Airborne fine particulate matter induces an upregulation of endothelin receptors on rat bronchi Полный текст

Airborne fine particulate matter (PM2.5) is a risk factor for respiratory diseases. However, little is known about the effects of PM2.5 on bronchi. The present study investigated the effect of airborne PM2.5 on rat bronchi and the underlying mechanisms. Isolated rat bronchial segments were cultured for 24 h. Endothelin (ET) receptor-mediated contractile responses were recorded using a wire myograph. The mRNA and protein expression levels of ET receptors were studied using quantitative real-time PCR, Western blotting, and immunohistochemistry. The results demonstrated that ETA and ETB receptor agonists induced remarkable contractile responses on fresh and cultured bronchial segments. PM2.5 (1.0 or 3.0 μg/ml) significantly enhanced ETA and ETB receptor-mediated contractile responses in bronchi with a markedly increased maximal contraction compared to the DMSO or fresh groups. PM2.5 increased the mRNA and protein expression levels of ETA and ETB receptors. U0126 (a MEK1/2 inhibitor) and SB203580 (a p38 inhibitor) significantly suppressed PM2.5-induced increases in ETB receptor-mediated contractile responses, mRNA and protein levels. SP600125 (a JNK inhibitor) and SB203580 significantly abrogated the PM2.5-induced enhancement of ETA receptor-mediated contraction and receptor expression. In conclusion, PM2.5 upregulates ET receptors in bronchi. ETB receptor upregulation is associated with MEK1/2 and p38 pathways, and the upregulation of ETA receptor is involved in JNK and p38 pathways.

Estuarine wetlands are major contributors to mercury (Hg) transformation into its more toxic form, methylmercury (MeHg). Although these complex habitats are important, estuarine Hg bioaccumulation is not well understood. The longnose gar Lepisosteus osseus (L. 1758), an estuarine predator in the eastern United States, was selected to examine Hg processes due to its abundance, estuarine residence, and top predator status. This study examined variability in Hg concentrations within longnose gar muscle tissue spatially and temporally, the influence of biological factors, potential maternal transfer, and potential negative health effects on these fish. Smaller, immature fish had the highest Hg concentrations and were predominantly located in low salinity waters. Sex and diet were also important factors and Hg levels peaked in the spring. Although maternal transfer occurred in small amounts, the potential negative health effects to young gar remain unknown. Fish health as measured by fecundity and growth rate appeared to be relatively unaffected by Hg at concentrations in the present study (less than 1.3 ppm wet weight). The analysis of biotic and abiotic factors relative to tissue Hg concentrations in a single estuarine fish species provided valuable insight in Hg bioaccumulation, biomagnification, and elimination. Insights such as these can improve public health policy and environmental management decisions related to Hg pollution.

While the spatial distributions of heavy metals in farmland soil of China have been comprehensively delineated, their temporal trends are rarely investigated but are important for environmental risk management. In this study, the current status and temporal trends of heavy metals in the farmland soil of Yangtze River Delta (YRD) were evaluated through field survey and meta-analysis. The field survey conducted in 2014 showed that the concentrations of Cd, Pb, Cu, Zn, and Ni in the farmland topsoil were 0.23 ± 0.14, 37.63 ± 15.60, 25.83 ± 41.62, 88.38 ± 43.30, and 29.21 ± 12.41 mg kg−1 (mean ± standard deviation), respectively. The heavy metals showed relatively higher concentrations on the borders among Zhejiang, Jiangsu, and Shanghai. In the meta-analysis, we selected 68 published studies related to heavy metal pollution in farmland topsoil of YRD from 2000 to the year (2014) when the field survey was conducted. The results show an increasing trend for Cd (p < 0.05; 0.0081 mg kg−1 year−1), a decreasing trend for Cu (p < 0.05; -0.80 mg kg−1 year−1), and no significant trend for Pb (p = 0.155), Zn (p = 0.746), and Ni (p = 0.305). The increasing rate of Cd from the meta-analysis is consistent with the rate (0.0013 mg kg−1 year−1) derived from the mass balance calculation for Cd, where atmospheric deposition originated from intensive coal combustion is considered as the main source of Cd in the topsoil. The decreasing trend of Cu is likely due to largely reduced application of copper-based agrochemicals. Environmental regulation and soil remediation are needed to protect food safety and ecosystem from heavy metal pollution, especially Cd.

Plastic is ubiquitous in global oceans and constitutes a newly available habitat for surface-associated bacterial assemblages. Microplastics (plastic particles <5 mm) are especially susceptible to ingestion by marine organisms, as the size of these particles makes them available also to lower trophic levels. Because many marine invertebrates harbour potential pathogens in their guts, we investigated whether bacterial assemblages on polystyrene are selectively modified during their passage through the gut of the lugworm Arenicola marina and are subsequently able to develop pathogenic biofilms. We also examined whether polystyrene acts as a vector for gut biofilm assemblages after subsequent incubation of the egested particles in seawater. Our results showed that after passage through the digestive tract of A. marina, the bacterial assemblages on polystyrene particles and reference glass beads became more similar, harbouring common sediment bacteria. By contrast, only in the presence of polystyrene the potential symbiont Amphritea atlantica was enriched in the investigated biofilms, faeces, and water. Thus, especially in areas of high polystyrene contamination, this polymer may impact the bacterial composition of different habitats, with as yet unknown consequences for the respective ecosystems.

The present study is the first assessment of occupational risk associated with artisanal cashew nut roasting using exposure and effect biomarkers, as well as a characterization and dispersion analysis of the released particulate matter (PM). A real-time particle monitor was used to quantify PM1.0, PM2.5 and PM10. Furthermore, the PM was sampled using a Handi-vol sampler, and the physicochemical characteristics were determined by SEM-EDS analysis. Trajectories, dispersion and deposition of the emitted material were calculated using the NOAA-HYSPLIT model. Urinary 1-hydroxypyrene (1-OHP) levels were analyzed by HPLC. DNA damage, chromosomal instability and cell death were measured by a buccal micronucleus cytome assay (BMCyt). The PM concentrations for all measurements in the exposed area were higher than in the non-exposed area. SEM-EDS analyses exhibited a wide variety of particles, and K, Cl, S and Ca biomass burning tracers were the major inorganic compounds. In addition, atmospheric modeling analysis suggested that these particles can reach regions farther away than 40 kilometers. Occupational polycyclic aromatic hydrocarbon exposure was confirmed by increases in 1-OHP levels in cashew nut workers. Frequencies of BMCyt biomarkers of genotoxicity (micronuclei and nuclear bud) and cytotoxicity (pyknosis, karyolysis, karyorrhexis and condensed chromatin) were higher in the exposed group compared with the controls. The influence of factors, such as age, on the micronuclei frequencies was demonstrated, and a correlation between 1-OHP and micronuclei was observed. To the best of our knowledge, no other study has demonstrated a correlation between these types of biomarkers. The use of exposure (1-OHP) and effect (BMCyt) biomarkers were therefore efficient in assessing the occupational risk associated with artisanal cashew nut roasting, and the high rates of PM2.5 are considered to be a potential contributor to this effect.

Dechlorane compounds, including Dechlorane Plus (DP), mirex, and Dechlorane (Dec) 602, 603 and 604, were measured in 61 serum samples collected from a Korean urban area (Seoul) in 2013. Dechlorane Plus and Dec 602 were dominant in most samples, whereas Dec 604 was not detected in any samples. The median value of DP was 0.75 ng/g lipid, which was comparable with the levels observed in Europe and Canada, but lower than that observed in China. Statistically significant correlations among Dechlorane compounds were observed. The median anti-isomer fractional abundance (fanti) was 0.74, and a negative correlation between fanti and the DP concentration in serum samples was observed. Polybrominated diphenyl ethers (PBDEs) were also measured for comparison with Dechlorane compounds, and BDE-153 was found the most abundant congener with a median value of 1.43 ng/g lipid. A time trend of Dechlorane compounds was investigated in 7 pooled serum samples to cover the period from 2006 to 2013. A distinct trend of Dechlorane compounds was not observed, while PBDEs steadily decreased with time.

Classical laboratory-based single-species sediment bioassays do not account for modifications to toxicity from bioturbation by benthic organisms which may impact predictions of contaminated sediment risk to biota in the field. This study aims to determine the effects of bioturbation on the toxicity of zinc measured in a standard laboratory bioassay conducted with chironomid larvae (Chironomus tepperi). The epi-benthic chironomid larvae were exposed to two different levels of sediment contamination (1600 and 1980 mg/kg of dry weight zinc) in the presence or absence of annelid worms (Lumbriculus variegatus) which are known to be tolerant to metal and to have a large impact on sediment properties through bioturbation.Chironomids had 5–6x higher survival in the presence of L. variegatus which shows that bioturbation had a beneficial effect on the chironomid larvae. Chemical analyses showed that bioturbation induced a flux of zinc from the pore water into the water column, thereby reducing the bioavailability of zinc in pore water to the chironomid larvae. This also suggested that pore water was the major exposure path for the chironomids to metals in sediment. During the study, annelid worms (Oligochaetes) produced a thin layer of faecal pellets at the sediment surface, a process known to: (i) create additional adsorption sites for zinc, thus reducing its availability, (ii) increase the microbial abundance that in turn could represent an additional food source for opportunistic C. tepperi larvae, and (iii) modify the microbial community’s structure and alter the biogeochemical processes it governs thus indirectly impact zinc toxicity.This study represents a contribution in recognising bioturbating organisms as “ecological engineers” as they directly and indirectly influence metal bioavailability and impact other sediment-inhabiting species. This is significant and should be considered in risk assessment of zinc levels (and other metals) in contaminated sediment when extrapolating from laboratory studies to the field.

Anthropogenic impacts have continuously intensified in mega urban centers with increasing urbanization and growing population. The spatial distribution pattern of such impacts can be assessed with soil halogenated flame retardants (HFRs) as HFRs are mostly derived from the production and use of various consumer products. In the present study, soil samples were collected from the Pearl River Delta (PRD), a large urbanized region in southern China, and its surrounding areas and analyzed for a group of HFRs, i.e., polybrominated diphenyl ethers (PBDEs), decabromodiphenyl ethane, bis(hexachlorocyclopentadieno)cyclooctane (DP) and hexabromobenzene. The sum concentrations of HFRs and PBDEs were in the ranges of 0.66–6500 and 0.37–5700 (mean: 290 and 250) ng g−1 dry weight, respectively, around the middle level of the global range. BDE-209 was the predominant compound likely due to the huge amounts of usage and its persistence. The concentrations of HFRs were greater in the land-use types of residency, industry and landfill than in agriculture, forestry and drinking water source, and were also greater in the central PRD than in its surrounding areas. The concentrations of HFRs were moderately significantly (r2 = 0.32–0.57; p < 0.05) correlated with urbanization levels, population densities and gross domestic productions in fifteen administrative districts. The spatial distribution of DP isomers appeared to be stereoselective as indicated by the similarity in the spatial patterns for the ratio of anti-DP versus the sum of DP isomers (fanti-DP) and DP concentrations. Finally, the concentrations of HFRs sharply decreased with increasing distance from an e-waste recycling site, indicating that e-waste derived HFRs largely remained in local soil.

This study investigated the occurrence of 8 polybrominated diphenyl ether (PBDE) congeners from homes (n = 20), offices (n = 20), air conditioners (n = 6), and computers (n = 6). High detection frequencies for most of the congeners were observed, indicating continued widespread use of Penta-, Octa- and Deca-BDE mixtures. The median concentrations of ∑PBDEs were 119 and 194 pg m−3 for home air and office air, respectively. Regarding dust, the median concentrations of ∑PBDEs were 239 and 437 ng g−1 for home and office dust, respectively. The ratios of the median concentrations of BDE-209 to ∑PBDEs were approximately 0.95 and 0.87 for home dust and office dust, respectively. The median concentrations of ∑PBDEs were 359 ng g−1 and 350 ng g−1 for dust on air conditioner filters and the back cabinet of the computer, respectively. The ratios of the median concentrations of BDE-209 to ∑PBDEs were approximately 0.58 and 0.46 for air conditioner and computer samples. Running air conditioners contributed to ΣPBDEs in office air through direct and indirect pathways. The daily intake of PBDEs was estimated to be 2630 pg (kg bw)−1 day−1 for toddlers in homes and 319 pg (kg bw)−1 day−1 for adults in homes and offices.

In this study, the effects of concentrations 0, 100, 250, 500, 750 and 1000 mg kg−1 of nanoscale zero-valent iron (nZVI) on germination, seedlings growth, physiology and toxicity mechanisms were investigated. The results showed that nZVI had no effect on germination, but inhibited the rice seedlings growth in higher concentrations (>500 mg kg−1 nZVI). The highest suppression rate of the length of roots and shoots reached 46.9% and 57.5%, respectively. The 1000mg kg−1 nZVI caused the highest suppression rates for chlorophyll and carotenoids, at 91.6% and 85.2%, respectively. In addition, the activity of antioxidant enzymes was altered by the translocation of nanoparticles and changes in active iron content. Visible symptoms of iron deficiency were observed at higher concentrations, at which the active iron content decreased 61.02% in the shoots, but the active iron content not decreased in roots. Interestingly, the total and available amounts of iron in the soil were not less than those in the control. Therefore, the plants iron deficiency was not caused by (i) deficiency of available iron in the soil and (ii) restraint of the absorption that plant takes in the available iron, while induced by (ⅲ) the transport of active iron from the root to the shoot was blocked. The cortex tissues were seriously damaged by nZVI which was transported from soil to the root, these were proved by scanning electron microscopy (SEM), transmission electron microscopy (TEM) and energy dispersive spectroscopy (EDS). This current study shows that the mechanism of iron deficiency in rice seedling was due to transport of active iron from the root to the shoot blocked, which was caused by the uptake of nZVI.
