Maternal transfer of phenol derivatives in the Baltic grey seal Halichoerus grypus grypus

Studies of circulating levels in difference sex and age classes, and maternal transfer of bisphenol A, 4-tert-octylphenol and 4- nonylphenol in the Baltic grey seal were performed from 2014-2017. Blood was collected from long-term captive adult males, pregnant females and pups. Milk was collected from nursing females. The aim of this study was not only to determine the concentrations of phenol derivatives, i.e. bisphenol A (BPA), 4-tert-octylphenol (OP) and 4-nonylphenol (NP), but also to try to evaluate the transfer of these compounds to the next generation in the final stage of foetal life and in the first few weeks of life in juvenile marine mammals. The measurements were carried out using high performance liquid chromatography. The obtained data show that all phenol derivatives are present in the blood of males, females and pups (range <0.07–101 ng·cm⁻³) and in female milk (range <0.1–406.3 ng·cm⁻³). The main source of phenol derivatives in organisms is food exposure. Gender, age, or number of births were not observed to have a significant effect on changes in phenol derivative levels in seal blood within the breeding group. In the prenatal stage of life, a small amount of BPA and alkylphenols was passed on to the offspring through the placenta. In the blood of the offspring the concentration of these compounds exceeded the concentration in the mother's blood 1.5-fold. During nursing, females detoxified their systems. Level of phenol derivatives in the pups blood increased linearly with its increasing concentrations in the mother's milk. On the other hand, the seafood diet which started after the physiological fasting stage of the pup, stabilised the levels of phenol derivatives below 10 ng ∙ cm⁻³.

Observed levels of lipid/lipoproteins are known to be associated with exposure to perfluoroalkyl substances (PFAS). In order to evaluate and update these associations among US children aged 6–11 years, data (N = 458) from National Health and Nutrition Examination Survey for 2013–2014 were used. The associations between the observed levels of total cholesterol, high density lipoprotein (HDL) cholesterol, and non-HDL cholesterol and selected PFAS were studied. PFAS data were available for perfluorononanoic acid (PFNA), perfluorohexane sulfonate (PFHxS), linear isomer of perfluorooctanoic acid (PFOA), linear isomer of perfluorooctane sulfonate (PFOS), monomethyl branch isomer of PFOS, and sum of PFAS. Regression models were fitted to evaluate these associations. A statistically significant (p = 0.03) positive association between the levels of linear isomer of PFOS and total cholesterol was observed. A 10% increase in the levels of linear isomer of PFOS measured in ng/L was found to be accompanied by a 0.03–0.42% increase in the levels of total cholesterol measured in mg/dL. For PFNA, girls in the first quartile of PFNA were found to have lower adjusted levels for total cholesterol than the girls in the fourth quartile of PFNA (152.6 vs. 164.7 mg/dL, p < 0.01). Also, non-Hispanic blacks in the first quartile of PFNA were found to have lower adjusted levels for total cholesterol than the non-Hispanic blacks in the fourth quartile of PFNA (143.4 vs. 160.5 mg/dL, p = 0.04). A negative association between branch isomer of PFOS and non-HDL cholesterol was also observed (β = −0.0066, p = 0.04). The adjusted levels of non-HDL cholesterol were higher in the second quartile of ∑PFAS than in the fourth quartile of ∑PFAS (103.0 vs. 97.5 mg/dL, p < 0.01). Linear PFOS and possibly PFNA are associated with total cholesterol in the most recent NHANES childhood sample. Concentrations of PFAS and associations with cholesterol have both decreased compared to previous literature.

This study investigated selected steroidal and phenolic endocrine disrupting compounds (EDCs) in the surface water of the Bahe River (China) using gas chromatography mass spectrometry (GC-MS). Their effect on the wild sharpbelly Hemiculter leucisculus was investigated. The concentrations of 4-t-octylphenol, nonylphenol, bisphenol-A, estrone, 17 β-estradiol, 17 α-Ethinylestradiol, and estriol were up to 126.0, 634.8, 1573.1, 55.9, 23.9, 31.5, and 5.2 ng L⁻¹ in the surface water, and up to 26.4, 103.5, 146.9, 14.2, 9.3, 13.8, and 1.3 ng g⁻¹ in the fish muscle tissue, respectively. High estrogen equivalent levels and hazard quotients were found in the middle and lower reaches of the river, and the pollution in these regions caused enhanced growth conditions, inhibition of gonad growth, and suppression of spermatogenesis in H. leucisculus. The up-regulation of Vitellogenin mRNA expression in male fish, collected from relatively heavily EDCs contaminated areas, indicates a potential estrogenic effect. The differential expression profiles of genes related to steroidogenesis at all sampling sites suggests that these endpoints may play an important role for the pollution monitoring of estrogenic EDCs in the Bahe River.

Rapid urbanization and economic growth has led to significant increase in municipal solid waste generation in India during the last few decades and its management has become a major issue because of poor waste management practices. Solid waste generated is deposited into open dumping sites with hardly any segregation and processing. Carbon dioxide (CO₂), methane (CH₄) and nitrous oxide (N₂O) are the major greenhouse gases that are released from the landfill sites due to the biodegradation of organic matter. In this present study, CH₄ and CO₂ emissions from a landfill in north-east India are estimated using a flux chamber during September, 2015 to August, 2016. The average emission rates of CH₄ and CO₂ are 68 and 92 mg/min/m², respectively. The emissions are highest in the summer whilst being lowest in winter. The diurnal variation of emissions indicated that the emissions follow a trend similar to temperature in all the seasons. Correlation coefficients of CH₄ and temperature in summer, monsoon and winter are 0.99, 0.87 and 0.97, respectively. The measured CH₄ in this study is in the range of other studies around the world. Modified Triangular Method (MTM), IPCC model and the USEPA Landfill gas emissions model (LandGEM) were used to predict the CH₄ emissions during the study year. The consequent simulation results indicate that the MTM, LandGEM-Clean Air Act, LandGEM-Inventory and IPCC models predict 1.9, 3.3, 1.6 and 1.4 times of the measured CH₄ emission flux in this study. Assuming that this higher prediction of CH₄ levels observed in this study holds well for other landfills in this region, a new CH₄ emission inventory (Units: Tonnes/year), with a resolution of 0.1⁰ × 0.1⁰ has been developed. This study stresses the importance of biodegradable composition of waste and meteorology, and also points out the drawbacks of the widely used landfill emission models.

The determination of values of abundance of antibiotic resistance genes (ARGs) per mass of soil is extremely useful to assess the potential impacts of relevant sources of antibiotic resistance, such as irrigation with treated wastewater or manure application. Culture-independent methods and, in particular, quantitative PCR (qPCR), have been regarded as suitable approaches for such a purpose. However, it is arguable if these methods are sensitive enough to measure ARGs abundance at levels that may represent a risk for environmental and human health. This study aimed at demonstrating the range of values of ARGs quantification that can be expected based on currently used procedures of DNA extraction and qPCR analyses. The demonstration was based on the use of soil samples spiked with known amounts of wastewater antibiotic resistant bacteria (ARB) (Enterococcus faecalis, Escherichia coli, Acinetobacter johnsonii, or Pseudomonas aeruginosa), harbouring known ARGs, and also on the calculation of expected values determined based on qPCR.The limits of quantification (LOQ) of the ARGs (vanA, qnrS, blaTEM, blaOXA, blaIMP, blaVIM) were observed to be approximately 4 log-units per gram of soil dry weight, irrespective of the type of soil tested. These values were close to the theoretical LOQ values calculated based on currently used DNA extraction methods and qPCR procedures. The observed LOQ values can be considered extremely high to perform an accurate assessment of the impacts of ARGs discharges in soils. A key message is that ARGs accumulation will be noticeable only at very high doses. The assessment of the impacts of ARGs discharges in soils, of associated risks of propagation and potential transmission to humans, must take into consideration this type of evidence, and avoid the simplistic assumption that no detection corresponds to risk absence.

To investigate the biological pathways involved in phthalate-induced developmental effects, zebrafish embryos were exposed to different concentrations of di-(2-ethylhexyl) (DEHP) and di-butyl phthalate (DBP) for 96 h. Embryonic exposure to DEHP and DBP induced body length decrease, yolk sac abnormities, and immune responses (up-regulation of immune proteins and genes). The lipidomic results showed that at a concentration of 50 μg/L, DEHP and DBP significantly reduced the levels of fatty acids, triglycerides, diacylglycerol, and cholesterol. These effects are partly explained by biological pathway enrichment based on data from the transcriptional and proteomic profiles. Co-exposure to DBP and ER antagonist did not significantly relieve the toxic symptoms compared with exposure to DBP alone. This indicates that phthalate-induced developmental abnormities in zebrafish might not be mediated by the ER pathway. In conclusion, we identified the possible biological pathways that mediate phthalate-induced developmental effects and found that these effects may not be driven by estrogenic activation.

Floods are threats to ecosystems that are caused by natural disasters such as typhoons and heavy rain, and to respond to these threats, resilience needs to be improved. In this study, the response of the social-ecological system of Haeundae-gu (Busan, Republic of Korea) to disasters is analyzed by using a causal loop diagram, and a resilience improvement plan is presented by simulating the disaster resilience using green infrastructure through the System Resilience Dynamics Model. First, the resilience values are highest when green infrastructure is applied at the maximum applicable ratio (30%) compared with no application. Second, in the public and private areas of Haeundae-gu, resilience according to green roof scenario was higher until approximately 8 h after the beginning of rainfall, but then the resilience according to infiltration storage facility scenario was higher. In the transportation and industrial areas, the overall resilience according to infiltration storage facility scenario was higher than the resilience according to porous pavement scenario. This study demonstrates that a resilience improvement plan based on simulation can support decision making to respond to disasters such as typhoons.

The industrialized portion of the Houston Ship Channel (HSC) is heavily contaminated with anthropogenic contaminants, most prominent of which are the polychlorinated biphenyls (PCBs). This contamination has driven adaptive evolution in a keystone species for Galveston Bay, the Gulf killifish (Fundulus grandis). We investigated the geographical extent of PCB impacts by sampling 12 sites, ranging from the heavily industrialized upper portion of the HSC to Galveston Island. At each site, PCB concentrations and profiles were determined in three environmental compartments: sediment, water (polyethylene passive samplers), and fish tissue (resident Gulf killifish). We observed a steep gradient of PCB contamination, ranging from 4.00 to 100,000 ng/g organic carbon in sediment, 290–110,000 ng/g lipid in fish, and 4.5–2300 ng/g polyethylene in passive samplers. The PCB congener profiles in Gulf killifish at the most heavily contaminated sites were shifted toward the higher chlorinated PCBs and were highly similar to the sediment contamination profiles. In addition, while magnitude of total PCB concentrations in sediment and total fish contamination levels were highly correlated between sites, the relative PCB congener profiles in fish and passive samplers were more alike. This strong correlation, along with a lack of dependency of biota-sediment accumulation factors with total contamination rates, confirm the likely non-migratory nature of Gulf killifish and suggest their contamination levels are a good site-specific indicator of contamination in the Galveston Bay area. The spatial gradient of PCB contamination in Galveston Bay was evident in all three matrices studied and was observed effectively using Gulf killifish contamination as an environmentally relevant bioindicator of localized contamination in this environment.

The objective of this study was to determine the effects of cooking and in vitro human digestion on the changes of five insecticides—fipronil, bifenthrin, 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane (DDT), 1,1-dichloro-2,2-bis(p-chlorophenyl)ethane (DDD), and 2,2-bis(p-chlorophenyl)-1,1-dichloroethylene (DDE)—in egg whites and yolks. Each insecticide was applied to egg whites and yolks at a concentration of 1000 μg/g. After cooking the egg whites and yolks, concentrations of bifenthrin, DDD, and DDE decreased (P < 0.05), whereas those of fipronil and DDT were unchanged (P > 0.05) in both egg whites and yolks. Next, an in vitro human digestion model that simulates all the steps of human digestion was employed. Until digestion in the small intestine, the concentrations of fipronil and DDT in the cooked egg whites and yolks were unchanged (P > 0.05), whereas those of bifenthrin, DDD, and DDE decreased (P < 0.05) at each digestion step. In the large intestinal digestion step with Escherichia coli and Lactobacillus sakei as enterobacteria, the concentrations of all the insecticides decreased (P < 0.05) in the cooked egg whites and yolks. Among the insecticides, bifenthrin showed the lowest concentration (P < 0.05). In conclusion, the use of bifenthrin as an insecticide would be comparatively less toxic than other insecticides in terms of environmental pollution and human health, because of its easy degradation.

Atrazine (ATR), a selective herbicide, is consistently used worldwide and has been confirmed to be harmful to the health of aquatic organisms. The release of neutrophil extracellular traps (NETs) is one of the newly discovered antimicrobial mechanisms. Although several immune functions have been analyzed under ATR exposure, the effect of ATR on NETs remains mainly unexplored. In the present study, we treated carp neutrophils using 5 μg/ml ATR and 5 μg/ml ATR combined with 100 nM rapamycin to elucidate the underlying mechanisms and to clarify the effect of ATR on phorbol myristate acetate (PMA)-induced NETs. The results of the morphological observation and quantitative analysis of extracellular DNA and myeloperoxidase (MPO) showed that NETs formation were significantly inhibited by ATR exposure. Moreover, we found that in the NETs process, ATR downregulated the expression of the anti-apoptosis gene B-cell lymphoma-2 (Bcl-2), increased the expression of the pro-apoptosis factors Bcl-2-Associated X (BAX), cysteinyl aspartate specific proteinases (Caspase3, 9), and anti-autophagy factor mammalian target of rapamycin (mTOR), decreased the expression of autophagy-related protein light chain 3B (LC3B) and glucose transport proteins (GLUT1, 4), disturbed the activities of phosphofructokinase (PFK), pyruvate kinase (PKM), and hexokinase (HK) and limited reactive oxygen species (ROS) levels, indicating that the reduced NETs release was a consequence of increased apoptosis and diminished ROS burst, autophagy and down-regulated glycolysis under ATR treatment. Meanwhile, rapamycin restored the inhibited autophagy and glycolysis and thus resisted the ATR-suppressed NETs. The present study perfects the mechanism theory of ATR immunotoxicity to fish and has a certain value for human health risk assessment.
