Determination and Validation of Nitrate Levels of Vegetables Grown in the Mediterranean Region

In our study, validation was carried out at 24 and 200 mg/kg enrichment for residue analysis of nitrate active substance only in lettuce samples. Analyses were performed by using high performance liquid chromatography (HPLC) in lettuce, spinach, salad and arugula samples. In order to eliminate the matrix effect, matrix calibration was used, ie calibration of lettuce samples. Validation of the method was performed with criteria such as precision, repeatability, repeatability limit, reproducibility, repeatability limit, accuracy, linearity of calibration and recovery. The calibration curve was found as linear in the range of 0.2-40 mg/kg. The recovery from the samples is between 97.3-100.2 %. The repeatability and reproducibility values for the two concentrations were 1.89 (matrix+24 ppm) and 3.65 (matrix+200 ppm), 3.951 (matrix+24 ppm) and 3.456 (matrix+200 ppm), respectively. The repeatability and reproducibility limit values are within acceptable limits. Accuracy, precision, combined uncertainty and extended uncertainty (95% confidence: k=2) were found as 0.015 (matrix+24 ppm) and 0.004 (matrix+200 ppm), 0.039 (matrix+24 ppm) and 0.006 (matrix+200 ppm), 0.041 (matrix+24 ppm) and 0.007 (matrix+200 ppm), 0.082 (matrix+24 ppm) and 0.014 (matrix+200 ppm). In our study, the highest nitrate ratio was determined in spinach with 3486 mg/kg in total 102 samples. Lettuce following spinach (2825 mg/kg) showed lower nitrate content, while lower concentrations of nitrate were detected in iceberg (1985 mg/kg) and arugula (1870 mg/kg), respectively. The results may be useful in estimating body intake levels for research on the effects of nitrite and nitrate on human health in vegetable producers and in our daily diet. In addition, the nitrate content of the vegetables analysed in the study is an indicator of the use of nitrogenous fertilizers in the Mediterranean region, but showed no problem for human and animal health.

Lettuce (Lactuca sativa L.) is a member of the family Compositae (Asteraceae). This plant, which is grown for its leaves, is grown in open field and greenhouse conditions almost everywhere in the world and in Turkey. With the present study virus diseases of lettuce and their prevalence in Konya province was revealed for the first time. For this purpose, leaf samples were taken from lettuce plants showing virus diseases symptoms with surveys carried out in Konya province lettuce planting areas from May to August in 2020. Then the collected lettuce leaf samples were tested in laboratory conditions by Double Antibody Sandwich Enzyme-linked Immunosorbent Assay (DAS-ELISA) method to reveal infections of Cucumber mosaic virus (CMV), Miraflori lettuce big vein virus (MiLBVV), Lettuce mosaic virus (LMV), and Tomato spotted wilt virus (TSWV). According to the information obtained from the results of the study, it was determined that 40 out of 97 (41.23%) lettuce plant samples and all (6) weed samples were infected with at least one of the viruses. In lettuce leaf samples; TSWV (27.83%), LMV (12.37%), CMV (10.31%) and MiLBVV (5.15%) infections have been detected. In weed samples; infections of CMV (83.33%), LMV (66.66%), and TSWV (50%) have been revealed. The infections of TSWV, LMV, CMV, and MiLBVV on lettuce plants in Konya province were reported firstly with the study.

The experiment was conducted in one of the unheated plastic polyethylene (PE) greenhouses of Ege University- Bayındır Vocational Training School -TURKİYE during the winter-early spring of 2019-2020. The goal of the trial was to evaluate the effects of liquid vermicompost (LVC) on Iceberg (Head salad) and Lolo rosso (Lactuca sativa) lettuce in a floating hydroponic system. Two types of growing pools were used as the growing media, one was made of galvanized steel and the other plastic polyethylene. Both pools were covered with a polyethylene at a height of 1 meter from the ground. Lettuce seedlings were transplanted into pools filled with water and a week later Liquid Vermicompost (LVC) and Hoagland Nutrient Solution (HNS) were added to the growing media according to the treatments. The trial was set up in a Randomized Block Design (RBD) with 3 treatments and 3 replications in both of the lettuce varieties. 1. Application: HNS +LVC; 2. Application (HNS); 3. Application (LVC). Lettuce leaves were harvested 84 days after the LVC and HNS applications and prepared for analysis. In this research, in order to determine the impact of liquid vermicompost on Iceberg and Lolo rosso lettuce varieties total yield, marketable yield, the number of leaves discarded, root length and root weights were determined after harvested plants. In both of the lettuce varieties, results indicated that the highest marketable yields were obtained in the first treatment (HNS+LVC). The highest root length and root weight were also determined in the first treatment. It can be concluded that adding liquid vermicompost (100 ml / 100 L water ratio) to the growing solutions of Iceberg (Head lettuce) and Lolo rosso (Lactuca sativa) lettuce varieties, high performances can be obtained.

In this study, the effect of agribor fertilizer (18% pure B) on yield features of sugar beet were investigated. The study was conducted on ustifluvent subgroup soils according to soil taxonomy of agricultural faculty of Gaziosmanpaşa university. Randomized block experiment design with three replications were used and three treatments of agribor fertilizer 0.18 kg da-1 B (1 kg da -1 agribor); 0.27 kg da-1 B (1.5 kg da -1 agribor); 0.36 kg da-1 B (2 kg da -1 agribor) and control (no fertilizer) were applied. The effects of applied doses on yield properties were studied. From yield properties for sugar beet tuber weight, green leaf weight, and proportion of polar sugar in tuber were investigated. Results showed that green leaf and proportion of polar sugar in tuber not statistically important but the first application (0.18 kg da-1 agribor fertilizer) gave an statistically important yield increase in sugar beet. Boron contents of soils and boron uptake of plant species should be considered at boron application.
