Chronic low-level perfluorooctane sulfonate (PFOS) exposure promotes testicular steroidogenesis through enhanced histone acetylation Полный текст

Perfluorooctane sulfonate (PFOS), an artificial perfluorinated compound, has been associated with male reproductive disorders. Histone modifications are important epigenetic mediators; however, the impact of PFOS exposure on testicular steroidogenesis through histone modification regulations remains to be elucidated. In this study, we examined the roles of histone modifications in regulating steroid hormone production in male rats chronically exposed to low-level PFOS. The results indicate that PFOS exposure significantly up-regulated the expressions of StAR, CYP11A1 and 3β-HSD, while CYP17A1 and 17β-HSD were down-regulated, thus contributing to the elevated progesterone and testosterone levels. Furthermore, PFOS significantly increased the histones H3K9me2, H3K9ac and H3K18ac while reduced H3K9me3 in rat testis. It is known that histone modifications are closely involved in gene transcription. Therefore, to investigate the association between histone modifications and steroidogenic gene regulation, the levels of these histone marks were further measured in steroidogenic gene promoter regions by ChIP. It was found that H3K18ac was augmented in Cyp11a1 promoter, and H3K9ac was increased in Hsd3b after PFOS exposure, which is proposed to result in the activation of CYP11A1 and 3β-HSD, respectively. To sum up, chronic low-level PFOS exposure activated key steroidogenic gene expression through enhancing histone acetylation (H3K9ac and H3K18ac), ultimately stimulating steroid hormone biosynthesis in rat testis.

Based on the wind erosion equation and the use of moderate resolution imaging spectroradiometer (MODIS) satellite remote sensing data combined with parameter normalization processing, an optimized high spatial-temporal resolution soil fugitive dust (SFD) emission inventory compiling method was proposed in this study. The "2 + 26" cities in northern China, where heavy pollution frequently occurs, were used as a case study. Using the optimized method, we estimated that the PM₅₀, PM₁₀, and PM₂.₅ emissions from SFD of "2 + 26" cities in 2018 were 2,014,927, 1,007,463, and 151,120 tons, respectively. The dust emissions and emission factors of each city presented significant differences and were generally of a greater level in high-latitude areas (such as cities in Hebei Province) than in low-latitude areas (such as cities in Henan and Shandong Province). Moreover, with an increase in latitude, vegetation cover factors generally exhibit an upward trend, while temperature and rainfall exhibit a downward trend. The dust emissions in the different months showed significant differences. The total dust emission reached the highest level in "late winter–early spring" season (February to April), and the monthly emission accounted for 15–17% of the annual emissions. While in the "summer–autumn" season (July to November), it is the lowest level of the whole year, monthly emissions accounted for 3–5% of the annual emissions. The emission inventory method proposed in this study can provide a reference for dust emission assessment and further pollution prevention and control work.

The current recovery of mountain lakes from atmospheric acidification is increasingly affected (both accelerated and/or delayed) by climate change. We evaluated long-term trends in the ionic composition of 30 lakes situated in the alpine zone of the Tatra Mountains, and compared the rates of their recovery with model (MAGIC) simulations done 20 years ago for the 2003–2020 period. The observed recovery was faster than the model forecast, due to greater reductions in acidic deposition than projected. Trends in water composition were further modified by climate change. Rising temperatures increased the length of the growing season and retention of inorganic N and SO₄²⁻ more in soil-rich compared with soil-poor catchments. In contrast, elevated precipitation and an increase in rainfall intensity reduced water residence time in soils, and consequently reduced N retention, especially in soil-poor catchments. It is likely that increases in rainfall intensity and annual number of days without snow, along with air temperatures fluctuating around the freezing point elevated the physical erosion of rocks, especially in high-elevation, steep, and scree-rich areas where rocks are not thermally insulated and stabilized by soils. Weathering of exposed accessory calcite in the eroded granodiorite bedrock was a source of Ca²⁺ and HCO₃⁻, while S-bearing minerals likely contributed to lake water SO₄²⁻ and partly mitigated its deposition-related decrease in scree-rich catchments. The extent of climate effects on changes in the water composition of alpine lakes recovering from acidic deposition thus depended on elevation and cover of soil and scree in catchments. Our results highlight the need for incorporating dominant climate-related process into existing process-based models to increase their reliability in predicting the future development of lake water composition.

Heart development requires a precise temporal regulation of gene expression in cardiomyoblasts. Therefore, the transcriptional changes in differentiating cells can lead to congenital heart diseases. Although the genetic mutations underlie most of these alterations, exposure to environmental contaminants, such as bisphenol A (BPA), has been recently considered as a risk factor as well. In this study we investigated the genotoxic and epigenotoxic effects of BPA throughout cardiomyocyte differentiation. H9c2 cells (rat myoblasts) were exposed to 10 and 30 μM BPA before and during the last two days of cardiac-driven differentiation. Then, we have analysed the phenotypic and molecular modifications (at transcriptional, genetic and epigenetic level). The results showed that treated myoblasts developed a skeletal muscle cell-like phenotype. The transcriptional changes induced by BPA in genes codifying proteins involved in heart differentiation and function depend on the window of exposure to BPA. The exposure before differentiation repressed the expression of heart transcription factors (Hand2 and Gata4), whereas exposure during differentiation reduced the expression of cardiac-specific genes (Tnnt2, Myom2, Sln, and Atp2a1). Additionally, significant effects were observed regarding DNA damage and histone acetylation levels after the two periods of BPA exposure: in cells exposed to the toxicant the percentage of DNA repair foci (formed by the co-localization of γH2AX and 53BP1) increased in a dose-dependent manner, whereas the treatment with the toxicant triggered a decrease in the epigenetic marks H3K9ac and H3K27ac. Our in vitro results reveal that BPA seriously interferes with the process of cardiomyocyte differentiation, which could be related to the reported in vivo effects of this toxicant on cardiogenesis.

Despite the increased attention for temporal aspects of stressor interactions and for effects of warming in ecotoxicological studies, we lack knowledge on how different exposure durations to warming may affect pesticide sensitivity. We tested how three types of exposure duration to 4 °C warming (acute, developmental and transgenerational exposure to 24 °C vs 20 °C) shape the effect of the pesticide chlorpyrifos on two ecologically relevant fitness-related traits of mosquito larvae: heat tolerance and antipredator behaviour. Transgenerational (from the parental generation) and developmental (from the egg stage) warming appeared energetically more stressful than acute warming (from the final instar), because (i) only the latter resulted in an adaptive increase of heat tolerance, and (ii) especially developmental and transgenerational warming reduced the diving responsiveness and diving time. Exposure to chlorpyrifos decreased the heat tolerance, diving responsiveness and diving time. The impact of chlorpyrifos was lower at 24 °C than at 20 °C indicating that the expected increase in toxicity at 24 °C was overruled by the observed increase in pesticide degradation. Notably, although our results suggest that transgenerational warming was energetically more stressful, it did reduce the chlorpyrifos-induced negative effects at 24 °C on heat tolerance and the alarm escape response compared to acute warming. Our results provide important evidence that the exposure duration to warming may determine the impact of a pesticide under warming, thereby identifying a novel temporal aspect of stressor interactions in risk assessment.

The frequency and duration of exposure to acrylamide (AA) from the environment and diet are associated with a range of adverse health effects. However, whether long-term AA exposure is related to diabetes mellitus (DM) remains unknown. Data from 3577 adults in the National Health and Nutrition Examination Survey (NHANES) 2005–2006 and 2013–2016 aged ≥ 20 years was analysed. The main analyses applied multivariate logistic regression and restricted cubic spline models to investigate the associations between DM and AA haemoglobin biomarkers, including haemoglobin adducts of acrylamide and glycidamide (HbAA and HbGA), the sum of HbAA and HbGA (HbAA + HbGA), and the ratio of HbGA to HbAA (HbGA/HbAA) levels. After multivariable adjustment, the odds ratios (95% confidence intervals) for DM comparing the highest with the lowest AA haemoglobin biomarker quartiles were 0.71 (0.55, 0.93), 0.92 (0.71, 1.18), 0.80 (0.62, 1.03) and 1.95 (1.51, 2.51) for HbAA, HbGA, HbAA + HbGA and HbGA/HbAA, respectively. The restricted cubic spline model demonstrated that HbAA was linearly and inversely associated with risk of DM (P for trend = 0.013), while HbGA/HbAA was nonlinearly and positively associated with the prevalence of DM (P for trend <0.001). These results support for epidemiological evidence that the HbAA and HbGA/HbAA are significantly associated with DM. Further studies are warranted to infer the causal role of AA exposure in the prevalence of DM.

It is highly desirable but remains extremely challenging to develop a facile strategy to prepare adsorbent for dealing with heavy metal pollution in water. Here, we report a facile approach for preparing sulfydryl-functionalized graphene oxide (S-GO) by cross-linking method with an unprecedented adsorption capacity and ultrahigh selectivity for efficient Hg(II) removal. The adsorbents exhibit a prominent performance in capturing Hg(II) from wastewater with a record-high adsorption capacity of 3490 mg/g and rapid kinetics to reduce Hg(II) contaminants below the discharge standard of drinking water (2 ppb) within 60 min under a wide pH range even in the coexistent of other interfering metal ions. In addition, the adsorbents can be also easily recycled and reused multiple times with no apparent decline in removal efficiency. Considering the broad diversity, we developed also a magnetic Fe₃O₄/S-GO adsorbent by a simple chemical cross-linking reaction to achieve rapid separation of S-GO from their aqueous solution. In addition, the adsorbents were successfully applied in dealing with the practical industrial wastewater. The results indicate the potential of rationally designed sulfydryl-functionalized graphene oxide for high performance Hg(II) removal.

A three-dimensional hydrodynamic-antibiotic model is developed to investigate the transport and dilution of sulfamethoxazole (SMX) in the Upper Gulf of Thailand (UGoT). The simulation produced a spatially averaged annual mean SMX concentration of 0.58 μgm−3, which varied slightly between seasons assuming a temporally constant river SMX loading observed in August. In contrast, the horizontal distribution of SMX concentrations strongly varied with season because of the changing residual currents. In addition, SMX is diluted to concentrations lower than 10% of those in river waters a short distance offshore of the estuaries. To better understand this behavior, we examined the relationship between salinity and SMX concentrations in the UGoT. The annual budget demonstrates that 98% of SMX in the UGoT is removed by natural decomposition. As the concentrations of fluvial pollutants in the UGoT depend on their river loading and decomposition rates, functions were derived to predict pollutant concentrations and flushing times based on the river input flux and half-life.

Despite the ubiquitousness of microplastics, knowledge on the exposure of freshwater fish to microplastics is still limited. Moreover, no standard methods are available for analyzing microplastics, and the quality of methods used for the quantification of ingested microplastics in fish should be improved. In this study, we studied microplastic ingestion of common wild freshwater fish species, perch (Perca fluviatilis) and vendace (Coregonus albula). Further, our aim was to develop and validate imaging Fourier-transform infrared spectroscopic method for the quantification of ingested microplastics. For this purpose, enzymatically digested samples were measured with focal plane array (FPA) based infrared microscope. Data was analyzed with siMPle software, which provides counts, mass estimations, sizes, and materials for the measured particles. Method validation was conducted with ten procedural blanks and recovery tests, resulting in 75% and 77% recovery rates for pretreatment and infrared imaging, respectively. Pretreatment caused contamination principally by small <100 μm microplastics. The results showed that 17% of perch and 25% of vendace had ingested plastic. Most of the fish contained little or no plastics, while some individuals contained high numbers of small particles or alternatively few large particles. Perch from one sampling site out of five had ingested microplastics, but vendace from all sampling sites had ingested microplastics. The microplastics found from fish were mostly small: 81% had particle size between 20 and 100 μm, and most of them were polyethylene, polypropylene, and polyethylene terephthalate. In conclusion, the implemented method revealed low numbers of ingested microplastics on average but needs further development for routine monitoring of small microplastics.

Selective removal of arsenic (As) is the key challenge for any of As removal mechanisms as this not only increases the efficiency of removal of the main As species (neutral As(III) and As(V) hydroxyl-anions) but also allows for a significant reduction of waste as it does not co-remove other solutes. Selective removal has a number of benefits: it increases the capacity and lifetime of units while lowering the cost of the process. Therefore, a sustainable selective mitigation method should be considered concerning the economic resources available, the ability of infrastructure to sustain water treatment, and the options for reuse and/or safe disposal of treatment residuals. Several methods of selective As removal have been developed, such as precipitation, adsorption and modified iron and ligand exchange. The biggest challenge in selective removal of As is the presence of phosphate in water which is chemically comparable with As(V). There are two types of mechanisms involved with As removal: Coulombic or ion exchange; and Lewis acid-base interaction. Solution pH is one of the major controlling factors limiting removal efficiency since most of the above-mentioned methods depend on complexation through electrostatic effects. The different features of two different As species make the selective removal process more difficult, especially under natural conditions. Most of the selective As removal methods involve hydrated Fe(III) oxides through Lewis acid-base interaction. Microbiological methods have been studied recently for selective removal of As, and although there have been only a small number of studies, the method shows remarkable results and indicates positive prospects for the future.
