In Vitro Regeneration of Tea (Camellia sinensis (L). O. Kuntze) By Somatic Embryogenesis from Immature Cotyledon Tissues Полный текст

Tea (Camellia sinensis) is the world's most popular beverage plant, as well as an important plantation crop with high commercial value. It has been maintained for centuries through conventional vegetative propagation. Tea clonal propagation in vitro has the advantage of producing a large number of elite plants. If an efficient in vitro regeneration technology is available, this technique could be exploited for selection of tea plants for desired trait. The selected plants could be later on multiplied through in vitro or ex vitro techniques. The study aimed to induced somatic embryogenesis from immature embryo explants to genetic variaton. Different concentrations of phenylboronic acid with benzyladenine and phenylboronic acid with kinetin were tested in MS medium with 30 g/L sucrose and 8 g/L agar. MS medium without any plant growth regulators was used as control group. Considering the embryo survival rate, 1.5 mg/ L-1 phenylboronic acid + 1 mg/ L-1 kinetin produced highest result as 87.3% while lowest was in control group as 36.7%. The highest plant regeneration rate was found in 1,5 mg/ L-1 phenylboronic acid + 1 mg/ L-1 kinetin and 1.5 mg/ L-1 phenylboronic acid + 1 mg/ L-1 benzyladenine medium respectively as 58.3% and 55.6%. Kinetin treatment with increasing phenylboronic acid concentrations gave the best results in terms of somatic embryo survival rate. Also, kinetin treatment produced better results when compared to benzyladenine concentrations.

Green tea obtained from the leaves of Camellia sinensis (L.) Kuntze having potential health benefits. It contains significant amounts of antioxidant and is considered as world healthiest drink. Polyphenols in tea are thought to exhibits anti-carcinogenic and anti-inflammatory effects. Green tea catechins acts as immune modulators in immune dysfunction and also play a role in protection from degenerative diseases. Intake of green tea catechins suppress high fat diet induced obesity, prevent lipid and glucose metabolism disorders and reduce risk of coronary heart disease. Green tea constituent epigallocatechin-3-gallate (EGCG) is a potential antioxidant that shows protective effects against photo toxicity and photo carcinogenesis. In vitro animal experiments have suggested that polyphenols in green tea protect from lung cancer by their antioxidant and antimutagenic properties. This study chiefly highlights the convenience of green tea on health and propagates its further prospects.

Listeria monocytogenes is a foodborne pathogen frequently isolated from food that causes different public health problems. In recent years, antibiotic resistance of pathogens has become an important problem affecting human health. For this reason, it is crucial to develop an alternative to antibiotics. Thus, the antibacterial effects of herbal extracts and essential oils are frequently investigated. In this article, the antibacterial activity of extracts obtained from 16 different herbs using ethanol, chloroform, acetone, and distilled water was evaluated against L. monocytogenes strains. The extract showing the highest antibacterial effect against L. monocytogenes was St. John’s Wort extracted in ethanol (31.72 ±0.52 mm). In addition, Myrtus leaf extracted in ethanol (27.2 ±0.52 mm) and St. John’s Wort extracted in acetone (25.6 ±0.52 mm) showed a high antibacterial effect against L. monocytogenes compared to other extracts. In the study, the solvent ethanol in which St. John’s Wort and Myrtus leaf were extracted showed the highest antibacterial activity. In contrast, the solvent that rosemary extract showed the highest antibacterial effect was acetone. However, the most antibacterial herb extracted in distilled water was peppermint (7.03 ±0.52 mm). The extracts of marjoram and yarrow did not show any antibacterial effect in any solvent used in the study. In conclusion, more studies are needed to determine the antibacterial effects of herbal extracts against pathogens in foods and their use.

In this study, the antioxidant activity values of infusions at different temperatures of the commercially sold rosehip tea bag. To determine the effect of different infusion temperatures on antioxidant activity, 7 different temperatures (0, 15, 30, 45, 60, 75, 90°C) also including cold infusions temperatures were selected. Infusions of the rosehip tea bags were prepared with packaged drinking water for 5 minutes (recommended time in tea bag packaging) at these temperatures. Antioxidant activity values of the infused extracts were determined by antioxidant activity determination methods: TPC (Total Phenolic Compound), CUPRAC (Cu (II) Ion Reducing Antioxidant Capacity), FRAP (Iron (III) Ion Reducing Antioxidant Power) and ABTS•+ (2.2-azinobis- (3-ethylbenzothiazoline-6-sulphonic acid)) radical scavenging assay. According to the results obtained by the antioxidant determination methods, the highest and lowest activity values for TPC, FRAP and ABTS were measured at 90°C and 0°C, respectively, and a positive correlation was found between temperature and antioxidant activity values. Contrary to the activity values determined in these methods, in the CUPRAC method, the highest and the lowest activity value was measured at 45°C and 90 °C, respectively. As determined in the CUPRAC method, infusions of rosehip tea at low temperatures may provide an advantage in terms of the solubility of some phenolics that can be denatured at high temperatures. In addition, rosehip tea infusions at lower temperatures will contribute to lower energy costs.

Bovine and caprine mastitis is a dreadful bacterial disease that drastically lowers milk production, and presently it is spreading as an endemic pathogen throughout the world, including Bangladesh. Moreover, mastitis is the main impediment to Bangladesh’s tremendously rising milk-related industry and general livestock economy. Therefore, the aim of this study is to demonstrate the overall prevalence of clinical and sub-clinical mastitis in both dairy cows and goats. It is the foremost meta-analysis study regarding mastitis prevalence in Bangladesh and the study gathered the articles from the electronic database on the topic bovine and caprine mastitis published between January 2005 and November 2020. Finally, 33 articles were selected for estimating the overall prevalence rate of mastitis in ruminant (cow and goat) via random effect model estimation. Analyzing the result, the study found 43.0% (95% Cl: 37.0 - 50.0%) and 31.0% (95% Cl: 15.0 - 47.0%) prevalence rate in dairy cattle and goat. Statistically, the occurrence rate in Bangladesh is significantly higher than other countries. Although many studies showed the seroprevalence across different districts, this study demonstrates the aggregate incidence rates across Bangladesh as a whole. This paper may also serve as a reference point for the implementation of any control strategy of mastitis in Bangladesh.

Royal jelly (RJ), is one of the important honey bee products and a functional food item in the regulation of diets and in the cosmetic industry. RJ has a potential towards various human disease treatments. The chemical content of RJ is influenced by some factors. In this study, the effect of the presence or absence of the queen on the amount of 10-hydroxy-2-decenoic acid (10-HDA) and soluble protein in RJ is determined. For this reason, colonies were prepared as queenless, queenright and starter-finisher. RJ yields in colonies queenless, queenright and starter-finisher were determined as 15.2 ± 0.89 g, 12.0 ± 0.90 g and 9.6 ± 0.72 g, respectively. Group queenless was different from the other two groups. While 10-HDA values of the groups were similar (queenless, queenright and starter-finisher, respectively; 2.0 ± 0.06%, 2.1 ± 0.06% and 2.0 ± 0.05%), the soluble protein amounts of the groups (queenless, queenright and starter-finisher, respectively, 9.65 ± 0.179%, 7.68 ± 0.184%, 7.50 ± 0.203%) were found different and significant from each other. As a result, the RJ production colony queenless or queenright affected the amount of soluble protein. The worker bees of queenless colonies secreted RJ containing more soluble protein.

Effect of Different Substrate Mixtures on Volatile Aroma Compounds and Antioxidant Activity of Maitake Mushroom Полный текст

In the present study, it was aimed to determine the volatile aroma composition and antioxidant activity of Maitake mushroom grown in different substrate mixtures comparatively. Five different substrate mixtures except control were prepared. Total polyphenols and antioxidant activities were specified by Folin-Ciocalteu, FRAP and DPPH methods. Furthermore, analyzes were carried out in both dried and frozen samples. Head Space Solid Phase Micro Extraction technique combined with Gas Chromatography-Mass Spectrometry (GC-MS) was used in the analysis of volatile compounds. In the present study, yield was obtained only from S4 (oak sawdust + wheat stalk + bran at 1:1:1 ratios) and S5 (poplar sawdust + wheat stalk + bran at 1:1:1 ratios) growing mixtures. Therefore, the studies have been continued by comparing only these two mixtures. While the yield in S4 mixture was 55.02 g 1 kg bag-1, it was determined as 124.82 g 1 kg bag-1 in S5 mixture. DPPH analysis results of frozen and dried samples were 7.99±0.08 and 8.19±0.05 µmol TE g-1 DM (S4) and 8.07±0.09 and 8.20±0.06 µmol TE g DM-1 (S5) respectively. In volatile aroma profile analysis, 22 and 32 compounds were detected in S4 and S5, respectively. Ketones were the most found compound groups and its ratio was 68.67% in S4 and 52.37% in S5. The highest percentage among ketones was obtained from 4-nonanone and 3-octanone compounds.

Mushrooms used as food have medicinal importance due to their antioxidant compounds. In this context, it is very important to determine the biological potential of fungi and to reveal these medicinal properties. In this study, it was aimed to determine the element contents, total antioxidant status, total oxidant status, oxidative stress index of Suillus collinitus (Fr.) Kuntze mushroom. In this context, the mushroom samples were extracted with ethanol in the Soxhlet extractor. Element contents were determined using atomic absorption spectrometry. Total antioxidant (TAS) and total oxidant (TOS) levels and oxidative stress index (OSI) were determined using Rel Assay commercial kits. As a result of the study, it was determined that the TAS value of S. collinitus was 2.467 ± 0.145 mmol/L, TOS value was 17.845 ± 0.273 µmol/L and OSI value was 0.677 ± 0.030. In addition, the Fe content (350.72 ± 10.23), Cu content (68.11 ± 2.51), Pb content (11.58 ± 2.43), Zn content (10.46 ± 1.28) and Ni content (1.47 ± 0.21 mg.kg-1) of S. collinitus measured. As a result, S. collinitus mushroom is thought to be a natural source of antioxidants. It has also been observed that the element contents are at normal levels.

Türkiye, bulunduğu coğrafik konumu ve arazi yapısından dolayı oldukça zengin doğal bitki çeşitliliğine sahiptir. Bu çalışmada doğal olarak yayılış gösteren Fabaceae familyasına ait endemik Astragalus vulnerariae DC. taksonunun yarı kontrollü serada ilkbahar ve sonbahar dönemlerinde çelikle üretimi yapılarak peyzaj çalışmalarında kullanım alanları belirlenmiştir. Bitki çelikleri köklendirilirken; kontrol grubu, 5.000 ppm ve 10.000 ppm’lik IBA konsantrasyonlarıyla üç uygulama yapılmıştır. Köklenme sonrası elde edilen verilere göre uygulamalar arasında istatistiki olarak anlamlı bir fark elde edilmiştir. A. vulnerariae çeliklerinde en iyi köklenme, ilkbahar döneminde, %60 oranında 5.000 ppm’lik IBA hormon uygulamasında görülmüştür. Hem estetik hem de fonksiyonel özellikleri ile dikkat çeken A. vulnerariae taksonunun peyzaj çalışmalarında, yer örtücü olarak kullanımı başta olmak üzere, kaya bahçeleri, çatı ve teras bahçeleri ve peyzaj onarım çalışmalarında kullanılabileceği belirlenmiştir.
