Occurrence of synthetic phenolic antioxidants and transformation products in urban and rural indoor dust

In this study, seven synthetic phenolic antioxidant (SPA) analogues were positively found in urban and rural indoor dust samples collected from Shandong province in China, among which the novel 2,4,6-tri-tert-butylphenol (AO 246), 2,6-di-tert-butyl-4-sec-butylphenol (DTBSBP), 2,4-di-tert-butylphenol (DBP) and 4,4'-butylidenebis (2-(1,1-dimethylethyl)-5- methyl-phenol) (AO 44B25) analogues accounted for 29% of total SPA concentrations (∑SPAs). Urban dust showed significantly higher ∑SPA levels (range: 1.56e3 - 2.03e4 ng/g) compared with those in rural indoor dust (668–4.39e3 ng/g, p < 0.05). 2,6-Di-tert-butyl-4-methylphenol (BHT) was the dominate analogue in the urban indoor dust, which constituted of 74% in ΣSPAs. While, varied composition profiles of SPAs were noticed in rural indoor dust, for instance, AO 246 (46%) and BHT (43%) had similar contributions to ∑SPAs. Three BHT transformation products (TPs) were also detected in most of the urban and rural dust samples (>97%), with individual residue level in the same order: 2,6-di-tert-butyl-1,4-benzoquinone (BHT-Q) > 2,6-di-tert-butyl-4-hydroxy- 4-methyl-2,5-cyclo-hexadienone (BHT-quinol) > 3,5-di-tert-butyl-4-hydroxybenzal-dehyde (BHT-CHO). Geometric mean values of total TP concentrations were 555 ng/g and 131 ng/g for urban and rural indoor dust samples, respectively. A preliminary estimated daily intake calculation at dust ingestion scenario suggested additional concerns might be paid to simultaneous exposure of several SPA analogues and TPs besides current focus on BHT exposure risks.

The main goal of the present study was to determine and validate an aquatic Maximum Acceptable Concentration-Environmental Quality Standard (MAC-EQS) value for the agricultural fungicide azoxystrobin (AZX). Assessment factors were applied to short-term toxicity data using the lowest EC50 and after the Species Sensitivity Distribution (SSD) method. Both ways of EQS generation were applied to a freshwater toxicity dataset for AZX based on available data, and to marine toxicity datasets for AZX and Ortiva® (a commercial formulation of AZX) obtained by the present study. A high interspecific variability in AZX sensitivity was observed in all datasets, being the copepoda Eudiaptomus graciloides (LC50,48h = 38 μg L⁻¹) and the gastropod Gibbula umbilicalis (LC50,96h = 13 μg L⁻¹) the most sensitive freshwater and marine species, respectively. MAC-EQS values derived using the lowest EC50 (≤0.38 μg L⁻¹) were more protective than those derived using the SSD method (≤3.2 μg L⁻¹). After comparing the MAC-EQS values estimated in the present study to the smallest AA-EQS available, which protect against the occurrence of prolonged exposure of AZX, the MAC-EQS values derived using the lowest EC50 were considered overprotective and a MAC-EQS of 1.8 μg L⁻¹ was validated and recommended for AZX for the water column. This value was derived from marine toxicity data, which highlights the importance of testing marine organisms. Moreover, Ortiva affects the most sensitive marine species to a greater extent than AZX, and marine species are more sensitive than freshwater species to AZX. A risk characterization ratio higher than one allowed to conclude that AZX might pose a high risk to the aquatic environment. Also, in a wider conclusion, before new pesticides are approved, we suggest to improve the Tier 1 prospective Ecological Risk Assessment by increasing the number of short-term data, and apply the SSD approach, in order to ensure the safety of aquatic organisms.

In response to the restrictions of polybrominated diphenyl ether (PBDE) flame retardants in various consumer products, alternative halogenated flame retardants have been subjected to increased use. Compared to aquatic ecosystems, relatively little information is available on the contamination of alternative flame retardants in terrestrial ecosystems, especially with regards to mammalian wildlife. In this study we used a top terrestrial carnivore, the bobcat (Lynx rufus), as a unique biomonitoring species for assessing flame retardant contamination in the Midwestern United States (U.S.) terrestrial ecosystems. Concentrations of ∑PBDEs (including all detectable PBDE congeners) ranged from 8.3 to 1920 ng/g lipid weight (median: 50.3 ng/g lw) in livers from 44 bobcats collected during 2013–2014 in Illinois. Among a variety of alternative flame retardants screened, Dechloranes (including anti- and syn-Dechlorane Plus and Dechlorane-602, 603, and 604), tetrabromo-o-chlorotoluene (TBCT), and hexabromocyclododecane (HBCD) were also frequently detected, with median concentrations of 28.7, 5.2, and 11.8 ng/g lw, respectively. Dechlorane analogue compositions in bobcats were different from what has been reported in other studies, suggesting species- or analogue-dependent bioaccumulation, biomagnification, or metabolism of Dechlorane chemicals in different food webs. Our findings, along with previously reported food web models, suggest Dechloranes may possess substantial bioaccumulation and biomagnification potencies in terrestrial mammalian food webs. Thus, attention should be given to these highly bioavailable flame retardants in future environmental biomonitoring and risk assessments in a post-PBDE era.

We explored acquired immunity resulting from vaccination in 3 to 7-year-old children, chronically exposed to multiple heavy metals and metalloids, in an e-waste recycling area (Guiyu, China). Child blood levels of ten heavy metals and metalloids, including lead (Pb), arsenic (As), mercury (Hg), chromium (Cr), cadmium (Cd), manganese (Mn), nickel (Ni), copper (Cu), zinc (Zn) and selenium (Se), and seven vaccine antibodies (diphtheria, pertussis, tetanus, hepatitis B, Japanese encephalitis, polio, measles) were measured. The exposed group had higher levels of blood Pb, Mn, Cu, Zn and Cr compared to the reference group (P < 0.05). Levels of all vaccine antibodies in the exposed group were significantly lower than in the reference group (P < 0.01). All vaccine antibodies negatively correlated with blood concentrations of Cu, Zn and Pb, based on spearman rank correlation analysis. Multiple logistic regression and univariate analyses identified the location of residence (Guiyu), high blood Pb (>10 μg/dL) and high blood Cu and Zn (upper median value of each group) to be inversely associated with seven antibody titers. Antibody titers increased with age, BMI, high blood Mn (>15 μg/L), and high blood Cd and Ni (upper median value of each group). Results suggest multiple heavy metal and metalloid exposure, especially to Pb, Zn and Cu, may be a risk factor inhibiting the development of child immunity, resulting in decreased child antibody levels against vaccines.

ZnO nanoparticles (ZnO-NPs) are emerging contaminants that raise the concerns of potential risk in the aquatic environment. It has been estimated that the environmental ZnO-NPs concentration is 76 μg/l in the aquatic environment. Our aim was to determine the aquatic toxicity of ZnO-NPs with chronic exposure at environmentally relevant concentrations using the nematode Caenorhabditis elegans. Two simulated environmentally relevant mediums—moderately hard reconstituted water (EPA water) and simulated soil pore water (SSPW)—were used to represent surface water and pore water in sediment, respectively. The results showed that the ZnO-NPs in EPA water has a much smaller hydrodynamic diameter than that in SSPW. Although the ionic release of Zn ions increased time-dependently in both mediums, the Zn ions concentrations in EPA water increased two-fold more than that in SSPW at 48 h and 72 h. The ZnO-NPs did not induce growth defects or decrease head thrashes in C. elegans in either media. However, chronic exposure to ZnO-NPs caused a significant reduction in C. elegans body bends in EPA water even with a relatively low concentration (0.05 μg/l); similar results were not observed in SSPW. Moreover, at the same concentrations (50 and 500 μg/l), body bends in C. elegans were reduced more severely in ZnO-NPs than in ZnCl2 in EPA water. The ATP levels were consistently and significantly decreased, and ROS was induced after ZnO-NPs exposure (50 and 500 μg/l) in EPA water. Our results provide evidences that chronic exposure to ZnO-NPs under environmentally relevant concentrations causes metabolic and locomotive toxicities implicating the potential ecotoxicity of ZnO-NPs at low concentrations in aquatic environments.

Coastal upwelling prevails in the coast of Hainan Island, the northern South China Sea (SCS) during summer. We studied the influences of the upwelling on the horizontal and vertical transport of terrigenous polycyclic aromatic hydrocarbons (PAHs). PAHs in dissolved and suspended particulate phase of water samples were determined in the upper (depth < 1 m) and water column (depth > 10 m). PAH levels decreased sharply from inshore to offshore to open sea. The results showed that terrestrial input was the main source of coastal PAHs. Perylene, an important indicator of land plant-derived PAH, showed the significant correlation with PAHs (p < 0.005). This implied that fluvial transport was the primary pathway of terrigenous PAHs into the coast of northern SCS. Variations of the concentrations, compositions and diagnostic ratios of PAHs, accompanied the partition equilibrium in the water column, could indicate the selective degradation of PAHs by the plankton affected by upwelling. Different from the “traditional” transport pathway of PAHs in the water column (surface enrichment-depth depletion distribution), the upwelling could provide the original driver to elevate the upward diffusion of sediment entrained contaminants towards the intermediate even the upper waters. It could also enhance the outward diffusion of terrigenous PAHs accompanied by the offshore transport of the upper waters. Therefore, the transport pathway of PAHs can be summarized by the coastal upwelling rising PAHs with their subsequent transport offshore and settling in the adjacent open sea.

We studied seasonal and physiological influences on mercury concentrations in western grebes (Aechmophorus occidentalis) and Clark's grebes (A. occidentalis) across 29 lakes and reservoirs in California, USA. Additionally, at three of these lakes, we conducted a time series study, in which we repeatedly sampled grebe blood mercury concentrations during the spring, summer, and early fall. Grebe blood mercury concentrations were higher among males (0.61 ± 0.12 μg/g ww) than females (0.52 ± 0.10 μg/g ww), higher among Clark's grebes (0.58 ± 0.12 μg/g ww) than western grebes (0.51 ± 0.10 μg/g ww), and exhibited a strong seasonal pattern (decreasing by 60% from spring to fall). Grebe blood THg concentrations exhibited a shallow, inverse U-shaped pattern with body size, and was lowest among the smallest and largest grebes. Further, the relationship between grebe blood mercury concentrations and wing primary feather molt exhibited a shallow U-shaped pattern, where mercury concentrations were highest among birds that had not yet begun molting, decreased approximately 24% between pre-molt and late molt, and increased approximately 19% from late molt to post-molt. Because grebes did not begin molting until mid-summer, lower grebe blood mercury concentrations observed in late summer and early fall were consistent with the onset of primary feather molt. However, because sampling date was a much stronger predictor of grebe mercury concentrations than molt, other seasonally changing environmental factors likely played a larger role than molt in the seasonal variation in grebe mercury concentrations. In the time series study, we found that seasonal trends in grebe mercury concentrations were not consistent among lakes, indicating that lake-specific variation in mercury dynamics influence the overall seasonal decline in grebe blood mercury concentrations. These results highlight the importance of accounting for sampling date, as well as ecological processes that may influence mercury concentrations, when developing monitoring programs to assess site-specific exposure risk of mercury to wildlife.

Perfluorooctanesulfonic acid (PFOS) is a ubiquitous environmental contaminant, previously utilized as a non-stick application for consumer products and firefighting foam. It can cross the placenta, and has been repeatedly associated with increased risk for diabetes in epidemiological studies. Here, we sought to establish the hazard posed by embryonic PFOS exposures on the developing pancreas in a model vertebrate embryo, and develop criteria for an adverse outcome pathway (AOP) framework to study the developmental origins of metabolic dysfunction. Zebrafish (Danio rerio) embryos were exposed to 16, 32, or 64 μM PFOS beginning at the mid-blastula transition. We assessed embryo health, size, and islet morphology in Tg(insulin-GFP) embryos at 48, 96 and 168 hpf, and pancreas length in Tg(ptf1a-GFP) embryos at 96 and 168 hpf. QPCR was used to measure gene expression of endocrine and exocrine hormones, digestive peptides, and transcription factors to determine whether these could be used as a predictive measure in an AOP. Embryos exposed to PFOS showed anomalous islet morphology and decreased islet size and pancreas length in a U-shaped dose-response curve, which resemble congenital defects associated with increased risk for diabetes in humans. Expression of genes encoding islet hormones and exocrine digestive peptides followed a similar pattern, as did total larval growth. Our results demonstrate that embryonic PFOS exposures can disrupt pancreatic organogenesis in ways that mimic human congenital defects known to predispose individuals to diabetes; however, future study of the association between these defects and metabolic dysfunction are needed to establish an improved AOP framework.

Simulating the storage of aerobic soils under water, the chemical speciation of heavy metals and arsenic was studied over a long-term reduction period. Time-dynamic and redox-discrete measurements in reactors were used to study geochemical changes. Large kinetic differences in the net-complexation quantities of heavy metals with sulfides was observed, and elevated pore water concentrations remained for a prolonged period (>1 year) specifically for As, B, Ba, Co, Mo, and Ni. Arsenic is associated to the iron phases as a co-precipitate or sorbed fraction to Fe-(hydr)oxides, and it is being released into solution as a consequence of the reduction of iron. The composition of dissolved organic matter (DOM) in reducing pore water was monitored, and relative contributions of fulvic, humic and hydrophylic compounds were measured via analytical batch procedures. Quantitative and qualitative shifts in organic compounds occur during reduction; DOM increased up to a factor 10, while fulvic acids become dominant over humic acids which disappear altogether as reduction progresses. Both the hydrophobic and hydrophilic fractions increase and may even become the dominant fraction.Reactive amorphous and crystalline iron phases, as well as dissolved FeII/FeIII speciation, were measured and used as input for the geochemical model to improve predictions for risk assessment to suboxic and anaerobic environments. The release of arsenic is related to readily reducible iron fractions that may be identified by 1 mM CaCl2 extraction procedure. Including DOM concentration shifts and compositional changes during reduction significantly improved model simulations, enabling the prediction of peak concentrations and identification of soils with increased emission risk. Practical methods are suggested to facilitate the practice of environmentally acceptable soil storage under water.

To support environmental management programs, there is an urgent need to know about the presence and understand the dynamics of major contaminants in seabird communities of key marine ecosystems. In this study, we investigated the concentrations and trophodynamics of trace elements in six seabird species and persistent organic pollutants (POPs) in three seabird species breeding on Grand Connétable Island (French Guiana), an area where the increase in human population and mining activities has raised concerns in recent years. Red blood cell Hg concentrations in adults were the highest in Magnificent frigatebirds Fregata magnificens (median: 5.6 μg g−1 dw; range: 3.8–7.8 μg g−1 dw) and lowest in Sooty terns Onychoprion fuscatus (median: 0.9 μg g−1 dw; range: 0.6–1.1 μg g−1 dw). Among POPs, dichlorodiphenyldichloroethylene (p,p’-DDE) was the most abundant compound in plasma of Cayenne terns Thalasseus sandvicensis (median: 1100 pg g−1 ww; range: 160 ± 5100 pg g−1 ww), while polychlorinated biphenyls (PCBs) were the most abundant compound class in plasma of Magnificent frigatebirds (median: 640 pg g−1 ww; range 330 ± 2700 pg g−1 ww). While low intensity of POP exposure does not appear to pose a health threat to this seabird community, Hg concentration in several adults Laughing gulls Leucophaeus atricilla and Royal terns Thalasseus maximus, and in all Magnificent frigatebirds was similar or higher than that of high contaminated seabird populations. Furthermore, nestling red blood cells also contained Hg concentrations of concern, and further studies should investigate its potential health impact in this seabird community. Differences in adult trophic ecology of the six species explained interspecific variation in exposure to trace element and POPs, while nestling trophic ecology provides indications about the diverse feeding strategies adopted by the six species, with the consequent variation in exposure to contaminants.
