Antibiotic resistome profile based on metagenomics in raw surface drinking water source and the influence of environmental factor: A case study in Huaihe River Basin, China

The contamination with antibiotic resistance genes (ARGs) in raw drinking water source may pose a direct threat to human health. In this study, metagenomics sequencing and analysis were applied to investigate the ARG pattern in 12 drinking water sources in upper and middle reach of Huaihe River Basin, China. Based on the redundant analysis and multi-linear regression model, location, specific microbial taxa, number of livestock and health facilities significantly influenced the ARG profile in drinking water sources. Besides the cluster effect of ARG in samples from plain and bedrock mountain areas, the samples from fracture aquifer areas also showed a distinctive biogeographic pattern with that from porous aquifer areas. Putative ARGs host Opitutus and Flavobacterium were the enriched biomarkers in plain and fracture aquifer area respectively, which mainly carried bacitracin, multidrug, beta-lactam and tetracycline ARGs. This result illuminated that both natural background and anthropogenic activities in the watershed influenced the ARG profile in natural freshwater system significantly. The low MGEs abundance and absence of pathogen revealed a low ARG dissemination risk in sampled drinking water sources, while Polynucleobacter was an abundant ARGs host and was significantly related to the ARG profile, which indicated that specific bacteria was responsible for ARGs propagation and accumulation in surface freshwater system. Further researches are needed to assess human exposure to raw drinking water source and the potential risk, as well as the species interaction in microbial community and its impact on ARG propagation under oligotrophic condition.

AmpC beta-lactamase genes are some of the most common antibiotic resistance genes and require special attention once they have become mobilised. The detection of these genes is well documented in clinical settings. However, there is insufficient knowledge of both plasmid and genomic AmpC genes in aquatic environments. This systematic review aimed to determine the extent of the knowledge gap in the literature regarding the prevalence of AmpC beta-lactamase genes in aquatic systems. Using selected criteria, a total of 27 databases were searched for applicable peer-reviewed journal articles. No date and language restrictions were applied. Journal articles that highlighted the detection of AmpC beta-lactamase genes in environmental aquatic systems, including wastewater treatment plants, were included. Of the 950 literature sources that were identified, 50 were selected for full text analysis based on predetermined criteria. Studies on AmpC genes detection were traced in 23 countries. These studies focused on surface water (24), wastewater (17), sea water (4) and both surface and wastewater (5). Most studies did not specifically aim to detect AmpC genes, but to detect antibiotic resistance genes in general. Presently no surveillance protocols, standardised detection methods or environmental limits exist for these genes and, due to a paucity of research in this field, it is unlikely that such systems will be implemented in the near future. The implications and dynamics of AmpC genes in aquatic systems remain unclear and require intense research to ensure the sustainability of environmental systems and human health.

This review evaluates the three dynamic models (biokinetic model: BK, physiologically based pharmacokinetic model: PBPK, and toxicokinetic-toxicodynamic model: TKTD) in our understanding of the key questions in metal ecotoxicology in aquatic systems, i.e., bioaccumulation, transport and toxicity. All the models rely on the first-order kinetics principle of metal uptake and elimination. The BK model basically treats organisms as a single compartment, and is both physiologically and geochemically based. With a good understanding of each kinetic parameter, bioaccumulation of metals in any aquatic organisms can be studied holistically and mechanistically. Modeling efforts are not merely restrained from the prediction of metal accumulation in the tissues, but instead provide the direction of the key processes that need to be addressed. PBPK is more physiologically based since it mainly addresses the transportation, transformation and distribution of metals in the organisms. It can be treated conceptually as a multi-compartmental kinetic model, whereas the physiology is driving the development of any good PBPK model which is no generic for aquatic animals and contaminants. There are now increasingly applications of the PBPK modeling specifically in metal studies, which reveal many important processes that are impossible to be teased out by direct experimental measurements without adequate modeling. TKTD models further focus on metal toxicity in addition to metal bioaccumulation. The TK part links exposure and bioaccumulation, while the TD part links bioaccumulation and toxic effects. The separation of TK and TD makes it possible to model processes, e.g., toxicity modification by environmental factors, interaction between different metals, at both the toxicokinetic and toxicodynamic levels. TKTD models provide a framework for making full use of metal toxicity data, and thus provide more information for environmental risk assessments. Overall, the three models reviewed here will continue to provide guiding principles in our further studies of metal bioaccumulation and toxicity in aquatic organisms.

Melatonin (Mel) serves as an important signalling molecule in various aspects of stress tolerance in plants. However, the function of Mel in pesticide metabolism remains unknown. Here, selecting the widely used fungicide carbendazim (MBC) as the model, we found that exogenous Mel had the ability to alleviate pesticide phytotoxicity and residues in tomato as well as in some other vegetables. Additionally, overexpression of the Mel biosynthetic gene caffeic acid O-methyltransferase 1 (COMT1) significantly enhanced the capacity of the tomato to reduce MBC phytotoxicity and residue. This outcome was mainly because of the Mel-induced antioxidant capability, as well as the key detoxification process. Indeed, levels of reactive oxygen species (ROS) and lipid peroxides significantly decreased after applying exogenous Mel or overexpressing COMT1, which resulted from direct ROS scavenging, and increased Mel levels significantly enhanced antioxidant enzymatic activity. More importantly, Mel activated the ascorbate-glutathione cycle to participate in glutathione S-transferase-mediated pesticide detoxification. A grafting experiment showed that rootstocks from COMT1 transgenic plants increased the Mel accumulation of wild-type scions, resulting in MBC metabolism in the scions. To our knowledge, this is the first report providing evidence of Mel-induced pesticide metabolism, which provides a novel approach for minimizing pesticide residues in crops by exploiting plant self-detoxification mechanisms.

Ecosystem services are present everywhere, green vegetation coverage (or green areas) is one of the primary sources of ecosystem services considering urban areas sustainability and peoples urban life quality. Urban vegetation cover loss decreases the capacity of nature to provision ecosystem services; the loss of urban vegetation is also observed within the Amazon. This study aims at identifying urban vegetation loss and relate it to the provision of ecosystem services of reduction of air quality, reduction of air pollution, and climate regulation. Urban vegetation coverage loss was calculated using NDVI on LANDSAT 5 imagery over a 23-year period from 1986 to 2009. NDVI thresholds were arbitrarily selected, and complemented by in locus observation, to establish guidelines for quantitative (area) and qualitative (density) evolution of green cover, divided in six different categories, named as water, bare soil, poor vegetation, moderate vegetation, dense vegetation and very dense vegetation. Data on air pollution, noise pollution and temperature were outsourced from previous works. Measurement show a significant loss of very dense, dense and moderate vegetation coverage and an increase in poor vegetation and bare soil areas, in accordance to increase in air and noise pollution, and local temperature, and provides positive refashions between the loss of urban green coverage and decrease in ecosystem services.

In the Kebili region of southern Tunisia, there is increasing demand of water from the Lower Cretaceous Continental Intercalaire (CI) aquifer and the Upper Cretaceous-Miocene Complex Terminal (CT) aquifer. The CI aquifer, given limited low recharge of water and increasing amounts of water extraction, has suffered intense overexploitation since the year 2000. Currently, the sustainability of CI resources is threatened by oil and brine contamination detected at a number of water wells in the Kebili region.Hydrocarbon pollution of the aquifers seems to be ubiquitous because the groundwaters sampled in El Fedjej and Nefzaoua basins exhibit bad water quality according to a number of toxicity indices. Geochemical data indicate that, on a regional scale, groundwater quality, salinity, and dissolved element concentrations are best correlated to petroleum contamination-extraction and to aquifer vulnerability to human perturbations rather than to multiple interactions within the hydrogeological system of the region.The analyses of petroleum compounds in sampled waters indicate that these waters are most consistent with increasing concentrations of organic pollutants; the organic matter is crude and unaltered, testifying to continuous flows of contaminants. The brine contamination, however, is limited to the Djemna water well where the water exhibits a salinity of 20 g L⁻¹. Combined, these findings suggest that groundwater composition in CI water wells in Kebili field is influenced by the migration of hydrocarbons and brine-enriched waters through fractures, and (or) by aquifer decompression.

Cement-solidification and chelator-stabilisation of municipal solid waste incineration fly ash (MSWI-FA) are two main treatment techniques to immobilise heavy metals. Differences in the long-term stabilities of those two methods of heavy-metal immobilisation were explored to aid in determining the better MSWI-FA treatment. However, few comparative studies have been conducted on 6-year-old cement-solidified FA (Ce-6-FA) and chelator-stabilised FA (Ch-6-FA). In this study, we compared the physicochemical and heavy metal leaching characteristics of Ce-6-FA and Ch-6-FA. The chemical speciation of heavy metals was modelled using geochemical software to assess long-term stability. The results showed weaker long-term stability in Pb immobilisation under the chelating system. The leaching concentrations of target heavy metals, acetic acid leaching tests, acid neutralising capacity, and pH-dependent leaching results indicated that Ce-6-FA had higher long-term stability than Ch-6-FA. A column experiment indicated that the cumulative release rates of Pb in Ce-6-FA and Ch-6-FA were 2.49% and 4.72%, respectively. The phase-controlled leaching of Pb in Ce-6-FA mainly occurred through Pb2(OH)3Cl and chloropyromorphite (Pb5(PO4)3Cl), whereas that in Ch-6-FA mainly occurred through Pb5(PO4)3Cl. The decomposition of heavy metal chelates in Ch-6-FA and salt generation in this process led to the release of Pb via the inorganic complex.

Civil aircraft emissions during landing and takeoff (LTO) are important air pollutant sources, but have been given insufficient attention in China. Accurate estimation of these emissions is limited by a lack of important parameters, such as detailed flight information and the dynamic time in climb and approach modes during LTO that are dependent on mixing layer height (MLH). We developed a flight-time/flight-height relationship using real-time height information in Aircraft Meteorological Data Relay data, and then calculated the actual time for each flight in those two modes based on the actual MLH from meteorological observation. Hourly emissions of civil aircraft were then estimated based on the database of each flight. Total emissions of NOx, CO, SO2, HC and PM from LTO cycles of domestic flights in China during 2015 were 37.78 Gg, 30.25 Gg, 12.00 Gg, 2.38 Gg and 0.75 Gg, respectively. Substantial monthly, daily and hourly variations of emissions due to the flight schedule as well as MLH were calculated. Large differences were found between the new estimation and emissions calculated based on traditional method. Compared with the emissions estimated based on default parameter obtained from International Civil Aviation Organization, the average difference of annual emission among airports with new estimation for various pollutants was approximately 30.3% in climb mode and 81.4% in approach mode; compared with the emissions estimated based on the method proposed by China National Guide, the average difference of annual emission among airports were 37.4% (NOx), 8.4% (CO), 73.1% (HC) and 58.1% (PM) during LTO process. The monthly airport-specific emissions per LTO were also proposed. These can provide necessary and meaningful support for the revision of the values in National Guide.

Hyperbranched polymers are nanomaterials belonging to the class of dendritic architectures with increasing applications in many diverse fields. We studied the toxicity of two hyperbranched polymers to the freshwater crustacean Daphnia magna. A hyperbranched hydroxyl-terminated polyester and a commercial hyperbranched polyamidoamine, Helux-3316 were tested for the acute immobilization of daphnids, the overproduction of reactive oxygen species and the activity of the antioxidant enzymes catalase and glutathione S-transferase. The effect for D. magna immobilization was higher for the hyperbranched polyamidoamine Helux-3316, which was attributed to the presence of primary amino groups on its surface. Following exposure to both hyperbranched polymers, a clear overproduction of reactive oxygen species took place accompanied by concentration-dependent enzymatic antioxidant response. Our results showed that the overproduction of reactive oxygen species activated antioxidant defence mechanisms and was responsible for the immobilization of daphnids exposed to both hyperbranched polymers. We showed evidence of the uptake of fluorescently labelled Helux-3316 that accumulated into the gastrointestinal tract of D. magna, and its removal via excretion within fecal pellets. This is the first work reporting the internalization of hyperbranched polymers in aquatic organisms.

This study analyzes microplastic ingestion by three deep-water elasmobranch species (Galeus melastomus, Scyliorhinus canicula and Etmopterus spinax) from the Tyrrhenian Sea, discriminating between stomach and intestine contents. The absence of significant differences in frequency and abundance of plastic items into stomachs seems to suggest that ecological diversity among the three sharks does not strongly influence the probability of plastic ingestion in the study area. On the other hand, the detected differences in the microplastic content into the intestine might be due to a different retention time of microplastics, suggesting how feeding habits could influence metabolic features, and therefore affect the recovery of ingested plastic items. This information would improve the future development of marine micro-litter monitoring systems, following the MSFD requirements. Moreover, this study shows that all the three examined elasmobranch species can give important information even with relatively small sample sizes (N ≈ 30), and they could be used as target species for monitoring micro-litter ingestion in deep-water habitats.
