Lead-based paints and children's PVC toys are potential sources of domestic lead poisoning – A review

Lead (Pb) both in paints and children's Polyvinyl chloride (PVC) toys is a major public health concern which has attracted attention of the international community. Concentrations of Pb both in lead-based paints and children's PVC toys have been assessed through various studies across the globe. Therefore, the purpose of this article was to summarize the results reported in these studies and provide some comprehension on their implications to human health for law enforcement as well as for awareness raising to the general public. Highlights on identified gaps have been provided to pave ways for further research interventions in order to establish comprehensive information on the subject.Regardless of regulatory limits on the content of lead, both in paints and children's PVC toys existing in different countries in the world, some of the reviewed articles have revealed significant levels of lead in these two items far above the permissible limits.High lead levels in paints have been recorded in China (116,200 ppm), Cameroon (500,000 ppm), South Africa (189,000 ppm), Tanzania (120,862.1 ppm), Uganda (150,000 ppm), Thailand (505,716 ppm) and Brazil (170,258.4 ppm) just to mention a few.Lead poisoning cases in children have been reported in several countries including France, Morocco, South Africa and United States. Countries where high levels of lead in children's PVC toys have been recounted include; China (860,000 ppm), South Africa (145,000 ppm), United States (22,550 ppm), Thailand (4,486.11 ppm), Palestine (6,036 ppm) and India (2,104 ppm).Awareness raising among parents is vital to impart them with knowledge on the matter so that they can take strenuous measures to protect their children from lead poisoning emanating from playing with toys and paint dust. Law enforcement on phasing out lead-based paints and control of lead content in children's PVC toys worldwide is also highly recommended.

Arsenic (As) in rice grains is a serious food safety concern. Some coexisting engineered nanoparticles (ENPs) were shown to alter the accumulation and speciation of As in rice grains. However, investigation on the effects of copper oxide nanoparticles (CuO NPs), a popular ingredient in pesticides, on the uptake and accumulation of As is rare. We explored the potentially different impact of CuO NPs and corresponding Cu(II) ions on the accumulation of two As species in rice seedlings in a hydroponic system. Rice seedlings were treated with a combinations of 1 mg/L of arsenite (As(III)) or arsenate (As(V)) and 100 mg/L of CuO NPs or Cu(II) for 6 days. Both forms of Cu significantly reduced the accumulation of total As in rice tissues, with Cu(II) exhibiting significantly greater effect than CuO NPs. As speciation in rice roots was markedly affected by both forms of Cu, and the impacts were Cu-form dependent. For example, the co-existence of As(V) with CuO NPs led to a 45% decrease of As(V) in rice roots, while the co-existence of As(V) with Cu(II) caused a 47% increase in As(V) in rice roots. As speciation in rice shoots was less affected by co-present Cu than in rice roots. Co-occurring As(III) or As(V) lowered Cu concentration in rice roots by 40% and 50% in treatments with CuO NPs, but did not affect Cu content in rice roots co-exposed to Cu(II). The study confirmed the reciprocal effect of co-occurring CuO NPs or Cu(II) and As in rice paddies and highlighted the unique “nano-effect” of CuO NPs. The results alsos showed that the initial oxidation state of As plays an important role in the interactions between As and Cu. The results shed light on the current debate on the safe applications of nano-enabled agrichemicals vs. conventional metal salts in agriculture.

Bisphenol A (BPA) is an endocrine disruptor whose ubiquitous presence in the environment has been related with impairment of male reproduction. BPA can cause both transcriptomic and epigenetic changes during spermatogenesis. To evaluate the potential effects of male exposure to BPA, adult zebrafish males were exposed during spermatogenesis to doses of 100 and 2000 μg/L, which were reported in contaminated water bodies and higher than those allowed for human consumption. Fertilization capacity and survival at hatching were analysed after mating with untreated females. Spermatogenic progress was analysed through a morphometrical study of testes and apoptosis was evaluated by TUNEL assay. Testicular gene expression was evaluated by RT-qPCR and epigenetics by using ELISA and immunocytochemistry. In vitro studies were performed to investigate the role of Gper. Chromatin fragmentation and the presence of transcripts were also evaluated in ejaculated sperm. Results on testes from males treated with the highest dose showed a significant decrease in spermatocytes, an increase in apoptosis, a downregulation of ccnb1 and sycp3, all of which point to an alteration of spermatogenesis and to meiotic arrest and an upregulation of gper1 and esrrga receptors. Additionally, BPA at 2000 μg/L caused missregulation of epigenetic remodelling enzymes transcripts in testes and promoted DNA hypermethylation and H3K27me3 demethylation. BPA also triggered an increase in histone acetyltransferase activity, which led to hyperacetylation of histones (H3K9ac, H3K14ac, H4K12ac). In vitro reversion of histone acetylation changes using a specific GPER antagonist, G-36, suggested this receptor as mediator of histone hyperacetylation. Males treated with the lower dose only showed an increase in some histone acetylation marks (H3K14ac, H4K12ac) but their progeny displayed very limited survival at hatching, revealing the deleterious effects of unbalanced paternal epigenetic information. Furthermore, the highest dose of BPA led to chromatin fragmentation, promoting direct reproductive effects, which are incompatible with embryo development.

This study examined the spatial and temporal variations of severe particulate pollution days (SPPDs) in China by using observed PM₂.₅ concentrations during April 2013 to February 2018 from the Ministry of Environmental Protection of China. SPPDs were defined as those with observed daily mean PM₂.₅ concentrations larger than 150 μg m⁻³. Observations showed that northern China had the highest number of SPPDs during the studied period. Since 2015, the number of SPPDs in northwestern China is comparable to or even higher than that observed in Beijing-Tianjin-Hebei (BTH). The highest numbers of SPPDs observed within BTH and the Yangtze River Delta (YRD) were 122 (33), 95 (17), 57 (15), 78 (18), and 31 (25) days in 2013, 2014, 2015, 2016, and 2017, respectively, indicating a general decreasing trend as a result of emission reduction measures. SPPDs occurred mainly from November to February in BTH and in December and January in the YRD. The major circulation patterns associated with large-scale SPPDs were analyzed by using principal component analysis. Five typical synoptic weather patterns were identified for BTH. The most dominant weather type (a cold high centered over the Xinjiang and Mongolian regions) for BTH was also responsible for most of the SPPDs in the YRD. These results have important implications for emission control strategies during SPPDs. Emission control measures can be applied once the dominant circulation patterns have been predicted.

Thirteen secondary organic aerosol (SOA) tracers of isoprene, monoterpenes and sesquiterpenes were measured for PM₂.₅ aerosols collected at the summit of Mt. Wuyi (1139 m, a.s.l.), to investigate their seasonality and formation mechanism. Concentrations of the isoprene and monoterpene SOA tracers were much higher in summer than those in other seasons. In contrast, β-caryophyllinic acid was found to be the lowest in summer. Concentrations of those BSOA tracers showed a positive correlation with temperature (R² = 0.52–0.70), and a negative correlation with relative humidity (R² = 0.43–0.78). Moreover, thermodynamic model (i.e., ISORROPIA-II) calculation results showed that acidity conditions are favorable for BSOA formation. Robust linear correlations between the BSOA tracers and anthropogenic pollutants such as SO₂ (R² = 0.53–0.7) and NO₂ (R² = 0.37–0.54) were observed for all the samples, suggesting that SO₂ and NOx can enhance BSOA production in the remote mountain area of southeast China, which is related to an acid-catalyzed heterogeneous chemistry. Moreover, we also found a significant correlation between the concentrations of the BSOA tracers and levoglucosan especially for β-caryophyllinic acid, indicating that biomass burning plumes from the distant lowland regions could influence the production of BSOA in the mountain free troposphere. Our results clearly demonstrated that anthropogenic emissions in China could enhance BSOA formation in the distant mountain regions.

F-53B (6:2 chlorinated polyfluorinated ether sulfonate) is currently recognized as a safe alternative to long-chain PFASs in China. However, an increasing number of studies have recently authenticated its biotoxicological effects. In this study, for evaluating the gut toxicity of F-53B in mammals, both female and male mice were orally exposed to 0, 1, 3, or 10 μg/L F-53B for 10 weeks. Our results showed that F-53B significantly accumulated in the colon, ileum and serum when exposed to 10 μg/L F-53B for 10 weeks. F-53B exposure not only increased the transcriptional levels of ion transport-related genes but could also interact with the CFTR protein directly. Interestingly, subchronic F-53B exposure also increased the transcription of mucus secretion-related genes, but the protein level of Muc2 decreased after F-53B exposure, indicating that there was a compensatory phenomenon after mucus barrier injury. Furthermore, F-53B exposure also induced colonic inflammation associated with gut microbiota dysbiosis in the colon. Taken together, our results indicated that the potential gut toxicity of F-53B and almost all of the changed parameters were significantly affected in both female and male mice, suggesting that F-53B could disturb the gut barrier without sex dependence in mice.

Urban stormwater is a major source of chemical pollution to receiving waters. Anthropogenic materials in the built environment can be an important source of chemicals to stormwater runoff. Roofing materials can leach significant amounts of metals, which vary over the life of the roof. We report concentrations of three metals (As, Cu, Zn) leaching into runoff from experimental panels of 14 roofing materials over 4.5 years of weathering. Ten roofing materials leached metals. Several leached >10 ppb during one or more study periods. The most common correlate with metal concentration was panel age, followed by precipitation amount. Extrapolating from these observations, we estimated the loading of metals from each roofing material during the first 10 years following installation. Eight materials were predicted to leach metals above background at the end of the 10 years. In combination with information on the prevalence of different roofing materials in the Puget Sound region of the Pacific Northwest, we estimated the relative amount of metals contributed from roofing materials in this basin. Most arsenic and copper was estimated to be contributed by residential roofing; nearly all arsenic from wood shakes manufactured with copper chromated arsenic, and copper contributed mainly from treated wood shakes followed by copper granule-containing asphalt shingles. Most zinc was estimated to be contributed by commercial roofs, including Zincalume and painted metal roofs. Overall our data shows that roofing materials can be an important long-term source of As, Cu, and Zn to stormwater runoff. Compared with atmospheric deposition, roof materials were a significant source, particularly of As and Cu. To get a complete picture of metals sourced from buildings, there is a need to study whole roof systems, including gutters, downspouts, and HVAC systems, as well as metals contributed from homeowner-applied treatments to their roofs.

Associations between single metal and fasting blood glucose (FBG) levels have been reported in previous studies. However, the association between multi-metals exposure and FBG level are little known. To assess the joints of arsenic (As), nickel (Ni), cadmium (Cd), selenium (Se), and zinc (Zn) co-exposure on FBG levels, Bayesian kernel machine regression (BKMR) statistical method was used to estimate the potential joint associations between As, Ni, Cd, Se, and Zn co-exposure and FBG levels among 1478 community-based Chinese adults from two counties, Shimen (n = 696) and Huayuan (n = 782), with different exposure profiles in Hunan province of China. The metals levels were measured in spot urine (As, Ni, and Cd) and plasma (Se and Zn) using inductively coupled plasma-mass spectrometry, respectively. The exposure levels of all the five metals were significantly higher in Shimen area (median: As = 57.76 μg/L, Cd = 2.75 μg/L, Ni = 2.73 μg/L, Se = 112.67 μg/L, Zn = 905.68 μg/L) than those in Huayuan area (As = 41.14 μg/L, Cd = 2.22 μg/L, Ni = 1.88 μg/L, Se = 65.59 μg/L, Zn = 819.18 μg/L). The BKMR analyses showed a significantly positive over-all effect of the five metals on FBG levels when metals concentrations were all above the 50th percentile while a statistically negative over-all effect when metals concentrations were all under the 50th percentile in Shimen area. However, a totally opposite over-all effect of the mixture of the five metals on FBG levels was found in Huayuan area. BKMR also revealed a non-linear exposure-effect of Zn on FBG levels in Huayuan area. In addition, interaction effects of As and Se on FBG level were observed. The relationship between single or combined metals exposure and FBG was different against different exposure profiles. Potential interaction effects of As and Se on FBG levels may exist.

Microcystin-leucine-arginine (MC-LR) can cause male reproductive disorder. However, the underlying mechanism are not yet entirely elucidated. In this study, we aimed to investigated the effects of MC-LR on the integrity of blood-testis barrier (BTB) and the related molecular mechanisms. Both in vivo and in vitro experiments revealed that MC-LR caused disruption of BTB and gap junctions between Sertoli cells respectively, which was paralleled by the alteration of connexin43 (Cx43). Our data demonstrated that MC-LR decreased gap junction intercellular communication (GJIC) and impaired Cx43 expression by activating the phosphatidylinositol 3-kinase/Akt cascades. In addition, a possible protective effect of Icariin (ICA), a flavonoid isolated from Chinese medicinal herb, against MC-LR toxicity was investigated. The ICA prevented the degradation of GJIC and impairment of Cx43 induced by MC-LR via suppressing the Akt pathway. Together, our results confirmed that the expression of Cx43 induced by MC-LR was regulated in vivo and in vitro, which was involved in the destruction of BTB. Additionally, ICA seems to be able to mitigate the MC-LR toxic effects.

Nitrogen deposition and tropospheric ozone are important drivers of vegetation damage, but their interactive effects are poorly understood. This study assessed whether long-term nitrogen deposition altered sensitivity to ozone in a semi-natural vegetation community. Mesocosms were collected from sand dune grassland in the UK along a nitrogen gradient (5–25 kg N/ha/y, including two plots from a long-term experiment), and fumigated for 2.5 months to simulate medium and high ozone exposure. Ozone damage to leaves was quantified for 20 ozone-sensitive species. Soil solution dissolved organic carbon (DOC) and soil extracellular enzymes were measured to investigate secondary effects on soil processes.Mesocosms from sites receiving the highest N deposition showed the least ozone-related leaf damage, while those from the least N-polluted sites were the most damaged by ozone. This was due to differences in community-level sensitivity, rather than species-level impacts. The N-polluted sites contained fewer ozone-sensitive forbs and sedges, and a higher proportion of comparatively ozone-resistant grasses. This difference in the vegetation composition of mesocosms in relation to N deposition conveyed differential resilience to ozone.Mesocosms in the highest ozone treatment showed elevated soil solution DOC with increasing site N deposition. This suggests that, despite showing relatively little leaf damage, the ‘ozone resilient’ vegetation community may still sustain physiological damage through reduced capacity to assimilate photosynthate, with its subsequent loss as DOC through the roots into the soil.We conclude that for dune grassland habitats, the regions of highest risk to ozone exposure are those that have received the lowest level of long-term nitrogen deposition. This highlights the importance of considering community- and ecosystem-scale impacts of pollutants in addition to impacts on individual species. It also underscores the need for protection of ‘clean’ habitats from air pollution and other environmental stressors.
