Associations of polycyclic aromatic hydrocarbons exposure and its interaction with XRCC1 genetic polymorphism with lung cancer: A case-control study Полный текст

Humans are extensively exposed to polycyclic aromatic hydrocarbons (PAHs) daily via multiple pathways. Epidemiological studies have demonstrated that occupational exposure to PAHs increases the risk of lung cancer, but related studies in the general population are limited. Hence, we conducted a case-control study among the Chinese general population to investigate the associations between PAHs exposure and lung cancer risk and analyze the modifications of genetic polymorphisms in DNA repair genes. In this study, we enrolled 122 lung cancer cases and 244 healthy controls in Wuhan, China. Urinary PAHs metabolites were determined by gas chromatography-mass spectrometry, and rs25487 in X-ray repair cross-complementation 1 (XRCC1) gene was genotyped by the Agena Bioscience MassARRAY System. Then, multivariable logistic regression models were performed to estimate the potential associations. We found that urinary hydroxynaphthalene (OH-Nap), hydroxyphenanthrene (OH-Phe) and the sum of hydroxy PAHs (∑OH-PAHs) levels were significantly higher in lung cancer cases than those in controls. After adjusting for gender, age, BMI, smoking status, smoking pack-years, drinking status and family history, urinary ∑OH-Nap and ∑OH-Phe levels were positively associated with lung cancer risk, with dose-response relationships. Compared with those in the lowest tertiles, individuals in the highest tertiles of ∑OH-Nap and ∑OH-Phe had a 2.13-fold (95% CI: 1.10, 4.09) and 2.45-fold (95% CI: 1.23, 4.87) increased risk of lung cancer, respectively. Effects of gender, age, smoking status and smoking pack-years on the associations of PAHs exposure with lung cancer risk were shown in the subgroup analysis. Furthermore, associations of urinary ∑OH-Nap and ∑OH-PAHs levels with lung cancer risk were modified by XRCC1 rs25487 (Pᵢₙₜₑᵣₐcₜᵢₒₙ ≤ 0.025), and were more pronounced in wild-types of rs25487. These findings suggest that environmental exposure to naphthalene and phenanthrene is associated with increased lung cancer risk, and polymorphism of XRCC1 rs25487 might modify the naphthalene exposure-related lung cancer effect.

The emissions of tyre wear particles (TWPs) into the environment are increasing and have negative impacts on the environment and human health. The aim of this study was therefore to establish a mass balance for vehicle tyres und TWP emissions in Austria using static material flow analysis, which enabled a quantification of mass flows of rubber including carbon black as the most mass-relevant tyre filler. Vehicle-specific and mileage-dependent emission factors were used to calculate the TWP emissions. The results for the year 2018 indicate that 80% of the tyre rubber remained in use, while 14% was re-treaded, recycled, incinerated or exported as end-of-life tyres and 6% was emitted as TWPs to air, soil or surface water. Of these 21,200 t/y released and dissipative lost TWPs, 6% were microscale, with a possible size between 0.1 and 10 μm, and 0.3% were nanoscale below 0.1 μm. The mass balance on the substance level shows that the TWPs contained 5,500 t/y of carbon black emitted in the form of airborne TWPs (6%) or entering in the soil or surface waters (47% each). Regarding air pollution from road vehicles, about 3,600 t/y were non-exhaust emissions, including tyre, brake and road-surface wear, which contributed to 9% of total dust emissions across Austria. Scenario analysis for 2050 with regard to e-mobility and the European Green Deal reveals that non-exhaust emissions can only be significantly reduced by a general reduction of the mileage or an environmentally friendly tyre design. This modelling approach provides a solid basis for decision makers in traffic planning as well as for chemical risk assessment. However, dynamic models with higher temporal and spatial resolution are needed to predict future mass flows of TWPs and their environmental fate, including their degradation products and possible accumulation effects.

The use of bird feathers to assess environmental contamination has steadily increased in ecotoxicological monitoring programs over the past decade. The Olrog’s Gull (Larus atlanticus) is a species endemic to the Atlantic coast of southern South America, constituting one of the three threatened gull species listed in the entire American continent. The aim of this study was to assess the exposure to Persistent Organic Pollutants (POPs) and chlorpyrifos in the Near Threatened Olrog’s Gull through the analysis of body feathers sampled at the Mar Chiquita coastal lagoon, the main wintering area of the species in Argentina, controlling for sex and age class. Chlorpyrifos showed the highest concentrations among all contaminants and groups of individuals (X¯ = 263 ng g⁻¹), while among POPs the concentration of organochlorine pesticides was higher than polychlorinated biphenyls and polybrominated diphenyl ethers, likely indicating the current use of these agricultural contaminant in the region. The highest values of total POP concentrations (males X¯ = 280 ng g⁻¹, females X¯ = 301 ng g⁻¹) were found in juvenile gulls, likely as a consequence of the incorporation of pollutants during the breeding season. Subadult and adult birds showed difference between sexes in the concentration of contaminants, with higher levels in males than females. The results highlight the need to include birds of different sex and age classes in order to better understand the variation in pollutants loads. The present study provides relevant information to improve the conservation status of the Olrog’s Gull and new insights about the environmental health of the Mar Chiquita coastal lagoon, Argentina, a MAB-UNESCO World Biosphere Reserve. However, there is a continued need for long-term monitoring programs focusing on this threatened species to understand the effects of pollutants on its population.

Sediment trapping as a tool to monitor microplastic influx was tested in an urban boreal lake basin. The one-year-long trap monitoring consisted of 5-month and 7-month periods representing growing season and winter season (including the spring flood event), respectively. Sediment accumulation rate (SAR), and organic content were determined, highest SAR – 14.5 g/m²/d – was measured during the winter period. Microplastics were extracted from the sediment applying heavy-liquid density separation method and collected under a microscope for further identification with FTIR spectroscopy. PE was identified as the most abundant synthetic polymer type, while PP and PET are also present. The annual microplastic flux rate is 32 400 pieces/m²/year, and highest accumulation does not coincide with the highest SAR, but occurs during the growing season. Changes in the microplastic accumulation rates are related to seasonal conditions. Highest microplastic concentration with respect to dry sediment weight (10 200 pieces/kg) was observed in a growing season sample, while highest concentration with respect to sediment volume (1800 pieces/l) was observed during winter. This finding underlines the problems related to reporting microplastic concentrations in various units. The results highlight that sediment trap monitoring is an efficient tool for monitoring microplastic accumulation rate in aquatic environments and provides an opportunity to better understand and define processes controlling microplastic accumulation.

Water pollution is one of the main challenges and water crises, which has caused the existing water resources to be unusable due to contamination. To understand the determinants of the distribution and abundance of antibiotic resistance genes (ARGs), we examined the distribution of 22 ARGs in relation to habitat type, heavy metal pollution and antibiotics concentration across six lakes and wetlands of Iran. The concentration of 13 heavy metals was determined by inductively coupled plasma atomic emission spectroscopy (ICP-AES) by Thermo Electron Corporation, and five antibiotics by online enrichment and triple-quadrupole LC-MS/MS were investigated. We further performed a global meta-analysis to evaluate the distribution of ARGs across global lakes compared with our studied lakes. While habitat type effect was negligible, we found a strong correlation between waste discharge into the lakes and the abundance of ARGs. The ARGs abundance showed stronger correlation with the concentration of heavy metals, such as Vanadium, than with that of antibiotics. Our meta-analysis also confirmed that overuse of antibiotics and discharge of heavy metals in the studied lakes. These data point to an increase in the distribution of ARGs among bacteria and their increasing resistance to various antibiotics, implying the susceptibility of aquatic environment to industrial pollution.

Pollutant emissions in ship exhaust have been continually increasing. SO₂ is one of the main gaseous pollutants in ship exhaust, resulting from the use of marine heavy fuel oil with high sulfur content. Therefore, it is necessary to detect the fuel sulfur content (FSC) to regulate ship exhaust emissions. Optical remote sensing methods, such as differential optical absorption spectroscopy (DOAS), light detection and ranging (LIDAR), and ultraviolet (UV) camera techniques, are regarded as simple and effective remote monitoring methods. One common technique is to estimate the SO₂ concentration in a ship plume using its local optical characteristics and use this to calculate FSC. One drawback of this technique is that there are always errors in the estimations of the SO₂ concentration despite the continuous improvement of such estimations. Another drawback is that calculating FSC from SO₂ often requires additional measurement methods. Here, a sulfur content prediction model based on a deep convolutional neural network using a UV camera is introduced. First, a ship benchmark test is performed. In the test, a large number of ultraviolet characteristic images of the ship exhaust plume are taken with a UV camera and the corresponding FSC data are collected. Next, a visual geometry group (VGG)-16 convolutional neural network model based on transfer learning is built. The model extracts all the features of the exhaust plume image as input data to the deep neural network and outputs the predicted FSC as a classification label. The results show that the model can predict the FSC value with high accuracy corresponding to the exhaust plume image. This study proves that it is theoretically feasible to apply a convolutional neural network to learn features of ultraviolet ship exhaust plume images for FSC predictions, which can provide guidance for the remote regulation of ship exhaust emissions.

HT-2 toxin (HT-2), a mycotoxin produced by Fusarium species, is detected in a variety of cereal grain-based human food and animal feed. Apart from its well-established immunotoxicity and haematotoxicity, it also causes reproductive disorders. In the present study, we revealed the adverse effects of HT-2 on early oogenesis at the foetal stage. Pregnant mice were orally administered with HT-2 for 3 days at mid-gestation. Oocytes from female foetuses exposed to HT-2 displayed defects in meiotic prophase, including unrepaired DNA damage, elevated recombination levels, and reduced expression of meiotic-related genes. Subsequently, increased oxidative stress was observed in the foetal ovaries exposed to HT-2, along with the elevated levels of reactive oxygen species, malondialdehyde, catalase, and superoxide dismutase 1/2, thereby resulting in impaired mitochondrial membrane potential and cell apoptosis. Furthermore, pre-treatment with urolithin A, a natural compound with antioxidant activities, partially reversed the delayed meiotic process by alleviating oxidative stress. Since early oogenesis is essential to determine female fertility in adult life, this study indicated that brief maternal exposure to HT-2 toxin may compromise the fertility of a developing female foetus.

PM₂.₅ has a major impact on the gastrointestinal system, but the specific mechanism behind this action is not fully understood. Current studies have focused on the relationship between PM₂.₅ and intestinal flora disorder, while ignoring the important influence of diet on gut microbes. In this study, SD rats were fed either a normal, high-fat, or high-carbohydrate diet for two months and exposed to PM₂.₅ (7 mg/kg b.w.) by intratracheal instillation. The results showed that the body and kidney weights of the rats in the high-fat diet group were significantly increased relative to those with a normal diet, and changes in the intestinal microbes and metabolites induced by PM₂.₅ were observed. Rats in the high-carbohydrate diet group had a significant response, and the diversity and richness indices of the flora were reduced (p < 0.05); additionally, intestinal Biffidobacterium and Lactobacillus were enriched, while many endogenous metabolites were found. Some amino acids derivatives and long-chain fatty acids were increased (p < 0.05). Both diet structure and PM₂.₅ exposure can affect the composition of gut microbiota, and intestinal metabolites may be associated with cell membrane damage when a high-carbohydrate diet interacts with PM₂.₅. This study considers multiple dietary factors to further supplement the evidence of intestinal damage via PM₂.₅.

Polystyrene nanoparticles (PS NPs), originated from breakdown of large plastic wastes, have already caused much concern for their environmental risks on health. This current study was aimed to reveal the toxicological mechanism of PS NPs on developing zebrafish and macrophage cells. To fulfill this purpose, 42 nm PS NPs were exposed to the early development stage of zebrafish for 5 days, the decreased heart rate and locomotor activity of zebrafish larvae were observed. The fluorescent PS NPs were used to precisely assess the accumulation of PS NPs in zebrafish larvae, and the results indicated that PS NPs not only accumulated in digestive system, but also infiltrated into the liver. More importantly, the transcriptomic analysis revealed that a total of 356 genes were differentially expressed and the KEGG class map showed significant differences in the MAPK pathway upon PS NPs treatment. Meanwhile, the induction of oxidative stress and inflammation were also observed in zebrafish larvae. Furthermore, PS NPs also induced oxidative damage and inflammatory response in RAW 264.7 cells, which activated p38 MAPK signal pathway and finally induced cell apoptosis. Our study provides a new understanding of MAPK signaling pathway involved in toxicity mechanism.

Agricultural plastic greenhouse (PG) production can extend the growing season of crops to satisfy domestic consumption in countries such as China. Workers in PGs have potential higher phthalate exposure risks than the general population as phthalate accumulation has been observed in greenhouse soil, air, and crops. To date, biomonitoring tests of phthalates for the working population have not been carried out. To address this shortage, we conducted a pilot study in Shaanxi Province, China, among 35 healthy PG workers by follow-up recording their seasonal dietary habits and work activities and urine sample collection and measurement between 2018 and 2019. The objectives were to uncover the association between phthalate metabolites and the population characteristics, seasonal and diurnal variations and causes, and to estimate exposure risks and contributions of exposure pathways from PG production systems. A total of 13 phthalate metabolite concentrations (Σ₁₃ phthalate metabolites) ranged from 102 to 781 (5th-95th) ng/mL (median: 300 ng/mL). Mono-n-butyl phthalate (MNBP) made up 51.3% of Σ₁₃ phthalate metabolites, followed by the sum of four di-2-ethylhexyl phthalate (DEHP) metabolites (24.2%), mono-2-isobutyl phthalate (MIBP) (13.4%), and mono-ethyl phthalate (MEP) (9.8%). The concentrations of MNBP and MIBP in summer were significantly higher than the levels in winter (p < 0.0001). A total of 62.3% of the PG worker population was shown to have exposure risks, and the proportion was as high as 79.4% in summer. Phthalate exposure of the workers from PG production systems constituted over 20% of the total creatinine-based daily intake, and consuming vegetables and fruit planted in PGs and inhalation in PGs were the two largest exposure pathways. Our findings demonstrate that it is important to protect workers in PGs from phthalate exposure risks, and phasing out the use of plastic materials containing phthalates in PGs is imperative, to guarantee food safety in PGs.
