Genetic Diversity of Some Tomato Cultivars and Breeding Lines Commonly Used in Pakistani Breeding Program

Genetic diversity present in gene pool is an important determination for breeding programs, and characterization is useful of building crop plant collections primarily based on the knowledge of the presence of valuable genes and traits. Developing successful varieties for increasing the future yield and quality of tomato depend mainly on the genetic diversity of parents used in the breeding program. Molecular characterization of 21 tomato genotypes used in in Pakistani breeding program was studied using random amplified polymorphic DNA (RAPD) markers. Total 102 bands were amplified among 21 genotypes using 20 RAPD primers. Overall 73.5% polymorphism was shown as 75 out of 102 loci were polymorphic. High degree of divergence between varieties was indicated by low level of monomorphic bands. The number of PCR products per primer varied from 2-8 with an average of 5.1 bands per primer. Primer GL J-20 and GL C-09 produced maximum number of bands whereas the primers GL A-09 produced the lowest. The polymorphism per RAPD primer ranged from 50% to 100% with an average of 73.5%. The accumulative analysis of amplified products generated by RAPD’s was enough to assess the genetic diversity among the genotypes. The information would be helpful for formulating future breeding and genome mapping programs. This study will also work as an indicator for tomato breeders to evolve varieties with genetic diverse back ground to achieve sustainability in tomato production in the country.

In this study, the effect of STAT5A gene was investigated on milk yield and components (fat, protein) in Jersey cows. Because, this gene is a member of the signal transducers and activators of transcription the factor (STAT) family and thought to be the impact on milk yield and components. As a material in the research, 100 Jersey cows raised in Samsun Karakoy State Farm were used. Milk samples were collected 12 times with one month intervals after calving, and these samples were performed by ultrasonic milk analyzer for milk fat and protein content. In addition, test day milk yield (TDMY) of each cow were also recorded in the control days. C/G polymorphism with PCR-RFLP method result in DNA isolation from blood samples taken from cows was determined at 8.exon at position 12195. Genotype distribution was detected as 69% CC, 28% CG and 3% GG. Allele frequencies were found as 0.83 for the C allele and 0.17 for G allele. As a result of the polymorphism identified in STAT5A gene, statistically no difference were detected among genotypes for TDMY, 305 day milk yield (305 DMY), milk fat and protein rates and milk fat and protein yield. Saffron, the dried stigmas of Crocus sativus, is extremely appreciated for its extraordinary color, taste and aroma. At the present time, nearly all the saffron harvest and post harvest processes are carried out manually. To increase the quality and development of economic role of saffron, it is essential to go beyond the traditional method of harvest of saffron. Considering that saffron components terminal velocities are different, a separator is planned and constructed to separate stigma from other parts of saffron flower. This separator is designed on the basis of aerodynamic and physical properties of saffron flower. The purpose of this study is to analyze the performance of a saffron separator for different air flows to increase the level of automation and efficiency of post-harvest operations. The results show that the maximum stigma separation happens when the air flow speed in outlet B is 3 m/s. finally, this data will be applied to find the optimum areas of outlet B and D, as two main parameters which have significant effect on the efficiency of saffron separator.

The aim of this study is to determine the slaughter and carcass traits of White and Multicolor Geese varieties reared in Yozgat province. 5 female and 5 male geese at six months old age from each variety were slaughtered and their slaughter and carcass traits were presented. Also, phenotypic correlations between carcass traits were calculated. The differences between the slaughter weight, hot and cold carcass weight, feather, head and foot weights of varieties were found insignificant, while male geese had higher values in terms of these traits. There were no significant differences between varieties and sexes in terms of hot and cold carcass yields and head ratio. Feather and foot ratios were found higher in White geese; whereas, abdominal fat ratio was found higher in Multicolor geese. There were insignificant differences between varieties in terms of carcass part weights, except back weight which was found higher in males. While heart weights of male geese were significantly higher, differences between varieties and sexes in terms of liver and gizzard weight were found insignificant. There were insignificant differences between the carcass parts and edible inner organ ratios of varieties, but breast ratio of females was found higher, while back ratio of males were found higher. Phenotypic correlation coefficients between slaughter weights, carcass weights and hot-cold carcass yields were found significant. Obtained data showed that native geese in the region are suitable for meat production in terms of slaughter and carcass traits.

Pomelo [Citrus grandis (L.) Osbeck] has the largest fruit size among citrus species. There are several cultivars for pomelo species which is sensitive to low temperatures thus its cultivation is possible in relatively preserved areas in subtropical regions. The interest to pomelo production has increased due to several reasons such as, its has a little or no bitterness in fruit flesh conversaly to grapefruit, avaibility of pigmented varieties, high concentration of vitamin C, total phenolics and carotenoids in fruit flesh, as well as its efficiency in destroying free radicals such as superoxide anion and hydrogen peroxide. Several pomelo varieties have been introduced to Turkey and adaptation studies have been established. In the present study, Reinking, Pomelo WN, Kao Panne and Citrus hassaku which is a spontaneous hybrid between pomelo and mandarin were investigated in terms of fruit yield and pomological characteristics. In addition, Henderson grapefruit was included in this study in order to obtain a comparasion between pomelo and grapefruit. A large variation was observed in fruit yield of cultivars established in 2011 and 2012. In 2011, the highest yield was determined in Henderson whereas it was obtained from Citrus hassaku in 2012. Pomelo varieties yielded between 5-60 kg fruit per tree and 8.6-50 kg fruit per tree in 2011 and 2012, respectively. In terms of pomological traits, Citrus hassaku had the highest fruit weight; Pink, Kao Panne and Reinking pomelos had the highest peel thickness and Pomelo WN yielded the highest TSS/TA ratio.

Concentrations and profile of polycyclic aromatic hydrocarbons(PAHs) and metals (Cd, Pb, Ni, Cr, Fe and Mn) were determined in selected brands of canned maize in the Nigeria market with a view to providing information on the hazards associated with the consumption of these products. The measurement of the concentrations of PAHs was carried out by using a gas chromatography equipped with flame ionization detector (GC-FID) after extraction by ultra-sonication with acetone/dichloromethane and clean-up. The 16 PAH concentrations varied between 45.1 and 335.7 µg/kg. The concentrations of the indicators for occurrence and effects of PAHs in food varied from 3.6 to 114.5 µg/kg for BaP, 6.4 to 168.2 µg/kg for PAH2, 11.8 to 232.7 µg/kg for PAH4 and 19.4 to 327.3 µg/kg for PAH8. Fire blight disease caused by Erwinia amylovora is a destructive bacterial pathogen mainly on pears, apples and quinces from Rosaceae family. In this study, it was aimed determination of total protein amounts in different apple cultivars (Braeburn, Fuji, Gala and Golden), pear cultivars (Santa Maria and Williams) and quince cultivars (Eşme and Ekmek) in the infections of two virulent E. amylovora strains (Ea234-1 and Ea240-3) according as the time. It was taken leaf samples after leaf inoculation with E. amylovora (108 CFU ml-1) at 24th, 36th and 72nd hours. For verification of the infections, re-isolations were made from bacteria inoculated plants and the agent was identified as E. amylovora by biochemical, physiological and molecular tests. In determining the amounts of total protein and in the SDS-PAGE analyses were used Bradford and Laemmli methods, respectively, and absorbance values of protein extracts derived from the leaf samples taken, were obtained at 595 nm wavelength. According to the findings obtained; after infection of E. amylovora in the apple varieties comparing to controls, total protein concentrations at 24th hours increased and a decrease in the amount of 36th to 72nd hours and Braeburn has the highest protein content was determined. In the pear varieties, while total protein concentrations at 24th and 36th hours increased, a decrease in the amount of 72nd hour, and Santa Maria variety has the highest protein content was detected. In the quince varieties, total protein concentrations at 72th hour increased and Eşme variety has the highest protein content was identified. As a result of SDS-PAGE analysis, protein fractions which have different molecular weights were obtained. The protein bands were defined approximately 55-70 kDa and 35-55 kDa molecule weight on apple and quince varieties, respectively and also approx. 55-70 kDa in pear varieties.

Meat and meat products are the basic building blocks of nutrition and are recognized as good sources of high biological value proteins, group B vitamins, minerals as well as some other bioactive compounds. The trend today is the development of novel food for special health use, called functional food, to promote human health and well-being of consumers. The trends are based on either reducing the content of unhealthy substances (like salt) or improving the content of substances with healthy benefits (like probiotics). Thus, it may also change the perspective of consumers towards meat products which associated with coronary artery disease. Meat is an ideal structure for probiotic microorganisms. Probiotic meat products are obtained by addition of probiotic to fermented meat products. These probiotic meat products are offered both healthy and improved taste and flavor, but also as safe food to consumer. Thus, when these probiotic meat products ensure flavor and nutritional to consumers, additionally they have a positive impact on their health.

A small scale pilot biodiesel production plant that has a volume of 65 liters/day has been designed, constructed and tested. The plant was performed using oil mixture (50% wild mustard seed oil + 50% refined canola oil) and methanol with sodium hydroxide (NaOH) catalyst. The fuel properties of biodiesel indicated as density at 15oC (889.64 kg/m3), kinematic viscosity at 40oC (6.975 mm2/s), flash point (170oC), copper strip corrosion (1a), water content (499.87 mg/kg), and calorific value (39.555 MJ/kg), respectively.

Food labels describe the content of a food and are intended to guide the consumer in food selection. The nutrition information provided must be in consistency with legal requirements and dietary recommendations. Selection of the specific nutrients or food components to be listed should further take into account label space, the analytical feasibility of measuring the particular nutritional component within the food matrix, and the relative costs of such analyses. Information provided on labels should be truthful and not mislead consumers. At the same time, labeling regulations should provide incentives to manufacturers to develop products that promote public health and assist consumers in following dietary recommendations. In some countries some segments of the population benefit from information about the composition of foods. In such cases, countries should consider the need to provide for appropriate labeling and its presentation relative to existing regulations. Different approaches and legal requirements have been established. These create difficulties in developing and harmonizing labels, which have broad international applications. For these reasons, the Codex Guidelines on food labeling play an important role to provide guidance to member countries when they want to develop or update their national regulations and to encourage harmonization of national with international standards. These Guidelines are based on the principle that no food should be described or presented in a manner that is false, misleading or deceptive. Exchange of information at the regional and sub-regional level is important, as each country can learn from the experience of others and regional co-ordination and co-operation can be developed.
