Short-term extractability of sulfadiazine after application to soils
2013
Müller, Tanja | Rosendahl, Ingrid | Focks, Andreas | Siemens, Jan | Klasmeier, Jörg | Matthies, Michael
The long-term environmental fate of the veterinary antibiotic sulfadiazine (SDZ) in soils is determined by a reversible sequestration into a residual fraction and an irreversible formation of non-extractable residues (NER), which can be described as first-order rate processes. However, the concentration dynamics of the resulting fractions of SDZ in soil show an unexplained rapid reduction of extractability during the first 24 h. We therefore investigated the short-term extractability of SDZ in two different soils under different SDZ application procedures over 24 h: with and without manure, for air-dried and for moist soils. In all batches, we observed an instantaneous loss of extractability on a time scale of minutes as well as kinetically determined sequestration and NER formation over 24 h. Data evaluation with a simple kinetic model led to the conclusion that application with manure accelerated the short-term formation of NER, whereas sequestration was very similar for all batches.

Coastal lagoons are constantly subjected to releases of chemical pollutants, and so organisms may be exposed to such toxicants. This study investigated through a multivariate approach the physiological status of bivalve Ruditapes philippinarum, farmed in Sacca di Goro lagoon. Biomarkers at different levels of biological organization (catalase, superoxide dismutase, genotoxicity, reburrowing behavior) were evaluated at three sites exposed to different environmental conditions. A seasonal trend was observed, and micronucleus frequency was significantly lowest at the relatively pristine reference site. Enzymatic activity toward oxyradicals be quite efficient since variations in responsiveness were not consistent. However, behavioral impairment was observed in reburrowing rates. Sediment concentrations showed low PAH levels and high natural levels of trace metals Cr and Ni. DistLM statistical analysis revealed a non-significant relationship between selected biomarkers and xenobiotics. Therefore other potentially toxic compounds in admixture at low doses may be involved in driving differing spatial distribution of physiological impairment.

2013
Zhang, Yan
Urban metabolism analysis has become an important tool for the study of urban ecosystems. The problems of large metabolic throughput, low metabolic efficiency, and disordered metabolic processes are a major cause of unhealthy urban systems. In this paper, I summarize the international research on urban metabolism, and describe the progress that has been made in terms of research methodologies. I also review the methods used in accounting for and evaluating material and energy flows in urban metabolic processes, simulation of these flows using a network model, and practical applications of these methods. Based on this review of the literature, I propose directions for future research, and particularly the need to study the urban carbon metabolism because of the modern context of global climate change. Moreover, I recommend more research on the optimal regulation of urban metabolic systems.

2013
Delgado-Moreno, Laura | Gan, Jay
Methods for determining bioavailability of organic contaminants suffer various operational limitations. We explored the use of stable isotope labeled references in developing an isotope dilution method (IDM) to measure the exchangeable pool (E) of pyrene and bifenthrin as an approximation of their bioavailability in sediments. The exchange of deuterated bifenthrin or pyrene with its native counterpart was completed within 48 h. The derived E was 38–82% for pyrene and 28–59% for bifenthrin. Regression between E and the sum of rapid and slow desorption fractions obtained from sequential desorption showed a slope close to 1.0. The ability of IDM to predict bioavailability was further shown from a strong relationship (r2 > 0.93) between E and bioaccumulation into Chironomus tentans. Given the abundance of stable isotope labeled references and their relatively easy analysis, the IDM has the potential to become a readily adoptable tool for estimating organic contaminants bioaccessibility in various matrices.

Lohmann, Rainer | Jurado, Elena | Dijkstra, Henk A. | Dachs, Jordi
Here we estimate the importance of vertical eddy diffusion in removing perfluorooctanoic acid (PFOA) from the surface Ocean and assess its importance as a global sink. Measured water column profiles of PFOA were reproduced by assuming that vertical eddy diffusion in a 3-layer ocean model is the sole cause for the transport of PFOA to depth. The global oceanic sink due to eddy diffusion for PFOA is high, with accumulated removal fluxes over the last 40 years of 660 t, with the Atlantic Ocean accounting for 70% of the global oceanic sink. The global oceans have removed 13% of all PFOA produced to a depth greater than 100 m via vertical eddy diffusion; an additional 4% has been removed via deep water formation. The top 100 m of the surface oceans store another 21% of all PFOA produced (∼1100 t).

2013
Smits, Judit E.G. | Nain, Sukhbir
This study evaluated the impact of oral perfluorooctanoic acid (PFOA) on Japanese quail at concentrations found in American and Belgian workers at PFOA manufacturing facilities. Three arms of the immune system were tested; T cell, B cell, and innate immunity. After 6 weeks exposure, quail were challenged with E. coli infection to test the ultimate measure of immunotoxicity, disease resistance. The T cell response was lower in the high exposure groups. Antibody mediated, and innate immune responses were not different. Growth rate was higher, whereas thyroid hormone levels were lower in PFOA-exposed birds. Morbidity/mortality from disease challenge was not different among the control and PFOA-exposed groups, and no overt PFOA toxicity was observed pre-disease challenge.Although PFOA at ‘worst case scenario’ levels caused T cell immunosuppression, this did not translate into increased disease susceptibility, demonstrating that immunotoxicity testing must be interpreted with caution since disease resistance is the ultimate concern.

2013
Wilschut, M. | Theuws, P.A.W. | Duchhart, I.
Many urban areas are polluted by industrial activities and waste disposal in landfills. Since conventional soil remediation techniques are costly and unsustainable, phytoremediation might offer an alternative. In this article, we explore how phytoremediation can be integrated into the transformation of urban post-industrial areas, while improving public space. Buiksloterham, a polluted and deprived industrial area in Amsterdam, serves as case study. Buiksloterham is polluted with heavy metals, with Zinc (Zn) concentrations being the highest. A regression-model for Alpine Pennycress (Thlaspi caerulescens) is used to estimate the time needed to remediate the site. This reveals a conflict in time between remediation and urban development. A research by design experiment shows how to overcome this conflict by dealing with polluted soil innovatively while emphasizing spatial and aesthetic qualities of the phytoremediation plant species. The resulting landscape framework integrates phytoremediation with biomass production and gives new ecological, economic and social value to Buiksloterham.

Several studies demonstrated high mercury (Hg) methylation and demethylation in the periphyton associated with floating roots in tropical ecosystems. The importance of aquatic plants on methylmercury production in three temperate ecosystems from south-western France was evaluated through Hg species concentrations, and Hg methylation/demethylation activities by using stable isotopic tracers (199Hg(II), Me201Hg). Hg accumulation and high methylation and demethylation yields were detected in plant roots and periphyton, whereas results for sediment and water were low to insignificant. The presence of sulfate reducing prokaryotes was detected in all compartments (T-RFLP based on dsrAB amplified through nested PCR) and their main role in Hg methylation could be demonstrated. In turn, sulfate reduction inhibition did not affect demethylation activities. The estimation of net MeHg budgets in these ecosystems suggested that aquatic rhizosphere is the principal location for methylmercury production and may represent an important source for the contamination of the aquatic food chain.

The effects of sulfathiazole (STA) on Escherichia coli with glucose as a growth substrate was investigated to elucidate the effect-based reaction of sulfonamides in bacteria and to identify biomarkers for bacterial uptake and effect. The predominant metabolite was identified as pterine-sulfathiazole by LC-high resolution mass spectrometry. The formation of pterine-sulfathiazole per cell was constant and independent of the extracellular STA concentrations, as they exceeded the modeled half-saturation concentration KMS of 0.011 μmol L−1. The concentration of the dihydrofolic acid precursor para-aminobenzoic acid (pABA) increased with growth and with concentrations of the competitor STA. This increase was counteracted for higher STA concentrations by growth inhibition as verified by model simulation of pABA dynamics. The EC value for the inhibition of pABA increase was 6.9 ± 0.7 μmol L−1 STA, which is similar to that calculated from optical density dynamics indicating that pABA is a direct biomarker for the SA effect.

2013
Oberholster, P.J. | Genthe, B. | Hobbs, P. | Cheng, P.H. | de Klerk, A.R. | Botha, A.-M.
Streams impacted by acid mine drainage (AMD) typically present water exhibiting low pH and high metal concentrations. These factors result in the environmental degradation of watercourses. The objective of this study was to develop and evaluate an ecotoxicological screening tool (EST) to prioritise future remediation of streams impacted by AMD. The Bloubank stream drainage system in South Africa, served as study area for this purpose. In the initial EST development phase physicochemical variables were assessed while in the second phase, epilithic filamentous green algae biomass (chl-a mg m−2), diatoms and filamentous green algae community structures were employed as bioindicators as well as Daphnia magna toxicity assays. Using a weight of evidence approach, the first three sites receiving AMD were critically and seriously modified, followed by site 4 that was modified. Sites 1–3 with EST scores ≤70% were assessed as priority candidates for future restoration.
