATCC 64065 (A. niger 2), Aspergillus foetidus NRRL 337) at three fermentation times. The concentration and speciation of organic acids of the filtrates was found to be a function of the substrate type, the fungal species and the fermentation time. Fermentation of rice resulted in the highest concentrations of citric acid while fermentation of corn cobs, apple pomace and hay tended to generate oxalic acid with an increasing fraction of this acid as the fermentation progressed. Batch extraction tests that employed the SSF filtrates revealed that soluble lead concentrations as high as 35 mg/l could be achieved. Filtrates containing elevated concentrations of citric acid resulted in the greatest lead extraction while oxalic acid inhibited solubilization. Due to the buffering of pH that was provided by the soil in the batch tests this factor did not appear to influence lead extraction. Lead extraction was observed over an extended period of time in a column test. Lead extraction was strongly influenced by the pH of the soil column and less strongly influenced by the organic acid content of the SSF filtrate. The speciation of organic acids was substantially modified from primarily citric acid in the SSF filtrate to gluconic acid in the soil column discharge.

A biomonitoring study using the ozone-sensitive bioindicator plant Nicotiana tabacum cv. Bel-W3 was conducted in the city of Valencia (eastern Spain) and surrounding areas in 2002. Plants were exposed to ambient air at seven sites, including four traffic-exposed urban sites, a large urban garden and a suburban and a rural station, for six consecutive 2-week periods using highly standardised methods. Foliar injury was registered at all stations in at least one of the exposure periods. The urban stations submitted to intense traffic showed lower ozone injury than the less traffic-exposed stations. Strong changes in the intensity of ozone injury were observed for the different exposure periods. Leaf injury was significantly related to both mean ozone values (24 and 12 h means) and cumulative exposure indices (AOT20, AOT40). However, correlation strength was moderate (r s = 0.39 to 0.58), suggesting that the plant response to ozone was modified by environmental factors. The use of sensitive bioindicators like tobacco Bel-W3 in cities provides complementary information to that of continuously operating air quality monitors, as the impact of ambient ozone levels is directly measured.

Gradient analyses were used to correlatively determine the importance of acid-related variables for littoral macroinvertebrate assemblages. To better ascertain the effects of acidity on macroinvertebrate assemblages we removed sites judged to be affected by other stressors such as agriculture, urbanization and liming. PCA of land use and water chemistry confirmed the presence of an acidity gradient; several acidity variables (e.g. pH and buffering capacity) were strongly correlated with the first PC axis, which explained Ca 32% of the variance in the environmental data. Partial constrained ordination of littoral macroinvertebrate assemblages with water chemistry, after removing the effect of other confounding variables (e.g. land use/type), showed that acidity variables accounted for significant amounts of among-lake variability in assemblage structure. Regression of canonical scores (a metric of community composition) and diversity with pH and alkalinity was used to visually determine ecological breakpoints or threshold values. Five classes were established for pH: pH < 5 (extremely acid), 5 < pH <= 5.6 (very acid), 5.6 < pH <= 6.2 (acid), 6.2 < pH <= 6.8 (weakly acid) and pH > 6.8 (neutral-alkaline). Similarly, three classes were determined for alkalinity/acidity: <0.02, 0.02-0.1 and >0.1 meq/L.

Temporal trends of non-sea salt (nss-) sulfate and nitrate were analyzed from nationwide precipitation chemistry measurements provided by the Ministry of the Environment (MOE) for the 1988-2002 fiscal years (April-March). The concentrations and deposition of nss-sulfate were found to be decreasing, and those of nitrate were stable or slightly increasing at most sites. These deposition trends were discussed from the viewpoint of emissions of SO₂ and NOX during the period of interest. Because monitoring techniques have changed in the number of active sites, samplers, and analytical methods during the operation period, the median of all annual depositions measured in Japan in a specific year was selected as the annual representative. The contribution of specific emission sources was also calculated for 1990 on the basis of the nss-sulfate and nitrate deposition in Japan obtained with a model simulation in which the model did not include volcanic emissions from Mt. Oyama, Miyakejima Island, which began to erupt suddenly and violently in 2000. For nss-sulfate, the calculated deposition agrees well with the intensity and trends of the median up to 1999. After 2000, a higher deposition than calculated in the preceding years was evident, which is attributable to the volcanic SO₂ from Mt. Oyama. For nitrate, both the calculated and observed depositions were slightly increasing; however, the calculation was found to exceed the observation.

A comparison was made of the ability of three different methods to describe the deposition and distribution of chloride from deicing salt in the roadside environment along a highway: direct sampling of airborne deposition (including snow ploughing) in containers; soil sampling and analysis of chloride content in the topsoil; and direct current resistivity measurements. Each method showed a distribution with significant decreasing values with increasing distance from the road. Two transport mechanisms, splash and spray, were identified when describing the airborne deposition. A mathematical model that includes these two transport mechanisms was adopted, and the total amount of airborne deposition on the ground 0–100 m from the road was estimated to approximately 45% of the salt applied on the road. The main part of the chloride spread by air and ploughing ended up within 10 m from the road. The soil sampling and resistivity measurements also showed the highest impact within this distance. The variation in chloride content in the soils reflected a poorer drainage ability of fine-grained soils compared to more coarse-grained soils. The resistivity measurements represented an integrated value of the differences in geology, water content and salinity. The increase in resistivity with distance from road in the topsoil was interpreted to reflect the distribution of chloride from deicing salt.

Nitrogen (N) budget was estimated with dissolved inorganic N (DIN) and dissolved organic N (DON) in a forested mountainous watershed in Tsukui, Kanagawa Prefecture, about 50 km west of Tokyo in Central Japan. The forest vegetation in the watershed was dominant by Konara oak (Quercus serrata) and Korean hornbeam (Carpinus tschonoskii), and Japanese cedar (Cryptomeria japonica). Nitrate (NO₃ -) concentration in the watershed streamwater was averagely high (98.0 ±± 19 (±± SD, n = 36) μmol L-¹) during 2001-2003. There was no seasonal and annual changes in the stream NO- ₃ concentration even though the highest N uptake rate presumably occurred during the spring of plant growing season, a fact indicating that N availability was in excess of biotic demands. The DON deposition rates (DON input rates) in open area and forest area were estimated as one of the main N sources, accounting for about 32% of total dissolved N (TDN). It was estimated that a part of the DON input rate contributed to the excessive stream NO- ₃ output rate under the condition of the rapid mineralization and nitrification rates, which annual DON deposition rates were positively correlated with the stream NO₃ - output rates. The DON retention rate in the DON budget had a potential capacity, which contributed to the excessive stream NO- ₃ output rate without other N contributions (e.g. forest floor N or soil N).

With a view to the selection of plants for the re-vegetation of contaminated, semi-arid land, two populations of the perennial species Bituminaria bituminosa (Fabaceae) from the south of Spain were studied: one (“LA”) from a non-contaminated soil and the other (“C2”) from a similar soil having elevated total levels of Pb and Zn (1,112 and 4,249 μg g-¹, respectively). For sand-cultured plants receiving nutrient solution, flow cytometry showed that heavy metals, at the concentrations measured in aqueous extracts from contaminated soils, had only slight genotoxic effects on root tip cell nuclei. Both populations were also grown in both soils, in two pot experiments. In the first, shoot biomass of LA and C2 in the contaminated soil was decreased to similar extents, with respect to the “clean” soil. Tissue heavy metal concentrations were unlikely to have been phytotoxic, except in the case of shoot Zn for population LA, but there were tissue deficiencies of P and K for populations LA and C2, respectively. In the second pot assay, the stimulation of growth by NPK fertiliser confirmed that even though this soil had high total heavy metal levels, nutrient availability was the principal factor limiting growth. The lesser transport of heavy metals (Cd, Mn and Zn) to the shoot by the population from the contaminated site is a factor that should be considered when selecting B. bituminosa lines for the phytostabilisation of such sites.

A plot-scale, rainfall-simulation study measured edge-of-field pollutant losses from conventional-till, strip-till, and no-till treatments in a burley tobacco production system. The field experiment results show that the conventional-till treatment yielded more total runoff than strip- and no-till treatments. Compared to the conventional-till treatment, both no-till and strip-till reduced the total mass losses of total suspended solids (TSS), total nitrogen (TN), total Kjeldahl nitrogen (TKN), nitrate (NO-N), ammonia (NH₃-N), total phosphorus (TP), orthophosphate (PO₄-P), and the insecticide chlorpyrifos in runoff. Although statistical analyses indicated that there was no significant difference between the no-till and strip-till practices, the no-till practice consistently yielded less edge-of-field pollutant loss than the strip-till practice. This research reinforces the body of knowledge documenting the effectiveness of conservation-tillage practices in reducing edge-of-field pollutant losses.

While developing a low-sulphate system combining indirect chromate-reduction by biologically-produced hydrogen sulphide and direct biological chromate-reduction to treat chromate-bearing waters, the aim of the present work was to evaluate the influence of sulphate and H₂ starvation on chromate reduction. Chromate-reduction was performed under continuous-feed conditions in a fixed-film column bioreactor originally inoculated with a bacterial consortium containing Desulfomicrobium norvegicum, and fed with H₂. With 500 mg l-¹ of sulphate in the feed solution, total chromate-reduction was observed in the effluent whereas sulphate-reduction was strongly decreased, as also confirmed by measurements of isotopic ratios for sulphur. In the absence of sulphate, a chromate-reduction activity was still observed but was lower than in the presence of sulphate, and chromate-reduction was H₂-dependent. Molecular biology techniques revealed the composition of the bacterial population in the effluent. D. norvegicum together with other micro-organisms of the Bacteria domain were detected. They include members related to the genera Acinetobacter, Acetobacterium and Rhodocyclus. Even when sulphate-reduction was strongly decreased, the presence of sulphate enhances the efficiency of the H₂-dependent chromate-reduction. A H₂- and CO₂-consuming bacterial population may be used in a globally autotrophic process to reduce chromate at low sulphate concentration, thus avoiding excess sulphide production.

In the study area physical and chemical factors control the composition of surface and groundwaters, which in turn determine the water quality of the “Biviere di Gela” lake. These factors combine to create diverse water types which change their compositional character spatially as rainfall infiltrates the soil zone, moves down a topographically defined flow path, and interacts with bedrock minerals. Low-salinity waters, which represent the initial stage of underground circulation, start dissolving calcium carbonate from the local rocks. The progressive increase in salinity, characterized by substantially higher Ca, SO₄, Na and Cl concentrations, suggests that dissolution of CaSO₄ and NaCl is an important process during water-rock interactions. The “Biviere di Gela” lake is often separated into two units (Lago Grande and Lago Piccolo). “Lago Grande” water is generally of Na-SO₄-Cl-type, whereas “Lago Piccolo” water is of Na-Cl-SO₄-type. Their total content of dissolved salts varies with season, the amount of rainfall, and inflow of ground and drainage water. Over time, an increasing trend towards greater salinity and also sudden changes in the relative abundances of Cl and SO₄ have been recorded for the “Lago Grande.” The isotope composition of the lake water appears to be affected by inflow of ground and surface waters and also by evaporative loss. The nitrate content of waters from the recharge basin is of particular concern because it contributes to lake eutrophication. The trace element contents do not evidence the presence of any significant metal contamination of lake waters, although a future potential hazard of metals bioaccumulation by the aquatic biota must be taken into consideration. Finally, a water balance for the basin shows that a drop in precipitation of about 20% might be critical for lake survival.
