[Departement_IRSTEA]Eaux [TR1_IRSTEA]BELCA | International audience | This study evaluated embryotoxicity and genotoxicity of two dissolved metals copper and cadmium (Cu and Cd) and two pesticides (metolachlor and irgarol) occurring in Arcachon Bay (SW France) in Pacific oyster (Crassostrea gigas) larvae and investigated the relationship between those two endpoints. Embryotoxicity was measured by calculating the percentage of abnormal D-shaped larvae and genotoxicity was evaluated with DNA strand breaks using the comet assay. After 24 h exposure, significant increases of the percentage of abnormal D-larvae and the DNA strand breaks were observed from 0.1µg/L for Cu, 10 µg/L for Cd and 0.01 µg/L for both irgarol and metolachlor in comparison with the controls. A strong positive relationship between embryotoxicity and genotoxicity was recorded for Cu, Cd and metolachlor. The current study suggests that copper, irgarol and metolachlor can induce larval abnormalities and DNA damage in a population of exposed oysters at environmentally relevant concentrations.

[Departement_IRSTEA]Eaux [TR1_IRSTEA]BELCA | Among the numerous PCB congeners, most of the dioxin-like PCBs (DL-PCBs) need to be characterized by hyphenated techniques. It has been shown in several instances that these congeners are well related to the total PCB content in fish.We examined datasets collected mainly in France, on freshwater and marine fish and sediments. A statistical model linking DL- and indicator PCBs was developed for a dataset composed of freshwater fishes, and proved to predict well DL-PCBs from indicator PCBs in all other fish sets, including marine ones. Type II error rates remained low in almost all fish sets. A similar correlation was observed in sediments. Non-dioxin-like PCBs elicit various adverse effects and represent 95% of the total PCBs. A European guideline for them is needed; the correlation between DL- and indicator PCBs could help develop this standard in the future.

International audience | The objective of this study was to assess the time variation of mineral and water stress levels across the life of a declining, Mg-deficient, spruce stand, in order to clarify the factors that caused the decline. Since 1985, strong soil acidification linked to a large leaching of nitrate and base cations was measured at the study site. In 1994, 5 trees were felled and tree rings were measured and analysed for Ca, Mg, K, Sr, 13 C/ 12 C and 87 Sr/ 86 Sr isotopic ratios. Strontium pools and fluxes as well as root Sr isotope ratio in relation to depth were also measured. Wood chemical concentrations and isotope ratios were strongly related to the dominance status of each tree. On average during the study period, the 87 Sr/ 86 Sr ratio of spruce wood decreased. Using a mechanistic model computing long term variations of 87 Sr/ 86 Sr ratio in trees and soils, we reproduced the observed trend by simulating soil acidification-increasing Sr drainage from the whole profile, and particularly from the organic horizon-, and root uptake becoming more superficial with time. Between 1952 and 1976, tree ring 13 C decreased strongly and continuously, which, in addition to other factors, might be related to an increase in water stress. Thus, a decrease in rooting depth, possibly related to soil acidification, appeared as a possible cause for the long term increase in water stress. The extreme drought event of 1976 appears to have revealed and triggered the decline.
