Perfluorinated substances and telomeres in an Arctic seabird: Cross-sectional and longitudinal approaches

Telomeres are non-coding DNA repeats located at the termini of eukaryotic chromosomes, regulated by dynamic processes balancing shortening and maintenance. Despite a mechanism to slow-down telomere shortening, cell division leads to progressive attrition of chromosomes, leading to the onset of cellular senescence or apoptosis. However, telomere restoration based on telomerase activity is the primary mechanism for telomere maintenance. Telomere length is associated to health and survival and can be impacted by a broad panel of environmental factors. However, the effect of contaminants on telomeres is poorly known for living organisms. The aim of this study was to investigate relationships between some poly- and perfluoroalkyl substances (PFASs), body condition and telomere length by using both a crosssectional and longitudinal approach in adult breeding Black-legged kittiwakes (Rissa tridactyla) from Svalbard. First, we examined the associations between absolute telomere length and PFASs contamination in a given year (cross-sectional approach). Second, we investigated the relationships between telomere dynamics and PFASs contamination within a two years’ time frame (longitudinal approach). Our results did not show any significant relationships of PFASs and body condition with absolute telomere length in a given year. Surprisingly, we found a positive and significant relationship between PFASs and telomere dynamics in both sexes with elongated telomere in birds bearing the highest concentrations of PFASs. Our study underlines (i) the need to investigate PFAS effects on telomere dynamics with a longitudinal approach and (ii) a potential positive effect of these contaminants on telomere length, with the most contaminated birds showing the slowest rate of telomere shortening or even displaying elongated ones. Our study is the first to report a relationship between PFASs and telomere length in freeliving vertebrates. A possible underlying mechanism and other potential confounding factors are discussed. Ambient air is a core media chosen for monitoring under the Stockholm Convention on POPs. While extensive monitoring of POPs in ambient air has been carried out in some parts of the globe, there are still regions with very limited information available, such as some developing countries as Nepal. This study therefore aims to target the occurrence of selected POPs in Nepal in suspected source areas/more densely populated regions. Four potential source regions in Nepal were furthermore targeted as it was hypothesized that urban areas at lower altitudes (Birgunj and Biratnagar located at approximately 86 and 80 m.a.s.l.) would be potentially more affected by OCPs because of more intensive agricultural activities in comparison to urban areas at higher altitudes (Kathmandu, Pokhara located 1400 and 1135 m.a.s.l). As some of these areas could also be impacted by LRAT, air mass back trajectories during the sampling period were additionally evaluated using HYSPLIT. The concentrations of overall POPs were twice as high in plain areas in comparison to hilly areas. DDTs and HCHs were most frequently detected in the air samples. The high p,p'-DDT/(pp'-DDE + pp'-DDD) ratio as well as the low o,p'-DDT/p,p'-DDT ratio observed in this study was inferred as continuing use of technical DDT. High levels of Sum26PCBs were linked to proximity to highly urbanized and industrial areas, indicating the potential source of PCBs. The measured concentrations of legacy POPs in air from this study is assumed to represent a negligible health risk through inhalation of ambient air, however, other modes of human exposure could still be relevant in Nepal. peer reviewed | Microplastics (MPs) are thought to be ingested by a wide range of marine organisms before being excreted. However, several studies in marine organisms from different taxa have shown that MPs and nanoplastics could be translocated in other organs. In this study, we investigated the presence of MPs in the livers of commercial zooplanktivorous fishes collected in the field. The study focuses mainly on the European anchovy Engraulis encrasicolus but concerns also the European pilchard Sardina pilchardus and the Atlantic herring Clupea harengus. Two complementary methodologies were used to attest the occurrence of MPs in the hepatic tissue and to exclude contamination. 1) MPs were isolated by degradation of the hepatic tissue. 2) Cryosections were made on the livers and observed in polarized light microscopy. Both methods separately revealed that MPs, mainly polyethylene (PE), were translocated into the livers of the three clupeid species. In anchovy, 80 per cent of livers contained relatively large MPs that ranged from 124 μm to 438 μm, showing a high level of contamination. Two translocation pathways are hypothesized: (i) large particles found in the liver resulted from the agglomeration of smaller pieces, and/or (ii) they simply pass through the intestinal barrier. We simulate oil spills of 1500 and 4500 m3/day lasting 14, 45, and 90 days in the spawning grounds of the commercial fish species, Northeast Arctic cod. Modeling the life history of individual fish eggs and larvae, we predict deviations from the historical pattern of recruitment to the adult population due to toxic oil exposures. Reductions in survival for pelagic stages of cod were 0–10%, up to a maximum of 43%. These reductions resulted in a decrease in adult cod biomass of < 3% for most scenarios, up to a maximum of 12%. In all simulations, the adult population remained at full reproductive potential with a sufficient number of juveniles surviving to replenish the population. The diverse age distribution helps protect the adult cod population from reductions in a single year's recruitment after a major oil spill. These results provide insights to assist in managing oil spill impacts on fisheries. | publishedVersion

A comprehensive approach to chemical accumulation and biological effects of short-term Cu exposure in juveniles of European seabass (Dicentrarchus labrax) has been achieved. Fish were exposed to 0.01–10 mg L− 1 nominal Cu concentrations for 24–96 h. Metal concentrations in water and gills, liver, muscle and brain tissues were studied along with oxidative stress biomarkers (superoxide dismutase, catalase, glutathione peroxidase, lipid peroxidation). Induction of oxidative damage was observed in all the organs with differential antioxidant responses; gills appearing as the most sensitive from low environmentally water Cu concentrations as 0.01 mg L− 1. Histopathological alterations were also observed in liver and gills, even without a significant Cu accumulation. The results show that the combination of oxidative stress parameters, particularly lipid peroxidation and glutathione peroxidase activities, and histopathological alterations provide a good model fish and reliable early biomarkers for monitoring Cu pollution in seawater and might call for the protection agencies to revise the Cu environmental standards.

Recent data indicate accumulation areas of marine litter in Arctic waters and significant increases over time. Beaches on remote Arctic islands may be sinks for marine litter and reflect pollution levels of the surrounding waters particularly well. We provide the first quantitative data from surveys carried out by citizen scientists on six beaches of Svalbard. Litter quantities recorded by cruise tourists varied from 9-524 g m-2 and were similar to those from densely populated areas. Plastics accounted for >80% of the overall litter, most of which originated from fisheries. Photographs provided by citizens show deleterious effects of beach litter on Arctic wildlife, which is already under strong pressure from global climate change. Our study highlights the potential of citizen scientists to provide scientifically valuable data on the pollution of sensitive remote ecosystems. The results stress once more that current legislative frameworks are insufficient to tackle the pollution of Arctic ecosystems.

This study reports the first evidence of the occurrence of perfluoroalkyl acids (PFAAs) in commonly consumed seafood from the coastal area of Bangladesh. Fifteen target PFAAs in 48 seafood samples (5 finfish and 2 shellfish species) were measured by HPLC-MS/MS. The results were comparable with other studies worldwide, particularly from China, Spain, Sweden, and USA. The majority of monitored PFAAs did not show clear seasonal variation. However, seafood from the southeast area (Coxs Bazar and Chittagong) showed relatively higher levels of PFAAs. Moreover, the dietary exposure assessment revealed that the daily intakes of PFAAs via seafood consumption were far less than the health-based guidelines, indicating low health risk for the Bangladeshi coastal residents."

peer reviewed | Swordfish (Xiphias gladius L., 1758) is an apex predator, highly migratory meso-pelagic fish widely distributed in the Atlantic Ocean and Mediterranean. As top predators, this fish may be the end reservoir of the bioaccumulation of trace elements in a food chain because they occupy higher trophic levels and are an important food source, causing them to be potentially hazardous to consume. This study aims to investigate the concentration of 18 trace elements of Swordfish, caught in the Mediterranean Sea and to discuss human exposure risks. The mean element levels in the fish muscles were clearly below the maximum allowable concentrations established by International food safety regulations. The data suggested that the risk is minor and acceptable for human health.
